Abstract

Tilapia (Oreochromis niloticus) bones are a fishery by-product whose use remains limited, despite containing proteins that can be processed into bioactive peptides with antioxidant activity. This study aims to evaluate the characteristics of protein hydrolysate and antioxidant activity from raw and cooked tilapia bones using the ABTS method. Enzymatic hydrolysis was performed using papain at concentrations of 4%, 6%, and 8% (b/v) at pH 7 and 55°C for 4 hours. The parameters analyzed included qualitative amino acid tests, dissolved protein levels determined by the Kjeldahl method, degree of hydrolysis based on dissolved proteins in 10% TCA, and antioxidant activity expressed as IC₅₀ values. The results showed that all samples tested positively for amino acids, including aromatic amino acids. The lowest IC₅₀ value was observed in the treatment of a ripe sample with an enzyme concentration of 8% (6.89 mg/mL), indicating the highest antioxidant activity among the treatments. This is in line with an increase in the degree of hydrolysis to 84.53% under the same conditions, as well as an increase in dissolved protein levels to up to 32.78%. These findings indicate that tilapia bone protein hydrolysate has the potential to serve as a natural source of antioxidants, particularly under optimal hydrolysis conditions.

Abstract

Penelitian ini bertujuan untuk membandingkan aktivitas antioksidan ekstrak metanol kulit batang gandaria (Bouea macrophylla Griff.) menggunakan dua metode pengujian, yaitu DPPH (2,2-diphenyl-1-picrylhydrazyl) dan ABTS (2,2′-azinobis(3-ethylbenzothiazoline-6-sulfonic acid)). Ekstraksi dilakukan secara maserasi menggunakan pelarut metanol. Aktivitas antioksidan ditentukan berdasarkan nilai IC50, yaitu konsentrasi yang diperlukan untuk meredam 50% radikal bebas. Hasil uji menunjukkan bahwa ekstrak memiliki aktivitas antioksidan sangat kuat dengan nilai IC50 sebesar 12,259 ± 0,232 ppm untuk metode DPPH dan 15,831 ± 1,550 ppm untuk metode ABTS. Nilai IC50 yang lebih rendah pada metode DPPH menunjukkan bahwa ekstrak memiliki kemampuan yang lebih tinggi dalam menangkap radikal bebas pada metode tersebut. Hasil ini menunjukkan bahwa kulit batang gandaria berpotensi sebagai sumber antioksidan alami yang efektif dengan kemampuan merangkal radikal bebas DPPH dan ABTS sangat kuat.

Abstract

The objective of this research is to investigate the influence of the Project-Based Learning (PjBL) model, when integrated with the STEM approach, on students’ collaborative abilities in the topic of atomic structure. The study is motivated by the persistent issue of low collaboration levels among chemistry learners, particularly when dealing with abstract subject matter. Employing a quantitative methodology with a quasi-experimental design—specifically the Non-Equivalent Control Group Design—the research was conducted with two tenth-grade classes at SMA Negeri 1 Suwawa, designating one as the experimental group and the other as the control group. Data were gathered through observation sheets that measured five aspects of collaboration: cooperation, responsibility, communication, adaptability, and compromise. The findings revealed a notable enhancement in collaborative skills within the experimental class following the application of the STEM-integrated PjBL model. The average final observation score for this group reached 76%, surpassing the control group’s 61%. The N-Gain score for the experimental group fell within the medium category (0.48), whereas the control group’s score was classified as low (0.17). Improvements were recorded across all collaboration indicators. Overall, the results suggest that the STEM-integrated PjBL model is effective in fostering collaboration skills and can be recommended as an alternative instructional approach for other chemistry topics to advance 21st-century skill development.

Abstract

This research aims to determine students' conceptual understanding ability of hydrocarbon material using a three-tier multiple-choice diagnostic test instrument. The research was conducted at SMA Negeri 2 Wonosari using a descriptive qualitative approach. The research was carried out in the even semester of the 2024/2025 academic year. The research subjects were the students of class XI B at SMA Negeri 2 Wonosari, with a sample size of 28 students. The instrument used consisted of 15 three-tier multiple-choice questions covering answers, reasoning, and the student's confidence levels. The validity and reliability of the instrument were tested through expert analysis and empirical testing using the Rasch model. Data collection was carried out using the three-tier multiple-choice test. After obtaining the students' responses, the answers were analyzed and categorized based on conceptual understanding levels: Understanding the Concept (UC), Negative Misconception (NM), Positive Misconception (PM), and Not Understanding the Concept (NUC). The results indicate that the average percentage of conceptual understanding was 29.05%, categorized as low; negative misconceptions were 23.10%, also categorized as low; postitive misconceptions were 10.24% categorized as very low; and not understanding the conceptions 37.62%, categorized as low. In conclusion, students of class XI B at SMA Negeri 2 Wonosari showed the lowest average level of conceptual understanding compared to those experiencing misconceptions or lacking scientific understanding of the concept.

