Abstract

Objective: This study aims to investigate the effect of alprazolam administration on sperm viability of mice (Mus musculus) strain Balb/C following CUMSfor 53 days. Method: This study used a true experimental design through randomization. Control group kept undisturbed without treatment and therapy throughout 60 days. K1 and K2 were exposed to a 53 day period of CUMS along with 7-day pretreatment adaptation. K2 was adapted for a week, stressed for 53 days along with cotreatment of alprazolam (4 mg/kg BW). Results: K1 and K2 showed insignificant improvement of sperm viability (p > 0.05).Meanwhile, K0 and K1 showed significant value compared to K2 (p<0.05). K2 showed little increase of sperm viability in CUMS group along with subsequent treatment of alprazolam. Conclusion: Overall, the results indicate an insignificant effect of alprazolam on sperm viability of CUMS-induced mice.

Abstract

Chronic pain is a common case and become a serious problem. Chronic pain management are inconclusive.One of the common treatment option is NSAID. Treatment Failed are around 34-79% of total case. Thesetreatment failed suspected as NSAID resistance. Treatment failed can be caused by a some molecule thatmake NSAID loss of efficacy. Cdk5 is one of the molecule that active in chronic pain condition. Cdk5 canincrease transmembrane insertion and activate TRPV1. The aim of this research is analyzed the role of cdk5and TRPV1 in NSAID resistance of chronic pain rat. This research used 42 rats as a subject and dividedinto 6 groups with random allocation method and factorial design. Meloxicam treatment was given orallyevery day for 7 days after rats have a chronic pain (28 days). Chronic pain induction used a CFA injection.Results : cdk5 and TRPV1 expression at the dorsal root ganglia of chronic pain groups are increase, nosignificant different of pain threshold and inflammation sign between treatment and no treatment groupsafter chronic pain occurred. Conclusion : chronic pain can induce cdk5 and TRPV1 expression, and inducedby meloxicam treatment. Cdk5 and TRPV1 have a positive correlation with meloxicam resistance.

Abstract

Background: Hyperglycemia is a common effect of uncontrolled DM. The main impact of DM on male infertility is the result of hyperglycemia which causes an increasing in free radicals which cause oxidative stress in the testes, and then attacks Sertoli cells. Increasing of free radicals will cause disruption and damage to this cell, including apoptosis and atrophy and then caused decreasing in the number of Sertoli cells. Kebar grass contains antioxidants compounds, such as flavonoids and vitamin E. There are synergy effects between vitamin E and flavonoids which reinforce both in working to improve infertility as an antioxidant. Objective: To compared the number of Sertoli cells between groups which receiving extract kebar grass (Biophytum petersianum Klotzsch) and the control group in diabetes mellitus mice (Mus musculus) model. Methods: The research subjects used DM male mice which were divided into 5 groups: 2 control groups (K- and K+) and 3 treatment groups (P1, P2, P3). Day 1 to day 5, all groups were induced by STZ. The 11th day to the 45th day, the control group (K+) is given metformin 2 mg/head /day, the P1, P2 and P3 groups is given kebar grass with dose of 67,5; 135 and 270 mg/kg/day and metformin 2 mg/head/day. The 11th day to the 31th day, the control group (K-) is given CMC-Na 1% suspension. The 46th day is done surgery and sampling testicular tissue. Examination of the number of Sertoli cells is done through examination of the histological picture of testicular tissue that has been given Haematoxylin-Eosin (HE) staining. Abstract

