Abstract

The Leydig cells play crucial role in steroidogenesis and spermatogenesis. Those processes need complex communication in hormonal and testicular to maintain male reproductive function. Abnormal condition induced by reactive oxygen species reduce cell viability through lipid peroxidation and apoptotic pathway. Antioxidant ameliorate ROS elevation and prevent cell damage. Specifically, Leydig cells are vulnerable to ROS exposure and decline its function in mediating spermatogenesis. Therefore, it is needed to improve Leydig cells viability within antioxidant supplementation. This study aimed to determine the protective effect of antioxidant on Leydig cells induced by reactive oxygen species. This type of study is a literature review. Various studies have been reviewed through critical appraisal tool Olsen-Baisch Scoring for integrated review. Furthermore, this study highlighted the importance of the mechanism of antioxidant as protective agent of Leydig cells. Supplementation of antioxidant with the correct administration, dosage, and duration is potential to balance reactive oxygen species level and protect Leydig cells.    Keywords             : Leydig cell, antioxidant, reactive oxygen species, infertility

Abstract

Environmental pollution is one of the factors that contribute to the decline in male fertility. Lead is one of six air pollutants harmful to the reproductive system. One parameter of infertility in men is an increase in reproductive function observed with decreased morphological abnormalities of spermatozoa. The purpose is to analyze the effect of giving various dosages of Solanum betaceum extract on spermatozoa morphology of mice exposed to lead acetate. This study was true experimental using a randomized post-test only control group design. The total sample was 40 male mice Balb/ C taken by simple random sampling technique. Treatment and maintenance of experimental animals for 35 days. Statistical tests showed there were significant differences with p-value 0.005. Solanum betaceum extract can be used as a protective to improve the normal morphology spermatozoa of mice that exposed to lead acetate.

Abstract

Lead is a toxic material that can have negative effects on reproductive organs. Lead exposure reduces the activity of endogenous antioxidant enzymes and increases the number of oxidants in the body. High free radicals will affect spermatogenesis and result in decreased motile spermatozoa. Antioxidants are known to protect the motility of spermatozoa, and adequate antioxidants can be found in Dutch eggplants (Solanumbetaceum). This study aimed to evaluate the effect of S. betaceum on spermatozoa motility after exposure to lead acetate. This study is a true experimental design with a randomized post-test-only control group design. Forty male Balb/C mice 12 weeks old were randomly divided into 5 groups: two control groups (C0, C1) and three treatment groups (T1, T2, T3).The C0 received distilled water, and the C1 received 75 mg/kg BW lead acetate. The T1, T2 and T3 received 100, 200 and 400 mg/kg BW of S. betaceum, respectively, an hour before exposed lead acetate. The data were analyzed using one-way ANOVA with a significant level of p <0.05. A significantly increasein the mean total motility of spermatozoa in T1, T2, and T3 was reported.This study indicates that S. betaceum have a protective effect on spermatozoa motility when exposed to lead acetate.

Abstract

BACKGROUND: Cancer stem cells (CSCs) eradication might serve as a robust approach for cancer eradication. MCF-7 as breast cancer continuous cell line is known to contain breast CSCs (BCSCs) for its capability to maintain its original tumor population. CSCs enriched culture is a fundamental tool for CSCs targeted therapy development. Effective and unsophisticated CSCs dedifferentiation protocol for producing CSCs enriched culture is needed.METHODS: MCF-7 cells were cultured initially in Dulbecco's Modified Eagle Medium (DMEM) low glucose medium then changed to DMEM:F12. Serum starvation was performed during each medium refreshment gradually with fetal bovine serum (FBS) concentration of 10%, 5%, 2.5% until reaching 1% FBS concentration. Stable MCF-7 culture was then adapted to serum free culture system, containing DMEM:F12, epidermal growth factor (EGF), basic fibroblast growth factor (bFGF), and B27 supplement as dedifferentiation protocol for 18 days. Cluster of differentiation (CD)44 and CD24 double staining immunocytochemistry was performed to evaluate cell stemness.RESULTS: The population of cells expressing BCSCs markers (CD44+/CD24low) in non-adherent single cells subpopulation was significantly increased after the dedifferentiation procedure (70.39%) compared to control groups (0.71%) (p<0.05). In contrast, the expression of BCSCs marker in adherent single cells subpopulation and for both adherent and non-adherent mammosphere the BCSCs markers showed a stable expression.CONCLUSION: BCSCs enrichment of breast cancer cell cultures from MCF-7 breast cancer cell line can be performed. Breast cancer cell plasticity is observed during the dedifferentiation protocol. Development of dedifferentiation inducing protocols can serve as an important foundation for breast cancer therapy development through BCSCs elimination.KEYWORDS: breast neoplasms, cell line, dedifferentiation, immunohistochemistry, neoplastic stem cells

