Abstract

The increasing resistance of pathogenic microbes to antibiotics is a major public health concern, necessitating the discovery of effective antimicrobial compounds. The aim of this study was to assess the bioactive metabolites produced by Aspergillus fumigatus LBKURCC269 and Bacillus paramycoides LBKURCC218 under three fermentation conditions: monoculture of each microorganism and their co-culture. Metabolite analyses initiated with gas chromatography-mass spectrometry (GC-MS) and liquid chromatography-high-resolution mass spectrometry (LC-HRMS) followed with molecular networking–Global Natural Products Social Molecular Networking (GNPS) and molecular docking. Antimicrobial activity of the extracts was then conducted. Metabolite analysis using GC-MS identified key antimicrobial compounds, including 2,6-bis(1,1-dimethylethyl)-4-methylphenol, pentadecanoic acid, cyclopropane pentanoic acid, and 3-piperidinol. LC-HRMS, combined with multivariate analysis and GNPS molecular networking, revealed additional antimicrobial compounds, including novel pyrazine derivatives induced in co-culture fermentation. Molecular docking analysis of 3-(propan-2-yl)-octahydropyrrolo[1,2-a]pyrazine-1,4-dione demonstrated its potential as an antimicrobial agent by inhibiting topoisomerase IV and cytochrome monooxygenase with binding affinity of -5.34 kcal/mol and -5.6 kcal/mol, respectively. The antimicrobial assays showed that the co-culture fermentation extract had the strongest activity, with inhibition zones of 20.33±0.59 mm (Escherichia coli), 14.33±0.59 mm (Staphylococcus aureus), and 25.67±0.59 mm (Candida albicans). This study highlights the advantages of co-culture fermentation in enhancing the discovery of antimicrobial compounds. The findings underscore the potential of this approach to simplify chemical isolation and accelerate the identification of novel antimicrobial agents for pharmaceutical development.

Abstract

Background: Haemodialysis is often accompanied by shivering, especially in patients with catheter access, which can reduce comfort, impair treatment efficacy, increase the risk of complications and contribute to inadequate dialysis. Addressing this issue through non-pharmacological means, such as a warming gown, offers a potentially effective, safe, and economical solution to improve patient outcomes. Purpose: To develop a warming gown as an innovation to reduce the incidence of shivering in chronic kidney disease patients undergoing haemodialysis with a haemodialysis catheter. Methods: This study employed a two-stage Research and Development design. In the first stage, a reusable, adaptive warming gown for HD catheter patients was developed and validated (S-CVI/Ave = 0.99). Second, a quasi-experiment was conducted with 60 patients recruited through total sampling. Subsequently, patients were randomly allocated to either the intervention group (warming gown) or the control group (blanket) using computer-generated randomisation based on their identification numbers. Shivering (Crossley and Mahajan scale) and body temperature (digital thermometer) were measured at 0, 15, 30, 60, and 120 minutes. Data were analysed using Wilcoxon, Friedman, and Bonferroni-corrected repeated Mann–Whitney tests. Results: The intervention group showed a significant reduction in shivering levels from 2.63 ± 1.27 to 0.37 ± 0.49 (p < 0.001) and an increase in body temperature from 36.36 ± 0.52 to 36.84 ± 0.29 (p < 0.001). In contrast, the control group showed no significant changes (p > 0.05). Conclusions: The warming gown was proven effective in reducing shivering and increasing body temperature in haemodialysis patients, offering advantages in comfort, safety, and cost efficiency.

Abstract

ABSTRACT Headaches are one of the most common medical complaints in the world, affecting the quality of life and productivity of individuals. One of the main causes of headaches is impaired brain tissue perfusion. Non-pharmacological interventions, such as acupressure, are becoming increasingly popular, but there are still few studies explaining the underlying physiological mechanisms. to evaluate the effectiveness of acupressure in improving tissue perfusion and reducing headaches, as well as to identify the underlying physiological mechanisms and relevant acupressure points in headache treatment. This study uses a literature review approach with the PRISMA protocol. Data were collected from electronic databases such as PubMed, Google Scholar, and ScienceDirect with a publication range of 2019–2024. The selected articles include experimental studies, clinical trials, and literature reviews related to acupressure and brain tissue perfusion. Nine articles that met the inclusion criteria showed that acupressure at specific points, such as Tai Yang, He Gu (LI4), and Feng Chi (GB20), effectively increases brain tissue perfusion. The mechanism of acupressure involves the release of nitric oxide as a vasodilator, stimulation of the autonomic nervous system, and the release of endorphins. The research results also noted a reduction in pain intensity, headache frequency, and an increase in cerebral blood flow. Acupressure is an effective and cost-efficient non-pharmacological intervention for alleviating headaches and enhancing cerebral tissue perfusion. This intervention has great potential to be integrated into modern nursing practice and healthcare services. Keywords: Acupressure, Brain Perfusion, Headache, Acupressure Points, Migraine.  ABSTRAK Nyeri kepala merupakan salah satu keluhan medis paling umum di dunia, yang memengaruhi kualitas hidup dan produktivitas individu. Abstract