Background: In addition to the influence of diabetes on male reproduction lowers testosterone levels of FSH and LH, the second number of hormones is reduced will interfere with the process of spermatogenesis. If the spermatogenesis process is interrupted then the number of germ cells decreases, this will cause a decrease in the weight of the testes. Objective: To analyze the effect of extract grass administration to the weight of the testicles diabetes Mellitus mice between the treatment group and the control. Methods: The research used DM male mice which is divided into 5 groups each consisting of 6 mice. Day 1 till day 5 all STZ-induced groups. On the 11th day to the 45th day, the control group (K +) was given metformin 2mg/head/day. The control group (K-) was given CMC-Na 1%. Group P1, P2 and P3 was given Kebar grass with dose of 67, 5mg/day, 135mg/kg/day, and 270mg/kg/day and metformin 2mg/head/day. The 46th was done with the release and sampling of the right and left testicles. Testicular weighing used analytic scales 0.001gram. Results: Statistical tests showed that there was a discrepancy in the p-value = 0.000 (0.05) on testicular weight. Conclusion: Increased testicular weight model of diabetes mellitus that gets the grass extractions of Kebar. Proses spermatogenesis terganggu maka jumlah sel germinal mengalami penurunan, hal ini akan menyababkan berat testis menurun. Tujuan: menganalisisi pengaruh pemberian ekstrak rumput kebar terhadap berat testis mencit model diabetes mellitus antara kelompok perlakuan dan kontrol. Metode: penelitian menggunakan mencit DM yang dibagi menjadi lima kelompok setiap kelompok terdiri dari 6 ekor mencit. Hari ke 1 sampai hari ke 5 semua kelompok diinduksi STZ. Hari ke 11 sampai hari ke 45 kelompok kontrol (K+) disonde metformin 2mg/ekor/hari. Kelompok kontrol (K-) disonde CMC Na 1%. Kelompok P1,P2 dan P3 disonde ekstrak rumput kebar dengan dosis, 67,5mg/kgBB/hari, 135mg/kgBB/hari, 270mg/kgBB/hari dan metformin 2mg/ekor/hari. Abstract

Infertility is a 10-15% problem experienced by men around the world. One of the causes of infertility in men is caused by an increase in reactive oxygen species (ROS) by toxic substances such as heavy metal lead (Pb). This research was conducted to find the potential of Dutch eggplant fruit (Solanum betaceum) in various doses to increase the concentration of mice spermatozoa (Mus muscullus) exposed to lead acetate. Dutch eggplant has a good phenolic content as an antioxidant to prevent free radicals. The study design used was Posttest only control group design with a sample of 40 male mice. The treatment time in all groups is 35 days. Mice were divided into 5 groups that were given oral treatment. The control group was given 75 mg/kgBW of lead acetate treatment. The treatment group was given lead acetate 75mg / KgBB and Dutch eggplant extract at various doses of 100 mg/kgBW (P1), 200 mg/kgBW (P2), and 400 mg/kgBW (P3). The results showed that Dutch eggplant extract (Solanum betaceum) with various doses could significantly increase spermatozoa concentration. The optimal dose of Dutch eggplant extract (Solanum betaceum) in mice exposed to lead is 100 mg/kg/day in the treatment group (P1). Penelitian ini dilakukan untuk menemukan potensi buah terong belanda (Solanum betaceum) dalam berbagai dosis terhadap peningkatan konsentrasi spermatozoa mencit (Mus musculus) yang dipapar timbal asetat. Terong belanda mempunyai kandungan fenolik yang baik sebagai antioksidan untuk mencegah radikal bebas. Desain penelitian yang digunakan Posttest only control group design dengan sampel 40 mencit jantan. Waktu perlakuan pada semua kelompok yaitu 35 hari. Mencit dibagi menjadi 5 kelompok yang diberikan perlakuan secara oral. Kelompok kontrol diberikan perlakuan timbal asetat 75mg/KgBB. Kelompok perlakuan diberikan timbal asetat 75mg/KgBB dan ekstrak terong belanda dengan berbagai dosis 100mg/KgBB (P1), 200mg/KgBB (P2), dan 400mg/KgBB (P3). Hasil penelitian menunjukkan bahwa ekstrak terong belanda (Solanum betaceum) dengan berbagai dosis mampu meningkatkan konsentrasi spermatozoa secara signifikan. Dosis optimal pemberian ekstrak terong belanda (Solanum betaceum) pada mencit yang dipapar timbal adalah 100 mg/kgBB/hari pada kelompok perlakuan (P1). Abstract

Lead exposure as environmental pollution can cause male reproductive problems. Provision of natural antioxidants can be an alternative solution to protect male reproductive function. This research was conducted to investigate the potential of Solanum betaceum extract as a protective agent for Leydig cells in mice exposed to lead acetate. The entire research process with a replication unit of 40 male mice. Mice were divided into five groups with each treatment given orally. Abstract

One of the environmental factors that cause male infertility is heavy metal pollution such as lead. Lead can interfere with male fertility. Flavanoids are antioxidants that can prevent the negative effects caused by lead. The flavonoids contained in the Solanum betaceum extract have the potential to prevent the adverse effects of lead on the reproductive tract of mice because it can prevent oxidative stress. So it can affect the thickness of the seminiferous tubule epithelium and the diameter of the seminiferous tubules of mice exposed by lead acetat. The research method was a pure experimental laboratory (true experimental) randomized posttest only control group design approach. The total sample of 40 heads was divided into 5 groups. The study used 5 treatments. The concentration of Solanum betaceum for research group 100 mg/kg BW/day, 200 mg/kg BW/day, 400 mg/kg BW/day. Data analysis was performed analytically using One Way Anova. The results showed the mean ± standard deviation of the highest seminiferous tubular epithelium was highest 76.63 ± 7.17 and the mean ± standard deviation of the highest seminiferous tubule diameter was highest at 219.98 ± 17.06 in the P3 group. Abstract

