Abstract

Cynometra ramifolia Linn is known as Sala plant. This plant, especially in Solo City categorized as rare plant and traditionaly used to cure uric acid, diabetes, hypertension, and others pain. It is interesting to know the other activities by done the test for human cancer cell-lines. This research objective to identify the cytotoxicity effects of ethanol extract from C.ramiflora the stem bark and leaves against T47D, HeLa and WiDr cancer cell lines. The cytotoxicity test of ethanol extract from C.ramiflora the stem bark and leaves determined by MTT assay by calculating the level of IC50  which was based on the percentage of the cell death following the 24 hours incubation with the extract. The results  showed that ethanol extract of stem bark has cytotoxic effect to HeLa, T47D and WiDr cell-lines with the IC50  of 1000; 0,90 and 6,29 g/mL respectively. The leave cytotoxic effect to HeLa, T47D and WiDr cell-lines with the IC50 of 1,92;  6,37 and 0,41 g/mL respectively. This research indicated that the ethanol extract isolated from C. Ramiflora leaves a selective cytotoxicity effect to WiDr cell line.

Abstract

Air sungai Bengawan Solo digunakan untuk berbagai keperluan, khususnya pertanian dan perikanan. Keberadaan industri-industri di sekitar aliran sungai diduga telah mencemari kandungan airnya. Penggunaan air tercemar ini untuk kebutuhan pertanian dan perikanan dapat menimbulkan masalah kesehatan karena bisa terjadi bioakumulasi. Penelitian ini bertujuan untuk mengevaluasi kandungan cemaran logam Pb dan Cd pada tanaman kangkung dan tanah yang diairi sungai Bengawan Solo. Sampel tanah dan tanaman kangkung diambil secara acak dari satu lahan yang menggunakan pengairan sungai Bengawan Solo. Destruksi sampel dilakukan prosedur Standar Nasional Indonesia. Analisis logam Pb dan Cd larutan sampel dilakukan dengan menggunakan Spektrofotometri Serapan Atom (SSA). Kadar logam terukur dibandingkan dengan batas aman kandungan logam dalam pangan menurut Standar Nasional Indonesia dan dihitung nilai bioaccumulation factor (BAF). Hasil analisis menunjukkan konsentrasi Pb dan Cd dalam sedimen dan tanaman kangkung berturut-turut adalah 6,94±0,60 ppm; 0,23±0,04 ppm; dan 1,19±0,18 ppm; 0,32±0,01 ppm. Berdasarkan nilai ambang batas keamanan untuk Pb 0,5 ppm dan Cd maka disimpulkan tanaman kangkung tidak aman untuk dikonsumsi. Nilai bioaccumulation factor CdPb, maka disimpulkan bahwa kemampuan tanaman kangkung menyerap logam Cd lebih besar dari pada Pb.  Kata kunci : Tanah, Tanaman kangkung (Ipomoea aquatica), kadar Pb, kadar Cd, Bioaccumulation Factor (BAF).

Abstract

Dewandaru (Eugenia uniflora L.) secara tradisional digunakan sebagai penurun panas dan sakit perut. Penelitian membuktikan aktivitas biologis dewandaru sebagai antibakteri, antidiabetes, dan antioksidan. Senyawa yang diduga bertanggungjawab adalah flavonoid. Penelitian ini dilakukan untuk mengetahui jenis senyawa flavonoid yang terdapat dalam daun dewandaru (Eugenia uniflora L.). Ekstraksi dilakukan dua tahap yaitu tahap penghilangan lemak dengan metode sokletasi menggunakan pelarut kloroform dan tahap kedua maserasi dengan etanol 70%. Ekstrak difraksinasi dengan etil asetat dan air, kemudian fraksinya diperiksa dengan KLT menggunakan fase gerak asam asetat 15% dan BAW. Fraksi etil asetat diisolasi dengan KLT preparatif. Bercak pita yang memiliki harga Rf dan warna yang sama dengan deteksi awal diambil dan disari. Kemudian dianalisis menggunakan spektrofotometer UV-Vis, pereaksi diagnostic NaOH, NaOAc, H3BO3, AlCl3, HCl, KLT dua dimensi, hidrolisis isolat fraksi etil asetat. Berdasarkan pergeseran panjang gelombang spektra UV-Vis dengan dan tanpa pereaksi   diagnostik serta uji KLT didapatkan struktur parsial yang diduga kuat 5,7,3’,4’-tetra hidroksi flavonol atau kuersetin. Kata Kunci : dewandaru (Eugenia uniflora L.), flavonoid, KLT, spektrofotometer UV-Vis.kombinasi ekstrak; tikus jantan dan betina galur SD.

