Abstract

Kegiatan pengabdian masyarakat ini dilakukan di Pesantren Al Fatih di Desa Klampar Kecamatan Proppo, Kabupaten Pamekasan Madura. Tujuan kegiatan pengabdian kepada masyarakat ini adalah untuk meningkatkan kualitas pengelolaan usaha peternakan sapi pedaging, dan untuk meningkatkan peran dan fungsi pondok pesantren sebagai pendidikan non formal guna mendukung ketahanan pangan serta penguatan penerapan biosecurity dalam upaya pencegahan PMK. Metode yang digunakan dalam kegiatan ini melalui observasi lokasi, diskusi dan wawancara, Participatory Rural, Participatory Technology Development, Educative, Forum Group Discussion (FGD). Hasil kegiatan menunjukkan bahwa peserta sangat antusias saat pelaksanaan. Edukasi yang diberikan pada peserta memberikan peningkatan pengetahuan tentang biosecurity yang signifikan, dari nilai pre-test 68.86 point, sedangkan nilai post-test 78.16 point, sehingga terjadi peningkatan 14%.  Pengetahuan yang diaedukasikan meliputi; biosecurity PMK, sanitasi kandang, biosecurity keluar masuk wilayah farm/peternakan dan isolasi sapi yang sakit. Kesimpulan dalam kaegiatan ini adalah, peserta mampu memahami tentang materi peningkatan kualitas ternak melalui biosafety dan biosecurity PMK dan menerapkan teknologi tepat guna tentang formulasi pakan, dan sanitasi kandang untuk pencegahan dan reinfeksi PMK.

Abstract

Streptococcus mutans is the prior bacteria causing dental caries. Suruhan (Peperomia pellucida) may prevent dental caries due to its antibacterial bioactive content. This study aims to identify the bioactive profile of Suruhan ethanol extract and its antibacterial activity against Streptococcus mutans. Bioactive compounds were analyzed using Gas Chromatography-Mass Spectrometry (GC-MS). Antibacterial activity was tested through an inhibition test using the well-diffusion method, the Minimum Inhibitory Concentration (MIC) test, and the Minimum Bactericidal Concentration (MBC) test. The most detected compounds in Suruhan ethanol extract were n-eicosane, n-hexadecane, and glycerol. There are also antibacterial bioactives such as phenols, flavonoids, alkaloids, and terpenoids. Inhibition test of 500 mg/mL extract showed a clear zone of 8.25 mm diameter. The MIC and MBC values were 50 mg/mL and 100 mg/mL, respectively. Based on the results, Suruhan ethanol extract shows potential as an antibacterial, although its inhibitory effectiveness could be better.

Abstract

The meats physical quality is reflected by meat characteristics and reference for consumers to choose good quality of beef. Several factors that affect the meat quality are interaction genetic and environmental factors during maintenance until post slaughter. The research was conducted to evaluate the meat physical quality with the breeds factor (Red Brahman Crossbred and Droughtmaster), genders (steer and heifer), and ages (PI0, PI2, and PI4). The research materials were 120 samples of Longissimus dorsi ribs 12th from AM FARM abattoir and tested physical quality meat on QC Laboratory of KASA Company. The data were analyzed by ANOVA and Least Significant Difference if there were any significant. The variables observed were pH, meat color, WHC, cooking loss, and tenderness. The results showed that breeds factors have no significant difference on meat physical quality. The factor of genders nested to breeds has very significance (P<0,01) on meat physical quality except for pH and tenderness there wasn’t significant differences. The factor of ages nested to genders nested to breeds has very significant (P<0,01) on meat physical quality except that pH. The conclusion that the evaluation of meat physical quality can be predicted by the relationships of breed, genders, and age factors. Keywords: Meat physical quality, breeds, genders, ages