Abstract

Introduction: Ketogenic diet seems to be in a great demand nowadays as a lot of people are interested in adopting it into their lifestyle. It is also found that the ketogenic diet shows several beneficial effects including cancer prevention. However, the detail mechanism still remains unknown. Therefore, this review was aimed to find out the effect of ketogenic diet on Bcl-2 (B-cell lymphoma-2) expression in cancer.Methods: We searched published literatures in PubMed through 2011-2020 using specific keywords. The literatures were filtered according to inclusion and exclusion criteria. Animal model, total sample size, underlying condition/inflammatory process occurs, details of the intervention/diet including diet contents in control group and high-fat group, and the duration of the intervention, Bcl-2 results, and p-value were extracted.Results: 7 studies were included in this review. Bcl-2 expression found decrease in 5 out of 6 studies. Similar result also obtained in Bcl-2 protein level, which measured by western blot. Bcl-2 protein level shows a decrease in 2 out of 3 studies.Conclusion: This review shows that high-fat diet that contained in ketogenic diet most likely lead to decrease in Bcl-2 expression. Therefore, indicating the ability of ketogenic diet to affect cancer progression by inducing apoptosis process.

Abstract

COVID19 pandemic became the primer focus of health problem by the government since the case was increasing every month in 2020. The impact of this pandemic was spread quickly across many areas including Surabaya city. Youth generation often found to be a silent source of infection. Furthermore, young people behavior often ignoring health prevention policy makes the infection more easily to spread. This condition might be because of their knowledge regarding COVID19 was relatively not enough. Factor that may affect their knowledge are varies including knowledge of city of residence. This study aims to evaluate the correlation between knowledge about city of residence with youth generation knowledge of COVID19 as this factor has not yet been studied before in Indonesia. We conducted an analytical cross-sectional with an online survey for youth generation across Surabaya to understand their knowledge concerning Surabaya and COVID19. A total of 57 participants were voluntarily enrolled in this study. The result data were analyzed statistically. There is significant positive correlation between participants’ knowledge of Surabaya city with knowledge of COVID19 (r= 0.314; p= 0.017). Furthermore, knowledge of COVID19 was significantly different between youth who have better knowledge of Surabaya than lower knowledge (p= 0.021). Knowledge of Surabaya city was correlated with knowledge of COVID19. Youth who have better knowledge regarding their city of residence seem to have better knowledge of COVID19.  Knowledge concerning city of residence proven to be important in order to increase knowledge of COVID19 in youth generation.

Abstract

Kanker kolorektum merupakan salah satu kanker yang tersering di dunia. Target molekuler untuk pengobatan kanker kolorektumyaitu Epidermal Growth Factor Receptor (EGFR) dengan pemberian antibodi monoklonal anti-EGFR. Pemberian pengobatan ini tidakdapat memberikan efek dampak di pasien dengan status gen KRAS bentuk mutan, sehingga perlu dilakukan pemeriksaan status mutasigen KRAS. Telitian berupa deskriptif dengan pendekatan potong lintang yang bertujuan untuk mendapatkan data status mutasi genKRAS kodon 12 dan 13 di pasien adenocarcinoma colorectal. Deteksi mutasi KRAS dilakukan dengan teknik Polymerase Chain ReactionRestriction Fragment Length Polymorphism (PCR RFLP) yang dikonfirmasi dengan sekuensing. Sampel telitian adalah 30 blok parafinyang diperoleh dari Rumah Sakit Dr.Soetomo Surabaya masa waktu Januari-Desember 2013. Setelah dilakukan ekstraksi DNA terdapat21 sampel yang dapat digunakan untuk pemeriksaan lanjutan. Hasil PCR RFLP menunjukkan terdapat 7/21 mutasi pada kodon12 dan tidak terdapat mutasi gen KRAS pada kodon 13. Mutasi pada kodon 12 yaitu GGT>GCT, GGT>GGA dan GGT>GAT yangmenyebabkan perubahan asam amino Gly12Ala, Gly12Gly dan Gly12Asp. Simpulan telitian ini adalah mutasi gen KRAS kodon 12 padaadenocarcinoma colorectal di Rumah Sakit Dr. Soetomo Surabaya sebanyak 33%.

