Abstract

Abstrak— Mikoriza adalah sistem perakaran yang bersimbiosis mutualisme dengan jamur. Penanaman dengan sistem hidroponik mampu membuat mikoriza melakukan kolonisasi lebih cepat sehingga dapat memaksimalkan penyerapan nutrisi P untuk membantu pertumbuhan tanaman. Penelitian ini untuk mengetahui  konsentrasi optimum pemberian pupuk P cair  terhadap pertumbuhan marigold  (Tagetes erecta L.)  yang telah terinfeksi oleh mikoriza dengan mengukur tinggi tanaman dan panjang akar. Perlakuan yang digunakan adalah dengan memberikan konsentrasi pupuk P (fosfat) cair yang berbeda-beda pada tanaman Tagetes erecta yang telah diinfeksikan ke mikoriza dengan beberapa konsentrasi yaitu 0 ppm, 5 ppm, 10 ppm, 20 ppm, 30 ppm dan 40 ppm secara hidroponik. Masing-masing perlakuan diulang sebanyak 4 kali. Hasil analisis ANOVA one-way menunjukkan bahwa adanya pengaruh perlakuan P cair terhadap pertumbuhan vegetatif tanaman Tagetes erecta. Perlakuan 5 ppm memberikan pengaruh optimum terhadap tinggi tanaman dan panjang akar tanaman.

Abstract

Caulerpa lentillifera is one of the most potential green seaweed to explored. It is abundantly available and cultivated in several region in Indonesia. Seaweed is well-known as a low lipid content but it is arranged by polyunsaturated fatty acid. Generally, organic solvent is used for lipid extraction. In an extraction method needs pre-treatment such as enzyme assisted extraction for degrading its cell wall and increasing solvent access to entry the cell. This research was designed to study the optimum condition of lipid enzyme assisted extraction process using cellulase from fresh green macroalga C. lentillifera. The optimization was carried out by Response Surface Methodology (RSM) using Central Composite Design (CCD) model with 15 runs. The aim of this study was to analyze the effect of some independent variables namely enzyme concentrations, hydrolysis temperatures, and hydrolysis times respectively to the dependent variables of lipid content and antioxidant activity. The optimum condition obtained from this experiment was 2% enzyme concentration, 30 °C hydrolysis temperature, and 1 h. The optimum condition could then be verified by making 2 or more replications of the chosen treatment approached the predicted result based on software Design Expert vers. 10 prediction. After methylation, extracted fatty acids were identified as palmitic acid and lauric acid using GC-MS. Extraction optimization enables to explore C. lentillifera’s lipid based on influence factors.

Abstract

Abstract Giant prawn (Macrobrachium rosenbergii) and vaname shrimp (Litopenaeus vannamei) are types of shrimp that became excellent commodities in the fisheries sector. However, one of the obstacles in the vaname shrimp aquaculture is a disease caused by the infection of Taura Syndrome Virus (TSV). One of the consequence of raising the vaname shrimp in Indonesia is the possibility of spreading TSV infection in another shrimp species. TSV infection in giant prawns in Indonesia has not been reported. The aims of this study were: 1. To determine the resistance of giant prawns toward TSV infection and 2. To detect molecularly using RT-PCR technique the presence of viruses TSV on vaname shrimp or giant prawns infected with 3 different doses (0.05 ml; 0.10 ml and 0.15 ml) of TSV inoculum using a pair of specific primers for TSV 9992F (5'-AAG CTT GCG TAG ACA GCC-3') and 9195R (5'-TCA AGA ATG GCT TCC TGG-3'). The research results showed that vaname shrimp mortality infected by 0.05 ml; 0.10 ml and 0.15 ml TSV inoculums were 14.28%, 42.86%, and 57.14%, respectively. Whereas the giant prawns mortality that were infected using the same dose of TSV inoculums were 0%, 8.33%, and 8.33%, respectively. Positive result was detected molecularly only from haemolymph of vaname shrimp infected using 0.15 ml of TSV inoculum. On the other hand, positive results were detected in pleopod and the gill of vaname shrimp infected using 0.05 ml; 0.10 ml or 0.15 ml of TSV inoculums. In giant prawns, infection using 3 different doses of TSV inoculums causes negative result molecularly. Based on all of the facts, it can be concluded that, giant prawns has the higher resistance to TSV infection than that of vaname shrimp.