Abstract

Iron was silvery white, clay, and can be formed.  it was usually obtained as hematite in nature. Iron was a chemical element that could be found in almost all places on the face of the earth, in all parts of the geological layer, and all bodies of water. In surface water, Fe was rarely found more than 1 mg / L, but in the water, Fe content could be much higher. When it accumulated in the body, iron could cause health problems, such as skin irritation, respiratory problems, and cause cancer in a long time. This study aims to determine the reduction of iron content in well water at the Universitas Muhammadiyah Surakarta (UMS) using the Spectrophotometry method with phenanthroline reagents. The samples were taken as many as five points in three different regions, namely Tanuragan (near the river), Menco and Nilasari (dense settlements), and Gonilan (near the rice fields) showed positive results containing iron with the highest levels in the sample without filtering at 9,478 ppm. These levels decreased after filtration using four filters, namely filters made of carbon, cotton, foam, carbon three layers in each sample. The iron content reduction was best to use cotton filters with 100% reduction in percent.

Abstract

Effect  of  water  extract  of  black  seed  on  uric  acid  metabolism  in hyperuricemia mice models were studied. The influence of water extract of black seed  on  uric  acid  biosynthesis  was  xamined  using  mice  treated  by  250  mg/ body  weight  potassium  oxonate  intra  peritoneal.  The result  showed  that  water extract of black seed (200 mg/kg body weight, per oral) have a significant effect in reduction in plasma uric acid levels in comparison to negative control, but less active  than  positive  control  (allopurinol).  The  levels  of  uric  acid  in  mice  plasma for negative control, black seed extract and positive control were 3.100 ± 0.346 mg/dL,  0.200  ±  0.100  mg/dL  dan  1.200  ±  0.561  mg/dL,  respectively.  The standardization  performed  by  determined  specific  and  non  specific  parameters based  on  National  Bureau  of  Food  and  Drug,  Republic of  Indonesia.  Specific parameters  of  water  extract  of  black  seed  are  phenolic  content,  and  flavonoid content  are  0.664  ±  0.035  %  and  0.400  ±  0.018  %,  respectively.  Non  specific parameters of water extract of black seed are loss of weight, water content, ash content,  ash  insoluble  acid  content,  heavy  metals  contamination  (Pb  and  Cd), and  aflatoxin  residue  are  14.93-14.99%,  6.09-7.89%, 7.12-7.49%,  5.20-6.66%, 6.405 +0.211 ppm, 0.0096 +0, not detected, respectively.Key words: antihyperuricemia effect, water extract of black seed, standardization

Abstract

Due to severe side effect and non-specific chemotherapeutic agent, screening and discovery for cancer therapy are still working, especially from natural resources. Traditionally, people used herbal medicine either to prevent or cure diseases. One of herbal that commonly used in Indonesia is Alpinia galangal. Previous study stated that active compound is acetoxy chavicol acetate (ACA) and active as anticancer. This research aimed to determine ACA concentration and cytotoxic activity of Alpinia galanga extract (AGE) from three local markets on HeLa, MCF7 and T47D cell lines. The galangal used from three local markets namely Pasar Legi Surakarta, Beringharjo Yogyakarta, and Wonogiri. The extraction was performed by maceration using 96% ethanol as solvent. ACA quantitation using UV spectrophotometer at λ = 208.5 nm. Samples were prepared by liquid-liquid extraction using an ethyl acetate. Cytotoxic activities were performed by MTT assay. The result showed that the concentration of ACA of AGE from the three local markets were 3.798; 0.035; and 0.009 % w/w, respectively. Cytotoxic activity, describes as IC50 value, on HeLa cell line of AGE from three local markets, in order were 13.26; 36.32 and > 100 µg/ mL. Meanwhile, AGE from Pasar Legi on MCF7 and T47D cell lines have IC50 value of 15.80; 12.50 µg/ mL, respectively. In contrast, two other samples have IC50 values of greater than 100 µg/ mL. The highest activity was from the highest concentration of ACA on the samples.Key words: Alpinia galanga, HeLa, T47D, MCF7 and Acetoxy chavicol acetate

Abstract

Previous study stated that the herba of Ruellia tuberosa L has inhibition activity on α-glucosidase enzymes. The leaves, flowers, and roots of Ruellia tuberosa L contain flavonoids but have different activities. This study aims to determine the inhibitory activity by the leaves, flowers and roots of Ruellia tuberosa on the α-glucosidase enzyme. Extraction was performed by maceration method using ethanol 96% as solvent. Inhibition activity was measured based on the formation of a yellow solution of p-nitrophenol compound as a result of the reaction between p-nitrophenyl-α-D-glucopyranoside (as substrate) and α-glucosidase enzyme at a wavelength of 405 nm. The system reaction of inhibition study is divided into two reactions, namely S0 (contains substrates without enzyme) and S1 (contain substrate and enzyme). The results showed that inhibition activities of leaves, flower and root extract were 9.97 ± 4.97 % at 7.8125 µg/mL concentration of extract; 13.98 ± 5.18 % at 7.8125 µg/mL concentration of extract; 99.08 ± 0.12 % at 31.25 µg/mL concentration of extract, respectively. Based on data, the extract of leaves, flowers and roots of Ruellia tuberosa L. has the potential as an inhibitor of the enzyme α-glucosidase.