Abstract

AbstractIntroduction: Infertility is one of the common clinical symptoms in women with endometriosis. Limited ways to diagnose endometriosis, symptoms of endometriosis considered normal and overlap with other diseases that cause delaying treatment that increase the duration of infertility. This study purpose to analyze the differences in duration of infertility to women with endometriosis and without endometriosis. Method: This study is observational analytical research with retrospective case-control designed by looking at medical records in Fertility Clinic Graha Amerta RSUD Dr. Soetomo Surabaya. Total samples in this research are 50 women with infertility, it is divided into 25 people with endometriosis and 25 people without endometriosis. In the medical records, we can see the duration of infertility in each sample. Results: The duration of infertility >3 years occurs in women with and without endometriosis (72% and 80%). In bivariate analysis using the chi-square test, there was no difference that showed increasing duration of infertility (P = 0.508) between women with endometriosis and without endometriosis. Conclusion: There was no differences in the duration of female infertility with endometriosis and without endometriosis

Abstract

Abstract Background : Low pregnancy planning can cause unwanted pregnancy, abortion, loss of reproductive health, domestic violence until death. 40% of 85 million pregnancies in the world are unplanned and 38% end with abortions, miscarriage and unplanned childbirth. Unplanned pregnancy are associated with an increased risk of birth outcomes, including preterm birth, low birth weight (LBW), high risk of congenital anomalies, and developmental delay. Most woman who have plan to get pregnant don’t prepare their pregnancy as well. Women who have children (Multiparous) having unreadiness of pregnancy as same as women who do not have children (Nulliparous). Therefore, pregnancy planning is required to avoid risks so that women become more prepared on facing the pregnancy. This study aimed to analyze the differences behaviour toward readiness of pregnancy among pregnancy women based on parity. Method : This study used observational analytical research with cross sectional approach. The sampling technique used was total sampling which was conducted for 2 months and the sample obtained 92 pregnant women (include nulliparous, primiparous and multiparous) who met the inclusion criteria. The collected data was analyzed by Fisher Exact levels with a degree of α = 0.05. Result : The study showed there were 12 respondents who ready to face pregnancy. Meanwhile, there were 80 respondents who were not ready enough to face pregnancy, it was based on the result of Fisher Exact test with the value obtained was P value = 0.223 (P > 0.223). Conclusion : This showed that there was no difference of attitudes about pregnancy readiness of nulliparous, primiparous and multiparous woman

Abstract

ABSTRACTBackground : K1 antenatal care visits at gestational age less than 13 weeks has a very important meaning for pregnant women as well as maternal and child health programs because of their contribution to the prevention of maternal deaths. Coverage of K1 in Puskesmas Watukapu  still has a gap of 28.05%.This study aims to determine the factors that influence the K1 antenatal care visit. Methode : The research method was analytic observational with cross sectional design. The sample in this study were all pregnant women trimesters 2 and 3 who came to visit the POLI KIA Puskesmas Watukapu.The sample size of 70 respondents was taken by non-probability sampling technique with consecutive sampling.The independent variable was K1 antenatal care visit, while the dependent variable were age, knowledge, education, parity, attitude, husband's support and family support.Research instrument with questionnaire. Bivariate data analysis used chi square test and multivariate test used  multiple logistic regression  test.  Results:this study found that the proportion of K1 antenatal care visits was 62.4%. Bivariate statistical test results knowledge P = 0,000 (p <0,005), attitude P = 0,135 (P> 0,005), husband support P = 0,000 (P <0,005), family support P = 0,004 (P <0,005), age P = 0,331 (P> 0.005), education P = 0.004 (P <0.005), parity P = 0.015 (P <0.05).  Multivariate test results with  P value <0.005 was  parity with a P value of 0.017.Conclusion: There is a correlation between knowledge, education, parity, husband's support and family support to the awareness of K1 antenatal care visits with the most dominant factor is parity.
