Abstract

This study aims to examine the quality of frozen semen in Tris egg yolk (TEY) extender and skimmed milk extender with or witout omega-3. A total of 18 Simmental bulls belong to Lembang Artificial Insemination Centre were divided into three groups. Each was fed with standard feed (R1), standard feeds supplemented with minimal Se and Zn (R2) and standard feed supplemented with maximal of Se and Zn concentration. Semen were collected using an artificial vagina and were evaluated macro- and microscopically. The semen then were divided into four tubes and each diluted with Skimmed, SkimmedOmega 3, TEY or TEY-O. The semen was then packed into a 0.25 ml straw and equilibrated at 5 oC for 4 hours, then frozen above liquid nitrogen vapor, and stored in liquid nitrogen container (-196 oC). The qualities of frozen semen were evaluated on the motility, individual score, viability and integrity of the plasma membrane of sperms. Sperm motility of bulls fed with standard feed (R1) in TEY extender and R3 in TEY and TEY-Omega-3 extender were higher (p <0.05) compared to the other combinations. No difference was found on the individual score. The viability of sperms in bulls fed with standard feed in SkimmedOmega-3 extender was higher than the other treatments and the highest sperm plasma membrane integrity was demonstrated by sperm in bull feeding with R2 in TEY extender.

Abstract

This study was conducted to determineSDS concentration also to compare Tris egg yolk and citrateegg yolk on the quality of ettawa grade chilled semen. The study consist of two experiments. The firstexperiment was to determine the best SDS concentration in Tris egg yolk diluents and the second experimentwas to compare the SDS suplementation in tris and citrate egg yolk in the quality of ettawa grade chilledsemen. The semen were collected from three bucks, immediately after collection the semen evalutedmacro and microscopycally. Semen demonstrate more than 70% motility and less than 10% spermatozoaabnormality, individually divided into four tube, each tube was diluted in egg yolk supplemented with 0;0,025; 0,05; and 0.075% SDS then stored at 5°C for 72 hours. The quality of chilled semen was observefor motility and viability every 12 hours. Result demonstrated that 0.05% SDS was the best concentrationcompared to others. In the second experiment,the semen were diluted in four different diluents, it wereTris egg yolk (TEY), Tris egg yolk with 0.05% SDS (TEYS), citrate egg yolk (CEY) and citrate egg yolk with0.05% SDS (CEYS). Result demonstrated that the mean value of motility and viability of spermatozoa inTEYSdiluents (67.08 ± 6.43% and77.07 ± 6.78%) was higher (p<0.05) than CEYS (60.42 ± 9.05% and72.31± 7.45%), TEY (59.23 ± 9.41% and71.21 ± 8.56%) and CEY (53.45 ± 11.33% and67.74 ± 8.90%). In conclusion,supplementation of 0.05% SDS in TEY was maintained best sperm quality compared to other diluentsduring preservation.

Abstract

One of the factorthat influencethe process offertilization on Echymiperakalubu is the quality of the spermwhich produced by males. The ability of spermtofertilize anovumis not only owned by the spermatozoa which derived from the results of ejaculation, butis also owned by spermatozoa from the caudaepididymis.This study aimed to determine sperm concentration and motility from caudaepididymis.Twenty-one males E. kalubuwere used which was 370-1500g. Based onthis weight, it was divided into three groupings, they were prepubertal (370-640 g), young adult (>640-850g) and adult (>850g).The results showed that the sperm concentration of the prepubertal was1.75±1.14x106/ml,the young adult was 8.86±4.21x106/ml and increased to12.56±6.33x106/mlin the adult group.Sperm motility also increased with increasing maturity. The prepuberta lgroup showed that sperm motility was33.33±25.17%, increased to41.67±12.58% (young adult) and3.54±57.50% followed by the adult.