Abstract

Malaria is a disease caused by Plasmodium parasite transmitted by female Anopheles mosquito. Jung rahab plant (Baeckea frutecens) is known have an ability to inhibit the growth of Plasmodium falciparum. However, the inhibition of the parasite growth is still unknown. This research aimed to determine fractionation of aqueaous extract of jung rahab leaves that is contain antimalaria compound with a potential inhibition activity. Jung rahab plant was extracted using decoctition. The analysis included phytochemical screening, column chromatography fractionation, antimalaria activity, and compound identification by using UV-Vis, FT-IR, and GCMS. Jung rahab aqueous extract fraction resulted a potential antimalaria activity with IC50 105,9 ppm and fractionation result based on UV-Vis, FT-IR, and GCMS as pyrogallol.Keywords: antimalarial, β-hematin, Baeckea frutecens, extraction, IC50ABSTRAKMalaria merupakan penyakit yang disebabkan oleh parasit Plasmodium yang ditransmisikan oleh nyamuk Anopheles betina. Daun jung rahab (Baeckea frutecens) diketahui memiliki kemampuan untuk menghambat pertumbuhan Plasmodium falciparum. Akan tetapi, mekanisme penghambatan pertumbuhan parasit tanaman ini masih belum diketahui. Penelitian ini bertujuan untuk mencari fraksi lain dari ekstrak air daun jung rahab yang mengandung senyawa antimalaria dengan aktivitas penghambatan yang lebih tinggi. Ektraksi daun jung rahab dilakukan dengan metode dekoktasi, sedangkan analisis yang dilakukan adalah skrining fitokimia, fraksinasi menggunakan kromatografi kolom, uji aktivitas antimalaria, dan identifikasi senyawa hasil fraksinasi dengan UV-Vis, FT-IR, dan GCMS. Berdasarkan hasil penelitian ini, fraksi ekstrak air daun jung rahab memiliki aktivitas antimalaria potensial dengan nilai IC50 sebesar 105,9 ppm dan hasil fraksinasi senyawa berdasarkan UV-Vis, FT-IR, dan GCMS sebagai pirogalol.Kata kunci: antimalaria, β-hematin, Baeckea frutecens, ekstraksi, IC50­

Abstract

ABSTRACTMangrove endophytic fungi potentially produce secondary metabolites such as antioxidant. Antioxidants are compounds that are widely applied in the industry as an ingredient in the manufacture of products in the food and health sector. In this study, we investigated the antioxidant  potential of secondary metabolites from fungal endophytic of mangrove species Aegiceras corniculatum. A total of eight endophytic fungi were successfully isolated, two isolates from leaf, three isolates from fruit and three isolates from twig.  Between the isolates obtained, only one isolated was active as the antioxidant with IC50 value 19.28 µL/mL eventhough still lower than the standard ascorbic acid (6.08 µL/mL). The results of chemical identification using GC-MS showed several chemical compounds that have antioxidant activity including phenol,3,5-bis(1,1-dimethylethyl), hexadecanoic acid, hexadecanoic acid methyl ester, malic acid, N-aminopyrrolidine,  9-octadecanoic acid, methyl ester (E), benzeneethanol, 4-hydroxy, 1,2-benzenedicarboxylic acid, d-tyrosine, bis(2-methylpropyl) ester 1-nonadecene dan heneicosane.  The selected fungal endophytic isolated were identified using molecular Internal Transcribed Spacer (ITS) marker and has a high taxonomy similarity with Microdochium sp.Keywords: Aegiceras corniculatum, antioxidant, endophytic fungi, mangrove, Microdochium sp ABSTRAKKapang endofit dari mangrove memiliki potensi dalam menghasilkan metabolit sekunder seperti antioksidan. Antioksidan merupakan senyawa yang banyak diaplikasikan dalam industri sebagai  bahan untuk pembuatan produk di bidang pangan maupun kesehatan. Penelitian ini bertujuan untuk  mengetahui potensi antioksidan kapang endofit dari mangrove species Aegiceras  corniculatum. Sebanyak 8 isolat kapang endofit berhasil diisolasi, yaitu 2 isolat berasal dari daun, 3 isolat berasal dari buah dan 3 isolat berasal dari ranting.  Di antara 8 isolat tersebut, hanya 1 isolat yang dihanyakan aktif sebagai antioksidan dengan nilai IC50  sebesar 19,28 µL/mL, meskipun nilai ini masih lebih rendah dibandingkan vitamin C yaitu sebesar 6,08 µL/mL. Hasil identifikasi kimia dengan menggunakan GC-MS menunjukkan beberapa senyawa kimia yang memiliki aktivitas aktioksidan di antaranya phenol,3,5-bis(1,1-dimethylethyl), hexadecanoic acid, hexadecanoic acid methyl ester, malic acid, N-aminopyrrolidine,  9-octadecanoic acid, methyl ester (E), benzeneethanol, 4-hydroxy, 1,2-benzenedicarboxylic acid, d-tyrosine, bis(2-methylpropyl) ester 1-nonadecene dan heneicosane. Abstract