Fermentation can be an effective step in addressing postharvest handling challenges and enhancing nutritional value through the conversion of perishable crops into more stable products. One of the traditional fermented foods originating from Kuantan Singingi Regency, Riau Province, Indonesia, is cangkuok semaung (CS). This food is prepared by fermenting all fruit components of the semaung (Pangium edule). This research aimed to isolate and identify lactic acid bacteria (LAB) from CS and investigate their antimicrobial properties. The bacteria were isolated from the food, purified using the quadrant streak method on the De Man–Rogosa–Sharpe agar, and continued with morphological analysis and antimicrobial activities evaluation. The findings of this study revealed that 45 isolates were gram-positive bacteria identified as Lactobacillus sp. Among the LAB isolates, the highest antimicrobial activity against Escherichia coli, Staphylococcus aureus, Acinetobacter baumannii, and Klebsiella pneumoniae were shown by isolates of S12, S14, S27, and S24, respectively. These results suggest that LAB from CS, especially isolates S12 and S14, have promising antimicrobial properties and show potential as probiotic candidates that merit further investigation into possible applications in food and health sectors.

Abstract

Atherosclerosis in diabetes mellitus (DM) patients is prevented by using inulin to increase glucose metabolism and reduce cholesterol synthesis. Therefore, this research aimed to analyze dahlia tuber inulin extract to determine the effect on coronary artery histopathology in type 2 DM rats. A total of 20 male Wistar rats were divided into five groups, namely negative control without treatment, DM, DM + 0.5 mg/gBW/day inulin extract, DM + 1 mg/gBW/day inulin extract, and DM + 1.5 mg/gBW/day inulin extract. Furthermore, DM induction was carried out by administering 60 mg/kgBB streptozotocin and 120 mg/kgBB nicotinamide. The results of coronary artery histopathology readings showed that the highest and lowest scores of atherosclerosis were in Groups II (0,91±0,17) and V (0,08±0,1), respectively. Data analysis using Kruskal Wallis test reported significant results (p<0.05), while Mann-Whitney test showed differences between control and DM, DM, and inulin groups (III, IV, V). In this context, the administration of dahlia tuber inulin extract to rats induced by Type 2 DM prevented atherosclerosis in histopathological image of coronary artery.

Abstract

As the prevalence of type 2 diabetes mellitus continues to increase over the years, research into antidiabetic drugs needs to continue. Probiotics are non-pathogenic microorganisms that have potential as α-glucosidase inhibitors. This study aimed to characterize and determine the α-glucosidase inhibitor activity of LBSU9 isolate, a probiotic isolated from "trites," a traditional food of North Sumatra. The results showed that LBSU9 had bacillus morphology, Gram positive, negative catalase test, TSIA (Triple Sugar Iron Agar) A/A test and non-hemolysis. LBSU9 had good tolerance to simulated gastric acid and bile salts, with growth percentages of 66.54% and 64.74%. LBSU9 also has potent antimicrobial activity against S.aureus and E.coli, with 20 mm and 17 mm inhibition zones, respectively. The alpha-glucosidase inhibitor activity of LBSU9 was 98.4% greater than that of acarbose, which was 97%. Based on the results of this study, it is concluded that LBSU9 isolate has the potential to be a complementary therapy to prevent or treat type 2 diabetes mellitus

Abstract

Introduction: Improving pathophysiology knowledge and critical thinking skills of nursing students remains as a challenge faced by nursing educators. Thus, an innovative learning method is required. Concept map by adding cases might assist increasing students' pathophysiology knowledge and critical thinking skills. Methods: A quasi-experimental study with pre- and post-test with control group design involved 104 first-year nursing students of Universitas Jenderal Soedirman, Purwokerto, Central Java in April-June 2023. Participants were divided into two classes at the beginning of the first semester and randomly allocated into the intervention (54 respondents) and the control (50 respondents) group. The intervention group was provided typical learning added with a case-based concept map development weekly for 7 weeks, while the control group underwent regular lectures based on the syllabus. Pathophysiology knowledge was measured by using Multiple Choice Questions, while critical thinking was evaluated by using the Critical Thinking Disposition Inventory questionnaire. Independent-t test and paired test were applied to evaluate the effectiveness of implementing the case-based concept map. Results: The mean score for knowledge the pathophysiology of the disease in the intervention group was significantly higher than the those in the control group (t=5.24, p=0.000). However, there was no difference in the average scores of critical thinking between two groups discovered in the study (t=0.90, p=0.37). Conclusions: Case-based concept map can be recommended as an innovative and effective learning method for teaching disease pathophysiology to nursing students. However, the case-based concept map in this study is still not consistent in improving students' critical thinking skills.