Abstract

ABSTRAKSapi madura merupakan sumber daya genetik rumpun asli lokal Indonesia. Pengukuran morfometrik dan fenotip kualitatif bermanfaat untuk menentukan asal usul dan hubungan filogenetik antara species bangsa dan tipe ternak. Profil morfometrik dapat memperkirakan karakteristik fenotipik atau ciri khas sapi sonok diantara sub populasi sebagai pusat pembiakan  dan menggali potensi sapi sonok sesuai kelompok bibit  sesuai standart bibit sapi Madura. Penelitian selama 3 bulan dengan lokasi  kecamatan Pasean, Batumarmar, dan Waru  Kabupaten Pamekasan. Materi 162 ekor sapi madura tipe sonok peserta kontes. Metode penelitian adalah metode survei, penentuan lokasi berdasarkan purposive sampling, teknik pengambilan data secara purposive sampling.  Data morfologi kuantitatif dianalisis menggunakan Analisis Komponen Utama (AKU), fenotipe kualitatif secara diskriptif.  Hasil penelitian menunjukan karakteristik kualitatif telah sesuai dengan SNI No.7651-2 (2013) dan SNI No.7651-2:2020. Hasil pendekatan Analisis Komponen Utama (AKU) sapi Madura tipe sonok dilokasi Waru, Pasean, dan Batumarmar menjadi kelompok tersendiri. Penciri spesifik sapi madura tipe sonok adalah arah tanduk keatas mengarah kedalam, mata sipit, punuk besar, bergelambir dengan lipatan-lipatan dan diantara kedua kaki depan terdapat jaringan otot yang menonjol.Kata Kunci: sapi madura, morfometrik, sonokMorphometric characteristics of Sonok Type Madura CowsABSTRACTMadura cattle are the genetic resources of Indonesia native cattle breeds. Morphometric and qualitative phenotype measurements are important in determining the origin and phylogenic relationships between species, breed, and type of cattle. Morphometric profiles can be used to estimate phenotypical charecteristic or special features of Sonok cattle among the sub population as the breeding center and explore the potential of Sonok cattle as standard for Madura cattle breed.  This research was conducted for 3 months at Pasean, Batumarmar, and Waru sub districts of Pamekasan District. The materials used for this research were 162 Sonok type Madura cattle which are enrolled for cattle contest. The methods used were survey by purposive sampling both on location and data selection. Quantitative morphology data was analysed by main coponent analysis and qualitative phenotypedata was analysed descriptively. The results of this research indicate that the qualitative characteristics of Sonok type Madura cattle is as required by SNI No.7651-2 (2013) and SNI No.7651-2:2020. Main component analysis results show that Sonok type Madura cattle located at Waru, Pasean, and Batumarmar are specific group of cattle. The specific characteristics of sonok type Madura cattle are the inner-towards horns, slit-eye, big hump, and flaky skin in between the forelegs with protruding muscle fibers.Keywords: madura cattle, morphometrics, sonok

Abstract

The objective of this study was to determine the effect of sex and age of slaughter on the carcass characteristics of white Brahman crossbred cattle in feedlot Lampung, Indonesia. A total of 126 white brahman cattles consisting of 63 steers (21 PI0, 21 PI1, 21 PI2) and 63 bulls (21 PI0, 21 PI1, 21 PI2) were selected for this study. The slaughter was conducted using non stunning method using box mark 4. Data analysis was done using factorial completely randomized design and Duncan's continued test. The results showed that the slaughter age factor significantly (P <0.05) had a significant effect on slaughter weight, carcass weight, carcass percentage, meat weight, fat weight, fat percentage, bone weight, bone percentage, meat bone ratio (MBR), meat fat ratio (MFR) and rib eye area (REA). The interaction of sex and slaughtering age factors showed a significant effect (P <0.05) on slaughter weight, carcass weight, meat weight, meat percentage, bone percentage, and MBR. Sex and age of slaughtering had no significant effect (P> 0.05) on the thickness of the back fat of the white Brahman crossbred cattle. It can be concluded that the factors of sex and age have an effect on the carcass characteristics of the white Brahman crossbred cattle.Keywords: Carcass Characteristics, White Brahman Crossbred, Steer, Bull, Age

