Abstract

The study is about to improve the students listening skill and pronunciation by using podcast as one of material sources taken from the internet. The objectives of this study are to identify some steps that are going to be implemented in teaching listening and speaking using podcast to improve the students’ listening skill and their pronunciation of English Department students of Dian Nusantara University, and to analyze the effectiveness of using podcast in their learning. The researchers apply the concept of ADDIE (Analysis, Design, Development, Implementation, and Evaluation) paradigm in this study because it is more effective and systematic and supported new learning models. The participants are the fourth- semester students of English Literature students. There are two methods of collecting data in this research. They are quantitative and qualitative data. Each week, quantitative data were gathered via a series of tests, including a listening comprehension and a pronunciation examination. For qualitative data, questionnaires were used to elicit student perspectives on podcasts' effectiveness in improving listening comprehension and pronunciation. Preliminary comparative data will be gathered through a pre-test. This study uses descriptive statistical methods to analyze the data. The result is then analyzed using descriptive statistics to know the difference of scores before and after the tests. The students' ability to pronounce words has improved. The majority of the pre-test pronunciation errors are no longer present in the post-test. Furthermore, the use of podcasts as a learning medium received positive responses in the student survey. Students are encouraged to improve their listening skills because podcasts appeal to them as a learning medium.

Abstract

Antibiotics are chemical or biological substances that have the ability to kill pathogens selectively. Currently, high-performance liquid chromatography (HPLC) is used routinely in the detection of antibiotics. However, the cost of analysis and running time are bottlenecks for HPLC to be used for routine tests to detect antibiotics. Alternative methods need to be developed to overcome this issue. In this study, the development of a penicillin G specific biosensor by using a DNA aptamer and gold nanoparticles (AuNPs) was done. Optimal aptasensor conditions were achieved with the concentrations of NaCl and aptamer at 0.25 M and 2 μM, respectively. An aptasensor of this type showed LOD for penicillin G at 3 mg/L and was able to detect penicillin G in the range of 3 to 27 mg/L. The established aptasensor showed specific sensitivity toward penicillin G after testing with several antibiotics, i.e., ampicillin, kanamycin, chloramphenicol, and erythromycin. The aptasensor could detect the presence of penicillin G from culture medium of wild-type, ultraviolet irradiation mutant, gamma irradiation mutant, and ultraviolet irradiation and gamma irradiation mutant strains of P. chrysogenum, at detection concentrations of 9.75 ± 0.004; 25.25 ± 0.005; 37.5 ± 0.005; and 45 ± 0.004 mg/L, respectively.

Abstract

Dari kacamata kesehatan, kandungan nutrisi jamur maupun fungsinya sebagai obat telah dikenali oleh Negara China dari 2000 tahun yang lalu. Jamur memiliki kandungan air yang tinggi, sehingga jamur segar mempunyai kandungan makronutrient dan energi yang rendah. Jamur merupakan sumber yang baik untuk protein yang dapat dicerna, dilain pihak kandungan lemaknya rendah. Kandungan lemak yang ada pada jamur biasanya terdiri dari phospholipid, sterol, sterol ester, mono-, di-, triglycerides, termasuk juga asam lemak. Sedangkan komponen bioaktif yang ada pada jamur biasanya terdiri dari polisakarida dan proteoglycans. Komponen bioaktif dari jamur mempunyai kemampuan sebagai antioksidan yang kuat. Pada perkembangannya penyakit yang akut dan kronis dapat mengkonsumsi antioksidan sebagai salah satu langkah untuk meningkatkan kesehatan. Berdasarkan hal ini, perubahan kebiasaan makanan dengan meningkatkan konsumsi jamur, dapat meningkatkan jumlah komponen bioaktif pada tubuh sehingga dapat mengurangi resiko penyakit kronis.

Abstract

Ekstrak biopigmen biru Fikosianin Spirulina plantesis dapat digunakan sebagai pewarna alami, contohnya untuk minuman. Penelitian dilakukan tiga tahap yaitu formulasi minuman ringan karbonasi, analisis kimia formulasi terpilih dan uji stabilitas penyimpanan. Formula minuman ringan karbonasi terpilih menurut panelis yaitu konsentrasi ekstrak biopigmen biru fikosianin 4% (F1) dengan warna yang disukai.Sedangkan rasa dan aroma dinilai agak disukai. Formula terpilih berdasarkan uji hedonik, dilanjutkan pada penelitian tahap dua yaitu analisa kimia formula terpilih meliputi kadar total fikosianin sebesar 3,21 mg/mL, kadar protein (Bradford) sebesar 67,15 ppm, aktifitas antioksidan (IC50) sebesar 545,6 μL, intensitas warna sebesar 1,302 (λ=615) dan 1,296 (λ=625) dan pH sebesar 3,30.Penentuan stabilitas dihitung dengan metode waktu paruh.Waktu paruh parameter kadar total fikosianin pada suhu refrigrator yaitu 67 hari atau 2 bulan, dan suhu ruang yaitu 12 hari. Intensitas warna pada suhu refrigrator yaitu 87 hari atau 3 bulan, dan suhu ruang yaitu hanya 13 hari. Semakin tinggi waktu paruh, maka semakin lama degradasi fikosianin yang akan terjadi.

