Abstract

Kelainan kongenital merupakan suatu kelainan struktur maupun kelainan fungsi yang timbul akibat gangguan pada periode intrauterine. Penegakkan diagnosa kelainan kongenital dapat dilakukan baik sebelum lahir, saat lahir, ataupun setelahnya. Angka kejadian kelainan kongenital bervariasi di setiap negara, di Indonesia angka kejadian kelainan kongenital mencapai 5 dari 1000 kelahiran (5%). Kelainan kongenital dapat terjadi pada semua sistem tubuh. Berbagai studi mengindikasikanbahwa sistem kardio-toraks, sistem saraf pusat dan kelainan kromosom merupakan jenis kelainan kongenital yang banyak dijumpai di seluruh dunia. Penelitian ini merupakan studi observasional deskriptif yang bertujuan untuk mengidentifikasi angka kejadian kelainan kongenital pada bayi baru lahir di RSMH Palembang periode Januari-November 2015 serta untuk mengidentifikasi jenis kelainan kongenital yang paling banyak dijumpai di RSMH Palembang. Seluruh bayi yang lahir dan dirawat serta dinyatakan menderita kelainan kongenital oleh dokter spesialis anak yang berkompeten di RSMH Palembang pada periode penelitian dijadikan sampel. Dari hasil penelitian didapatkan sebanyak 108 bayi yang terdiagnosa dengan berbagai jenis kelainan kongenital pada periode Januari-November 2015 di RSMH Palembang. Adapun jenis kelainankongenital yang dijumpai pada 108 sampel penelitian meliputi kelainan pada sistem susunan saraf pusat, sistem kardiotoraks, sistem gastro-intestinal, sistem kranio-fasial, sistem muskuloskeletal dan juga kelainan kromosom. Kasus-kasus kelainan kongenital pada sistem gastro-intestinal merupakan jenis kelainan kongenital yang paling banyak dijumpai diRSMH Palembang (50%), diikuti oleh kasus-kasus kelainan kongenital pada sistem kardio-toraks (29,63%). Abstract

Introduction Candida is a yeast fungus classes most commonly found in the oral cavity, gastrointestinal tract, reproductive tract and skin, especially the species Candida albicans. Alternative medicines such as herbs are easily available and affordable by the community, such as keghenyat leaves (Acmella uliginosa). Aim of StudyTo determine the activity of Keghenyat leaves fraction (Acmella uliginosa) compared to Nystatin against Candida albicans in vitro. MethodsIn vitro study was held in Laboratory of Genetics and Biotechnology Department of Biology, Faculty of Science and Medical Faculty, Sriwijaya University in December 2015 and January 2016. Research stages: extraction, fractionation, sensitivity test using Nystatin and Candida albicans, fractions antifungal activity test, the determination of minimum inhibitory concentration (MIC), bioautografi test and determination of compound classes. One final stages of testing the active fraction Keghenyat leaves (Acmella uliginosa) with Nystatin. Data were analyzed using ANOVA and Post Hoc Duncan test and linear regression using SPSS 20. ResultsHexane faction of Keghenyat leaves (Acmella uliginosa) is active against the fungus Candida albicans, has a Minimum Inhibitory Concentration (MIC) of 0.625% (6250μl) against the fungus Candida albicans antifungal contain active compounds that terpenoids and flavonoids, amounting to 84.68 mg / ml equivalent to 1 mg / ml Nystatin against the fungus Candida albicans. ConclusionThere is a significance different MIC between hexane fraction of Keghenyat leaves (Acmella uliginosa) and Nystatin, Nystatin have more high activity.

Abstract

Background Male participation in KB is still relatively low when compared to the participationof women. Researchers have to do research to find the contraception drug. Temu putih (Curcuma Zedoaria (Berg) Roscoe) is one of traditional herb that used as antifertility. Aim of Study Aim of this study to examine change in the amount, motility, morphology, and viability spermatozoa male rats (rattus norvegicus) due to temu putih fraction suplementation. Methods This study was an experimental study using a completely randomized design (CRD), post test with control group design. The sample in this study was 30 male rats, 10 weeks old, weight 150-200 gram. Rats were given temu putih fraction (n hexan, etylacetate and methanol-water) at dose of 300 mg/kgBB/day for 48 days. Temu putih was extracted by ethanol and did fractionation by liquid-liquid methods. The results of this study were assayed by SPSS 18. Results The amount, motility, morphology and viability of spermatozoa in the group of metanol fraction of water decreased compared with the control group (p= 0,000), motilitas of spermatozoa in the group of metanol water fraction decreased compared with the control group. Conclusion Temu putih fraction can reduce the amount, motility, morphology, and viability of spermatozoa in male rats. Abstract