Abstract

Background: Recently, many people have considered electronic cigarettes to be a safer alternative to conventional tobacco cigarettes, even though they still contain harmful substances such as nicotine, which is associated with a decrease in hippocampal neurons. Objective: This study aimed to examine the effect of exposure to e-cigarettes on hippocampal neurons in rats. Methods: An experimental study was performed using 26 tissue samples obtained from Wistar rats. The rats were randomly assigned to four groups and treated over an eight-week period as follows: the control group (K) (n=7) had no vapor exposure; the V0 group (n=6) received vapor without nicotine (0 mg/ml); the V6 group (n=7) was exposed to vapor and 6 mg/ml nicotine; and the V12 group (n=6) was exposed to vapor and 12 mg/ml nicotine. Hippocampal neurons were examined using hematoxylin-eosin staining at 400x magnification across 7 fields of view. Data were analyzed using the Kruskal-Wallis test with the Mann-Whitney post-hoc.  Results: The number of hippocampal neurons was significantly reduced in both the V12 and V0 groups compared to that in the control group (p<0.05). The lowest number of hippocampal neurons was observed in the V0 group (0 mg/ml nicotine), followed by the V12 group (12 mg/ml), and then the V6 group (6 mg/ml).Conclusion: The present study showed that the variation of nicotine in vapor exposure decreases hippocampal neurons in rats.

Abstract

ABSTRACT. Knowledge of farmers about estrus detection is one of the factors that affect fixed-time artificial insemination (AI). This study aims to evaluate the success rate of AI using Y sexing frozen semen based on the estrus character of cattle. Forty-five Limousin Crossed Cows involved in this research with a Body Condition Score of 3-5 (1-9 scale), 1.8-7 years old. Y sexing frozen semen is produced by the Singosari Center for Artificial Insemination using Percoll's Gradient Density Centrifugation method. Observation of estrus character was done before insemination with the deep insemination technique of AI. Artificial insemination was carried out using double doses at the 2nd and 8th hours after estrus. Rheinbio vitamins as BioATP+ are injected after AI. The results showed that the Conception Rate and Pregnancy Rate based on the estrus character was higher as indicated by the red colour of the vulva was 17.78% and 22.22%; very swollen vulva was 20% and 22.22%; abundant cervical mucus was 15.56% and 17.78%, and vaginal temperatures ranging from 38.0-38.5oC was 13.33% and 20%. In conclusion, the low percentage of pregnancy in this study was caused by various factors, mainly the genetic quality, early embryonic efficacy, maintenance management especially feed, and there were cows that experienced reproductive disorders.(Evaluasi keberhasilan inseminasi buatan menggunakan semen beku sexing berdasarkan karakter estrus yang berbeda)ABSTRAK. Pengetahuan peternak mengenai deteksi estrus merupakan salah satu faktor yang berpengaruh terhadap ketepatan waktu inseminasi. Penelitian ini bertujuan untuk mengevaluasi tingkat keberhasilan IB menggunakan semen sexing Y berdasarkan karakter estrus ternak. Akseptor yang digunakan pada penelitian ini berjumlah 45 ekor Sapi Persilangan Limousin dengan BCS 3-5 dan umur berkisar antara 1,8-7 tahun. Semen beku sexing Y merupakan hasil produksi Balai Besar Inseminasi Buatan (BBIB) Singosari menggunakan metode Sentrifugasi Densitas Gradien Percoll (SGDP). Pengamatan karakter estrus dilakukan sebelum inseminasi. Inseminasi Buatan dilakukan dengan menggunakan double dosis pada jam ke-2 dan jam ke-8 dengan teknik deep insemination. Vitamin Rheinbio sebagai BioATP+ diinjeksikan setelah IB. Hasil penelitian menunjukkan bahwa keberhasilan kebuntingan ditinjau dari nilai conception rate (CR) dan pregnancy rate (PR) berdasarkan karakter estrus ternak lebih tinggi ditunjukkan pada kondisi warna vulva merah merata yaitu 17,78% dan 22,22%; vulva yang sangat bengkak yaitu 20% dan 22,22%; banyak mengeluarkan lendir servik yaitu 15,56% dan 17,78%, serta suhu vagina yang berkisar antara 38,0-38,5oC yaitu 13,33% dan 20%. Persentase kebuntingan yang rendah pada penelitian ini disebabkan oleh berbagai faktor, utamanya kualitas genetik ternak, kematian embrio dini, manajemen pemeliharaan khususnya pakan, dan terdapat ternak yang mengalami gangguan reproduksi.

