Abstract

On the freezing semen chicken cryoprotectants required to overcome the damage of spermatozoa due to cold shock. This study aims to get the best concentrations of cryoprotectants glycerol concentration of 5%, 7% and 9% in freezing Sentul chicken semen. The semen used in this study came from three chickens Sentul and be repeated nine times. Semen was collected by  messase methods for three times a week. Semen was evaluated macroscopic and microscopic. Furthermore spermatozoa diluted with egg yolk and the addition of three concentrations of cryoprotectants glycerol (5%, 7% and 9%). Semen diluted 0:25 ml is packed into straw. Then equilibrated at a temperature of 5°C for two hours. After equilibration to evaluate the motility and viability of spermatozoa. Furthermore, frozen in liquid nitrogen vapor for 10 minutes. Frozen semen is then stored in liquid nitrogen containers with temperature -196°C. After 24 hours, semen is thawed at 37°C for 30 seconds. The results showed that the percentage of sperm motility and viability of frozen semen cock Sentul using glycerol cryoprotectants 5% better P (0.05) compared with the use of glycerol 7% and 9%. The use of glycerol 5% at this stage of equilibration and storage can reduce the damage of spermatozoa in the semen of chicken Sentul. Neither glycerol 5% could increase recovery rate after thawing

Abstract

Chicken Kampung Broiler (KB) is the result of a cross between a local chicken (chicken) with broiler imported chicken (broiler chicken). Testing the quality of frozen chicken semen from crossing needs to be done to get high productivity. This study aims to obtain a concentration of cryoprotectant Dimethyl Acetamide (DMA) the best of the concentration of 5%, 7% and 9% in chicken semen freezing KB. The semen used in this study came from three chickens of KB and repeated as many as nine times. Semen is collected by the method of message three times a week. Semen is evaluated macroscopically and microscopically. Subsequently spermatozoa were diluted with yolk diluent and the addition of three DMA cryoprotectant concentrations (5%, 7%, and 9%). Semen diluted 0:25 ml is packed into straw. Then equilibrated at a temperature of 5 ° C for two hours.  After equilibration evaluation of motility and viability of spermatozoa. It is then frozen on liquid nitrogen vapor for 10 minutes. Frozen semen is then stored in liquid nitrogen containers at a temperature of -196 ° C. Abstract

Quails are classified according to their immunity to different IgY concentrations. Quail IgY concentrations range from 0 to 1200 µg ml-1, and these IgY concentrations may affect production and reproductive performance. This study aimed to analyze IgY concentrations in male quail and to compare production and reproductive performance at different IgY concentrations. Forty-two male quail at five weeks of age participated in the study, including 29 quail with low IgY concentration (210-393 µg ml-1)   and, 13 with intermediate IgY concentration (403-564 µg ml-1). The observed productive performances were feed intake, initial body weight, final body weight, weight gain, feed conversion, morbidity, and mortality. In addition, this study observed testicular weight and macroscopic and microscopic semen quality for reproductive traits. Differences in production and reproductive performance of each group were analyzed using an independent-sample t-test. The result showed that male quail with different IgY concentrations were equal in all productive traits (P0.05). This means that male quail with low IgY and medium IgY concentrations are equally good. Testicular weight, semen color and pH were identical at different IgY concentrations. Quails with low IgY concentrations have better sperm consistency, which is related to the weight of the testes, which are heavier in quails with low IgY concentrations than in those with moderate IgY concentrations. This consistency is related to the concentration of sperm in the semen. The result concludes IgY concentrations in male quail did not affect production performance. Quails with low IgY concentration had thicker sperm consistency and higher sperm concentration.