Abstract Background Receptor for advanced glycation endproducts (RAGE) is a transmembrane protein that belongs to the immunoglobulin superfamily. As its name implicates, it can bind to advanced glycation endproducts, the resulting product of non-enzymatic glycosylation, and it also has the ability to interact with multiple ligands having common motifs as a so-called multi-ligand receptor. The ligands include high-mobility group protein (B)1 (HMGB1), S-100 calcium-binding protein, amyloid-β-protein, Mac-1, and phosphatidylserine. Interaction between RAGE and its ligands activates various cellular processes, including inflammation, proliferation, apoptosis, autophagy, and migration. Methods The proccess of isolation protein of RAGE was initiated with extraction protein and purification of RAGE protein. After that, the immunization of Rats was be done to produce Anti-RAGE. The confirmation of Anti RAGE was be done by SDS PAGE and Immunobloting. Results The production of Anti RAGE was enough pure compared by Anti RAGE commercial. Anti RAGE was protein that have molecular weight around 35 kD. Conclusion The methods that used in this study effective to develop production of anti-RAGE. Abstract

ABSTRACT Background. Disorders of Sex Development (DSD) is a term used to describe a congenital condition where sex chromosomes, gonads, and or anatomy of a person are atypical. It is estimated to affect 1:4,500 people worlwide with varied clinical findings, hence the integrated and diverse interventions. Objective. This study wished to observe the many interventions DSD patients received in Dr. Mohammad Hoesin Hospital as one of the national reference hospitals in Indonesia for South Sumatera region. Methods. Medical records of 173 patients with clinical findings in accordance with DSD spectrum during 2013-2017 was observed in order to identify both surgical and medical interventions applied. Results. Sixty two patients were diagnosed of having hypospadias (35.84%), making it the most common clinical finding among DSD patients in this hospital. Surgical intervention was the most favored intervention, with 155 patients (89.6%) underwent various surgeries, ranging from urethroplasty to vaginoplasty and many more. Five patients(2.9%) were treated with either hydrocortisone and florinef, or both. One patient diagnosed with Congenital Adrenal Hyperplasia (CAH) had both surgical and medical intervention. Conclusion. DSD gives a wide phenotypic spectrum, making its management both challenging and complicated. Surgical, medical, and or a combination of both are the indispensable interventions when dealing with DSD patients. Study focusing on psychological intervention for DSD patients is further needed. Abstract

Abstract Background. The increasing prevalence of depression gives rise to challenges in not only elucidating its diverse causes, but also in finding an effective treatment. One of the factors linked to depression is the imbalance of serotonin, norepinephrine, and dopamine neurotransmitters. Cinnamon (Cinnamomum burmannii) as one of the world’s wellknown cooking ingredients is believed to be able to regulate the neurotransmitters imbalance with the help of terpenoids and flavonoid polyphenols as one of its content. Objective. This study aims to determine the effectiveness of cinnamon extract as an antidepressant in depressed induced animal model. Methods. An experimental in vivo with pre-post control group design was conducted in twenty five Wistar strain white rats that were divided into 5 treatment groups that received fluoxetine as positive control, aquades, and different dose of cinnamon extracts (50 mg/kgBW, 100 mg/kgBW, and 200 mg/kgBW). Depression induction method used was 3-minute Tail Suspension Test, done for 14 days. The antidepressant effectiveness test was carried out by calculating the immobility time duration with Forced Swimming Test method and was further analyzed using one-way ANOVA test. Results. One-way ANOVA test results showed that there were differences in the mean duration of immobility time between treatment groups after being given cinnamon extract (p value = 0,000). Groups that were given 100 mg/kgBW cinnamon extract and 200 mg /kgBW showed a p value>0.05 when compared with positive control group receiving Fluoxetine although displayed a similar reduced immobility time. Conclusion. Cinnamon (Cinnamomum burmannii) extract showed a promising potential as an effective antidepressant tested in animal model. Keywords: cinnamon, extract, depression, immobility time, rat