Abstract

ABSTRACT. Penelitian ini bertujuan untuk mengevaluasi pejatan sapi Madura dengan menggunakan metode pendugaan nilai pemuliaan berdasarkan bobot badan dan ukuran tubuh. Pengambilan sampel dilakukan secara Purposive sampling dengan menggunakan data recording keturunan 7 pejantan sapi (684, Adikara/160726, 685, Kelesap/160725, Muntahai/16011001, Banteng/934, 386) untuk bobot badan (BB), tinggi badan (TB), panjang badan (PB) dan lingkar dada (LD) kelahiran tahun 2014-2020. Analisis data dengan koreksi umur 1 tahun ke 365 hari, nilai heritabilitas, nilai pemuliaan, dan korelasi rangking nilai pemuliaan. Hasil analisis nilai heritabilitas bobot badan dan ukuran tubuh sapi Madura umur 1 tahun tergolong kategori sedang hingga tinggi yaitu 0,57 (BB), 0,48 (TB), 0,83 (PB), dan 0,61 (LD). Sebanyak 42,85% bobot badan dan panjang badan pejantan sapi memiliki nilai pemuliaan positif, sedangkan pada tinggi badan dan lingkar dada memiliki nilai pemuliaan positif sebesar 28,57%. Korelasi rangking nilai pemuliaan bobot badan dengan ukuran tubuh sapi Madura umur 1 tahun tergolong kategori tinggi yaitu 0,86 (BB-TB), 0,68 (BB-PB) dan 0,86 (BB- LD). Kesimpulan dari penelitian ini yaitu pejantan sapi di UPT Pembibitan dan Kesehatan Hewan Provinsi Jawa Timur yang memiliki nilai pemuliaan positif pada bobot badan dan ukuran tubuh adalah pejantan Ke lesap/160725 dan Adikara/160726 sehingga pejantan tersebut dapat digunakan sebagai tetua dalam program seleksi untuk meningkatkan mutu genetik dan menghasilkan sapi unggul.(Evaluation of sire breeding values on Madura cattle based on body weight and body measurement of their offspring at one-year-age)ABSTRAK. This study aimed to evaluate Madura cattle by using the method of estimating breeding value based on body weight and body measurement. The sampling was carried out by Purposive sampling using data recording offspring the 7 cattle (684, Adikara/160726, 685, Kelesap/160725, Muntahai/16011001, Banteng/934, 386) for Body Weight (BW), Withers Height (WH), Body Length (BL), and Chest Girth (CG) of born in 2014-2020. The data were analyzed by correction of 1 year to 365 days of age, heritability value, breeding value, and correlation ranking of breeding values. The results of the analysis of heritability values of body weight and body measurement of Madura cattle at 1 year of age rating on medium to high category were 0,57 (BW), 0,48 (WH), 0,83 (BL), and 0,61 (CG). A total of 42,85% body weight and body length of cattle at 1 year age have a positive value, while the body height and chest girth has a positive breeding value of 28,57%. The correlation between body weight and body measurement at 1-year-age Madura cattle classified as high category was 0,86 (BW-WH), 0,68 (BW-BL) and 0,86 (BW-CG). The conclusion this study the cattle at the UPT Breeding and Animal Health of East Java Province which has positive values on body weight and body measurement are the Ke lesap/160725 and Adikara/160726 males, while sires can be used as elders in the selection program to improve genetic quality and to produce superior cattle.

