Abstract

Background Cervical squamous cell carcinoma (CSCC) represents 70-80% of all cervical cancer cases. This cancer ranks 4th most common death cause due to malignancy in women. Cervical SCC patients were found at an older age and were detected at an advanced stage. High risk human papillomavirus (HPV) infection is main cause of SCC. Protein cyclin-dependent kinase inhibitor 2A also called p16 is a well-known tumor suppressor protein involved in cell cycle regulation, senescence, apoptosis, cell invasion and angiogenesis. This p16 protein can be used as surrogate marker for HPV infection. From several previous studies, there were differences between p16 expression relationship with differentiation degree and cervical SCC tumor stage results. Relationship research between p16 expression and differentiation degree and tumor stage of cervical SCC patients has never been conducted in West Sumatra. Methods This research was retrospective observational cross-sectional study with 35 cervical SCC cases that have been diagnosed at Anatomical Pathology Laboratory, RSUP Dr. M. Djamil Padang period January 2018-December 2019. Samples were obtained by simple random sampling from paraffin blocks derived from tumor tissue. The histopathological slide was reassessed to assess differentiation degree and tumor stage data were obtained from the patient's medical record. p16 expression in tumor cells was seen by immunohistochemical examination. To determine the correlation, bivariate statistical analysis was performed using the chi-square test with significance p<0.05 level. Results p16 high expression was found in 22 cases, tended to be higher in degree of well differentiation (40.9%) and moderate differentiation (50.0%) and was higher at an advanced stage (90.9%). Statistically, there is no correlation between p16 expression and differentiation degree with p value=0.138. There was no correlation between p16 expression and tumor stage with p value=0.337. Conclusions This study concluded that p16 expression was not associated with differentiation degree and tumor stage in cervical SCC.

Abstract

BackgroundEndometrial carcinoma is the fourth most common malignancy among women worldwide with increasing incidence and death rate every year. One of the types of endometrial carcinoma is endometrioid carcinoma, originated from atypical hyperplasia and develop into carcinoma. Lack of intercellular cohesiveness in the epithelial tumors such as endometrioid carcinoma occur due to lack of expression of E-cadherin. It can also causetumor invasion and metastatic through the epithelial-mesenchymal transition (EMT) process. Tumor differentiation and tumor budding are presumed to be histopathologic representations due to lack of cohesiveness and the occurrence of the EMT process so that these two things need to be related with the expression of E-cadherin on tumor cells.MethodsAn observational study was conducted using a cross-sectional approach with 46 cases of endometrioid endometrial carcinoma.Samples were obtained from 4 Anatomical Pathology Laboratories in West Sumatra during 2016-2019 in the form of paraffin blocks and HE slides that obtained from hysterectomy surgery. Reevaluation of tumor grade and tumor budding in HE slides was performed. E-cadherin expression in tumor cells was seen by immunohistochemical staining. Bivariate statistical analysis was performed using Fisher's Exact test and the results were considered significant if the p value was <0.05.ResultsSpecimens with negative E-cadherin expression were more common in grade 2 tumors (41.7%) and grade 3 tumors (50.0%) and in specimes with positive tumor budding (91.7%). Statistical analysis revealed a significant correlation between E-cadherin expression with tumor grade (p=0.000) and tumor budding (p=0.000).ConclusionExpression of E-cadherin has a significant correlation with the tumor grade and tumor budding in endometrioid endometrial carcinoma.

