Abstract

Background: A stress, sleep deprivation, can cause imbalance of reactive oxygen species (ROS) that can damage DNA, lipids and proteins of spermatozoa, consequently leading to infertility.Objective: The aim of this study is to investigate differences of morphology and DNA fragmentation with some models of sleep deprivation in spermatozoa of white male Wistar rat strain (Rattus novergicus).Methods: This study was an experimental study with post-test-only with control group design. There were 30 male Wistar rats which were randomly divided into 5 groups: control group, paradoxical sleep deprivation (PSD) group, total sleep deprivation (TSD) group, sleep recovery (SR) group, PSD group + SR group, and TSD group + SR group. Stress induction was conducted for 5 days. Morphological abnormality of spermatozoa was observed by creating sperm removal tinged by eosin and nigrosine. DNA fragmentation was performed by determining DNA fragmentation index (DFI). Morphological data of spermatozoa were analysed by One-Way Annova and Post-Hoc Turkey test, and DNA fragmentation of spermatozoa was analysed by Kruskal Wallis and Mann Whitney test to determine differences among the groups.Results: The highest mean of the abnormal morphology was in PSD Group (89.5 %, p=0.004). The normal mean of DNA fragmentation was found in control group, PSD+SR group and TSD+SR group, PSD and TSD groups. Bad DNA fragmentation index was found in PSD and TSD groups (30.16% and 31.4%, p=0.023).Conclusion: There were differences of morphology and DNA fragmentation of the spermatozoa in the male Wistar rat strain induced by various models of sleep deprivation.

Abstract

Background: Methicillin-Resistant Staphylococcus aureus (MRSA) has become a major pathogene and its incidence continues to increase in various countries. The resistance to methicillin is caused by the mecA gene that encodes the expression of Penicillin Binding Protein 2a (PBP2a) so it has a low affinity for beta-lactam antibiotics. One of the factors that can increase the incidence of MRSA is inappropriate antibiotics use. Objective: This study is to identify presence of the mecA gene in MRSA and to compare histories of inappropriate antibiotic uses of healthcare workers and communities in Banyumas.Methods: This was a cross-sectional study conducting 120 samples from nasal swabs. Isolates of MRSA tested by bacteriological examinations and PCR of the mecA genes. Histories of antibiotic uses were assessed by using questionnaires and then analysed descriptively by using Fisher Exact test (SPPS, version 20).Result: One sample of S. aureus from the microbiology examination (0.83%) was an MRSA although there was no a mecA gene identified by using the PCR method. 66.1% of the healthcare workers and 88,3% of the communities used antibiotics inappropriately. The results showed that there was a statistically significant difference in the histories of irrational uses of antibiotics (p?0.05).Conclusion: A number of the communities who used inappropriate antibiotics was higher than the health workers. Furthermore, one MRSA strain with a negative mecA gene was found in the health worker group.

Abstract

Introduction: The prevalence of autism spectrum disorders has increased in the last three decades. The genetic etiological component of ASD is quite strong and complex. Although genetic factors clearly contribute to ASD risk, environmental, prenatal and post-natal factors are also involved. Father's age and mother's age at the conception of children contribute to the occurrence of autism spectrum disorders in children.Objective: To explore the association between maternal and paternal age and risk of autis, spectrum disorders (ASD) in Banyumas district, Central Java, IndonesiaMethods: An analytical study with case control approach was conducted. This study used case control method that include 43 children with ASD as case group and 189 normal children as control group. Consecutive cases were selected over 4 months, with every weekend allocated to visit the school of children with special needs. Controls were selected from normal children who came with their parents and were also visited by our team at local schools, until the required sample size was reached.Results: This study showed that paternal and maternal age at conception onset were not associate with the occurence of asutism spectrum disorders in children based on the statistical annalyzes in every range of paternal and maternal age results p value > 0,05.Conclusion: Paternal and maternal age were not related with the occurence of autism spectrum disorders

