Abstract

Penelitian ini bertujuan untuk mempelajari karakteristik semen dan mengevaluasi hubungan antara konsentrasi testosteron dengan libido pejantan sapi Simental. Sebanyak 12 ekor sapi Simental, 9 ekor dengan motilitas spermatozoa ≥70% (A) dan 3 ekor dengan motilitas spermatozoa <70% (B) berdasarkan data sekunder dari Balai Inseminasi Buatan (BIB) tahun 2018-2019. Semen ditampung menggunakan vagina buatan pada pagi hari. Evaluasi kualitas semen segar hanya dilakukan dengan penilaian morfologi spermatozoa. Sampel darah disentrifugasi (3000 rpm, 10 menit), supernatan dimasukkan ke dalam microtube dan disimpan pada suhu -20 °C. Analisis testosteron menggunakan metode ELISA. Data kualitas semen segar sapi pejantan Simental dianalisis secara deskriptif. Korelasi antara karakteristik semen, libido dan konsentrasi hormon testosteron menggunakan korelasi Pearson’s. Hasil analisis menunjukkan terdapat perbedaan signifikan (P<0,05) antara kualitas semen segar kelompok pejantan A dengan pejantan B dalam parameter motilitas, konsentrasi, dan morfologi spermatozoa normal. Volume semen dan skor libido pada semua pejantan tidak menunjukkan perbedaan signifikan. Konsentrasi testosteron pada kelompok pejantan A lebih tinggi (42,57 ng/mL) dibandingkan dengan kelompok pejantan B (33,26 ng/mL). Konsentrasi testosteron menunjukkan korelasi positif (P<0,01) dengan karakteristik semen seperti motilitas spermatozoa (0,813), morfologi spermatozoa normal (0,639), dan libido (0,952). Hasil penelitian ini dapat digunakan sebagai pertimbangan untuk dilakukannya pengukuran konsentrasi testosteron dalam proses seleksi sapi jantan menjadi pejantan.

Abstract

Artificial insemination (AI) has been widely applied to improve genetic quality in cattle worldwide. One of the most important factors in AI program is the quality of semen.  Many procedures in semen processing have been developed to preserve the sperm quality. The Singosari AI center produces the frozen semen through cryopreservation method. During this process, the number of semen collected will be rejected if the quality is low. Fresh semen with less than 70% of sperm motility, before freezing semen with less than 55%of sperm motility and less than 40% of sperm post-thawing motility evaluation will be rejected, so that it was only the viable sperm will be processed to be frozen semen commercial. Therefore, comparative study was carried out to analyze the freezing capability of sperm.

Abstract

The objective of this study was to evaluate effect of genetic and non-genetic factors on semen characteristics including ejaculate volume, sperm concentration, total sperm number and sperm moetility of Bali cattle. Semen data were collected from the National Centre of Artificial Insemination at Singosari, Malang, East Java, Indonesia. A total of 3,847 ejaculates of 17 Bali bulls from 2014 to 2016 were collected and evaluated. Data were analyzed by restricted maximum likelihood (REML) method using mixed models which the bull was a random effect, while age of bull, season of collection, frequency of ejaculation and collection intervals were the fixed effects. Results showed that age significantly affected all semen characteristics (P<0.01). Season affected only on sperm motility (P<0.01). Effect of frequency of ejaculation and collection intervals was significant on all studied variables (P<0.01), except sperm motility. Repeatability of ejaculate volume, sperm concentration, total sperm number and sperm motility was 0.43; 0.35; 0.32 and 0.31, respectively. It is concluded that age, frequency of ejaculation and collection intervals were the most factors affected semen characteristics of Bali cattle. Repeatability estimations of semen characteristics were moderate to high.

Abstract

The objective of the research was to analyze the feeding effect of bean sprout waste and omega-3 to the spermatozoa production  of garut rams. Twelve garut rams were divided into three groups of feeding; grass + concentrate (R1), bean sprouts waste + concentrate (R2)  and bean sprouts waste + concentrate + omega-3 (R3). The semen were collected by using artificial vagina once a week. Semen evaluated macroscopically              (volume, color, consistency and pH) and microscopically (mass movement, motility, viability, concentration, concentration per ejaculate and morphology of spermatozoa).The research was designed by using a completely randomized design (repeated measurement) and statistically analyzed using SAS 9.1 software. The results showed macroscopy no differences  was found on microscopyc evaluation except for semen volume. Microscopyc evaluation showed that motility, viability and spermatozoa concentration as well as total number per ejaculete feed with R2dan R3 were highger (P<0.05) than R1. However supplementation of omega-3 was no better than feed containing bean sprouts waste.

Abstract

The success of artificial insemination is very much determined by the quality of spermatozoa. The detection or identification of damaged chromatin of spermatozoa DNA is very important to forsee the adverse clinical outcome. However, the method of identification is still depended on expensive imported kits. Therefore, the objective of this research was to developed an identification kit to determine the quality of livestock spermatozoa DNA chromatin.This study consist of three step. Step 1) Determination of low melting point agarose (LMP-agarose) concentration which is 0,6%, 0,7% and 0,8%. 2) Comparison of three lysis solution (LS) which is LS I (0.4M Tris, 0.8M DTT, 1% SDS, pH 7.5), LS II (0.4M Tris,2 M NaCl, 1% SDS , pH 7.5), and LS III (0.4M Tris-HCl, 2M NaCl, 1% SDS 0,05 M EDTA, pH 7.5). 3) Comparison different staining which is Eosin yellow and Methylene blue. The results showed that 0.6% LMP-agarose demonstrated the best concentration to “trapped the spermatozoa” compared for sheep and goats. whereas the three concentration of spermatozoa cows can not be used to trap spermatozoa cow. The best formulation to lysis the membrane was LS III (0.4M Tris -HCl, 2M NaCl, 1% SDS 0,05 M EDTA). The best staining was eosin yellow and methylene blue with 2:1 ratio.