Abstract

This study investigated the effect of glycerol and dimethylacetamide (DMA) in beltsville thawing solution (BTS) extender. Semen samples were collected from three boars of the Duroc breed (n=5) and were evaluated microscopically and macroscopically. Semen that have been evaluated and meets the following criteria: has motility characteristics >70%, concentration >300 million/ml and abnormality 20%; was accepted as appropriate samples. Semen samples were divided into four tubes and were diluted with 5 ml of BTS. The diluted semen was left for two hours in the temperature range from 20 to 22°C before was centrifuged for 15 minutes (500G). The pellet of centrifuged semen was taken as much as 1 ml with its supernatant. The BTS glycerol (BTSG), BTS-DMA (BTSD), BTS-Glycerol-DMA (BTSG-D) were added into the pellet. The diluted semen was packed into straw of 0.5 ml and was equilibrated for 2 hours in 4 °C. The equilibrated semen was frost with the steam of nitrogen and stored in liquid nitrogen. The quality of frozen semen was assessed at least 24 hour after freezing process. The results indicate that the sperm motility after thawing in BTS extender BTSD is 52.14±0.4%, ,higher than the other two extenders (P<0.05). Thus, it is conclude that the DMA concentration in BTS extender maintains the quality of frozen semen from the Duroc breed better than other extenders.

Abstract

Sperm cryopreservation is the process of preserving sperm cells at low temperatures, so that its frozen semen can be used in the future. The quality of the frozen sperm is affected by the diluent. The objective of this study was to compare the effects of commercial diluents on acrosome status, malondialdehyde (MDA) and aspartate aminotransferase (AspAT) enzyme concentration of thawed Limousin and Simmental bull semen. Semen was collected twice weekly using an artificial vagina. The fresh semen processed into frozen semen had  sperm motility of 70%. The one-step procedure was used for the dilution methods. Andromed®, Optixcell® and Steridyl® were used as diluents. Data were analyzed by analysis of variance (ANOVA) followed by Tukey HSD 5% confidence interval. The result showed no interaction (P0.05) between two factors on acrosome status. The sperm acrosome damage of Simmental in Steridyl® was significantly lower than others (P0.05), although all diluents showed low sperm acrosome damage. Also, no interaction between the type of diluent and breed on MDA and AspAT enzyme concentrations was detected (P0.05). The results suggest that three commercial diluents have equal efficacy in protecting acrosome status and maintaining MDA and AspAT enzyme concentrations of frozen Limousin and Simmental bull semen. Therefore, all commercial diluents can be an alternative for Limousin and Simmental frozen semen.

Abstract

Indonesia has two National Artificial Insemination Centers (AIC) and more than 15 Regional Artificial Insemination Centers (RAIC) spread across several provinces. Bulls in the AIC must have a high libido and produce good quality semen. This study examines the correlation between libido with semen quality and testosterone concentration to determine potential frozen semen production from Bali bulls in South Sulawesi RAIC. Ten Bali bull were used in this study. Semen collection was carried out twice a week with semen evaluation following the RAIC protocol. At the same time, blood samples and libido measurements were carried out from each bull. The frozen semen production potential was calculated by multiplying the semen volume, motility, and sperm concentration. The results showed that the quality of fresh semen and testosterone concentrations did not differ between high and low libido of Bali bulls. Libido has a positive correlation with semen volume (r= 0.52) and sperm motility (r= 0.62), while testosterone concentration has a negative correlation with semen volume (r= -0.65), sperm motility (r= -0.60), and libido (r= -0.48). Bulls with high and low libido have good frozen semen production potential, ranging from 19,755 – 21,640 straws per year. Bali bulls in RAIC have fresh semen quality and testosterone concentrations under normal conditions, with high potential for frozen semen production, although only 60% of Bali cattle have high libido and 40% have low libido.

Abstract

The superior Balitbangtan Kampung Chicken (KUB) chickens have different phenotypes. It was reported that the chicken phenotype was related to semen quality. This study aimed to determine the post-thawing characteristics and quality of KUB chicken semen with different phenotypes frozen in Ringer's lactate egg yolk (RLEY) diluent with the addition of fructose or glucose. Semen was collected using the massaging method from 20 KUB chickens with a single comb phenotype and black or dark brown feather color with a red feather neck (SCNR), green-black single comb with white feather neck collar (SCNW), pea comb and black feathers or dark brown fur with a red neck (PCNR), and a green-black pea comb with a white neck (PCNW). Semen from each chicken phenotype was divided into three parts or frozen in three types of diluents: RLEY, RLEY+fructose (RLEYF), and RLEY+glucose (RLEYG). The highest sperm motility was found in the diluent with the addition of glucose in the SCNR and PCNW phenotypes (P0.05). The highest sperm viability was shown in the RLEYG diluent in the PCNW phenotype (P0.05). The highest abnormality was found in the RLEY and RLEYF diluents in the SCNW, PCNR, and PCNW groups, whereas in the RLEYG group, it was only found in the PCNR group. From the results of this study, it can be concluded that the type of glucose and chicken phenotype influences the quality of post-thawing semen. The best is found in diluents with glucose attachments in the SCNR phenotype.

