Abstract

This study was conducted to evaluate the success rate of non-penetrative pre slaughter stunning (NPPSS) and the factors that influence it in 460 Australian Brahman cross cattle. Observations were made on the handling of cattle, the implementation of NPPSS, the slaughtering process until the animal was declared dead. The results showed that the stunning success rate of NPPSS (SSR) was 74.35%. Ordinal regression analysis of the six observed parameters, three parameters have a significant influence on SSR: shooting placement area (ASP), shooting placement distance (DSP), and the presence of frontal and nuchal eminence (FE, NE). The ASP at the point of the cross line between two lines from the center of the dorsal eye to the center of the contralateral horn base, DSP at a low position (DSP<3 cm), and presence of FE gave a relatively low of SSR. Thus, it can be concluded that the SSR of the use of NPPSS in Indonesia is relatively low and is influenced by ASP, DSP, and the presence of FE and NE.

Abstract

Global demand for the water monitor’s skin, Varanus salvator, has made it a valuable wildlife commodity. Leathercraft manufacturing must consider not only beauty but also the strength and flexibility of the leather, which is determined by its structure in the skin. Therefore, this study analyzed and evaluated the fiber type characteristic of the water monitor’s skin. Skin samples were collected from 10 Sumatra water monitors with a Snout-Vent Length size of 39-89 cm and were divided into small (39-59 cm) and large (60-89 cm) groups. The skins from the dorsocervical, lumbosacral, and ventral regions were proceeded for histological sections. Histochemical approaches utilized were Hematoxylin Eosin, Picrosirius Red, and Elastin Verhoeff’s Hematoxylin staining methods. Thick fibers are the main component in the skin, ranging from 69-73%, respectively, while thin fibers varied greatly and were observed predominantly in the reticular dermis. Fiber size in the reticular dermis of small lizards was lower than that of larger ones. Elastic fibers were observed abundantly at the border of the reticular dermis and subcutaneous layer in both small and large lizards. Moreover, the skin of the small-sized lizard also has a lower morphometric than that of a large-sized lizard, both in thickness and fiber type percentage. Therefore, the skin of small-sized lizards was considered less tough than that of large-sized lizards.

Abstract

Abstract

The Sailfin lizard is a lizard that has a semi-aquatic life and has a sail fin on its tail. These lizards can be found in habitats close to water such as rivers, lakes, and estuaries of mangrove forests. This study aimed to observe the morphofunctional characteristic of the appendicular skeleton of Sailfin Lizard (Hydrosaurus amboinensis), associated with their function and behavior. The appendicular skeleton of the Sailfin lizard was studied morphofunctional by observing and measuring the bones that make up the extremities. The naming of bones and their parts is based on the Nomina Anatomica Veterinaria 2017 and other journals. The clavicle and interclavicle bones are located anterior to the scapula coracoid which has a convex shape. The patella ulnar bone is found at the humeroulnar joint and the radius et ulna bones have a relatively long antebrachial interosseous space. On the hind legs, the coxal bone has a relatively broad acetabulum, two lunula bones and patellar mineralization are found at the craniodorsal of the femorotibiofibular joint, and tibia et fibula bones have a relatively long crural interosseous space. The palmar part of the carpal bone is found in the palmar sesamoid bone, while the tarsi bones have a fused astragalus and calcaneus bones to become astragalocalcaneus bones. The pedis skeleton's size is relatively long compared to the manus skeleton and has a different middle phalanx bone. The extremity skeletal structure allows the Sailfin Lizard to do movements such as walking, running, climbing, running on water, and swimming.Key words: appendicular skeleton; morphofunction; Sailfin lizard.

Abstract

The present study aimed to standardize cardiothoracic ratio (CTR) in clinically healthy Sunda porcupine. This study was an explorative study using four Sunda porcupines, male and female, with an estimated age of 6 months-2years. The sampled animals were radiographed in ventrodorsally projection using a digital X-ray machine with 50 kVp and 6 mAs. Each animal was injected with xylazine as sedation agent at 2 mg/kg BW, intramuscularly route in the dorsal part of animals tail. The radiogram showed that Sunda porcupine heart tends to be round and more massive, characterized as the convex edge of the heart. The cardiothoracic ratio (CTR) showed that the ratio of the broadest expansion of the heart relative to the thoracic width in a ventrodorsally projection ranged from 0.48-0.55 with an average 0.52. Standard heart size was less than half of the largest thoracic diameter (normal CTR0.5). Sunda porcupine heart tends to be round and larger characterized as the convex edge of the heart. The CTR values obtained were 0.48-0.55.

