Abstract

Highlights: 1. The use of re-heated cooking oil can give negative impact on health,  especially on the cardiovascular system. 2. Unlike other types of cooking oil, repeated heating of canola oil did not show significant change in aortic wall thickness. Abstract Background: Structural alteration of blood vessels such as formation of atheroma or changes in the thickness of vessel walls, are heavily involved in pathogenesis of cardiovascular disease. Several studies have shown that canola oil has a positive effect on such diseases by reducing LDL and cholesterol levels. However, there may be several negative impacts on reheating canola oil upon administration, similar to other oils. Although canola oil can improve lipid profiles, studies related to how canola oil alters the structure of blood vessels are limited. Objective: The aim of this study was to investigate the effect of heated canola oil on intimal-to-medial thickness (IMT) of Thoracic aorta in high-fat diet rats. Material and Method: A total of 27 rats were divided into 3 groups, the K+, P1, and P2. Rats among the three groups were given a high-fat diet for 14 days, accompanied by consumption of canola oil without heating in the P1, and with repeated heating in the P2. Thoracic aorta was taken on the 15th day and then processed into histological preparations. IMT was measured using CellSens software on a microscope with a magnification of 400. The difference between groups was tested using the one-way ANOVA test on SPSS. Result: The mean and standard deviations of each group in a row were K+ (134.96 and 21.27) P1(132.04 and 27.30) and P2 (152.05 and 31.75). There was no significant difference in IMT between groups (p >0.05). However, the P2 group showed the highest mean of IMT. Conclusion: The consumption of canola oil with or without repeated heating did not result in the changes of the IMT in rats fed with a high-fat diet.

Abstract

Background : Male infertility affects millions globally, with 12-15% of Indonesian couples facing conception challenges, 50% of which are attributed to male factors. Despite extensive research, 70% of male infertility cases remain idiopathic. Diabetes, a prominent factor in male infertility, affects 9.3% of the global population, with projections estimating 700 million cases by 2045. Recent studies suggest the Mediterranean diet positively impacts semen parameters, particularly in diabetic individuals. This review aims to explore the Mediterranean diet's effects on fertility in male diabetic patients. Reviews : The literature review employed three databases, focusing on in-vitro, in-vivo, pre-clinical, and clinical studies from 2014-2024. Diabetes, a chronic disease characterized by abnormal blood sugar levels, affects insulin production or effectiveness, leading to complications if left uncontrolled. Erectile dysfunction (ED), prevalent in diabetic men, is attributed to nerve and blood vessel damage from high blood sugar levels. Treatments for ED include PDE5 inhibitors, but alternative plant-based therapies are being explored. Platelet-rich plasma (PRP) has shown promise in animal studies but lacks human trial data. The Mediterranean diet, rich in plant-based foods and healthy fats, may help prevent sexual function deterioration in diabetic individuals. Comprehensive nutrition education and government efforts are crucial in promoting healthy lifestyles and ensuring access to fresh foods. Summary : In conclusion, the Mediterranean diet shows promise as a dietary intervention for improving male fertility in diabetic populations. Further research is warranted to confirm these findings and to explore the mechanisms underlying the effects of the Mediterranean diet on male fertility.

Abstract

Background: Colorectal cancer is the second leading cause of death in the world. Currently, resection and adjuvant chemotherapy are the main therapies for colorectal cancer. Resection is an invasive procedure and chemotherapy often causes side effects due to non-specific work targets. Purpose: This study examines the potential of soursop leaf extract and curcumin with magnetic and mucus-penetrating nanoparticles as an alternative therapy for colorectal cancer. Review: Soursop leaf extract has acetogenin agent as an anti-tumor, anti-inflammatory, and cytotoxic agent that acts specifically on target organs. Curcumin from turmeric extract has antioxidant, anti-inflammatory, antimutagenic, antiangiogenic, and anticancer effects. Curcumin works molecularly on cyclooxygenase-2 (COX-2) to prevent inflammation, thereby inhibit the growth of cancer cells and reducing the risk of metastasis. Curcumin also plays a role in the inhibition of nuclear factor κ-light-chain-enhancer of activated B (NF-κB) cells, thus inhibits carcinogenesis. For drug delivery, magnetic nanoparticles and mucus-penetrating nanoparticles could be used. The magnetic field of magnetic nanoparticles generates force and torque to increase the temperature and movement of the organelles, so that apoptosis occurs from cancer cells. Mucus-penetrating nanoparticles are more resistant to mucus degradation because they can avoid mucoadhesive effects, penetrate the mucus adherent layer, and are easily absorbed by the intestinal epithelium. Conclusion: Soursop leaves, curcumin, magnetic nanoparticles, and mucus-penetrating nanoparticles are potential to be an alternative therapy for colorectal cancer.

Abstract

Pancreatic cell damage in diabetes mellitus is closely linked to inflammation and apoptosis. This study aimed to investigate the protective effects of phloroglucinol on pancreatic cells in a streptozotocin-induced diabetic model by assessing its anti-inflammatory and anti-apoptotic mechanisms. Phloroglucinol ligand and the structures of Bax, Bcl-2, and caspase-3 proteins were sourced from the PubChem database. Molecular docking was performed using Autodock Tools and docking results were analyzed with PyRx software. In addition, during the in vivo study, the BALB/c mice were grouped into four categories: healthy control, untreated streptozotocin-induced diabetic, and streptozotocin-induced diabetic treated with two doses of oral phloroglucinol at 100 mg/kg and 200 mg/kg body weight. After 28 days, pancreatic tissues were collected for flow cytometric analysis of NF-κB, IL-6, TNF-α, and apoptotic markers (Bax, Bcl-2, and caspase-3). The docking simulations revealed specific binding interactions: phloroglucinol interacted with Bcl-2 via amino acid residues of ALA90 and TYR139, with Bax via ALA42, LEU45, ALA46, LEU47, PRO130, and ILE133, and with caspase-3 through ARG64, SER120, GLN161, CYS163, and ARG207. The binding affinities for Bax, Bcl-2, and caspase-3 were -5.0, -4.7, and -4.9 kcal/mol, respectively. In vivo, results showed that streptozotocin significantly elevated inflammatory cytokines NF-κB, TNF-α, and IL-6, along with apoptotic markers in pancreatic cells (p<0.05) compared to healthy controls. Phloroglucinol administration at 200 mg/kg significantly reduced TNF-α, NF-κB and IL-6 levels. Phloroglucinol also prevented streptozotocin-induced pancreatic cell damage through anti-apoptotic effects by downregulating Bax and caspase-3 and upregulating Bcl-2. These findings suggest that phloroglucinol may offer protective benefits in diabetic conditions by modulating apoptotic and inflammatory pathways.