Abstract

Penelitian ini bertujuan untuk menghasilkan instrumen penelitian berupa lembar validasi produk multimedia pembelajaran fisika yang valid dan reliabel. Instrumen berupa lembar validasi produk multimedia pembelajaran fisika ini terdiri dari 55 item pernyataan yang berfungsi untuk menilai kelayakan produk multimedia pembelajaran fisika. Penelitian ini menggunakan model pengembangan 3D yang dimodifikasi dari Thiagarajan. Data validitas dan reliabilitas instrumen diperoleh dari validator yang terdiri dari tiga orang ahli. Pengumpulan data menggunakan teknik angket dengan skala 1 sampai 4. Teknik analisis data menggunakan formula Aiken untuk uji validitas instrumen dan Cronbach’s Alpha untuk uji reliabilitas instrumen. Hasil penelitian menunjukkan bahwa terdapat 8 item yang memperoleh nilai koefisien Aiken’s V sebesar 0,60<V<0,80 (validitas tinggi) dan 47 item memperoleh nilai 0,80<V<1,00 (sangat tinggi). Oleh karena itu dapat disimpulkan bahwa 55 item pada instrumen adalah valid. Hasil uji reliabilitas instrumen memperoleh nilai Cronbach’s Alpha sebesar 0,994 yang termasuk dalam kategori sangat reliabel. Dengan demikian, instrumen berupa lembar validasi produk multimedia pembelajaran fisika yang dikembangkan sangat valid dan reliabel sehingga layak digunakan untuk mengukur kelayakan  produk multimedia pembelajaran fisika.Development Of Validation Instruments Multimedia Products Physics LearningAbstractThis study aims to produce a valid and reliable instrument as a validation sheet for measuring the advisability of physics learning multimedia products. The instrument consists of 55 items. This study uses a 3D development model that is modified from Thiagarajan. Validity and reliability of instruments obtained from validators consisting of three experts. Data collection uses a questionnaire technique with a scale of 1 to 4. The data analysis technique uses the Aiken formula to test the validity of the instrument and Cronbach's Alpha to test the reliability of the instrument. The results showed that eight items scored Aiken's V coefficient of 0.60 <V < 0.80 (high validity), and 47 items scored 0.80 <V<1.00 (very high). Therefore, it can be said that 55 items on the instrument are valid. The results of the instrument reliability test obtained a Cronbach's Alpha value of 0.994, which was included in the very reliable category. Thus, the instrument is very valid and reliable, so it is suitable for measuring the advisability of physics learning multimedia products.

Abstract

Antioxidant and anticancer activities of sand sea cucumber Holothuria scabra (dried and fresh) extracts were studied. The highest extraction yield of sea cucumber H. scabra (3.9%) was obtained using dried H. scabra at 60oC. The highest antioxidant activity was found in fresh H. scabra extract at 60°C with an IC50 value of 629.89 ± 0.15 µg/mL using the ABTS method, and the highest antioxidant activity by DPPH method was found in dried H. scabra extract at 70°C with an IC50 value of 32017.18 ± 0.82 µg/mL. The best antioxidant activity based on FRAP and TBARS methods was found in fresh H. scabra extracts at 80°C, respectively. The highest total phenol and flavonoid contained in dried H. scabra extract were 317.54 ± 8.91 mg GAE/100 g sample and 247.56 ± 11.70 mg QE/100 g sample. H. scabra extracts inhibited more than 50% of the growth of the MDA-MB-231 cell line at concentrations of 25 and 50 μg/mL except for dried H. scabra extracts at 80°C. Similarly, the extracts showed the highest cytotoxic effect up to 100% at the highest concentration (100 μg/mL) except for dried H. scabra extracts at 70°C and 80°C.