Abstract

The presence of glycoconjugates in caput, corpus, and cauda epididymis of the Javan muntjacs was performed using lectin histochemical method. The epididymis organs were obtained from two adult male Javan muntjacs in hard antler stage and then immediately fixed in Bouins solution and processed in histological manner. Six types of biotinylated lectins namely Peanut agglutinin (PNA), Soybean agglutinin (SBA), Wheat germ agglutinin (WGA), Ricinus communis agglutinin (RCA), Concanavalin A (Con A), and Ulex europaeus agglutinin I (UEA I) were applied to detect the specific glycoconjugates in the epididymal tissue of muntjacs. The result showed the presence of glycoconjugates were detected by five types of lectins except UEA I in caput, corpus, and cauda epididymis with specific distribution and also the intensity of lectins binding. Glycoconjugates -galactose, -glucose, mannose, N-acetyl-galactosamine, N-acetyl-glucosamine, and sialic acid were stained with various intensity in epithelial cells, basal cells, stereocilia, spermatozoa, and stroma of caput, corpus, and cauda epididymis. The presence of sialic acid on the surface of spermatozoa detected by WGA are thought to be play a role on sperm protection during their maturation and transit along epididymal lumen. In conclusion, the difference of glycoconjugates distribution pattern and lectin binding in the epididymal caput, corpus, and cauda of the Javan muntjac indicates the regional differences in epididymal function in spermatozoa maturation.

Abstract

The morphological characteristics of the stomach and esophagus of five adults bandicoot (Echymipera kalubu) with average 1.160.29 kg in body weight and 38.24.76 cm in body length were studied macroscopically, microscopically and histochemically. The bandicoot esophagus were found relatively long about one third of body length. The esophageal glands were identified along the esophagus and the number decreased gradually from cranial to caudal region. In addition of smooth muscle, skeletal muscles were also examined in the external muscle layer of esophagus up to the esophageal junction. The stomach of the E. Kalubu had short lesser curvature and three glandular regions of cardiac, fundic and pyloric glands were observed, respectively. The cardiac glands area was small and concentrated in the cranial part of the stomach. The fundic glands area occupied about half parts of the stomach. Mucous surface epithelial cells, mucous neck cells, parietal cells and chief cells were found in the entire fundic gland. The parietal cells were the most abundant cells in the major curvature distributed from the basal to neck area of the glands, but few in the minor curvature. Chief cells were mostly distributed in the basal gland. The pyloric glands region was observed in caudal part of the stomach. Staining with alcian blue-periodic acid Schift (AB-PAS) showed various distribution and intensity of neutral and acid mucopolysaccharides in the esophageal and stomach mucosa. High concentration of neutral and acid mucopolysaccharides were detected in esophageal and stomach glands with various concentration. Morphological characteristics of the esophagus and stomach were assumed to be related to the digestive processes in the gastrointestinal tract of bandicoot.

Abstract

The objective of this study was to identify the type of specific glycoconjugates and its distribution in testicular spermatogenic cells in muntjak (Muntiacus muntjak muntjak) based on lectins histochemistry. An adult male muntjak aged 4-5 years old in hard antler period was used in this study. Testicular tissue was fixed in Bouin solution and processed histologically. Histochemistry method was performed using six types biotinylated lectins such as peanut agglutinin (PNA), soybean agglutinin (SBA), wheat germ agglutinin (WGA), ricinus communis agglutinin (RCA), concanavalin A (Con A), and ulex europaeus agglutinin I (UEA I) with 20 g/ml of concentration for PNA lectins and 15g/ml for other type of lectins. The results showed that glycoconjugates were detected by all type of lectins except UEA I in testicular spermatogenic cells with variation in distribution pattern and also the intensity of lectins binding. Glycoconjugates -galactose, -glucose, mannose, Nacetylgalactosamine, N-acetylglucosamine and sialic acid were stained intensely by lectins in golgy-cap phase and acrosomal phase of spermatids. Glycoconjugate N-acetylgalactosamine was the sugar residues which distributed abundantly that marked by positive reaction with PNA, SBA, and RCA lectins. In conclusion, glycoconjugates are detected in testicular spermatids cells of muntjak indicated that glycoconjugates have an important role in spermatogenesis particularly in spermiogenesis.Key words: glycoconjugates, lectins, spermatid, spermatozoa, muntjak