Abstract

ABSTRACT. Penelitian ini bertujuan mengevaluasi keberhasilan IB menggunakan metode dosis tunggal dan ganda pada sapi Madura. Penelitian menggunakan 25 ekor dengan metode IB dosis tunggal dan 25 ekor dengan metode IB dosis ganda. IB menggunakan semen beku sapi Madura yang diproduksi oleh Balai Besar Inseminasi Buatan (BBIB) Singosari, Malang. Teknik deposisi semen adalah 4+ (deep insemination) yaitu pada posisi cornua uteri selanjutnya dilakukan IB dosis tunggal (jam ke-8 setelah menunjukkan tanda-tanda berahi) dan IB dosis ganda (jam ke-2 dan ke-8 setelah menunjukkan tanda-tanda berahi). Akseptor diinjeksi dengan Bio ATP+ merek Rheinbio 10 ml per ekor dan pemberian pakan konsentrat merek Novo NC62 1 kg per ekor per hari selama tujuh hari setelah IB. Variabel penelitian meliputi Non Return Rate (NRR), Conception Rate (CR), dan Service Per Conception (S/C). Hasil penelitian menunjukkan bahwa IB dosis tunggal menghasilkan nilai NRR 1 sebesar 92%, NRR 2 sebesar 80%, CR sebesar 60%, dan S/C sebesar 1,71. Sedangkan pada IB dosis ganda menghasilkan nilai NRR 1 sebesar 92%, NRR 2 sebesar 84%, CR sebesar 68%, dan S/C sebesar 1,53. Kesimpulan penelitian adalah nilai NRR, CR, dan S/C pada sapi Madura dengan IB dosis ganda lebih baik daripada dengan IB dosis tunggal, dan IB dosis ganda dapat meningkatkan conception rate pada sapi Madura.(The success different of artificial insemination using single and double dosage methodof Madura cows)ABSTRAK. This study aims to evaluate the success of AI using methods single and double dosage plus in Madura cows. The study used 25 Madura cow using the AI method single dosage and 25 Madura cow using the AI method double dosage. AI uses frozen semen from Madura cows produced by the Singosari Center for Artificial Insemination. The semen deposition technique is 4+ (deep insemination) in the cornua uteri position then AI implemented with a single dosage (8th hours after showing signs of estrus) and double dosage (2nd and 8th hours after showing signs of estrus). The acceptors were injected with 10 ml of "Rheinbio'' brand Bio ATP+ per cow and 1 kg of concentrate "Novo NC62" brand per cow per day was given for seven days after IB. Research variables include Non Return Rate (NRR), Conception Rate (CR), and Service Per Conception (S/C). The results showed that AI of single dosage had value NRR 1 of 92%, NRR 2 of 80%, CR of 60%, and S/C of 1.71. While AI of double dosage had value NRR 1 of 92%, NRR 2 of 84%, CR of 68%, and S/C of 1.53. In conclusion, the value of NRR, CR, and S/C in Madura cows with AI double dosage was better than AI single dosage, and AI double dosage could increase the conception rate in Madura cows.