ABSTRACTPenicillium chrysogenum is one of the fungi which known to produce penicillin G antibiotics. The current superior strain P. chrysogenum producing high penicillin G has gone through various stages from the wild type isolates. The wild type isolates of P. chrysogenum from Indonesia also have great potential to be developed. Therefore, this study aimed to determine the changes of penicillin G levels from mutated P. chrysogenum isolate with both ultraviolet (UV) radiation and chemical mutagens ethyl methane sulfonate (EMS) compared to the wild type isolates. Random mutation through UV radiation, EMS chemical mutagen and their combination was carried out on wild type isolates of P. chrysogenum. Antibacterial activity was tested against Escherichia coli and Bacillus subtilis. The level of penicillin G produced was detected using HPLC with the Penicillin G as the standard. The results showed that the treatment of UV, EMS mutations and their combination can increase the antibacterial activity as well as levels of penicillin G than the wild type. Mutant C5-4.10 isolate resulted from the combination UV and EMS had the best antibacterial activity and produced penicillin G. level 2.9 times compared to the wild type.Keywords: antibiotic, HPLC, mutation, Penicillium chrysogenum, penicillin GABSTRAKPenicillium chrysogenum adalah salah satu kapang yang diketahui memproduksi antibiotik penisilin G. Strain P. chrysogenum unggul penghasil penisilin G tinggi yang saat ini tersedia merupakan galur yang telah melalui berbagai tahapan pemuliaan dari wild type-nya. Isolat P. chrysogenum asal Indonesia juga memiliki potensi besar untuk dikembangkan. Oleh karena itu, penelitian ini bertujuan untuk mengetahui perubahan kadar penisilin G dari isolat P. chrysogenum yang telah dimutasikan menggunakan radiasi sinar ultraviolet (UV) dan mutagen kimiawi etil metan sulfonat (EMS) dibandingkan dengan wild type nya. Mutasi acak menggunakan radiasi sinar UV, mutagen kimiawi EMS dan kombinasi keduanya dilakukan pada isolat P. chrysogenum. Uji aktivitas antibakteri dilakukan terhadap Escherichia coli dan Bacillus subtilis. Kadar penisilin G yang diproduksi dideteksi menggunakan HPLC dan dibandingkan dengan standar Penicillin G. Hasil penelitian menunjukkan bahwa mutasi menggunakan sinar UV, EMS dan kombinasi keduanya dapat meningkatkan aktivitas antibakteri serta kadar penisilin G dibandingkan dengan wild type nya. Isolat mutan C5-4.10 hasil mutasi kombinasi sinar UV dan EMS memiliki aktivitas antibakteri terbaik dan kadar penisilin G sebesar 2,9 kali lipat dibandingkan dengan wild type nya.Kata kunci: antibiotik, HPLC, mutasi, Penicillium chrysogenum, penisilin G

Abstract

Alpukat (Persea americana Mill.) merupakan tanaman yang banyak ditemukan di daerah tropis seperti Indonesia. Selama ini masyarakat hanya memanfaatkan daging buahnya saja untuk dikonsumsi, kemudian bagian bijinya dibuang. Penelitian ini bertujuan untuk mengisolasi, menguji dan mengidentifikasi senyawa aktif antioksidan dari ekstrak biji alpukat yang dilakukan dari Januari sampai Juni 2018 di Laboratorium Kimia Bahan Alam, Pusat Penelitian Bioteknologi-LIPI. Buah alpukat yang digunakan dalam penelitian diperoleh dari Pasar Induk Cibitung, Bekasi. Ekstraksi dilakukan secara bertingkat menggunakan pelarut n-heksana, etil asetat, etanol 96%, dan air. Uji aktivitas antioksidan dilakukan dengan metode peredaman radikal bebas menggunakan reagen 2,2-difenil-1-pikrilhidrazil (DPPH), penentuan struktur kimia senyawa aktif antioksidan menggunakan metode spektrofotometri UV-Vis, Spektroskopi Inframerah Transformasi Fourier (FTIR), dan GCMS.  Hasil pengujian aktivitas antioksidan menunjukkan bahwa ekstrak etanol 96% memiliki aktivitas antioksidan tertinggi pada isolat EtOH.4.3.1 sebagai isolat paling aktif dengan IC50 sebesar 23,07±1,63 μg.ml-1. Hasil identifikasi berdasarkan data spektrofotometri UV-Vis, Spektroskopi Inframerah Transformasi Fourier, dan GCMS menunjukkan bahwa isolat EtOH.4.3.1 biji alpukat mengandung senyawa 2-etilheksil-4-metoksisinamat, yang terbukti memiliki aktivitas sebagai antioksidan. Analisis lebih lanjut perlu dilakukan menggunakan spektrometri Resonansi Magnetik Inti (RMI) proton dan karbon untuk memastikan jumlah dan posisi proton serta karbon dalam struktur kimia tersebut.