Abstract

Highlights: Understanding the relationship between TIL infiltration and tumor stage could refine prognostic models and guide targeted therapies. Tumor-infiltrating lymphocytes density is considered an essential immune marker in ovarian cancer, but this study did not demonstrate a significant association with FIGO staging.   Abstract Introduction: Ovarian cancer is the fifth most common cancer among females. Prognostic factors such as stage and tumor-infiltrating lymphocytes (TILs) may influence outcomes. This study aimed to analyze the relationship between stromal TIL density and 2014 International Federation of Gynecology and Obstetrics (FIGO) staging in epithelial ovarian cancer. Methods: This cross-sectional analytical study included 44 cases (n=44) of epithelial ovarian cancer diagnosed at the Laboratory of Anatomical Pathology, Dr. M. Djamil General Hospital, Padang, Indonesia, in 2021–2022, selected using consecutive sampling. Stromal TIL density was evaluated on hematoxylin and eosin (HE) slides according to the International TILs Working Group guidelines and categorized as low (≤10%) or high (>10%). 2014 FIGO staging was obtained from medical records. Statistical analysis was performed using the International Business Machines Corporation (IBM) Statistical Package for the Social Sciences (SPSS) version 26, with Chi-square or Fisher’s exact test, and p<0.05 was considered significant. Results: Early-stage disease (stage I-II) comprised 25 cases (56.8%), of which 12 cases (48%) had low TIL density, and 13 cases (52%) had high TIL density. Advanced-stage disease (stage III-IV) comprised 19 cases (43.2%), of which 13 (68.4%) showed low TIL density, and 6 (31.6%) showed high TIL density. Chi-square analysis showed no significant association between TIL density and FIGO stage (p=0.295). Conclusion: No significant relationship was found between stromal TIL density and 2014 FIGO staging in epithelial ovarian cancer. Further research with larger cohorts and detailed TIL subtypes may clarify their prognostic relevance.

Abstract

Background: Hepatocellular carcinoma (HCC) is the sixth most common cancer globally and shows significant regional variations in clinicopathological characteristics. Limited data exist on HCC patterns in Indonesia, particularly in West Sumatra. This study aimed to characterize the clinicopathological features of HCC at Dr. M. Djamil Hospital, Padang, from 2021to2024.Methods: A descriptive retrospective study was conducted using medical records and histopathological re-evaluation of 46 HCC cases. Specimens were examined for demographic data, tumor characteristics, histopathological subtypes according to the WHO 2019 classification, differentiation grade, lymphovascular invasion, and background liver pathology. Results: The median age was 56 years, with male predominance (60.9%). Most specimens were resection specimens (60.9%) with tumor size ≥4 cm in 47.8% of cases and single nodules in 52.1%. Clear cell (39.1%) and macrotrabecular massive (37.0%) were the predominant subtypes, contrasting with global data showing NOS predominance. Most tumors were moderately differentiated (63.0%), with lymphovascular invasion present in 65.2% and cirrhotic background in 58.7% of casesConclusion: HCC in West Sumatra demonstrates distinct histopathological patterns, with a predominance of clear cell and macrotrabecular massive subtypes and high rates of lymphovascular invasion. These finding suggest histopathological features associated with aggressive tumor behavior (particularly macrotrabecular massive subtype and lymphovascular invasion), although no clinical outcome or survival analysis was performed.

Abstract

Bitter melon (Momordica charantia) has been empirically proven for antimicrobial properties that can be used as an alternative medicine, while the changes that occur in Escherichia coli-infected mouse intestine after administering bitter melon have not been widely studied. The aim of this study was to determine the effect of Momordica charantia administration on histopathology of intestine of E. coli infected mice. This research was post-test control randomized group study involving 20 mice’s small intestine paraffin blocks which were divided into 4 groups consisted of control and different dosage of bitter melon juice, observed subsequently under a microscope to measure the of the small intestine lining and assess inflammation using modified Barthel Mandja scoring. This study found significant changes the height of mucosa and villi of mice small intestine after administration of 100 grams of bitter melon juice and the muscle layer and crypts after administration of 200 grams of bitter melon juice. The Barthel Manja score showed decrease of inflammation degree among P2 group, while increased inflammation degree found in P3 group. It can be concluded that administration of bitter melon juice has the potential to improve the intestinal histopathology of mice infected with Escherichia coli.
