Abstract

The Effect of Ethyl Methane Sulphonate (EMS) on Growth and Variations of Marigold (Tagetes sp.) The aims of this research are to determine the variation of marigold (Tagetes sp) derived from seed treated with EMS and to recommend the EMS concentrations that are able to induce varietion. Seeds of marigold cv Narai Orange were soaked in water for 6 hours, followed by soaking in EMS at concentration of 0%, 0.3%, 0.6% and 0.9% for 4 hours. This study employed Randomized Complete Blok Design with 10 replicates and each replicate consisted of 10 plants. Six plants were randomly chosen for measurements. The total number of samples observed were 240 plants. Observations were made on the percentage of the growth, plant height, number of leaves, number of branches, diameter and weight of flowers. Data obtained from the observations were analyzed using Analysis of Variance (ANOVA), followed by DMRT (Duncan’s Multiple Range Test) if there is a significant difference between treatments. The EMS treatment reduced all characters observed. The EMS concentration of 0.6% showed plant that had yellow flowers. The 0.9% EMS treatment resulted in one plant with chimera, 6 dwarf plants, 2 plants with thin stems, and 1 short plant with many branches. Untreated plants did not show any variation.

Abstract

Induction of Mutation of Red Chili (Capsicum Annuum L.) Using Ethyl Methanesulfonate at Several Soaking Periods.One way to increase genetic variation is through induced mutation usingchemical mutagen. Ethyl methanesulfonate (EMS) is a chemical compound that can cause mutationsand commonly used in plant. In this study seeds of red chili were treated using EMS 1% through seedsoaking. Seeds of red chili were soaked with EMS 1% in phosphate buffer pH 7 for 6, 9, 12 and 15hours at room temperature. As control, seeds were soaked in phosphate buffer pH 7. This study aimsto evaluate, physiological and reproductive characters of plants after treated with EMS. Experimentwas conducted in an open field with 5 replicates for each treatment. Results showed that concentrationof chlorophyll a, b and total chlorophyll increased in plants derived from seed treated with EMS 1% for9 hours compared to control and other soaking periods. Soaking seeds with EMS 1% for 12 hoursincreased viability of pollen compared to control and other treatments. The first time of floweringoccurred earlier at 6 and 9 hours soaking period.

Abstract

The Lesser Sunda Islands of Indonesia are composed of small islands scattered from Bali to Timor Island. We analyzed a molecular phylogeny of Adiantum collected from Lesser Sunda Islands to reveal its phylogenetic relationships. A total of 12 species of Adiantum from this region and seven species from Java Island were collected and used in this study. Two cpDNA regions (rbcL and trnL-F) were chosen as markers and phylogenetic analyses were conducted using Neighbour-Joining (NJ) and Maximum Parsimony (MP) methods. The tree topologies reconstructed by NJ and MP from specimens used in this study and other species downloaded from GenBank are congruent in which trees are divided into five major clades. Adiantum species of Lesser Sunda Islands are not monophyletic and comprises three clades, i. e. Clade I composed of A. hispidulum group, Clade III composed of A. peruvianum group and Clade IV or A. caudatum group, each together with extra-Lesser Sunda samples. No sample from Lesser Sunda Islands examined is located in Clade II (A. tenerum group) and V (A. capillus-veneris group).