Abstract

There is an increasing amount of evidence that oxidative stress related to hypertension can damage the function of diverse structures such as aorta. It is a well-established fact that chlorogenic acid and anthocyanine found in purple sweet potato generates bioactive compound with antihypertensive and antioxidant activities. The present study sought to investigate antioxidant activity of extract ethanol of purple sweet potato (EP) in deoxycorticosterone acetate (DOCA–salt)–induced hypertensive rats (Rattus norvegicus). The rats were orally administrated a 95% ethanol extract of purple sweet potato (var. Ayamurasaki) (EP) in a daily dose of 200 and 400 mg/kg body weight for 4 weeks. Aorta total malondialdehyde (MDA) and histopathology of aorta abdominal were examined. Aorta injury was observed in DOCA-salt hypertensive group rats compared to normotensive group rats, as aorta MDA significantly increased  (P <0.05). In contrast, treatment of DOCA-salt hypertensive rats with different dose of EP significantly reduced the total aorta MDA, as well as repair kidney damage, suppressed smooth muscle cell proliferation and lessen aorta wall thickening compared to controls.   This is the first report that demonstrated blood pressure lowering and antioxidative effects of an ethanol extract of purple sweet potato, containing chlorogenic acid, in a DOCA–salt model of hypertension.

Abstract

Ubi Jalar mengandung komponen serat fraksi oligosakarida seperti raffinosa, verbakosa, dan stakhiosa yang berpotensi sebagai prebiotik, yaitu subtrat yang dapat menstimulasi pertumbuhan bakteri probiotik. Tujuan penelitian ini adalah untuk membandingkan viabilitas bakteri probiotik seperti L. plantarum dan L. casei pada medium fermentasi berbasis tepung ubi jalar dan untuk mengetahui perubahan komponen tepung ubi jalar dari tiga jenis ubijalar selama proses fermentasi.  Penelitian ini menggunakan dua faktor: jenis bakteri probiotik (L. casei dan L. plantarum) dan jenis tepung ubi jalar (tepung ubi jalar putih, kuning dan ungu). Data dianalisis dengan ANNOVA dengan nilai α<5%. Perlakuan terbaik dipilih dengan metode Multiple atrribute. Berdasarkan hasil penelitian didapatkan jenis ubi jalar mempengaruhi viabilitas L. casei dan L. plantarum selama proses fermentasi. Perbedaan jenis tepung ubi jalar secara signifikan berbeda nyata terhadap nilai pH, total asam, total BAL, gula reduksi sedangkan perbedaan jenis bakteri probiotik sangat mempengaruhi nilai pH, total asam, total BAL, total gula, serat kasar, gula reduksi, total nitrogen dan kadar pati. Perbedaan jenis tepung ubi jalar dan bakteri probiotik memiliki perbedaan signifikan (nilai α<1%)  terhadap pH, total asam, total BAL dan gula reduksi. Perlakuan terbaik adalah medium terfermentasi dari tepung ubi jalar ungu (10% b/v) oleh L. plantarum dengan total BAL yaitu 1,55x1010 CFU/ml, total asam 0,953%, nilai pH 4,267 total gula 1,647%, kadar pati 2,567%, serat kasar 0,066%, gula reduksi 3,139% dan total N 0,023%.

Abstract

Ihau fruit (Dimocarpus longan var. malesianus Leenh) is a typical Kalimantan fruit which has a longan-like morphology. The vitamin C content and various phytochemical compounds of ihau has the potential as a natural antioxidant and antibacterial agent. This study aimed to determine the potential antioxidant and antibacterial activity of ihau extract on Staphylococcus aureus bacteria in vitro with a post-test only control group design. Samples were extracted with 96% ethanol and water using the maceration method. Antioxidant activity measured using DPPH test. Antibacterial test using well diffusion method was performed in six treatment groups (ethanol extract concentration 50, 60, 70, 80, 90, 100mg/ml), positive control (Ampicillin) and negative control (sterile distilled water). The results showed that IC50 value for the ethanol extract of ihau was 681.05 g/ml and the aqueous extract was 698.3 g/ml. The antibacterial test indicated that ethanol extract had an inhibitory effect on the growth of Staphylococcus aureus at 70 mg/ml to 100 mg/ml concentration. It can be concluded that ihau fruit extract has antibacterial ability against gram positive bacteria (Staphylococcus aureus) but does not have antioxidant activity.Keywords - Ihau fruit, ethanol extract, antioxidant, antibacterial, Staphylococcus aureus.
