Abstract

The aim of this study was to examine the preservation of sentul crossbreed chicken semen in ringer lactate egg yolk diluent supplemented with various monosaccharide. Semen was collected from three roosters using massage method. Immediately after collection, the semen was evaluated macroscopically and microscopically. Semen with more than 70% motility was divided into four tubes. Each of them diluted with ringer lactate egg yolk glucose (RLEYG), ringer lactate egg yolk fructose (RLEYF), ringer lactate egg yolk xylose (RLEYX) and ringer lactate egg yolk mannose (RLEYM). Semen was stored in refrigerator (5o C) for sixty hours and evaluated every twelve hours for spermatozoa motility and viability. Results showed that no significant difference (P0.05) among diluents used on spermatozoa quality parameters after dilution and during preservation. Semen quality decrease during storage and at sixty hours of storage, the motility and viability of spermatozoa ranging from 48.332.56 to 55.422.26% and 58.592.87 to 64.832.42%, respectively. This research conclude that glucose, fructose, xylose and mannose can be used as energy source for roosters semen during preservation.

Abstract

Indonesian local sheep represent a particular species of sheep reared by farmers on small-scale farms. Semen freezing can be performed using either a homemade or commercial extender. The present study evaluated the effectiveness of several commercial extenders in freezing Indonesian local sheep. Semen was collected from three mature and healthy rams using an artificial vagina and evaluated. It was determined that only samples exhibiting sperm motility levels of greater than 70% would be included in the study. Subsequently, the semen from each ram was divided into three portions and diluted with Andromed, Sterydil, and OptiXcell, respectively. The frozen semen was then evaluated 24 hours post-freezing. The thawing process was conducted individually at 37°C for 30 seconds. The evaluation encompassed a range of metrics, including sperm motility, viability, and the presence of any abnormalities, in addition to the recovery rate. The results indicated that sperm frozen in Optixcell and Sterydil extenders exhibited superior motility, viability, and recovery rate than Andromed. The study also demonstrated no interaction between individual Ram and extender, and no differences were found between individuals. The study concluded that Optixcell and Sterydil extenders effectively froze Indonesian local sheep semen.

Abstract

One factor that affects frozen semen characteristics is bull age. Increasing age induces changes in sperm that reduce frozen semen quality. This study aimed to evaluate the frozen semen characteristics of Bali bulls aged 3, 8, and 13 years. Frozen semen was derived from fresh semen with sperm motility > 70%. The frozen semen characteristics tested included motility, viability, plasma membrane integrity, acrosome integrity, and sperm abnormalities. Sperm motility was analyzed by computer-assisted sperm analysis (CASA). Viability was assessed using eosin-nigrosin staining; plasma membrane integrity was assessed using the hypoosmotic swelling test (HOST); acrosome integrity was assessed using Giemsa staining; and sperm abnormalities were assessed using Williams staining. The plasma membrane integrity of the 13-year-old group was significantly lower (P < 0.05) than that of the younger age groups. However, there were no significant differences (P > 0.05) in the other parameters. We conclude that age affects the integrity of the sperm plasma membrane in frozen semen from Bali bulls.

Abstract

Penelitian ini mengevaluasi efektivitas sinkronisasi estrus menggunakan prostaglandin F2α (PGF2α) pada sapi Bali dan menilai hasil kehamilan setelah perlakuan awal dan resinkronisasi selanjutnya. Sepuluh sapi Bali menerima dua suntikan PGF2α, diberikan dengan jarak 11 hari, sesuai rekomendasi standar. Hewan yang menunjukkan estrus diinseminasi buatan, dan diagnosis kehamilan dilakukan 30–45 hari setelah inseminasi menggunakan ultrasonografi. Sapi yang gagal hamil selama siklus pertama menjalani protokol resinkronisasi dua minggu kemudian. Protokol sinkronisasi menghasilkan respons estrus 100%, dengan tingkat kehamilan awal 40% (4 sapi). Resinkronisasi enam sapi yang tidak hamil menghasilkan dua kehamilan tambahan (33,3%), sehingga menghasilkan tingkat kehamilan keseluruhan 60%. Temuan ini menunjukkan bahwa PGF2α efektif dalam mengatur siklus estrus dan meningkatkan efisiensi reproduksi sapi Bali, terutama bila diintegrasikan dengan strategi resinkronisasi.

Conventional assessments subject to subjectivity, human error, and high variability. Computer-assisted semen analysis (CASA) is considered more objective. This study aimed was to conduct a comparative analysis of motility and concentration assessment of frozen semen sperm using conventional and the portable CASA AndroScope. Twenty-one laboratory assistants from 21 national and regional frozen semen producers participated in this study. Sperm motility and concentration were assessed conventionally (using a Neubauer chamber) and AndroScope was used with five replicates. The results of the comparison of sperm motility scores revealed significant differences (p<0.05) between conventional examination and using AndroScope. The results of the comparison of the sperm concentration calculations revealed no significant difference (p>0.05) between the calculations performed using the Neubauer chamber and AndroScope. Assessment with the AndroScope is considered reliable and can replace conventional assessment in assessing motility and sperm concentration, thereby improving and standardizing the assessment performed by frozen semen producers.

Abstract

Sheep farming plays a crucial role in Indonesia's livestock industry; however, the effectiveness of artificial insemination (AI) is hindered by the less-than-ideal quality of cryopreserved semen. This research assessed the longevity and progressive motility recovery rate (RR) of frozen semen from local Indonesian rams, using a homemade Tris egg yolk (TEY) extender and three commercial extenders (AndroMed®, Steridyl®, BoviFree®). Semen from three rams (Batur, Garut, and Sakub) was processed, frozen in 0.25 mL straws (50 × 10⁶ sperm per straw), and its quality was evaluated using CASA. Longevity was monitored hourly from 0 to 4 h at 37°C. The data were analyzed using a Randomized Block Design with ANOVA, followed by Duncan’s Multiple Range Test. Sperm motility decreased over incubation time for all extenders, with notable differences at 4 h (total motility, p = 0.017; progressive motility, p = 0.023). Commercial extenders preserved higher total and progressive motility than TEY, with Steridyl® showing the highest mean RR (64.11%; mean all rams; RR analysis, n = 18), significantly outperforming Andromed®, BoviFree®, and TEY (p = 0.007). The superior results of Steridyl® align with its low-density lipoprotein content, which stabilizes the sperm membranes during cryopreservation. In summary, Steridyl® was the most effective extender for maintaining motility and RR of frozen semen from local Indonesian rams, potentially enhancing AI success in small ruminant programs.