Abstract

Kebar grass (Biophytum petersianum Klotzsch.) has long been known by the locals of West Papua as medicinal plant that improves fertility. The objectives of this research were to identify and determine the antioxidant activity of kebar grass crude extract through conventional method and to analyze for its antioxidant activity. Extraction method used was conventional method, performed by mixing dry kebar grass with water and boiled until the volume shrunk to 1/3 of initial volume. Analized parameters were crude extract yield, qualitative phytochemical, total phenol and antioxidant activity. Research result showed extract crude yield value of 18.32±0.01%. Qualitative phytochemical contents were dominated by flavonoid, tannin, and saponin. Total phenolic content (TPC) of crude extract was 147±1.24 mgGAE/gdw (0.688±0.01 mgGAE/gfw). Crude extract antioxidant parameter was measured as the value of inhibition, AEAC, TEAC, and IC50 where each value respectively was as followed 42.0±0.047%, 69.93±2.19 mgAE/gdw (8.30±0.26 mgAE/gfw), 65.70±1.54 mgTE/gdw (7.80±0.18 mgTE/gfw) and 0.129±0.003 mgdw (257.75±4.90 mg/L). The contribution of kebar grass TPC content to antioxidant activity is 47.38% (equal to ascorbic acid) and 44.51% (equal to trolox) which means that almost half of kebar grass crude extract’s phenolic compound act as antioxidant. The results of this research can show that the kebar grass has potential as a source of multiple antioxidants, which acts as an analogue of vitamin C and vitamin E.  The antioxidant activity of the kebar grass extract can be the scientific basis for the use of kebar grass as a medicinal plant for infertility problems (curative) and as a functional food ingredient for antioxidant sources that prevent a decrease in reproductive performance (preventive).

Abstract

Ayam KUB adalah ayam hasil seleksi dari Balai Penelitian Ternak Ciawi yang memiliki perbedaan fenotip seperti bentuk jengger dan warna bulu. Perbedaan fenotip berpengaruh terhadap konsentrasi testosteron sehingga memengaruhi kualitas semen segar ayam KUB. Penelitian ini bertujuan untuk mengkaji pengaruh fenotip terhadap kualitas semen segar dan testosteron serta korelasinya. Sebanyak 20 ekor ayam KUB umur 1,5 tahun terbagi atas 4 kelompok perlakuan fenotip masing-masing 5 ekor yaitu jengger tunggal bulu penutup merah (JTBM), jenggger tunggal bulu penutup putih (JTBP), jengger pea bulu penutup merah (JPBM), dan jengger pea bulu penutup putih (JPBP). Peubah yang diamati adalah volume, warna, pH dan konsistensi, gerakkan massa, motilitas, viabilitas, abnormalitas dan konsentrasi ejakulat, konsentrasi testosteron dan panjang taji. Hasil penelitian menunjukkan pejantan dengan fenotip JPBM memiliki volume ejakulat paling tinggi dan JTBM adalah yang paling rendah (P<0.05). Pejantan berjengger tunggal memiliki pH lebih tinggi dibandingkan pejantan berjengger pea. Pejantan JTBM juga memiliki motilitas dan konsentrasi ejakulat paling rendah dibandingkan dengan fenotipe lainnya dan untuk peubah warna semen, gerakan massa, viabilitas dan abnormalitas tidak berbeda pada semua fenotip. Panjang taji kiri pejantan JTBP lebih pendek dibandingkan ketiga pejantan lainnya (P<0.05). Ukuran taji berkorelasi positif dengan konsentrasi testosteron dengan kisaran nilai r = 0.33-0.56. Konsentrasi testosteron berkorelasi postif dengan warna (r=0.76), konsentrasi ejakulat (r= 0.44), konsistensi semen (r=0.75) dan motilitas spermatozoa (r= 0.46). Kesimpulan dari penelitian ini adalah pejantan JTBM memiliki kualitas semen segar kurang baik dibandingkan dengan ketiga kelompok pejantan lainnya. Ukuran taji bisa digunakan untuk memprediksi konsentrasi testosteron pada ayam jantan.