Abstract

The ethanolic extracts of the Zingiber zerumbet rhizome contains flavonoid and zerumbon compounds, the fish meat powder from Channa striata has an albumin content. The active compounds was contained in both materials had activity that can lower blood sugar levels in diabetic rats. This study aims to determine the hypoglycemic effect of each ingredients and the combination of Z. zerumbet rhizome extract and C. striata powder in alloxan-induced diabetic rats. This research uses pre and post test method with the control group design. The treatment was consisted of 5 groups, the first group was given aquades (as negative control), the second group was given glibenclamide with dose 5 mg/kgBW (as positive control), the third group was given 300 kg/kgBW of Z. zerumbet rhizome extract, the fourth group was given of C. striata fish powder 300 mg/kgBW, and the last group was given a combination materials with dosesrespectively, 300 mg/kgBW. The ingredient was given for 12 days and blood was taken on days 3, 6, and 12. The results shows the percentage of decrease in blood sugar of combination materials of 70.4% is greater than the single content of Z. zerumbet and C. striata, with the value of successive activity of 62.9% and 34.4%, respectively. The antidiabetic activity of combinations of materials had produced not different significant (p 0.05) compared with single of C. striata powder as well as the Z. zerumbet rhizome extract.

Abstract

ABSTRAK Lempuyang di masyarakat dikenal memiliki 3 spesies yaitu lempuyang gajah, wangi, dan emprit. Penelitian sebelumnya menunjukkan lempuyang gajah memiliki aktivitas antidiabetes pada tikus yang diinduksi aloksan. Skrining farmakologi terhadap lempuyang emprit (Zingiber amaricans BL) masih sangat terbatas. Penelitian ini bertujuan untuk mengetahui potensi ekstrak etanol rimpang lempuyang emprit terhadap kadar glukosa darah tikus diabetes yang diinduksi aloksan. Hewan uji yang digunakan adalah 20 ekor tikus jantan putih galur Wistar yang dibagi menjadi 5 kelompok yaitu kelompok kontrol positif menggunakan glibenklamid, kontrol negatif menggunakan akuades, dan 3 kelompok perlakuan ekstrak etanol rimpang lempuyang emprit dengan tingkatan dosis  0,4 g/kg BB, 0,6 g/kg BB dan 0,8 g/kg BB. Pengukuran kadar glukosa darah pada hari ke-0, ke-4 dan ke-19. Data diuji dengan metode analisis statistik menggunakan software SPSS versi 17 for windows. Hasil pengujian menunjukkan bahwa tiga tingkatan dosis ekstrak etanol rimpang lempuyang emprit dan glibenkamid mampu menurunkan kadar glukosa darah dengan bermakna. Kadar rerata glukosa darah pada hari ke-4 untuk masing-masing kelompok dosis 0,4 g/kg BB, 0,6 g/kg BB, 0,8 g/kg BB dan kontrol positif adalah 264,5±18,86 mg/dL, 261,5±30,34 mg/dL, 245,0±29,02 mg/dL dan 264,5±14,30 mg/dL menurun menjadi 105±44,06 mg/dL, 149±45,20 mg/dL, 123,75±21,40 mg/dL dan 112,5±17,75 mg/dL pada hari ke-19. Kata kunci : antidiabetes, tikus jantan, Wistar, Zingiber amaricans.

Abstract

Ekstrak etanol kulit buah rambutan (Nephelium lappaceum) dan ikan gabus (Channa striata)telah dilaporkan memiliki aktivitas antidiabetes secara in vivo. Tujuan dari penelitian ini untukmengetahui apakah kombinasi dari kedua ekstrak tersebut memiliki aktivitas antidiabetes yanglebih tinggi dibandingkan sediaan tunggalnya. Desain penelitian ini menggunakan metode preand post-test with control group. Penelitian ini menggunakan 15 tikus yang diinduksi aloksan150 mg/KgBB secara intraperitoneal, yang dikelompokkan menjadi 5 kelompok perlakuan, yaitukontrol negatif, kontrol positif glibenklamid 5 mg/KgBB, serbuk ikan gabus (SIG) 300 mg/KgBB,ekstrak kulit buah rambutan (EKBR) 300 mg/KgBB dan ekstrak kombinasi perbandingan 1:1. Setiapperlakuan diberikan secara oral setiap hari hingga 10 hari pengamatan. Ekstrak kombinasi mampumenurunkan 55,6% kadar glukosa darah. Serbuk tunggal ikan gabus dan ekstrak tunggal kulitbuah rambutan menurunkan glukosa darah berturut-turut 27,4% dan 71,2%. Berdasarkan analisisstatitistik, tidak ada perbedaan signifikan (p>0,05) antara ekstrak kombinasi terhadap perlakuantunggal SIG atau EKBR. Aktivitas kombinasi ekstrak memiliki kemampuan menurunkan kadargula darah yang lebih tinggi dibandingkan serbuk ikan gabus, tetapi lebih rendah dibandingkandengan ekstrak kulit buah rambutan.
