Abstract

ABSTRACT. Maggot Hermetia illucens (Maggot Black Soldier Fly, MBSF) is an alternative protein source besides soybean meal (SBM) which may be used as a feed for improving the quality of semen particularly in Garut rams to support prolific nature. The aims of this study were to analyzed and compare the impact of different protein sources in feed on semen quality of Garut rams, and to assess the prediction ability of Garut rams to serve ewe in small-scale breeders in West Java, Indonesia. This study was conducted using a completely randomized design with 3 treatments and 4 replications, consisted of Brachiaria humidicola (BH) grass and T1 (concentrate contains 20% of SBM), T2 (concentrate contains 10% of SBM and 10% of MBSF), and T3 (concentrate contains 20% of MBSF). The parameters measured were feed consumption, semen quality (macroscopic and microscopic characteristics), also a potential ability of rams to serve ewe. The results showed there were no significant effect on protein consumption, semen volume, semen pH, semen color and consistency, sperm mass movement, sperm motility, sperm concentration, sperm morphology, and prediction potential ability to serve ewe. However, the result showed a significant effect (P0.05) on sperm viability and sperm plasma membrane integrity. Sperm plasma membrane integrity of ram feed with T3 was better than T1 and T2 (P0.05). The prediction potential ability rams to serve ewes on MBSF treatment was 38 heads, while in T1 and T2 were 43 and 57 heads, respectively. In conclusion, MBSF can be an alternative source of protein besides SBM to improve the semen quality of Garut rams.ABSTRAK. Maggot Hermetia illucens (Maggot Black Soldier Fly; MBSF) adalah sumber protein alternatif selain bungkil kedelai (SBM) yang dapat dipergunakan sebagai pakan untuk memperbaiki kualitas semen terutama pada domba Garut untuk mendukung sifat prolifik. Tujuan penelitian ini adalah menganalisis dan membandingkan dampak pemberian sumber protein berbeda terhadap kualitas semen domba Garut dan untuk menilai kemampuan domba Garut pejantan dalam melayani betina pada peternakan rakyat di Jawa Barat, Indonesia. Penelitian ini menggunakan Rancangan Acak Lengkap (RAL) dengan 3 perlakuan dan 4 ulangan yang terdiri dari rumput Brachiaria humidicola (BH) dan T1 (konsentrat mengandung 20% SBM), T2 (konsentrat mengandung 10% SBM dan 10% MBSF), dan T3 (konsentrat mengandung 20% MBSF). Parameter yang diukur adalah konsumsi pakan, karakteristik semen (makroskopis dan mikroskopis) serta potensi domba jantan melayani betina. Hasil Penelitian menunjukkan tidak ada perbedaan signifikan pada konsumsi protein pakan, volume semen, pH semen, warna dan konsistensi semen, gerakan massa sperma, motilitas sperma, konsentrasi sperma, morfologi sperma, dan prediksi potensi pejantan dalam melayani betina. Namun, hasil penelitian menunjukkan terdapat perbedaan (P0.05) pada viabilitas sperma dan membran plasma utuh sperma. Membran plasma utuh pada perlakuan T3 lebih baik dibandingkan perlakuan T1 dan T2 (P0.05). Prediksi potensi betina terlayani dari pejantan yang diberi pakan MBSF adalah 38 ekor, sedangkan yang diberi SBM dan kombinasinya adalah 43 dan 57 ekor. Kesimpulan penelitian ini adalah MBSF dapat menjadi alternatif sumber protein selain bungkil kedelai dalam memperbaiki kualitas sperma domba Garut.

Abstract

Indo-Pacific bottlenose dolphin (Tursiops aduncus) is a marine mammal that lives in relatively small populations. The geographic ranges of this species are susceptible to the effects of human activities, thereby necessitating conservation efforts to prevent extinction. Therefore, this study aimed to evaluate the daily quality of dolphin sperm after several days of refrigeration. The sperm of two male dolphins were stored at 4oC for 4 days, and the quality was observed daily to determine the motility, viability, membrane integrity, and sperm abnormalities. Sperm samples were divided into four groups, consisting of two centrifuged followed by the removal of seminal plasma, and two groups without centrifugation, containing 100x106 and 200x106 sperm/ml each. After liquid storage, the motility of sperm was 63-75% with no significant reduction in the first 3 days. Sperm viability following storage was 65-75% and the percentage with abnormal morphology ranged from 2-6%. Furthermore, there was no significant increase in abnormal morphology of sperm on any day of storage for 3 days. Sperm membrane integrity was 36-49%, with no significant reduction in the membrane integrity in the first 2 days. There was no significant difference in sperm quality, although centrifugation and removal of seminal plasm had a slight effect. The results of this study showed that Indo-Pacific bottlenose dolphin sperm could be stored for a short period as liquid storage while maintaining a quality that allows for future use.