Abstract

Peningkatan populasi dan mutu ternak, khususnya sapi dan kerbau d ilakukan melalui program inseminasi buatan (IB). Angka kebuntingan hasil IB di Indonesia, relatif masih rendah, salah satunya diduga disebabkan oleh fertilitas pejantan. Program IB di Indonesia menggunakan semen beku yang diproduksi oleh Balai Inseminasi Buatan (BIB). Standar evaluasi semen bagi pejantan yang digunakan sesuai SNI masih mengacu pada konsentrasi, motilitas pra dan pasca thawing serta skor individu. Perkembangan terbaru menunjukkan bahwa parameter uji tersebut tidak lagi memadai. Protein dan gen pada spermatozoa dan seminal plasma pejantan dapat menjadi kandidat alat bantu “akurat” untuk menentukan fertilitas pejantan sesuai keberhasilan IB di lapangan. Policy brief ini diharapkan dapat menjadi bahan pertimbangan bagi Pemerintah dan BIB dalam p engembangan dan pengaplikasian penanda fertilitas berbasis molekuler sebagai alat bantu “akurat” dalam proses seleksi dan kebijakan pengafkiran pejantan sapi yang akan digunakan untuk IB. Dengan demikian, efisiensi penggunaan pejantan unggul di berbagai Ba lai Inseminasi Buatan, baik Nasional maupun Daerah di Indonesia dapat dioptimalkan.

Abstract

ABSTRACT. Maggot Hermetia illucens (Maggot Black Soldier Fly, MBSF) is an alternative protein source besides soybean meal (SBM) which may be used as a feed for improving the quality of semen particularly in Garut rams to support prolific nature. The aims of this study were to analyzed and compare the impact of different protein sources in feed on semen quality of Garut rams, and to assess the prediction ability of Garut rams to serve ewe in small-scale breeders in West Java, Indonesia. This study was conducted using a completely randomized design with 3 treatments and 4 replications, consisted of Brachiaria humidicola (BH) grass and T1 (concentrate contains 20% of SBM), T2 (concentrate contains 10% of SBM and 10% of MBSF), and T3 (concentrate contains 20% of MBSF). The parameters measured were feed consumption, semen quality (macroscopic and microscopic characteristics), also a potential ability of rams to serve ewe. The results showed there were no significant effect on protein consumption, semen volume, semen pH, semen color and consistency, sperm mass movement, sperm motility, sperm concentration, sperm morphology, and prediction potential ability to serve ewe. However, the result showed a significant effect (P0.05) on sperm viability and sperm plasma membrane integrity. Sperm plasma membrane integrity of ram feed with T3 was better than T1 and T2 (P0.05). The prediction potential ability rams to serve ewes on MBSF treatment was 38 heads, while in T1 and T2 were 43 and 57 heads, respectively. In conclusion, MBSF can be an alternative source of protein besides SBM to improve the semen quality of Garut rams.ABSTRAK. Maggot Hermetia illucens (Maggot Black Soldier Fly; MBSF) adalah sumber protein alternatif selain bungkil kedelai (SBM) yang dapat dipergunakan sebagai pakan untuk memperbaiki kualitas semen terutama pada domba Garut untuk mendukung sifat prolifik. Tujuan penelitian ini adalah menganalisis dan membandingkan dampak pemberian sumber protein berbeda terhadap kualitas semen domba Garut dan untuk menilai kemampuan domba Garut pejantan dalam melayani betina pada peternakan rakyat di Jawa Barat, Indonesia. Penelitian ini menggunakan Rancangan Acak Lengkap (RAL) dengan 3 perlakuan dan 4 ulangan yang terdiri dari rumput Brachiaria humidicola (BH) dan T1 (konsentrat mengandung 20% SBM), T2 (konsentrat mengandung 10% SBM dan 10% MBSF), dan T3 (konsentrat mengandung 20% MBSF). Parameter yang diukur adalah konsumsi pakan, karakteristik semen (makroskopis dan mikroskopis) serta potensi domba jantan melayani betina. Hasil Penelitian menunjukkan tidak ada perbedaan signifikan pada konsumsi protein pakan, volume semen, pH semen, warna dan konsistensi semen, gerakan massa sperma, motilitas sperma, konsentrasi sperma, morfologi sperma, dan prediksi potensi pejantan dalam melayani betina. Namun, hasil penelitian menunjukkan terdapat perbedaan (P0.05) pada viabilitas sperma dan membran plasma utuh sperma. Membran plasma utuh pada perlakuan T3 lebih baik dibandingkan perlakuan T1 dan T2 (P0.05). Prediksi potensi betina terlayani dari pejantan yang diberi pakan MBSF adalah 38 ekor, sedangkan yang diberi SBM dan kombinasinya adalah 43 dan 57 ekor. Kesimpulan penelitian ini adalah MBSF dapat menjadi alternatif sumber protein selain bungkil kedelai dalam memperbaiki kualitas sperma domba Garut.