Abstract

Latar Belakang : Kejadian penyakit genetik di Banyumas semakin meningkat. Hal ini ditandai dengan prevalensi gen pembawa penyakit thalasemia sebanyak 8% dari setiap penduduk sehat. Kejadian ini menandakan wilayah banyumas rentan terhadap penyakit genetik. Sebagian besar penyakit genetik tidak dapat disembuhkan. Tetapi dapat dicegah dengan skrining genetik dan premarital. oleh karena itu pengetahuan, sikap dan perilaku skrining genetik ini penting. Salah satu pengaruhi ketiganya ini adalah tingkat pendidikan. Sehingga penelitian ini perlu dilakukan. Tujuan: Untuk mengetahui hubungan antara tingkat pendidikan masyarakat dengan pengetahuan, sikap, dan perilaku skrining genetik dan premarital di Kabupaten Banyumas. Metode: penelitian ini menggunakan rancangan cross sectional dengan jumlah responden 400 orang. Metode penelitian yang digunakan untuk penelitian adalah purposive sampling dan pengambilan data menggunakan kuesioner hubungan antara tingkat pendidikan dengan pengetahuan, sikap, dan perilaku Skrining Genetik dan Premarital. Uji hipotesis penelitian ini dengan uji korelasi rank spearman. Hasil : Penelitian ini menunjukan terdapat hubungan yang signifikan antara tingkat pendidikan dengan pengetahuan dengan p-value (p=0,005), tidak terdapat hubungan yang signifikan antara tingkat pendidikan dengan sikap dengan p-value (p=0,454), dan tidak terdapat hubungan yang signifikan antara tingkat pendidikan dengan perilaku dengan p-value (0,927). Kesimpulan : Terdapat hubungan yang signifikan antara tingkat pendidikan terhadap pengetahuan skrining genetik dan premarital (P value<0,05) dan Tidak terdapat hubungan yang signifikan antara tingkat pendidikan dengan sikap dan perilaku skrining genetik dan premarital (P Value >0,05).

Abstract

Plasmodium knowlesi is a parasite of the genus plasmodium that naturally infects long-tailed macaques (Macaca fascicularis), but currently reported has ability to infect humans. The identification/detection of P. knowlesi can be done using RDT, microscopic, or molecular examinations using nested PCR. Nested PCR is the most sensitive and specific method of examination to date. This study aimed to detect P. knowlesi in humans by RDT, microscopic, and nested PCR examinations. The study was descriptive with a cross-sectional approach, carried out from March to July 2019. The samples in this study were 123 patients who were suspected of being infected with malaria and who underwent laboratory tests at the Muara Komam Health Center. Microscopic examination and RDT examination were carried out at the Muara Komam Health Center, while nested PCR was carried out at the Eijkman Molecular Biology Laboratory Jakarta. The results of RDT and microscopic examinations showed as many as 16 of 123 (13%) malaria-positive samples of P. falciparum and P. vivax, and 10 of 123 (8.1%) malaria-positive samples of P. falciparum and P. vivax. Nested PCR tests targeting the rRNA SSU gene were able to identify P. knowlesi by 6 out of 123 (4.87%). In conclusion, the study showed that Plasmodium knowlesi was detected in humans in Muara Komam, East Kalimantan through nested PCR examination.

Abstract

Brain tumor patients may experience stress due to changes in their health, daily life, and intense head pain. Prolonged stress can worsen their condition and cause depression, hindering their treatment process. The study consisted of two groups, each with 15 participants. However, after some withdrawals, the final sample for Group 1 was 11, and Group 2 was 7. The stress, anxiety, and depression levels were measured using the DASS 42 questionnaire before and after the intervention, which involved providing the Visiting Patient app to Group 1 and not to Group 2. Data was collected every 4 to 5 days using the same questionnaire. The app, which contains Islamic educational content on the positive aspects of illness and rewards, increases the patient's spiritual knowledge and helps them adopt a positive attitude towards their condition, reducing stress, anxiety, and depression. It allow them to cooperate better with their treatment process.

Abstract

Genetic screening is a complex procedural genetical test to understand the potential genetical risk that someone might have been possessed. Screening procedure which is conducted before a couple got married is called premarital screening. Screening behavior is influenced by many factors, including predisposition, enabling, and reinforcing factors. As a healthcare provider, medical and health students must support the personal medicine concept by increasing their knowledge, attitude, and behavior. The aim of this research is to understand the relationship between knowledge level with attitude, knowledge level with behavior, and attitude with behavior among medical and health students in genetic and premarital screening. A cross-sectional study was conducted among 379 medical and health students at their first, second, and third-year (2020, 2019, and 2018) study in Banyumas Regency, selected with proportional random sampling technique. Datas were collected using Knowledge, Attitude, and Behavior in Genetic and Premarital Screening among Medical and Health Students in Banyumas Regency Questionnaire. The hypothesis was analyzed using Fisher correlation. The p-value of the Fisher correlation analysis between knowledge and attitude (p = 0,060), knowledge and behavior (p = 1,000), and attitude and behavior (p = 1,000). Participants knowledge level are at good category (75,46%) and have positive attitude (99,47%). Most of the participants’ behavior had never done genetic or premarital screening (99,47%). There was no relationship between knowledge level and attitude, knowledge level and behavior, and also attitude and behavior (p<0,05).