Abstract

Anggur Bali (Vitis vinifera L. cv Alphonse Lavallee) merupakan salah satu tanaman buah yang banyak dibudidayakan di Kabupaten Buleleng, Bali. Kebutuhan akan bibit anggur Bali cukup tinggi, sehingga diperlukan teknik perbanyakan yang dapat menghasilkan bibit dalam jumlah besar. Salah satu teknik yang dapat digunakan adalah kultur jaringan tanaman. Penelitian ini bertujuan menganalisis respon eksplan cabang yang masih muda terhadap zat pengatur tumbuh (ZPT) NAA dan BAP pada kultur Anggur Bali. Cabang tanaman anggur yang masih muda diambil dari salah satu perkebunan Anggur Bali di Kecamatan Seririt, Kabupaten Buleleng, Bali. Dilakukan sterilisasi eksplan dan ditanam pada medium MS dengan penambahan zat pengatur tumbuh NAA dan BAP dalam beberapa kombinasi. Pengamatan dilakukan terhadap pembentukan kalus, tunas dan akar. Hasil menunjukkan bahwa pembentukan kalus, tunas dan akar Anggur Bali dengan teknik kultur jaringan dipengaruhi oleh zat pengatur tumbuh NAA dan BAP. Persentase tertinggi kultur yang membentuk kalus terjadi pada penambahan 1,5 mg/L NAA dan 2 mg/L BAP, tetapi skor kalus yang terbentuk tertinggi pada kombinasi 1 mg/L NAA dengan 4 mg/L BAP dan kombinasi 1,5 mg/L NAA dengan 4 mg/L BAP. Tunas terbentuk pada perlakuan 4 mg/L BAP tanpa NAA dan perlakuan 1 mg/L NAA dengan 4 mg/l BAP. Persentase kultur yang membentuk akar dan jumlah akar terbanyak terjadi pada pemberian kombinasi 1 mg/L NAA dengan 2 mg/L BAP. Perubahan kecil pada konsentrasi ZPT mengakibatkan perubahan respon tanaman, sehingga penelitian lebih lanjut diperlukan untuk mendapatkan kombinasi dan konsentrasi ZPT yang tepat untuk menghasilkan plantlet dalam jumlah besar.

Abstract

Penelitian ini bertujuan untuk menganalisis pengaruh pemberian kolkisin terhadap jumlah kromosom marigold serta dapat mengetahui konsentrasi kolkisin yang efektif untuk menginduksi poliploidi pada kecambah marigold.  Kecambah diberi direndam dalam kolkisin dengan konsentrasi 0,1% dan 0,2% selama 6 jam Sebagai kontrol digunakan akuades. Parameter yang diamati adalah jumlah kromosom ujung akar marigold pada masing-masing perlakuan. Pengamatan kromosom dilakukan dengan metode squash ujung akar. Pemberian kolkisin  menyebabkan penggandaan jumlah kromosom kecambah marigold. Rata-rata jumlah kromosom kecambah marigold pada kontrol adalah 2n=2x=20,5, sementara  rata-rata jumlah kromosom yang diberi perlakuan kolkisin 0,1% dan 0,2 % berturut-turut yaitu 2n=4x=42 dan 2n=4x=43.

Abstract

The immune system is important in dealing with foreign objects that enter the body. A good body defense system is very important in the existence of the corona virus disease 19 (COVID-19) pandemic caused by the SARS-CoV2. The immune system can be improved by consuming foods with balanced nutrition and various kinds of herbs, in the form of herbal medicine. Jamu is a product derived from medicinal plants that can function as an immune booster. The government has campaigned for the planting of family medicinal plants (TOGA) as an effort to maintain health. In Budaga Village, Klungkung Regency, several families have planted medicinal plants, but the types of are still limited and the processing carried out is still very simple, namely brewing the leaves or plant rhizomes. The purpose of this activity was to introduce cultivation techniques of various kinds of medicinal plants, as well as to train member of PKK of Budaga Village in processing medicinal plants into herbal jamu immune booster. The activity was held on September 1, 2021, attended by representatives of PKK in Budaga Village. The methods used are lectures, demonstrations and direct practice of seeding and planting medicinal plants and practice of preparing jamu. The results of this activity are that the community can cultivate medicinal plants and able to prepare jamu following recipe from Kementerian Kesehatan RI. This activity produced seeds of medicinal plants ginger, red ginger, Piper betle, lemongrass, turmeric, mint and three herbal products, namely jamu gula asam, kunyit asam and temulawak. Abstract

Advances in molecular biology has brought a new branch of multidiscipline study named biotechnology. To comprehend the complex nature of concepts in biology, practical and hands-on approaches are often required. This activity was a collaboration between Biology Teachers Working Group (MGMP Biologi) in Klungkung Regency with university as the facilitator, and aimed to train senior high school biology teachers in compiling learning modules as well as supplying them with the appropriate materials required in simple practical sessions, such as those in animal physiology and reproduction, as well as biotechnology. The event was held successfully, attended by total of 20 biology teachers from six senior high schools (SMA) in Klungkung District, Klungkung Regency, started by introductory lectures and followed by practical sessions on how to perform mice vaginal and sperm smear, human blood smear, and DNA extraction of plant tissues. Abstract