Abstract

Standar kualitas semen yang berperan penting dalam keberhasilan inseminasi buatan (IB) sebagai upaya peningkatan populasi dan mutu ternak sapi di Indonesia terbukti tidak seutuhnya mampu menentukan tingkat fertilitas sapi pejantan di Indonesia. Studi terbaru membuktikan bahwa meskipun sudah memenuhi standar kualitas semen yang sudah ditetapkan, namun tiga dari enam ekor sapi pejantan di Indonesia masih tergolong fertilitas rendah (<60%). Potamine-1 (PRM1) merupakan molekul pada inti spermatozoa yang telah terbukti memiliki hubungan erat dengan fungsi fertilitas dan kualitas spermatozoa pada berbagai sapi pejantan di Indonesia. Hal tersebut mendasari usulan pengembangan uji berbasis molekuler PRM1 sebagai penentu tingkat fertilitas yang diharapkan dapat digunakan untuk proses seleksi dan kebijakan pengafkiran sapi pejantan unggul di Indonesia. Melalui policy brief ini diharapkan usulan pengembangan uji tersebut dapat lebih tersampaikan dengan segala solusi yang diberikan, termasuk terkait pendekatan uji berbasis molekuler yang paling tepat digunakan, kajian-kajian lanjutan, dan berbagai harapan dukungan dari segala pihak demi terwujudnya pengembangan uji PRM1.

Abstract

Indonesian local sheep represent a particular species of sheep reared by farmers on small-scale farms. Semen freezing can be performed using either a homemade or commercial extender. The present study evaluated the effectiveness of several commercial extenders in freezing Indonesian local sheep. Semen was collected from three mature and healthy rams using an artificial vagina and evaluated. It was determined that only samples exhibiting sperm motility levels of greater than 70% would be included in the study. Subsequently, the semen from each ram was divided into three portions and diluted with Andromed, Sterydil, and OptiXcell, respectively. The frozen semen was then evaluated 24 hours post-freezing. The thawing process was conducted individually at 37°C for 30 seconds. The evaluation encompassed a range of metrics, including sperm motility, viability, and the presence of any abnormalities, in addition to the recovery rate. The results indicated that sperm frozen in Optixcell and Sterydil extenders exhibited superior motility, viability, and recovery rate than Andromed. The study also demonstrated no interaction between individual Ram and extender, and no differences were found between individuals. The study concluded that Optixcell and Sterydil extenders effectively froze Indonesian local sheep semen.

Abstract

One factor that affects frozen semen characteristics is bull age. Increasing age induces changes in sperm that reduce frozen semen quality. This study aimed to evaluate the frozen semen characteristics of Bali bulls aged 3, 8, and 13 years. Frozen semen was derived from fresh semen with sperm motility > 70%. The frozen semen characteristics tested included motility, viability, plasma membrane integrity, acrosome integrity, and sperm abnormalities. Sperm motility was analyzed by computer-assisted sperm analysis (CASA). Viability was assessed using eosin-nigrosin staining; plasma membrane integrity was assessed using the hypoosmotic swelling test (HOST); acrosome integrity was assessed using Giemsa staining; and sperm abnormalities were assessed using Williams staining. The plasma membrane integrity of the 13-year-old group was significantly lower (P < 0.05) than that of the younger age groups. However, there were no significant differences (P > 0.05) in the other parameters. We conclude that age affects the integrity of the sperm plasma membrane in frozen semen from Bali bulls.

Abstract

Bottlenose dolphins were often found in zoos and aquariums. Dolphin calves have a higher mortality rate in the first month after birth as they are unable to eat and breathe properly. The inability to thrive can affect growth until adulthood. This research was being conducted to better understand the nursing and breathing patterns of a bottlenose dolphin calf. Data were collected by observing the behaviour of a post-partum dolphin calf for 24 hours over 3 months (90 days). The behaviours observed were the calf's nursing (frequency, daily duration, and duration of suckling) and breathing (frequency). The results showed a decrease in the frequency of suckling and an increase in the duration of suckling, which indicated that the calf was becoming more successful at suckling. A constant decrease in respiratory frequency indicated a healthy growing calf due to an increase in lung capacity. These results indicated that the feeding and respiratory behaviour patterns of normally developing bottlenose dolphin calves showed a decrease in feeding frequency and an increase in feeding duration, as well as a decrease in respiratory frequency per week. Further studies on the feeding and respiratory behaviour of bottlenose dolphin calves.

