Abstract

The objective of this study was to examine the effects of repeated freeze-thaw cycles and short-term storage of fecal extracts at ambient temperature on the stability of fecal glucocorticoid (fGCM) and estrogen metabolite (fEM) levels from crested macaques.In total 100 aliquots of fecal extracts from fecal samples collected from female crested macaques (Macaca nigra) living at the Tangkoko-Batuangus Nature Reserve, North Sulawesi were used. We performed two different experiments: (1) An experiment to investigate if levels of fGCM and fEM measured from fecal extracts that were exposed to two, four, six and eight repeated freeze-thaw cycles (test groups) differ to control samples (i.e. fecal extracts always stored frozen); (2) An experiment to evaluate whether storing fecal extracts at ambient temperature for two, four, six, and eight days (test groups)affects the levels of fGCM and fEM compared to the control group (i.e. fecal extracts frozen immediately).Results showed that hormone levels were significantly increased (P0.05) after four freeze-thaw cycles for fGCM and after eight freeze-thaw cycles for fEM. By contrast, there was no significant difference (P0.05) in levels of fGCM and fEM between the test groups and the control group in fecal extracts stored at ambient temperature. In conclusion, our data show that more than two and six repeated freeze-thaw cycles should be avoided when measuring fGCM and fEM in crested macaque fecal extracts, respectively. We also demonstrate that storing fecal extracts at ambient temperature is possible for at least 8 days without taking a risk of affecting the stability of fGCM and fEM levels.

Abstract

The objective of this study was to identify the type of specific glycoconjugates and its distribution in testicular spermatogenic cells in muntjak (Muntiacus muntjak muntjak) based on lectins histochemistry. An adult male muntjak aged 4-5 years old in hard antler period was used in this study. Testicular tissue was fixed in Bouin solution and processed histologically. Histochemistry method was performed using six types biotinylated lectins such as peanut agglutinin (PNA), soybean agglutinin (SBA), wheat germ agglutinin (WGA), ricinus communis agglutinin (RCA), concanavalin A (Con A), and ulex europaeus agglutinin I (UEA I) with 20 g/ml of concentration for PNA lectins and 15g/ml for other type of lectins. The results showed that glycoconjugates were detected by all type of lectins except UEA I in testicular spermatogenic cells with variation in distribution pattern and also the intensity of lectins binding. Glycoconjugates -galactose, -glucose, mannose, Nacetylgalactosamine, N-acetylglucosamine and sialic acid were stained intensely by lectins in golgy-cap phase and acrosomal phase of spermatids. Glycoconjugate N-acetylgalactosamine was the sugar residues which distributed abundantly that marked by positive reaction with PNA, SBA, and RCA lectins. In conclusion, glycoconjugates are detected in testicular spermatids cells of muntjak indicated that glycoconjugates have an important role in spermatogenesis particularly in spermiogenesis.Key words: glycoconjugates, lectins, spermatid, spermatozoa, muntjak

Abstract

Chromolaena odorata L. is a widely used traditional herbal medicine known for its medicinal usage throughout the world. It has been used by the community for its anti-cancer, anti-bacterial, anti-inflammatory, antipyretic, anti-diabetic, and anti-diarrheal, and it also plays a role in hemostasis and wound healing. This study aims to analyze the chemical compounds of C. odorata leaves from Indonesia and to determine the gel stability of its ethanol extract. In this study, C. odorata leaves were collected from Lamkawe Village, Darul Imarah District, Aceh Besar Regency, Aceh, Indonesia. The extraction process was conducted using the maceration method. The active chemical compounds of the ethanol extract of C. odorata leaves were analyzed through phytochemical screening and gas chromatography-mass spectrometry (GC-MS) analysis. The results of the phytochemical screening indicated it contained terpenoids, saponins, flavonoids and phenolics. The most abundant secondary compounds identified were glycidyl palmitate (17.02%), n-9-octadecenoic acid (Z) (15.68%), hexadecanoic acid and l-(+)-ascorbic acid 2,6-dihexadecanoate (12.19%) and 16-hentriacontanone. The 7.5% ethanol extract of C. odorata leaves showed good stability.