Abstract

This research was aimed to study the effect of administration of glycerol and dimetilacetamida (DMA) in BTS® and MIII®extender supplemented with sodium dodecyl sulphate (SDS) on boar frozen semen. A number of four boars were used in this study for semen collection (n=20). The collected semen was evaluated both macroscopically and microscopically. In this study, only the semen that demonstrated70% sperm motility, 200.106/mL sperm concentration, and20% sperm abnormalities were used and divided into eight tubes. A number of 4 tubes were diluted with 5 mL of BTS, and the rest with 5 mL MIII. The sampel was stored at 20-22° C for 2 hours, followed by centrifugation for 15 minutes (at 2000 rpm), and taken of pellet with 1 ml supernatant. The pellet that was resulted from centrifugation using BTS, then re-diluted with BTS-glycerol 5% (BTSG), BTS DMA 5% (BTSD), BTS-glycerol 5% and SDS (BTSG-S), BTS-DMA 5% and SDS (BTSD-S). Four other pellets that were centrifuged with MIII also re-diluted with MIII-glycerol 5% (MIIIG), MIII-DMA 5% (MIIID), MIII-glycerol 5% and SDS (MIIIG-S), MIII-DMA 5% and SDS (MIIID-S). Next, all of diluted semen were inserted into 0.5 mL straw and equilibrated for 2 hours (4° C), then frozen and stored in liquid nitrogen. The evaluation of frozen semen quality was conducted at 24 hours after frozen. The result of this study showed that post-thawing motility of spermatozoa in BTSD-S (40.17±0.2%) was found higher (P0.05) compared to seven other dilution processes. Therefore, it is concluded that the concentration of 5% DMA that supplemented with SDS in BTS dilution much better for maintaining boars frozen semen quality.

Abstract

The aim of this study was to examine the preservation of sentul crossbreed chicken semen in ringer lactate egg yolk diluent supplemented with various monosaccharide. Semen was collected from three roosters using massage method. Immediately after collection, the semen was evaluated macroscopically and microscopically. Semen with more than 70% motility was divided into four tubes. Each of them diluted with ringer lactate egg yolk glucose (RLEYG), ringer lactate egg yolk fructose (RLEYF), ringer lactate egg yolk xylose (RLEYX) and ringer lactate egg yolk mannose (RLEYM). Semen was stored in refrigerator (5o C) for sixty hours and evaluated every twelve hours for spermatozoa motility and viability. Results showed that no significant difference (P0.05) among diluents used on spermatozoa quality parameters after dilution and during preservation. Semen quality decrease during storage and at sixty hours of storage, the motility and viability of spermatozoa ranging from 48.33±2.56 to 55.42±2.26% and 58.59±2.87 to 64.83±2.42%, respectively. This research conclude that glucose, fructose, xylose and mannose can be used as energy source for roosters semen during preservation.

Abstract

The aims of this study were to investigate the freezing capability of Pasundan bull spermatozoa in Tris-egg yolk (TEY), Tris-soy (TS), and AndroMed® as diluents. Semen were collected twice a week from four Pasundan bulls aged 3-5 years old using an artificial vagina and evaluated macro- and microscopically. Semen had ≥70% sperm motility, ≥800x106/mL sperm concentration, and less than 20% sperm abnormalities were divided into three parts and each of them diluted with TEY, TS, or AndroMed®. After an equilibration step at 5°C for four hours, diluted semen were packaged in 0.25 mL straw, frozen in liquid nitrogen for ten minutes and kept in liquid nitrogen container until examination. Motility test on fresh, diluted, equilibrated, and after-thawed semen was done using Androvision®. The results showed that after thawing motility of sperm diluted in AndroMed® (58.64±0.72%) was higher than in TEY (49.45±1.22%) and TS (39.34±6.33%). Sperm motility of Pasundan bulls diluted in these three diluents reduced around 33.27±2.45% during freezing process.