Abstract

Age is among the factors affecting reproductive organs, particularly semen characteristics. Semen characteristics are also influenced by composition of seminal plasma that is ejaculated with the sperm. Therefore, this study aimed to investigate characteristics of fresh semen and seminal plasma composition from Balinese cattle aged 3, 8, and 13 years. Semen characteristics observed were concentration, progressive motility, viability, plasma membrane integrity, acrosome integrity, and sperm abnormalities. Biochemical composition of seminal plasma analyzed was fructose, malondialdehyde (MDA), total antioxidant capacity (TAC), glutamic oxaloacetic transaminase (GOT), glutamic pyruvic transaminase (GPT), total protein (TP), albumin, cholesterol, and triglycerides. Mineral compositions observed were Na, K, Ca, Mg, Zn, Cu, Fe, and Se. The results showed that there were no significant differences (p > 0.05) in semen characteristics across age groups, meaning that semen of Balinese cattle can still be processed into frozen semen even at an older age, provided it meets the SNI 4869-1:2024 standards. Furthermore, the concentrations of each seminal plasma biochemical component were as follows: fructose (377.5–615.4 mg/dL), MDA (9.7–10.36 nmol/mL), TAC (66.22–67.22%), GOT (203–265 U/L), GPT (19–35 U/L), TP (4.3–5.5 g/dL), albumin (1.7–1.9 g/dL), cholesterol (11–35 mg/dL), and triglycerides (822–1.078 mg/dL). Surprisingly, the concentration of triglycerides in semen plasma in this study was very high compared to those reported in previous studies on several cattle breeds. Seminal plasma mineral concentrations included Na (1.500–1.900 mg/kg), K (600–700 mg/kg), Ca (600–800 mg/kg), Mg (38.85–45.16 mg/kg), Zn (5.33–9.64 mg/kg), Cu (0.06–0.12 mg/kg), Fe (11.85–15.88 mg/kg), and Se (0.92–1.48 mg/kg). These findings indicate that age did not affect semen characteristics, biochemical, and mineral composition of seminal plasma in Balinese cattle.

Abstract

Local sheep are small ruminants that are kept by many farmers. The reproductive success of the animals is influenced by the ration offered. The aim of this study is to evaluate the reproductive performance of multiparous ewes with different flush management. For the study, 16 multiparous ewes were used in a 4 x 4 randomized group design. Treatments consisted of P1 = no flushing, P2 = flushing before and after mating, P3 = flushing before and after mating and at mid-pregnancy, P4 = flushing before and after mating, at mid-pregnancy and at the end of pregnancy. The variables observed were the percentage of pregnancy, number of embryos, embryo diameter, amnion diameter, embryo durability, number of offspring and duration of pregnancy. The results showed that the management of flushing rations influenced the pregnancy rate and amniotic diameter. The number and diameter of embryos, gestation length, number of offspring and embryo lifespan did not differ between treatments (P>0.05). This study concludes that the management of flushing rations affects reproductive performance in terms of pregnancy rate and amniotic diameter.

Abstract

The aims of this study were to investigate the freezing capability of Pasundan bull spermatozoa in Tris-egg yolk (TEY), Tris-soy (TS), and AndroMed as diluents. Semen were collected twice a week from four Pasundan bulls aged 3-5 years old using an artificial vagina and evaluated macro- and microscopically. Semen had 70% sperm motility, 800x106/mL sperm concentration, and less than 20% sperm abnormalities were divided into three parts and each of them diluted with TEY, TS, or AndroMed. After an equilibration step at 5C for four hours, diluted semen were packaged in 0.25 mL straw, frozen in liquid nitrogen for ten minutes and kept in liquid nitrogen container until examination. Motility test on fresh, diluted, equilibrated, and after-thawed semen was done using Androvision. The results showed that after thawing motility of sperm diluted in AndroMed (58.640.72%) was higher than in TEY (49.451.22%) and TS (39.346.33%). Sperm motility of Pasundan bulls diluted in these three diluents reduced around 33.272.45% during freezing process.

Abstract

This research was aimed to study the effect of administration of glycerol and dimetilacetamida (DMA) in BTS and MIIIextender supplemented with sodium dodecyl sulphate (SDS) on boar frozen semen. A number of four boars were used in this study for semen collection (n=20). The collected semen was evaluated both macroscopically and microscopically. In this study, only the semen that demonstrated70% sperm motility, 200.106/mL sperm concentration, and20% sperm abnormalities were used and divided into eight tubes. A number of 4 tubes were diluted with 5 mL of BTS, and the rest with 5 mL MIII. The sampel was stored at 20-22 C for 2 hours, followed by centrifugation for 15 minutes (at 2000 rpm), and taken of pellet with 1 ml supernatant. The pellet that was resulted from centrifugation using BTS, then re-diluted with BTS-glycerol 5% (BTSG), BTS DMA 5% (BTSD), BTS-glycerol 5% and SDS (BTSG-S), BTS-DMA 5% and SDS (BTSD-S). Four other pellets that were centrifuged with MIII also re-diluted with MIII-glycerol 5% (MIIIG), MIII-DMA 5% (MIIID), MIII-glycerol 5% and SDS (MIIIG-S), MIII-DMA 5% and SDS (MIIID-S). Next, all of diluted semen were inserted into 0.5 mL straw and equilibrated for 2 hours (4 C), then frozen and stored in liquid nitrogen. The evaluation of frozen semen quality was conducted at 24 hours after frozen. The result of this study showed that post-thawing motility of spermatozoa in BTSD-S (40.170.2%) was found higher (P0.05) compared to seven other dilution processes. Therefore, it is concluded that the concentration of 5% DMA that supplemented with SDS in BTS dilution much better for maintaining boars frozen semen quality.