Abstract

The aim of this study was to evaluate the effect of different IgY concentrations on the quality of semen (macroscopic and microscopic) and testicular morphometry (weight, length and width of the testes) of Sentul cock testis and its ability to fertilize hens. This study used 4 months of 20 Sentul cocks consisting 10 high IgY concentration cocks and low IgY concentration cocks. This study used a complete randomized design (RAL) with 2 treatments namely low and high IgY concentration and10 times repetitions. Data were analyzed using t-test minitab program versions 16. Based on the result, different IgY concentrations did not affect the quality of semen and testicular morphometrics, but high IgY concentration cocks were able to fertilize more hens than the low IgY concentrations one by using dilution methods. This proves that high IgY concentration cocks are more effective and efficient cocks or stud.

Abstract

One of the factorthat influencethe process offertilization on Echymiperakalubu is the quality of the spermwhich produced by males. The ability of spermtofertilize anovumis not only owned by the spermatozoa which derived from the results of ejaculation, butis also owned by spermatozoa from the caudaepididymis.This study aimed to determine sperm concentration and motility from caudaepididymis.Twenty-one males E. kalubuwere used which was 370-1500g. Based onthis weight, it was divided into three groupings, they were prepubertal (370-640 g), young adult (>640-850g) and adult (>850g).The results showed that the sperm concentration of the prepubertal was1.75±1.14x106/ml,the young adult was 8.86±4.21x106/ml and increased to12.56±6.33x106/mlin the adult group.Sperm motility also increased with increasing maturity. The prepuberta lgroup showed that sperm motility was33.33±25.17%, increased to41.67±12.58% (young adult) and3.54±57.50% followed by the adult.

Abstract

This study investigated the effect of glycerol and dimethylacetamide (DMA) in beltsville thawing solution (BTS) extender. Semen samples were collected from three boars of the Duroc breed (n=5) and were evaluated microscopically and macroscopically. Semen that have been evaluated and meets the following criteria: has motility characteristics >70%, concentration >300 million/ml and abnormality 20%; was accepted as appropriate samples. Semen samples were divided into four tubes and were diluted with 5 ml of BTS. The diluted semen was left for two hours in the temperature range from 20 to 22°C before was centrifuged for 15 minutes (500G). The pellet of centrifuged semen was taken as much as 1 ml with its supernatant. The BTS glycerol (BTSG), BTS-DMA (BTSD), BTS-Glycerol-DMA (BTSG-D) were added into the pellet. The diluted semen was packed into straw of 0.5 ml and was equilibrated for 2 hours in 4 °C. The equilibrated semen was frost with the steam of nitrogen and stored in liquid nitrogen. The quality of frozen semen was assessed at least 24 hour after freezing process. The results indicate that the sperm motility after thawing in BTS extender BTSD is 52.14±0.4%, ,higher than the other two extenders (P<0.05). Thus, it is conclude that the DMA concentration in BTS extender maintains the quality of frozen semen from the Duroc breed better than other extenders.

Abstract

Sperm cryopreservation is the process of preserving sperm cells at low temperatures, so that its frozen semen can be used in the future. The quality of the frozen sperm is affected by the diluent. The objective of this study was to compare the effects of commercial diluents on acrosome status, malondialdehyde (MDA) and aspartate aminotransferase (AspAT) enzyme concentration of thawed Limousin and Simmental bull semen. Semen was collected twice weekly using an artificial vagina. The fresh semen processed into frozen semen had  sperm motility of 70%. The one-step procedure was used for the dilution methods. Andromed®, Optixcell® and Steridyl® were used as diluents. Data were analyzed by analysis of variance (ANOVA) followed by Tukey HSD 5% confidence interval. The result showed no interaction (P0.05) between two factors on acrosome status. The sperm acrosome damage of Simmental in Steridyl® was significantly lower than others (P0.05), although all diluents showed low sperm acrosome damage. Also, no interaction between the type of diluent and breed on MDA and AspAT enzyme concentrations was detected (P0.05). The results suggest that three commercial diluents have equal efficacy in protecting acrosome status and maintaining MDA and AspAT enzyme concentrations of frozen Limousin and Simmental bull semen. Therefore, all commercial diluents can be an alternative for Limousin and Simmental frozen semen.

Abstract

Indonesia has two National Artificial Insemination Centers (AIC) and more than 15 Regional Artificial Insemination Centers (RAIC) spread across several provinces. Bulls in the AIC must have a high libido and produce good quality semen. This study examines the correlation between libido with semen quality and testosterone concentration to determine potential frozen semen production from Bali bulls in South Sulawesi RAIC. Ten Bali bull were used in this study. Semen collection was carried out twice a week with semen evaluation following the RAIC protocol. At the same time, blood samples and libido measurements were carried out from each bull. The frozen semen production potential was calculated by multiplying the semen volume, motility, and sperm concentration. The results showed that the quality of fresh semen and testosterone concentrations did not differ between high and low libido of Bali bulls. Libido has a positive correlation with semen volume (r= 0.52) and sperm motility (r= 0.62), while testosterone concentration has a negative correlation with semen volume (r= -0.65), sperm motility (r= -0.60), and libido (r= -0.48). Bulls with high and low libido have good frozen semen production potential, ranging from 19,755 – 21,640 straws per year. Bali bulls in RAIC have fresh semen quality and testosterone concentrations under normal conditions, with high potential for frozen semen production, although only 60% of Bali cattle have high libido and 40% have low libido.
