Abstract

ABSTRACT. Due to their phytochemical constituents, Dewa leaves (Gynura japonica (Thunb.) Juel.) are often used in traditional medicinal herbs. However, in vitro and in vivo of antioxidative and anti-aging studies of Dewa leaves on yeast as a eukaryotic cell model have not been widely carried out. This study aims to determine the antioxidant, anti-aging, and antibacterial activities derived from G. japonica leaves extract. Extraction was conducted using 70% and 96% ethanol solvents, total phenolic content (TPC) was assayed using Folin-Ciocalteu method, flavonoid contents (TFC) was assayed using aluminum chloride method, antioxidant activity was tested using DPPH (1,1-diphenyl-2-picrylhydrazyl) and ABTS (2,2-azinobis-3-Ethylbenzothiazoline)-6-sulfonic acid) radicals. Subsequently, an anti-aging activity test was performed on the yeast Saccharomyces cerevisiae as a model organism following antibacterial activity. The antibacterial activity test was carried out using well diffusion agar, and the Minimal Inhibitory Concentration (MIC) was determined using the microplate method. We obtained that G. japonica leaves extracted from 70% and 96% methanol solvents, have TPC of 13.14 and 22.11 mg GAE/gr extract, and TFC of 8.04 and 14.09 mg QE/gr extract, respectively. As for DPPH and ABTS antioxidant activity, D70 showed the best activity with IC50 values of 1411.36±56.35 µg/mL and 2516.10±18.77 µg/mL, respectively. The anti-aging test showed that both 70% and 96% ethanol extracts were able to maintain the yeast cell viability under H2O2 oxidative stress. Further, 70% and 96% ethanol extract also showed antibacterial activity at the best value against Staphylococcus aureus with a MIC value of 390.62 µg/mL, it means that Gynura leaves extract has a potency as antibacterial agent. The results of this study indicate that the ethanol extract of G. japonica leaves can be developed for further investigation as an antioxidant and antibacterial therapeutic agent. Keywords: Anti-aging, antibacterial, antioxidant, Gynura japonica, Saccharomyces cerevisiae

Abstract

Earlier research reported that GABA had a correlation with diabetes in processes of glucose homeostasis. This study aims to identify the validity of B-receptor Rattus norvegicus as GABA protein target, modeling the structure and knowing the binding affinity between GABA and B-receptor Rattus norvegicus through molecular docking.   The research was carried out using in-silico method.  The interaction of GABA with the target protein was determined using SuperPred, followed by modeling the protein target using SwissadMe. Ramachandran Plot and Errat Procheck are used to determine the validity of the protein target. Molecular docking was determined using Pyrx and PyMol. The results showed that GABA binding to the B- receptor Rattus norvegicus has biological activity as glucose oxidase inhibitor and antidiabetic.  The conclusion are: B-receptor Rattus norvegicus is a valid protein target for binding to GABA; there are four 3-dimensional models of B-receptor Rattus norvegicus and the best model has 98.43% sequence identity; the binding affinity of GABA (ligand) on B-receptor Rattus norvegicus from the best model is -3.4 kcal/mol energy, 1.773 RMSD lower bound, and 1.81 RMSD upper bound. It is suggested that this research might be used as an empirical basis to further investigate GABA as antidiabetic.

Abstract

This study aimed to compare the growth of green spinach (Amaranthus hybridus L.) cultivated in two different soil types originating from Java and Kalimantan, and to examine the effect of soil pH on plant height. The experiment was conducted over a 21-day period (3–24 November 2025) using a comparative experimental design with a total of eight samples, consisting of four plants in each treatment group. The parameters observed included soil pH, plant height, number of leaves, leaf width, and leaf color. Environmental variables—such as light intensity, water volume, pot size, and seed quantity—were controlled to ensure consistent experimental conditions. The data were analyzed using statistical assumption tests, including the Shapiro–Wilk normality test and Levene’s homogeneity test, followed by linear regression analysis and analysis of covariance (ANCOVA). The results indicated that the data were normally distributed (p = 0.328) and homogeneous (p = 0.575). Regression analysis showed that soil pH had a significant effect on plant height (p = 0.026), whereas soil type did not have a significant effect after controlling for soil pH (ANCOVA, p = 0.089). The mean difference in plant height between soils from Java and Kalimantan was 2.90 cm; however, this difference was not statistically significant. These findings suggest that the growth of green spinach—particularly plant height—is more strongly influenced by environmental factors, especially soil pH, than by differences in soil origin.

Abstract

This study aims to analyze the initial profile of high school students' creative thinking skills in biology learning. The research is descriptive quantitative research using an open-ended test method. The subjects of this study were tenth-grade high school students. The results of the analysis show that the average creative thinking skills of high school students is 37.85%, which is classified as not creative. Specifically, the lowest percentages were found in the aspects of originality, elaboration, and flexibility, while the aspect of fluency showed relatively higher achievements. The conclusion of this study is that the profile of students' creative thinking skills in biology learning is low. These findings indicate that the dominance of convergent learning through closed questions and student passivity in class are factors that inhibit the development of divergent thinking potential. Therefore, it is necessary to apply active learning models and use more exploratory evaluation instruments to stimulate students' ability to produce unique, flexible, and in-depth ideas.