Male infertility with unknown definite etiology can be associated with lead exposure. Lead that accumulates in the body can cause oxidative stress and spermatogenesis disruption. To minimize the impact of lead exposure, it is necessary to use conventional methods with natural materials. One of them is the use of Solanum betaceum which have been proven to have secondary metabolic components that are antioxidants. This study aims to prove the effect of Dutch Eggplant (Solanum betaceum) extract to testicular weight of mice (Mus musculus) exposed to lead. This research was an true experimental research using male Mus musculus. Mice were 40 and divided into five groups randomly. Abstract

Background: Administration of antioxidants has been proven to reduce the production of free radicals in the body. One of the natural ingredients that contain antioxidants are Kebar grass. Kebar grass (Biophytum petersianum Klotzch) contains three types of chemical compounds that could potentially impact spermatogenesis. Those are flavonoids, saponins and tannins so it needs to investigate the effect on the testicles with a model of diabetes mellitus (DM). Purpose: Analyzed the Effectivness of Biophytum petersianum Klotzch extract to the epithelium thickness of the seminiferous tubules of mice (Mus musculus) model of diabetes mellitus. Methods: This research was a post test only control group design using Balb / C 45 male tail number, divided 5 groups were randomly selected. K-: DM mice group was given CMC Na 1% for 21 days. K +: DM mice group was given CMC Na 1% + metformin 2 mg / gBW for 35 days. (P1, P2, P3): DM group of mice given doses of 67.5 kebar seaweed extract, 135, 270 mg / kg + metformin 2 mg / gBW 35 days. Valid to 45 surgery and sampling and examination of thickness of seminiferous tubule epithelium. Results: The observation of thick epithelium of the seminiferous tubules was significant difference (p = 0.005) with a mean negative control group (K) 65 235 ± 5012, positive control(K +) 71 216 ± 12 514, (P1) 76 756 ± 6144, (P2) 80 660 ± 4140, (P3) 81 418 ± 7319. Conclusion Extract grass kebar (Biophytum petersianum Klotzch) efectivness in improving a thick epithelium of the seminiferous tubules in male mice model of diabetes mellitus. Salah satu bahan alami yang mengandung antioksidan adalah rumput kebar. Rumput kebar (Biophytum petersianum Klotzch) mengandung tiga jenis senyawa kimia yang berpotensi memberikan pengaruh terhadap proses spermatogenesis, yaitu flavonoid, saponin dan tannin sehingga perlu dijadikan penelitian untuk melihat potensi pada testis dengan model diabetes melitus. Tujuan : Menganalisis efektivitas pemberian ekstrak rumput kebar (Biophytum petersianum Klotzch) terhadap tebal epitel tubulus seminiferus pada mencit (Mus musculus) jantan model diabetes melitus. Metode : Penelitian ini merupakan penelitian eksperimental dengan post test only control group design menggunakan mencit Balb/C jantan sejumlah 45 ekor, dibagi 5 kelompok dipilih secara acak. K- : kelompok mencit DM diberikan CMC Na 1% selama 21 hari. K+ : kelompok mencit DM diberikan CMC Na 1% + metformin 2 mg/gBB selama 35 hari. (P1, P2, P3) : kelompok mencit DM diberikan ekstrak rumput kebar dosis 67,5, 135, 270 mg/kgBB + metformin 2 mg/gBB 35 hari. Hari ke 36 dilakukan pembedahan dan pengambilan sampel dan melakukan pemeriksaan tebal epitel tubulus seminiferus. Hasil : Hasil pengamatan tebal epitel tubulus seminiferus didapatkan ada perbedaan yang signifikan (p=0,005) dengan rerata kelompok kontrol negatif (K-) 65.235 ± 5.012, kontrol positif (K+) 71.216 ± 12.514, ( P1) 76.756 ± 6.144, (P2) 80.660 ± 4.140, (P3) 81.418 ± 7.319. Kesimpulan : Ekstrak rumput kebar (Biophytum petersianum Klotzch) efektif dalam meningkatkan tebal epitel tubulus seminiferus pada mencit jantan model diabetes melitus .