Abstract

Mangrove endophytic fungi have the potential to produce secondary metabolites with antibacterial properties. This study aimed to obtain endophytic mold isolates associated with mangrove plants and their antimicrobial activity. Seventeen endophytic molds were isolated from 7 mangrove species originating from Berau, East Kalimantan, Indonesia. Three extracts from the mold isolates OJ-B, OJ-Bu, and 6Xg-Bh showed the highest antibacterial activity with an inhibition zone of 9.63–25.4 mm and with MIC (minimum inhibitory concentration) and MBC (minimum bactericidal concentration) values ??of around 0.625–5 and 2.5–10 mg mL–1, respectively. LC-MS (liquid chromatography-mass spectrometry) analysis identified several compounds with antibacterial potential, namely beauvericin, choline, nicotinic acid, pyridoxine, quinoline, cyclo (phenylalanyl-prolyl), fusarin c, oleamide and borrelidin. The 3 species were molecularly identified as Diaporthe phaseolorum, Fusarium proliferatum and Phomopsis sp. Thus, the endophytic molds from several of the mangrove species produced bioactive compounds as antibacterial.  Kapang endofit mangrove berpotensi menghasilkan metabolit sekunder yang bersifat antibakteri. Penelitian ini bertujuan mendapatkan isolat kapang endofit yang berasosiasi dengan tanaman mangrove beserta aktivitas antimikrobanya. Tujuh belas kapang endofit diisolasi dari 7 spesies mangrove asal Berau, Kalimantan Timur, Indonesia. Tiga ekstrak dari isolat kapang OJ-B, OJ-Bu, dan 6Xg-Bh memiliki aktivitas antibakteri tertinggi dengan zona hambat 9.63–25.4 mm dan dengan nilai MIC dan MBC masing-masing sekitar 0.625-5 dan 2.5–10 mg mL–1. Analisis LC-MS menunjukkan beberapa senyawa dengan potensi antibakteri yaitu beauvericin, choline, nicotinic acid, pyridoxine, quinoline, cyclo (phenylalanyl-prolyl), fusarin c, oleamide dan borrelidin. Uji molekuler menunjukkan 3 spesies terindentifikasi sebagai Diaporthe phaseolorum, Fusarium proliferatum dan Phomopsis sp.  Hasil menunjukkan kapang endofit dari beberapa spesies mangrove tersebut mampu menghasilkan senyawa bioaktif sebagai antibakteri.

Abstract

An increase in blood glucose levels in people with diabetes can cause an increase in free radicals, which can worsen the disease. Thus, drugs that have antidiabetic and antioxidant activities are needed. The cinnamon plant is high in antioxidants and has long been used as a source for a diabetes drug. The utilization of endophytic fungi isolated from cinnamon plants as antidiabetic and antioxidant has never been reported. This study aims to investigate the antidiabetic as well as antioxidant activity from the extract of endophytic fungi from the cinnamon plant. The antidiabetic activity was tested using the α-glucosidase enzyme inhibition method, while antioxidant activity was tested using the DPPH free radical scavenging method. Total phenol content was measured based on the Follin-Ciocalteu reagent reaction. All endophytic fungal extracts from the cinnamon leaves, twigs, flowers, and fruit have antidiabetic and antioxidant activity as well as high total phenol content. The three parameters measured showed a positive correlation. Endophytic fungal extract of Cb.D6 isolate derived from the leaf had the highest antidiabetic and antioxidant activity among the other isolates amounting to 92.41% and 90.28%, respectively. In addition, the total phenol content of Cb.D6 isolates was also the highest with 357.83 mg equivalent to gallic acid/g extract. Therefore, the endophytic fungal extract of Cb.D6 isolate has the potential to be developed as a source of the antidiabetic and antioxidant ingredients.

Abstract

This study aims to determine whether the differences in communication skills of students who use the Peer Assessment technique with students who use Teacher Assessment and how much the influences to students. The research method was using pre-experimental design with Intact-Group Comparison. The sample of this research is grade VII students of SMP Negeri 19 Pontianak. The results of observational data analysis of students' communication skills in the experimental class and the control class using the Independent Samples T-test, showed the differences in communication skills between the class using Peer Assessment technique and the class using Teacher Assessment. The result obtained, the average communication skills of students who use the Peer Assessment technique is 66.6 % with a good category and students who use Teacher Assessment is 59.3 % with a fairly good category. Based on the calculation of the effect size obtained results indicate that there is an effect of Peer Assessment on students' communication skills by 0.9 with a high category. Keywords: Students' communication skills, peer Assessment.

Abstract

Cancer prevalence is steadily increasing each year and becomes the second leading cause of death worldwide. In Indonesia, breast cancer had a prevalence of 16.7% in 2018. Free radicals contribute to the occurrence of breast cancer, while antioxidants play a vital role in protecting cells and repairing the damage caused by free radicals. Ihau, an endemic fruit in Kalimantan Island, contains phytochemical compounds with potential antioxidant and anticancer properties. Utilizing local food as natural antioxidants could serve as an alternative for breast cancer prevention and treatment. This study aims to assess the anticancer potential of Ihau fruit extract on the MCF-7 breast cancer cell line. A post-test-only control group design method using the MTT assay was used, with two treatment groups: water and 96% ethanol extract, and four different concentrations (125, 250, 500, and 1000 ppm), and each was replicated three times. Statistical analysis using the ANOVA test found no significant difference among all concentrations. The IC50 values of the cytotoxic activity of water and ethanol extracts were 1,197.7 ppm and 1,148 ppm, respectively. It can be concluded that both water and ethanol extract of Ihau fruit exhibited very weak cytotoxic activity.