Abstract

Diabetes mellitus remains a major global health challenge, prompting the exploration of natural products as sources of safer antidiabetic agents. This study aimed to identify bioactive compounds from the roots of Acalypha indica and evaluate their potential as inhibitors of glutamine–fructose-6-phosphate amidotransferase (GFAT), an enzyme involved in glucose metabolism. Compound profiling was conducted using liquid chromatography–high resolution mass spectrometry (LC-HRMS), while the inhibitory potential against GFAT was assessed through in silico docking. This integrated approach has not been previously reported. Metabolite profiling using LC-HRMS tentatively identified thirteen carboxylic acid derivatives, including hydroxycinnamic acid, gluconic acid, trans-aconitic acid, and glucoheptonic acid. molecular docking analysis revealed strong binding affinities of several compounds toward GFAT.  Glucoheptonic acid exhibited the lowest binding energy (−8.0 kcal/mol), followed by xylaric acid A (−7.9 kcal/mol) and trans-aconitic acid (−7.5 kcal/mol). These compounds interacted with key active-site residues Gln421, Ser420, Ser422, and Lys675, suggesting a potential enzyme inhibition mechanism. In silico pharmacokinetic analysis indicated that 2-hexylpentanedioic acid exhibited favorable ADME properties, with high gastrointestinal absorption, no major inhibition of CYP450, and low predicted acute toxicity risk. These findings highlight Acalypha indica roots as a promising source of bioactive carboxylic acid derivatives, with glucoheptonic acid emerging as the most promising candidate for further in vitro evaluation as a GFAT inhibitor

Abstract

Kegiatan pengabdian kepada masyarakat merupakan bagian integral dari Tri Dharma Perguruan Tinggi, yang melibatkan lembaga, dosen, dan mahasiswa dalam berkontribusi kepada masyarakat. Dalam konteks ini, dilakukan pengabdian melalui pengenalan lingkungan sebagai laboratorium alam yang bertujuan untuk meningkatkan pengetahuan dan minat siswa dalam mempelajari IPA. Tujuan dari kegiatan pengabdian ini adalah memberikan pengetahuan, pemahaman, kepedulian, dan minat siswa dalam mempelajari ilmu pengetahuan alam. Proses pelaksanaan kegiatan meliputi perencanaan, pelaksanaan, dan evaluasi kegiatan. Hasil yang diperoleh dari kegiatan ini antara lain adalah peningkatan minat siswa dalam belajar IPA sains dan memahami lingkungan dapat dijadikan sebagai laboratorium alam. Respon siswa menunjukkan rata-rata persentase mencapai 88,67%, Indikator dengan persentase tertinggi adalah keinginan siswa untuk mengikuti kegiatan serupa, yaitu sebesar 93,33%, diikuti oleh ketertarikan siswa terhadap pembelajaran sains sebesar 92,50%. Dengan demikian, dapat disimpulkan bahwa kegiatan pengabdian ini berhasil meningkatkan pengetahuan, sikap, dan minat siswa dalam memanfaatkan lingkungan sebagai sumber belajar

Abstract

Kitosan, biopolymer kationik alami dari deasetilasi kitin eksoskeleton hewan laut, memiliki sifat biokompatibilitas, biodegradabilitas, dan non-toksisitas yang menarik untuk berbagai aplikasi. Penelitian ini bertujuan untuk mengkarakterisasi kitosan yang diisolasi dari limbah cangkang sotong yang diperoleh dari Pantai Tanjung Kramat, Kota Gorontalo dengan focus rendemen, kadar air, dan analisis Fourier Transform Infrared (FITR). Proses isolasi melibatkan tahap demineralisasi, deproteinasi, dan deasetilasi. Hasil penelitian menunjukkan rendemen kitosan tertinggi 28% pada perlakuan HCl 5%, sedangkan kadar air seluruh sampel berkisar antara 2,00-2,21% dan memenuhi standar SNI 7949:2013 (?12%). Analisis FTIR mengidentifikasi gugus fungsi utama dan derajat deasetilasi (DD) tertinggi mencapai 80,17% pada perlakuan HCl 7%, memenuhi standar mutu (?75%). Penelitian ini memberikan wawasan penting mengenai potensi cangkang sotong sebagai sumber kitosan berkualitas.

Abstract

This study aimed to develop a Higher Order Thinking Skills (HOTS)-based formative test instrument on chemical equilibrium material and to analyze the quality of the instrument based on validity, reliability, difficulty level, discrimination power, and distractor effectiveness using the Rasch Model. This study employed a Research and Development method involving 564 students as research subjects. The developed instrument consisted of HOTS multiple-choice questions at cognitive levels C4, C5, and C6. Instrument validity was analyzed through expert validation, while empirical analysis was conducted using Winsteps and Microsoft Excel. The results showed that the instrument obtained a validity percentage of 88.95%, categorized as very valid. Rasch Model analysis indicated that all items met the item fit criteria with a Cronbach Alpha reliability value of 0.78, categorized as good. The item difficulty levels consisted of easy, moderate, and difficult categories, with moderate items being dominant. All items demonstrated good discrimination power as indicated by positive Pt Measure Corr values. In addition, most distractors functioned effectively because they were selected by students. The distribution of HOTS items consisted of 44.44% C4 level, 33.33% C5 level, and 22.22% C6 level. Therefore, the HOTS-based formative test instrument on chemical equilibrium material was considered feasible for measuring students’ higher-order thinking skills.
