Abstract

Lipopolysaccharide (LPS) is a major component of a gram-negative bacterial wall that is widely used and well-established to induce inflammation in vitro. In addition, the in vitro model using RAW 264.7 cells is the most commonly applied in screening the anti-inflammatory and elucidating the pathophysiology of inflammation-based disease, as well.  However, there is still limited data on the efficacy of different doses of LPS in inducing inflammation in RAW 264.7 cells. This study aimed to evaluate the effect and safety of LPS at various doses in RAW 264.7 cells. RAW 264.7 cells were exposed to LPS at different dose ranges (10ng/mL-10µg/mL) for 24 hours. The nitric oxide (NO) release as inflammatory responses and viability test were evaluated using Griess assay and CCK-8 assays, respectively. The result showed that NO production was increased at different doses of LPS compared to the control although not significant. Whereas, All LPS-treated RAW 264.7 cells tended to increase but not significantly compared to the control groups. This study showed that the LPS treatment effectively induced inflammation in RAW 264.7 cells as shown by NO production and was considerably safe as the viability was comparable between LPS and control group for RAW cells 264.7 at least up to 10µg/mL for 24 hours.

Abstract

Obesity has been an alarming global health issue that relates to nutrition intake. In Indonesia, the prevalence of obesity among individuals aged over 18 years showed upward trend from 2013 to 2018 as reported by the Basic Health Research. Obesity is a key factor contributing to the development of metabolic syndrome. Addressing this issue requires adequate education to the public on obesity prevention through various methods. This community service program was conducted in the form of webinars, held on July 4, 2021, and July 11, 2021 on Zoom and was attended by 424 participants. Several topics were discussed in the webinars, including deep dive into obesity, its management, dietary planning, and an introduction to the My Weight Loss Diet application. A notable improvement in participants understanding was evident as seen from increased post-test scores. My Weight Loss Diet application is expected to assist participants in regulating their food intake in order to prevent obesity.

Abstract

Background: Consumption of beverages and processed foods with high fructose content has increased recently. This condition increases the risk of metabolic syndrome, which include insulin resistance, impaired lipid profile, and elevated blood pressure. Moringa leaves (Moringa Oleifera), with their bioactive compounds that function as antioxidants, are known to improve fat metabolism and blood glucose. Objectives: The purpose of this study is to analyze the effect of Moringa Oleifera leaf powder and quercetin in controlling metabolic syndrome conditions by improving lipid profiles and aortic thickness in prediabetes mellitus (pre DM). Methods: This experimental research used a post-test-only control group design approach. A total of 35 male Wistar rats were divided into 5 groups such as group K0 (standard feed), E1 (66% fructose probe for 16 weeks), E2 (66% fructose probe for 12 weeks), E3 (66% fructose probe + quercetin 50 mg/kgBW), and E4 (66% fructose probe + Moringa leaf powder 500 mg/kgBW). The examination method for lipid profiles involved serum extraction and enzymatic tests, weighing for white fat tissue mass, while the aorta was examined using a light microscope and histological image analysis. Data analysis used the One-Way ANOVA test. Results: This research showed there are no difference in triglycerides, Low-Density Lipoprotein (LDL) cholesterol, and white adipose tissue significantly between the treatment groups. Level of High-Density Lipoprotein (HDL) cholesterol, total cholesterol, and aortic thickness between the treatment groups are significant differences, with the best results sequentially in groups E2, E1, and K0. Conclusions: The administration of Moringa leaf powder for 4 weeks showed improvements in lipid profiles and aortic thickness in prediabetes mellitus conditions.

Abstract

Adolescence is a period of rapid growth that a healthy lifestyle must support. School as an environment where teenagers spend most of their time has a School Health Unit (UKS), an essential part of health services at school. UKS can be used as a health education for adolescents at school. However, in reality, UKS needs to be actively used and involved. This community service healthily provided interactive education and achieved optimal growth for students through using lectures, focus group discussion, roleplay, and student projects. At the end of the activity, it was reported that there was a significant improvement in students' knowledge. This activity has also involved the role of parents and schools with UKS as part of the sustainability guarantor of healthy lifestyles for students. The provision of diverse education strategies can effectively influence students' health knowledge.

Abstract

Pendahuluan: Jumlah anak stunting di Desa Paspan, Banyuwangi, masih mengalami peningkatan, sehingga diperlukan optimalisasi upaya pencegahan untuk menghindari dampak negatif terhadap kehidupan anak. Pengetahuan dan persepsi ibu mengenai stunting merupakan faktor penting dalam upaya pencegahan kondisi ini. Tujuan penelitian mengkaji hubungan antara pengetahuan ibu tentang stunting dengan persepsi keluarga terhadap stunting pada ibu yang memiliki anak dengan stunting di Desa Paspan, Banyuwangi. Metode: Penelitian kuantitatif cross-sectional dilakukan di Desa Paspan, Banyuwangi, pada bulan Desember 2022. Kriteria inklusi dan eksklusi digunakan dan didapatkan 26 responden dari 32 populasi ibu anak dengan stunting. Data dikumpulkan melalui kuesioner yang terdiri atas sepuluh pertanyaan tentang pengetahuan mengenai stunting dan sebelas pertanyaan tentang persepsi keluarga mengenai stunting. Uji korelasi gamma, dengan tingkat signifikansi 0,05, digunakan untuk mengkaji hubungan antara dua variabel.  Hasil: Mayoritas responden memiliki tingkat pengetahuan yang baik mengenai stunting (96%), sementara 84,6% memiliki persepsi yang cukup terhadap stunting. Hasil analisis uji korelasi gamma menunjukkan bahwa tidak terdapat hubungan yang signifikan antara pengetahuan ibu tentang stunting dengan persepsi keluarga terhadap stunting (p = 0,340; r = 1,000). Kesimpulan: Pengetahuan ibu mengenai stunting tidak selalu berkorelasi dengan persepsi keluarga terhadap stunting. Oleh karena itu, upaya pencegahan stunting memerlukan pendekatan holistik yang mempertimbangkan faktor sosial, budaya, dan lingkungan guna meningkatkan pemahaman serta mendorong perubahan perilaku masyarakat.