Abstract

ABSTRAK. Sexing spermatozoa bertujuan untuk mengatur jenis kelamin sesuai harapan. Salah satu metode sexing adalah dengan menggunakan Sentrifugasi Gradien Densitas Percoll. Tujuan dari penelitian ini adalah untuk mengetahui berbagai parameter motilitas menggunakan Computer-assisted Sperm Analysis dan proporsi spermatozoa X dan Y menggunakan metode Sentrifugasi Gradien Densitas Percoll dengan volume awal yang berbeda. Penelitian dilakukan di Loka Penelitian Sapi Potong Grati, Pasuruan, pada bulan Januari sampai Maret 2020. Materi yang digunakan adalah Semen Sapi Peranakan Ongole berumur 5 tahun dan bobot badan 700 kg sebanyak 3 ekor, motilitas masa 2+ dan motilitas individu 70%. Metode yang digunakan adalah eksperimental menggunakan Rancangan Acak Kelompok (RAK) dengan 3 perlakuan volume saat sexing yaitu P1= 1 ml, P2= 1,5 ml dan P3= 2 ml dengan ulangan 11 kali. Ulangan juga berfungsi sebagai kelompok (block). Hasil penelitian menunjukkan bahwa setelah dilakukan sexing dan pendinginan hingga 5oC persentase motilitas adalah P1:71,02+10,08 %; P2: 79,63+8,65 % dan P3: 83,38+6,67 %, sedangkan motilitas progresif pada P1: 47,68+8,71%; P2: 59,61+7,26 % dan P3: 62,21+6,74%. Curvilinear Velocity (VCL) pada P1: 50,97,73 m/s, P2: 55,25,03 m/s dan P3: 53,25,97%. Straight-line Velocity (VSL) pada P1: 20,88,19 m/s; P2: 21,66,02 m/s; P3: 22,15,77 m/s, sedangkan Average Path Velocity (VAP) pada P1: 30,27,82 m/s; P2: 32,56,14 m/s dan P3 : 31,56,18 m/s. Linearity (LIN) pada P1: 40,212,26%;P2: 39,110,31%;P3: 39,87,25%. Straightness (STR) pada P1: 67,211,20%; P2: 65,710,06%;P3: 67,27,92% sedangkan Wobble (WOB) pada P1: 599,49%; P2: 58,88,63% dan P3: 59,17,74%. Kesimpulan dari penelitian adalah motilitas dan motilitas progresif spermatozoa pada sampel dengan volume awal 2 ml lebih baik dibandingkan dengan sampel dengan volume awal 1 ml dan 1,5 ml.(Characteristics of Sexing Spermatozoa Motility in Ongole Cattle with Different Initial Volume) ABSTRACT. Sperm sexing is a technique of sorting a specific type of sperm cell to fertilize the egg cell. One of the sexing methods that can be used for spermatozoa sexing is percoll gradient density centrifugation. This research aims to find out various motility parameters using Computer-assisted Sperm Analysis and the proportion of spermatozoa X and Y using density gradient centrifugation method percoll with different initial volume. The study was conducted at Grati Beef Cattle Research Station, Pasuruan, from January to March 2020. Materials of the study were Semen of 3 Filial Ongole Cattles, aged 5 years, with 700 kg body weight, mass motility 2+, and individual motility 70%. The method used was experimental using a Randomized Group Design with 3 treatment volume when sexing i.e. T0= 1 ml, T1= 1.5 ml dan T2= 2 ml with repetition 11 times. The repetition also functions as groups (blocks). The results showed that after sexing and cooling up to 5oC, the percentage of motility is T0:71.02+10.08 %; T1: 79.63+8,65 % and T2: 83.38+6.67 %. Progressive motility on T0: 47.68+8.71%; T1: 59.61+7.26 % and T2: 62.21+6.74%. Curvilinear Velocity (VCL) on T0: 50.97.73 m/s, T1: 55.25.03 m/s and T2: 53.25.97%. Straight-line Velocity (VSL) on T0: 20.88.19 m/s; T1: 21.66.02 m/s; T2: 22.15.77 m/s. The Average Path Velocity (VAP) on T0: 30.27.82 m/s; T1: 32.56.14 m/s and T2 : 31.56.18 m/s. Linearity (LIN) on T0: 40.212.26%; T1: 39.110.31%;T2: 39.87.25%. Straightness (STR) on T0: 67.211.20%; T1: 65.710.06%; T2: 67.27.92%. Wobble (WOB) on T0: 599.49%; T1: 58.88.63% and T2: 59.17.74%. The conclusion of the study is the motility and progressive motility of spermatozoa in samples with an initial volume of 2 ml is better than samples with an initial volume of 1 ml and 1.5 ml.
