Abstract

Ikan gabus (Channa striata) memiliki kadar protein yang tinggi dengan kandungan utamanya adalah albumin. Sehingga diperlukan metode dan kondisi ekstraksi yang sesuai agar mendapatkan rendemen dan kandungan albumin yang optimal. Penelitian ini bertujuan untuk mendapatkan metode ekstrkasi yang optimal antara perebusan biasa, microwave-assisted extraction (MAE) dan ultrasonic-assisted extraction (UAE) dengan parameter kadar albumin. Ekstraksi dilakukan dengan perbandingan daging ikan gabus dan akuades (1 : 1). Variasi waktu ekstraksi yaitu 5, 10 dan 15 menit. Kadar albumin ditetapkan dengan metode Lowry. Rendemen ekstrak yang dihasilkan dari 3 metode ekstraksi pada waktu ekstraksi 5 menit berurutan 2,419; 1,293 dan 1,575 %b/b. Kurva baku Albumin didapatkan persamaan regresi linier yaitu y = 3,38x + 0,7396 dengan nilai r = 0,997. Kadar albumin yang diperoleh dari 3 metode ekstraksi pada waktu ekstraksi 10 menit secara berurutan adalah 47,009 + 1,046; 44,792 + 5,372 dan 28,946 + 2,260 %b/b. Berdasarkan data tersebut, metode ekstraksi yang memberikan kadar albumin yang terbesar adalah metode perebusan dengan waktu 10 menit.

Abstract

Artocarpus heterophyllus Lam leaves are recognized as a promising source of bioactive compounds with potential antidiabetic activity. This study aimed to evaluate the effects of vacuum liquid chromatography (VLC) fractionation on the distribution of phenolic and flavonoid compounds and their α-glucosidase inhibitory activity. Crude ethanolic extract was fractionated by VLC into polar, semipolar, and nonpolar fractions. Total phenolic content (TPC) and total flavonoid content (TFC) were determined using the Folin–Ciocalteu and aluminum chloride colorimetric assays, respectively, while α-glucosidase inhibitory activity was evaluated using a p-nitrophenyl-α-D-glucopyranoside-based assay. The crude extract exhibited the highest TPC (4.70 ± 0.37 mg GAE/g extract) and TFC (5.63 ± 0.11 mg QE/g extract), whereas both parameters decreased markedly after fractionation. Consistently, the crude extract showed the strongest α-glucosidase inhibitory activity, as indicated by the lowest IC₅₀ value, while the nonpolar fraction exhibited the weakest activity. These findings demonstrate that VLC fractionation redistributed rather than enhanced the bioactive constituents responsible for α-glucosidase inhibition. The crude extract therefore represents a promising natural source of α-glucosidase inhibitors and supports the potential utilization of Artocarpus heterophyllus Lam for the development of plant-derived antidiabetic agents.

Abstract

This study aims investigate the potential of phytochemical compounds from citrus mistletoe leaves (Dendrophthoe glabrescens) leaves as anticancer candidates using an in silico approach. Phytocemical constituents identified by LC-MS and screened using Lipinski’s Rule of Five. Followed by Absorption, Distribution, Metabolism, Excretion and Toxicity (ADMET) prediction using ProTox-3.0. The docking protocol was validated with root mean square deviation (RMSD) values of 1.80 Å for epidermal growth factor receptor (EGFR) (PDB ID: 1M17) and 0.69 Å  for estrogen receptor-α (ER-α) (PDB ID: 1A52). Tenylidone exhibited the strongest binding affinity toward EGFR (-8.12 kcal/mol), followed by quercetin (−7.48 kcal/mol) and kaempferol (−7.43 kcal/mol). For estrogen receptor alpha, fisetin showed the strongest affinity among the tested D. glabrescens compounds (−8.21 kcal/mol), followed by nomelidine (−8.16 kcal/mol) and tenylidone (−7.95 kcal/mol). Kaempferol showed the most favorable predicted acute toxicity profile among the leading compounds, with an estimated LD50 of 3919 mg/kg and toxicity class 5. Overall, tenylidone and fisetin were prioritized for epidermal growth factor receptor and estrogen receptor alpha, respectively, while kaempferol showed a favorable predicted safety profile.

Abstract

Oxidative stress caused by the accumulation of free radicals is a key factor in the development of degenerative diseases. Jackfruit leaves (Artocarpus heterophyllus), which are rich in polyphenolic compounds, have the potential as natural antioxidants. This study aims to activate the antioxidant activity of jackfruit leaf extracts and fractions. The study was conducted by extraction using the maceration method with 70% ethanol solvent and fractionated by vacuum liquid chromatography into polar, semi-polar and non-polar fractions. Antioxidant activity was measured in vitro, using 3 methods including DPPH, ABTS and FRAP which were read with an ELISA Reader with vitamin C as the reference standard. The results showed that the crude extract had strong antioxidant activity with an IC₅₀ value of 31.93 µg/mL (DPPH) and 29.59 µg/mL (ABTS), as well as the highest antioxidant equivalent ascorbic acid (AEAC) levels in the FRAP test. The polar fraction showed moderate antioxidant activity, while the semi-polar and non-polar fractions showed weak activity. These findings suggest that polar compounds such as flavonoids and phenolics play a role in the antioxidant effects of jackfruit leaves. In conclusion, jackfruit leaf extract and its polar fractions have significant antioxidant activity and show promise for further development as natural antioxidant agents in nutraceutical formulations aimed at preventing oxidative stress-related diseases.