Abstract

Background: Obesity is linked to the rising incidence of type 2 diabetes, with excessive dietary fat intake being one of the primary causes. The development of fat animal models has been discovered to be a standard experimental strategy based on replicating human behaviour in food consumption. Objectives: This study aims to investigate how long-term consumption of a high-fat, high-fructose (HFHF) diet affects the indicators of diabetes mellitus in Sprague Dawley (SD) rats, such as insulin sensitivity, by measuring the HOMA-IR, counting beta cells, and analyzing the histology of the pancreas. Methods: This experiment was conducted with 36 male SD rats in normal and HFHF groups. The normal groups had a modified AIN-93 M, while the HFHF group received a high-fat diet with 30% fructose-based water. Feed and beverage intakes were monitored every 24 hours to calculate daily caloric consumption (energy intake) for 17 weeks. Results: The results demonstrated a significant difference between the normal and HFHF groups in the HOMA-IR levels (insulin sensitivity) and number of pancreatic beta cells (p-value<0.05). This implied that following 17 weeks of HFHF intake, the HOMA-IR level of insulin sensitivity was reduced. However, the islet of Langerhans in pancreatic histopathology seemed damaged in the HFHF rats, as evidenced by the changes in their shape and lower beta cell number. Conclusions: Consuming the HFHF diet over an extended period increased glucose level, decreased insulin sensitivity, and damaged pancreatic histopathology.

Abstract

Background: In 2023, obesity prevalence in Indonesia was reported at 15.7% in men and 31.2% in women. Eating out has become increasingly common among university students in major cities like Malang due to busy schedules, easy access to food accessibility, and lifestyle changes. Objectives: This study aims to examine the relationship between Body Mass Index (BMI), body fat percentage, and eating habits among university students. Methods: This cross-sectional observational study was conducted at Universitas Brawijaya, Malang, involving 385 respondents. The measured variables included BMI, body fat percentage, and eating habits. Eating habits were assessed using a questionnaire covering five indicators: Takeaway food consumption, eating at home, dining at restaurants, consuming supermarket food, and consuming traditional market snacks. Data were analyzed using the Chi-square test and Logistic Regression. Results: Most respondents were female, lived in dormitories or rented housing, and had a family income below five million rupiahs per month. Among the five eating habit indicators, only restaurant dining showed a significant correlation with BMI and body fat percentage (p-value<0.05), while takeaway food consumption, eating at home, consuming supermarket food, and consuming traditional market snacks were not significantly related (p-value>0.05). Conclusions: Eating habits, in general, showed no significant relationship with BMI and body fat percentage, except for restaurant dining, which was significantly correlated with increased body fat percentage.

Abstract

Background: Type-2 diabetes mellitus (T2DM) can have a notable impact on the psychological and physical well-being of individuals, which in turn affects the management of the condition. Men and women experiencing stress and adopting distinct coping strategies. However, research focusing specifically on T2DM in men is still limited.Purpose: The present study intends to investigate the distress and coping strategies adopted by male T2DM outpatients in Malang, East Java, Indonesia.Methods: This study employed a qualitative research design and conducted in-depth interviews to 24 male T2DM outpatients. The interview guidelines were formulated using the Indonesian version of the Diabetes Distress Scale (DDS17) questionnaire, which comprised four domains: physician distress, emotional burden, regimen distress, and interpersonal distress. A thematic analysis was performed to analyze the results gathered during the research and compile them into a final report.Results: The study revealed that individuals diagnosed with T2DM experienced a range of emotional and practical difficulties, including feelings of fear, anxiety, and a lack of understanding. Disease burden, a lack of understanding of both diabetes and healthcare services, difficulties managing their diet, routine medication, financial concerns, and fatigue also contributed to the distress. To cope with distress, the informants identified eight distinct coping strategies. Of these, the most effective strategy was receiving support from family members, followed by acceptance, self management, positive attitude, understanding of their illness, joining the diabetes community, spirituality, and getting more information about T2DM.Conclusion: The findings of this study indicated that men experience eight distinct types of stress and utilise comparable coping strategies associated with T2DM. Emotional distress represents the predominant pressure, while family support constitutes the primary coping strategy. These results are important for nurses and other healthcare professionals in supporting patients.

Abstract

The prevalence of diabetes mellitus has been increasing. This study aimed to develop Food for Special Dietary Use (FSDU) for individuals with diabetes. Different combinations of food ingredients were added to the basic formula, consisting of brown rice and mushroom flour in a ratio of 70% and 11%. The combinations used in the formulation were F1 = 9% moringa and fish flour, F2 = 10% carrot and fish flour, F3 = 11% moringa and tempeh flour, and F4 = 12% tempeh flour. The results showed that the FSDU contained energy, fat, and fiber ranging from 391.33–404.33 Kcal, 5.53–7.75 g, and 4.22–7.60 g per 100 g product, respectively. These values were slightly higher than the respective value of a commercial product. The panelists expressed positive acceptance of the developed FSDU products, with F2 chosen as the most preferred. The developed FSDU exhibited lower scores for feeling of hunger, desire to eat, and prospective food consumption compared to the commercial product. The product provided a feeling of fullness similar to the commercial counterpart. In conclusion, the FSDU based on brown rice and oyster mushroom with an additional 10% carrot fish flour would be the most preferred option for further use as FSDU for diabetics.