Abstract

Indo-Pacific bottlenose dolphin (Tursiops aduncus) is a marine mammal that lives in relatively small populations. The geographic ranges of this species are susceptible to the effects of human activities, thereby necessitating conservation efforts to prevent extinction. Therefore, this study aimed to evaluate the daily quality of dolphin sperm after several days of refrigeration. The sperm of two male dolphins were stored at 4oC for 4 days, and the quality was observed daily to determine the motility, viability, membrane integrity, and sperm abnormalities. Sperm samples were divided into four groups, consisting of two centrifuged followed by the removal of seminal plasma, and two groups without centrifugation, containing 100x106 and 200x106 sperm/ml each. After liquid storage, the motility of sperm was 63-75% with no significant reduction in the first 3 days. Sperm viability following storage was 65-75% and the percentage with abnormal morphology ranged from 2-6%. Furthermore, there was no significant increase in abnormal morphology of sperm on any day of storage for 3 days. Sperm membrane integrity was 36-49%, with no significant reduction in the membrane integrity in the first 2 days. There was no significant difference in sperm quality, although centrifugation and removal of seminal plasm had a slight effect. The results of this study showed that Indo-Pacific bottlenose dolphin sperm could be stored for a short period as liquid storage while maintaining a quality that allows for future use.

Abstract

Standar kualitas semen yang berperan penting dalam keberhasilan inseminasi buatan (IB) sebagai upaya peningkatan populasi dan mutu ternak sapi di Indonesia terbukti tidak seutuhnya mampu menentukan tingkat fertilitas sapi pejantan di Indonesia. Studi terbaru membuktikan bahwa meskipun sudah memenuhi standar kualitas semen yang sudah ditetapkan, namun tiga dari enam ekor sapi pejantan di Indonesia masih tergolong fertilitas rendah (<60%). Potamine-1 (PRM1) merupakan molekul pada inti spermatozoa yang telah terbukti memiliki hubungan erat dengan fungsi fertilitas dan kualitas spermatozoa pada berbagai sapi pejantan di Indonesia. Hal tersebut mendasari usulan pengembangan uji berbasis molekuler PRM1 sebagai penentu tingkat fertilitas yang diharapkan dapat digunakan untuk proses seleksi dan kebijakan pengafkiran sapi pejantan unggul di Indonesia. Melalui policy brief ini diharapkan usulan pengembangan uji tersebut dapat lebih tersampaikan dengan segala solusi yang diberikan, termasuk terkait pendekatan uji berbasis molekuler yang paling tepat digunakan, kajian-kajian lanjutan, dan berbagai harapan dukungan dari segala pihak demi terwujudnya pengembangan uji PRM1.

Abstract

Indonesian local sheep represent a particular species of sheep reared by farmers on small-scale farms. Semen freezing can be performed using either a homemade or commercial extender. The present study evaluated the effectiveness of several commercial extenders in freezing Indonesian local sheep. Semen was collected from three mature and healthy rams using an artificial vagina and evaluated. It was determined that only samples exhibiting sperm motility levels of greater than 70% would be included in the study. Subsequently, the semen from each ram was divided into three portions and diluted with Andromed, Sterydil, and OptiXcell, respectively. The frozen semen was then evaluated 24 hours post-freezing. The thawing process was conducted individually at 37°C for 30 seconds. The evaluation encompassed a range of metrics, including sperm motility, viability, and the presence of any abnormalities, in addition to the recovery rate. The results indicated that sperm frozen in Optixcell and Sterydil extenders exhibited superior motility, viability, and recovery rate than Andromed. The study also demonstrated no interaction between individual Ram and extender, and no differences were found between individuals. The study concluded that Optixcell and Sterydil extenders effectively froze Indonesian local sheep semen.
