Abstract

Antioksidan merupakan substansi yang diperlukan untuk menetralisir radikal bebas dan mencegah kerusakan sel normal, protein, dan lemak. Radikal bebas berlebih dalam tubuh menyebabkan stres oksidatif. Salah satu parameter stres oksidatif adalah meningkatnya kadar malondialdehid (MDA). Ekstrak ikan gabus berperan sebagai antioksidan. Penelitian dilakukan untuk mengetahui efek pemberian suplemen ekstrak ikan gabus sebagai antioksidan pada tikus yang mendapatkan obat rifampisin-isoniazid dosis tinggi. Tikus dikelompokan menjadi 4 yaitu kelompok I (kontrol), kelompok II (ekstrak ikan gabus), kelompok III (rifampisin-isoniazid), dan kelompok IV (rifampisin-isoniazid dan ekstrak ikan gabus). Kadar MDA dalam darah diukur menggunakan metode Tiobarbiturat Acid Reactive Substance (TBARS) dengan spektrofotometer UV-Vis 532 nm. Analisis data menggunakan uji Anava dan uji Kruskal Wallis. Hasil analisis kadar rata-rata MDA darah dan hati kelompok I, II, III, dan IV berturut-turut pada darah 0.67±0.12; 0.38±0.20; 4.01±1.57; 2.68±1.72 μg/mL, dan hati 3.73±0.80; 2.96±1.94; 31.31±4.45; 17.69±10.55 μg/g. Hasil menunjukkan kadar MDA darah kelompok III dan IV signifikan terhadap kelompok II, pada hati kelompok III signifikan terhadap kelompok I dan II sedangkan kelompok IV signifikan terhadap kelompok I, II, dan III. Suplemen ekstrak ikan gabus dosis tinggi berpotensi sebagai antioksidan pada tikus yang mengalami stres oksidatif.

Abstract

Rifampisin merupakan salah satu obat anti-tuberkulosis. HPLC merupakan metode pilihan karena kepekaan, reprodusibilitas dan spesifitasnya. Namun metode terbaru dari kromatografi cair membutuhkan penambahan tetra-butyl ammonium hydroxide yang dapat memperpendek umur kolom. Penelitian ini bertujuan untuk mengembangkan suatu metode kromatografi cair dengan waktu pengerjaan yang efisien dibandingkan metode-metode sebelumnya. Sistem kromatografi yaitu HPLC Alliance 2695, C18 (100 x 4,6 mm; 5 µm), fase gerak sistem gradient berupa asetonitril dan akuades dengan laju alir 1,0 mL/menit dan dideteksi pada 254 nm. Hasil penetapan parameter validasi didapatkan bahwa metode akurat dengan %recovery 99,96 ± 0,04% dan RSD 0,64 ± 0,30%. Parameter linieritas didapatkan koefisien korelasi 0,998, kerberulangan dan presisi antara didapatkan RSD masing-masing yaitu 1,51% dan 1,54%. Nilai LoQ didapatkan dari persamaan regresi linier residual yaitu 0,0164%. Metode yang divalidasi memenuhi parameter validasi akurasi, presisi dan linieritas.

Abstract

Tumbuhan Sala (Cynometra ramiflorra Linn.) merupakan salah satu tumbuhan mangrove, yang memiliki beberapa manfaat antara lain antioksidan, antihiperglikemia, dan antiinflamasi. Uji toksisitas subkronis digunakan untuk mengetahui toksisitas suatu senyawa yang dilakukan selama 90 hari. Penelitian ini bertujuan untuk mengetahui pengaruh penggunaan ekstrak etanol daun Sala terhadap parameter kimia urin dan histopatologi ginjal. Penelitian ini merupakan penelitian eksperimental dengan rancangan Post Test Controlled Group Design, yaitu melihat perubahan yang terjadi pada tikus setelah diberi perlakuan. Sampel penelitian terdiri dari 20 ekor tikus putih jantan yang dibagi 4 kelompok dosis perlakuan. Kelompok pertama merupakan kelompok kontrol yang diberi CMC-Na (Sodium Carboxy Methyl Cellulose), kelompok kedua, ketiga, dan keempat diberi ekstrak etanol daun Sala dengan dosis 500, 1000, dan 1500 mg/kgBB. Parameter yang diamati yaitu kimia urin dan histopatologi ginjal. Parameter kimia urin dievaluasi menggunakan urin tampungan pagi hari pada hari ke-0, 45, dan 90 menggunakan reagen strip. Hasil parameter kimia urin tikus untuk kelompok kontrol dan perlakuan selama 90 hari memberikan pengaruh pada parameter protein, glukosa, keton, bilirubin, urobilinogen terhadap fungsi ginjal, namun tidak mempengaruhi pH dan nitrit. Hasil histopatologi menunjukkan terjadinya peradangan pada ginjal tikus yang diberi ekstrak etanol daun Sala dengan dosis 1000 dan 1500 mg/kgBB.Kata kunci: Cynometra ramiflora Linn., toksisitas subkronis, kimia urin

Abstract

Abstract

Antibacterial activity in vivo study of ethanol extract nonpolar fraction of inggu stem (Ruta angustifolia [L.] Pers) was conducted on Staphylococcus aureus and Streptococcus mutans. It had a purpose to determine the effect of non polar fraction of ethanol extract of inggu stem on animals that had been infected by bacteria Staphylococcus aureus and Streptococcus mutans. Inggu stem powder was macerated with ethanol 96% and fractionated by vacuum liquid chromatography using hexane : chloroform 6:4, 5:5, 4:6, 3:7. Inggu stem was tested with in vivo antibacterial activity on mice with various doses of 0.3, 1.2 and 2.14 g/kg. Intraperitoneal fluid of mice had been treated was cultured on agar media and colonies of formed bacteria were counted. Antibacterial activity on Staphylococcus aureus bacteria has an average percentage of inhibition for colony are respectively 73, 81 and 97 %., whereas that of the Streptococcus mutans bacteria are 41,81 and 97% respectively. Identification of thin layer chromatography fraction in nonpolar compounds of ethanol extract of inggu stem is indicated by flavonoids, terpenoids, and alkaloids.