Abstract

A wound is a condition where tissue damage occurs due to disruption of tissue continuity caused by injury or surgery. Wound healing is a normal biological process in the human body which aims to restore the function and integrity of damaged tissue. In the process, wound healing involves cellular and biochemical responses, including antioxidants and cytokines, which occur locally and systemically. Interleukin-2 (IL-2) is a cytokine that plays a role in wound healing. It is produced by CD8+ and CD4+ T cells in an active state. In the wound healing process, IL-2 plays an important role in activating macrophages and natural killer (NK) cells and encouraging the proliferation of B cells and T cells. In this review, the role of the IL-2 in wound healing is discussed comprehensively.

Abstract

Herbal antioxidants have gained attention recently as possible treatments for disorders of male reproductive function linked to oxidative stress because of their ability to neutralize free radicals.The investigation into herbal flavonoids for preserving oxidative stress-impaired testicular physiology has been effectively conducted through the visualization of bibliometric analysis. Bibliometric data of published papers collected from the PubMed and Scopus databases in 2018-2023. Network visualization was performed using VOSviewer software on the set keywords that occur (n=15). The literature review was conducted according to the research trends identified by the clusters that appeared in the network visualization. The result revealed that a total of 266 records have been published based on the specified search set keywords since 2018. Through the analysis of keywords, inclusion and exclusion criteria, and the relevant content of papers, 11 articles were identified as suitable for conducting a literature review. Network visualization revealed that the research falls into two general categories: (1) toxicity to the testicles caused by oxidative stress and (2) prevention of oxidative stress impact on tissue through herbal medicine. Examination of the paper's content revealed that certain antioxidant compounds from plants can stabilize free radicals in the testicles. These antioxidants include vitexin, diosmin, hesperidin, phenolic acids, naphthoquinones, fisetin, pachypodol, sciadopitysin, carvacrol, and morin. However, further research is needed to compare active antioxidant content in different herbal plants and identify optimal concentrations for therapeutic effects.

Short communication: Disparities in early luteal phase progesterone concentrations between repeat breeder and fertile Aceh cattle. Asian J Agric 9: 854-860. Repeat breeding remains a major reproductive constraint in cattle, often associated with altered luteal function and inadequate progesterone production during early estrus cycles. Understanding hormonal differences between fertile and repeat breeder cows is essential for improving reproductive efficiency in Aceh cattle (Bos indicus). This study aimed to investigate disparities in progesterone concentrations during the early luteal phase between repeat breeder and fertile Aceh cows. Six multiparous Aceh cows, aged 3-5 years and weighing 150-250 kg, were selected as study subjects. The cows were divided into two groups: repeat breeders (n=3) and fertile cows (n=3). To ensure synchronized estrus timing, all cows received 5 ml PGF2α (Enzaprost®-T, Dinoprost 5 mg) using the by double injection method. Estrus detection was performed following PGF2α injection in the morning and afternoon. Blood samples were collected on days 3 and 6 post-estrus for progesterone analysis. Progesterone concentrations were determined measured using the Enzyme-Linked Immunosorbent Assay (ELISA) method, and the data were analyzed using the Mann-Whitney U test. Results indicated that progesterone concentrations on days 3 and 6 were 5.68±4.98 and 6.61±3.04 ng/mL in fertile cows, whereas in repeat breeder cows, the concentrations were only 0.35±0.07 and 2.74±0.58 ng/mL, respectively. A significant difference in progesterone concentration was observed on day 3 between the two groups (p < 0.05). The conclusion indicates that repeat breeder Aceh cows exhibit lower progesterone concentrations during the early luteal phase compared to fertile cows. These findings contribute significantly to the understanding of hormonal differences between the two groups, which may serve as a foundation for developing more effective and sustainable breeding strategies.