This study aimed to evaluate the resolution of PCR-RFLP and sequencing of trnS/trnfM in detecting genetic diversity of seagrass Enhalus acoroides at Sanur Beach, Sindhu Beach and Semawang Beach. This research used six samples E. acoroides from each location. DNA extraction method followed Doyle and Doyle. PCR amplifications were done using primers P672/P673, P674/P675 and ITS4/ITS5 followed by digestion using restriction enzymes EcoRI, PstI, HindIII, BamHI, Rsa, Mva and HinfI. Sequencing analysis used PCR products of primers trnS/trnfM. Testing of DNA sequences of E. acoroides were conducted using BLAST (Basic Local Alignment Search Tool). DNA sequences were further analyzed using MEGA 5.2 software (Molecular Evolutionary Genetic Analysis) to evaluate variations of DNA. The sequence alignments were done using ClustalW software to determine the homology between the DNA sequences. The results showed that 18 samples of E. acoroides from Sanur, Sindhu and Semawang Beach have no polymorphism based on restriction enzyme analyses. Furthermore, sequencing of trnS/trnfM region of 18 samples E. acoroides showed that the sequences were identical.

Abstract

Body scrub cream is used to clean and nourish the skin. Several illegal creams were reported to contain harmful chemicals. Some plant extracts can replace chemical compounds because they can lighten the skin. Enhalus acoroides is one of the seagrass species that is widely distributed in Indonesia. Leaf extract of E. acoroides was found to contain phenolic, tannin, and flavonoid. Due to the bioactive compounds in the E. acoroides extract can be added to cosmetic products such as body scrub cream. This study aimed to analyze the effect of the addition of encapsulated E. acoroides extract on the characteristics of the body scrub cream and determined the concentration of the encapsulated E. acoroides extract, which produced the best body scrub cream. The extraction was done using chloroform: ethanol (9:1) and encapsulation using maltodextrin and tween. The body scrub cream was made in two phases, the oil phase and the water phase. The concentrations of the encapsulated E. acoroides extract tested were 0%, 0.5%, 1%, 1.5%, 2%, 2.5%. The characteristics evaluated were homogeneity, pH, viscosity, spreadability, stickiness, IC50, and organoleptic test for color and scent. The results showed that the body scrubs were homogenous. The pH values were 5.43-6.66, and viscosity was 34400-46400cp which are in the range of SNI for cosmetic products. The spreadability values were 4.03-5.5cm, and stickiness was 4.8-6.91sec. The antioxidant capacity (IC50) ranged from 147.38 to 157.64 ppm, and the overall acceptance scores were 5 to 6.80. Based on the effectivity index and statistical analyses of physical-chemical characteristics, the best concentration of encapsulated E. acoroides extract was 2%.  These findings indicate the potential of encapsulated E. acoroides extracts to improve skin health.

Abstract

Tanaman anggrek merupakan tanaman hias yang memiliki nilai komersial tinggi dan memiliki keunikan khas dalam estetika. Konservasi anggrek telah dilakukan di Kebun Raya Eka Karya Bali. Ada 265 anggrek yang dikumpulkan di Kebun Raya Eka Karya Bali. Koleksi anggrek membutuhkan karakterisasi morfologis dan stomata. Tujuan penelitian ini adalah untuk menganalisis karakteristik morfologi dan stomata delapan spesies anggrek yang dikumpulkan di Kebun Raya Eka Karya Bali. Karakter morfologi yang diamati meliputi karakter batang, daun, bunga dan habitat. Hasil penelitian menunjukkan bahwa dari delapan spesies anggrek, empat merupakan anggrek epifit, dan empat spesies dapat ditemukan hidup secara epifit dan terrestrial. Delapan spesies anggrek tersebut memiliki bentuk yang bervariasi secara morfologis dari ukuran daun, diameter batang, bentuk daun, warna daun, bentuk bunga, dan warna bunga. Berdasarkan karakteristik stomata, di antara delapan spesies anggrek, terdapat tiga spesies dengan tipe stomata parasitik dan lima spesies dengan tipe anomositik.