Abstract

Insulin resistance causes 90% of type 2 diabetes mellitus (DM) cases and this is due to an increase in the activity of glycogen synthase kinase-3 (GSK-3). Dephosphorylation and activation of glycogen synthase prevent type 2 diabetes by inhibiting the work of GSK-3 beta isoform primarily. The study was done in unix (mac and linux) operating system with the sequence of steps were optimization of geometry of ligand, ligand and protein preparation, validation of docking methods, molecular docking and analysis, and 3D modeling of ligand-protein interactions. Optimization of target ligand geometry using Gaussian showed that molecular docking score increased 62.5% compared with geometry optimization using MarvinSpace. Polyhydroxy compound zerumbon derivats: ZER04, ZER05 and ZER06 potential to be tested further in the laboratory as an anti- type 2 diabetes both in vitro and in vivo. This is based on the ability of the compounds in term of molecular interactions that better than the native ligand of target protein 4PTE, 4AFJ, 3ZRK, 3GB2, 3DU8 and 1Q3W (60% of the total protein targets tested) as an inhibitor of glycogen synthase kinase- 3 types of beta (GSK-3b).Keywords: polyhydroxyzerumbone, glycogen synthase kinase-3 beta, docking molecular, DOCK6

Abstract

IPTEKDA LIPI XVIII activities in 2015 was intended to improve the quality and capacity of production in SMEs CV. Arba'in Jaya Mandiri Surakarta, through the introduction of some technology included medicinal herb powder mixer machines, mixer machines for honey, and semi-manual capsule filling. While in the second phase, will be introduced vacuum evaporator machine and application software for the development marketing online of herbal medicinal and honey products Arba'in. The results had been achieved in the first phase of IPTEKDA LIPI 2015, ie an increased in productivity and efficiency of production procesing as well as improving the quality of herbal medicine production. The mixing process of herbal ingredients were more efficient with the capacity of mixing up to 25 kg / process, from only 3-5 kg / process before. The process of mixing herbs-based honey, had showed a more homogeneous mixing and faster. Through the introduction and applications of technology production procesing, it was hoped will provide multifier effect for SMEs, employment, supplier partners and the communities.Keywords : SMEs CV. Abstract

The rind of rambutan fruit (Nephelium lappaceum L.) has been reported to have a very strong antioxidant activity with IC50 value of 7.74 μg / mL. The fruit contains  avonoids and phenolic . This compounds are potentially as hypolipidemic effect. This study aims to determine the hypolipidemic effect of 96% of ethanolic extract of rind of rambutan fruit in wistar strain male rats. This research method is the pre- and post-control group design. 25 rats were divided into 5 groups. Group I (negative control) were treated by a solution of 0.5% CMC-Na, group II (positive control) were treatedby cholestyramine 0.8 kg / kg, whereas group III, IV and V treated by 96% of ethanolic extract of rind of rambutan with a dose of 125 , 250, and 500 mg / kg respectively. hiperkolesterlemia induction is done by giving a high cholesterol diet and feed for 4 weeks. High-cholesterol diet consists of cooking oil, quail egg yolk, water, PTU and feed high cholesterol consisting of margarine and standard feed. Treatment of mice was conducted for 2 weeks. The results showed that the 96% of ethanolic extract of rind of rambutan fruit with a dose of 125, 250, and 500 mg / kgbw have antihyperkolesterol activity 21.39 ± 6.61%, 31.15 ± 18.15%, and 60.75 ± 8, 26%.Keywords: Antihypercholestrol, Nephelium Lappaceum L., High-cholesterol diet

Abstract

Antihyperuricemia activity in vivo testing has been done of single and combined extracts of Salam (Syzigium polyanthum Walp) and Belimbing Wuluh leaves (Averrhoa bilimbi L.), and also standardized extracts. Salam leaves extraction and Belimbing Wuluh leaves carried by the water solvent. Dried extract of each ingredient had been done antihyperuricemia preclinical in vivo testing against male white mice strain Balb-C induced by potassium oxonate dose of 250 mg / kg b.w. Antihyperuricemia preclinical in vivo testing results showed that the single extracts of Salam and Belimbing Wuluh at a dose of 200 mg / kg b.w. had lower activity levels of uric acid in the blood serum of male white mice, respectively 0.640 and 0.680. Abstract

Iron was silvery white, clay, and can be formed.  it was usually obtained as hematite in nature. Iron was a chemical element that could be found in almost all places on the face of the earth, in all parts of the geological layer, and all bodies of water. In surface water, Fe was rarely found more than 1 mg / L, but in the water, Fe content could be much higher. When it accumulated in the body, iron could cause health problems, such as skin irritation, respiratory problems, and cause cancer in a long time. This study aims to determine the reduction of iron content in well water at the Universitas Muhammadiyah Surakarta (UMS) using the Spectrophotometry method with phenanthroline reagents. The samples were taken as many as five points in three different regions, namely Tanuragan (near the river), Menco and Nilasari (dense settlements), and Gonilan (near the rice fields) showed positive results containing iron with the highest levels in the sample without filtering at 9,478 ppm. These levels decreased after filtration using four filters, namely filters made of carbon, cotton, foam, carbon three layers in each sample. The iron content reduction was best to use cotton filters with 100% reduction in percent.