Abstract

ABSTRACT. Background. Hypospadias is one of the most common urogenital anomaly in males. The etiology is poorly understood, but is believed to be multifactorial where Intra Uterine Growth Restriction (IUGR) is one of the suspected risk factors, leading to low birth weight and low birth length in affected patients. Objective. This study aims to identify the birth anthropometry characteristics of hypospadias patients who visited RSUP. Dr. Mohammad Hoesin Palembang. Materials and Methods. This was an observational descriptive study with cross sectional design. This study included hypospadias patients with detailed birth characteristics in RSUP Dr. Mohammad Hoesin Palembang during the period of 2012-2015. Results. Out of 106 hypospadias patients, only 19 (17.92%) of them presented with detailed birth characteristics. There were 12 (63,2%) patients with low birth weight and 7 (36,8%) patients with normal birth weight. There were 10 (52,6%) patients with low birth length and 9 (47,4%) patients with normal birth length. The mean birth weight is 2442 grams and the mean birth length is 46,9 centimeters. Conclusion. The majority of hypospadias patients in this study showed a low birth weight and low birth length, signalling an accordance to the existing theory. Abstract

ABSTRACT Background. Disorders of Sex Development (DSD) is a condition where the development of sex chromosomes, gonads, and/or one’s anatomy is atypical. Its causes are often due to genetic mutations, although some are also linked to environmental risk factors. These multiple aetiologies lead to varied clinical findings, ranging from obvious ambiguous genitals to subtle ones in different regions worldwide, signalling a hint of geographical variability. Objective. This study wishes to observe the variations of clinical findings of DSD patients geographically in South Sumatera. Methods. This was an observational study using patients’ medical records in RSUP Dr. Mohammad Hoesin Palembang. Both inpatients and outpatients during five-year period span (2013-2017) with clinical findings suited DSD criteria based on Chicago Consensus in 2006 were included in this study. Results. One hundred and forty nine patients from cities and regencies in South Sumatera province and other provinces like Jambi, Lampung, Bengkulu, Bangka-Belitung, and even Riau were included in this study. Among sixteen clinical findings identified, hypospadias ranked first (59.06%), both in general, and in each regions as well. When set by side with other regions, Palembang city as the capital city of South Sumatera province displays twelve out of sixteen clinical findings documented in this study, showing a lot more variety. Conclusion. Every regions show difference clinical findings. Some regions housed clinical findings that were not found in other regions. However, hypospadias is the most commonly found clinical findings in all regions. It is suspected due to its correlation with certain environmental risks, that the occurence of it becomes rather often, compared to other DSD conditions. Future studies considering risk factors involvement in order to elucidate both differences and similarities found in each regions are strongly suggested. Abstract

ABSTRACT Background. Trinucleotide Repeat Expansion (TRE) in human DNA could lead to various diseases. An expanded CAG repeat (>31 or 37 repeats, depends on the ethnicity) in Androgen Receptor gene is suggested to be associated with the occurrence of isolated hypospadias. In an effort to identify the exact numbers of repeats, sequencing has been the most favored method to be conducted despite its cost. Objective. This study wished to investigate the possibilities of using Polymerase Chain Reaction (PCR) method to screen expanded repeats in isolated hypospadias, as one of the TRE diseases. Materials and Methods. Numbers of CAG repeat in twelve hypospadias patients and one normal male was first predicted from the visualization of PCR products in 3% agarose gel electrophoreses with 20 bp ladder marker before it was finally sequenced. Results. Two samples gave the same exact result, while the rest showed a range of 1-11 bp differences. Statistically, there was a significant difference between the mean of CAG repeats from PCR method (M=26.1667, SD=6.71272) and the mean of CAG repeats from sequencing (M=23.75, SD=5.70685); t(11)= 4.570, p=0.001. Furthermore, the sensitivity of PCR was 100% and the specificity was 83.33%. Conclusion. It can be concluded that PCR method could be used as a screening method in identifying TRE with large numbers of repeats. However, PCR in TRE disease with small numbers of expanded repeats needs to be followed by sequencing in order to obtain the exact numbers of repeats. Abstract

Background. Autism Spectrum Disorder (ASD) is a complex neurodevelopmental condition characterized with disorders in cognitive, behaviour, and social interaction. It is estimated to affect 1:160 children worlwide. Despite its increasing trend, the exact cause of it is yet to be known. Objective. This study wished to identify the geographical distribution of ASD cases in Dr. Mohammad Hoesin Hospital as one of the national reference hospitals in Indonesia for South Sumatera region. Methods. Medical records of the patients who are diagnosed with ASD during 2014-2018 was observed in order to identify the patients’ residential area. Results. There were 107 patients who were clinically diagnosed of having ASD during 2014-2018 in Dr Mohammad Hoesin Hospital, Palembang. Based on its residential area, ASD cases were mostly found in Palembang as the capital city of South Sumatera Province. Conclusion. ASD is found in small and big cities across the South Sumatera province, implying its common occurrence. The varied prevalence could be signifying the role of different related risk factors identified in the area.
