Abstract

Sunda porcupine (Hystrix javanica) is one of the endemic porcupine species in Indonesia. The importance of wild species preservation brings with it the requirement for more basic aspects of anatomy. The knowledge of the normal radiological anatomy of the lung is of crucial importance for the making of a correct diagnosis of lung diseases. This study aimed to provide information about the radiographic anatomy in the lung lobation of sunda porcupine. This study was an explorative study using four sunda porcupines. Thoracic radiograph was retrieved using conventional and digital X-ray machine. Standard projections for thoracic radiography used lateral and ventrodorsal projection. The evaluation result of thorax radiogram showed that interlobar fissures separate the different lobes. The porcupine right lungs consist of four lobes, i.e. cranial lobe (cranial and caudal segment), middle lobe, caudal lobe, and accessory lobe, while left lung consisted of only two lobes, i.e. cranial lobe (cranial and caudal segment) and caudal lobe.

Abstract

The aim of this study was to evaluate the concentration, progressively motile, and percent live sperm in the various regions of epididymis and ductus deferens after preservation at 4oC. Epididymis and ductus deferens were collected from 21 epididymis and ductus deferens of domesticated cat (Felis catus) by castration. One testicle of pair (control testicle) was analyzed at the day of castration, while the other testicle of the pair was stored at 4oC up to 7 days. The sperm concentration, percentage of sperm motility and live sperm were examined daily until day-7 of preservation. The sperm concentration was higher (p<0.05) in cauda epididymis (23.99x106 sperm/ml) and ductus deferens (25.42x106 sperm/ml) than caput (11.51x106 sperm/ml) and corpus epididymis (14.82x106 sperm/ml). The percentage of sperm motility and live sperm decreased (p<0.05) during preservation period. However, the percentage of motile (11.33 to 16.00 %) and live (15.05 to 20.20 %) sperm could be found in preserved epididymis and ductus deferens up to day-7. These results show that motile and live sperm can be collected from cat’s epididymis and ductus deferens up to day 7 after preservation at 4oC.

Abstract

Hematology value is important to evaluate individual health status. This study was purposed to observe the hematological values of male and female Cynopterus titthaecheilus. The study was conducted on 16 C. titthaecheilus (six adult males, six adult females and four juveniles) to study the haematological profiles. Blood samples were taken using tuberculin needles and analyzed by hematology analyzer for hematological values. Females C. titthaecheilus had a higher number of white blood cells than the males, but no differences in the white blood cells differential. Males C. titthaecheilus had a higher number of platelets than the females. Generally, the juveniles had a higher number of leucocytes as well as number of platelets than the adults. Based on the stress index value, it was concluded that young bats had a lower stress level compared to adult bats.

Abstract

Alpha tocopherol, as the most active component of Vitamine E, is identified as antioxidant present in some parts of the skin and the concentration is vary in several regions of the body due to the skin characteristic of the region. Sunda porcupine (Hystrix javanica) is one of the rodents in Indonesia with quill as its spesific skin characteristic. The porcupine quill are distributed in various shapes and sizes in body parts, while dorsal regions and the tail is the body part that has the most developed quill form. This study aimed to measure alpha tocopherol content in the dorsal region skin of male and female Sunda porcupine. The study used skin samples of thoracodorsal and lumbosacral regions of 6 adult Sunda porcupine (3 males and 3 females) and 2 frozen specimens (1 male and 1 female). Samples were prepared and processed for analysis through high performance liquid chromatography. The results showed that the level of tocopherol in the dorsal thoracic region of males and females was much higher than that of the lumbosacral region. The results of this study indicate the characteristics of the skin region affect the levels of alpha tocopherol in the region.

Abstract

Antioxidants and free radicals have long been known to be the main factors in the occurrence of degenerative diseases. Various studies related to antioxidants and free radicals which have implications for oxidative stress have increased in the last decade. Knowledge of stress oxidative physiology in various animals help in understanding the pathophysiology of diseases associated with oxidative stress. Bats are claimed to be the best known animals in term of survival compared to other mammals. Bats are reported to produce low reactive oxygen species (ROS) but high endogenous antioxidants that can prevent oxidative stress. Bats high defense against oxidative stress has implications for their extreme longevity, the role as a reservoir of viruses, and the potential as experimental animals.

Abstract

One of the factorthat influencethe process offertilization on Echymiperakalubu is the quality of the spermwhich produced by males. The ability of spermtofertilize anovumis not only owned by the spermatozoa which derived from the results of ejaculation, butis also owned by spermatozoa from the caudaepididymis.This study aimed to determine sperm concentration and motility from caudaepididymis.Twenty-one males E. kalubuwere used which was 370-1500g. Based onthis weight, it was divided into three groupings, they were prepubertal (370-640 g), young adult (>640-850g) and adult (>850g).The results showed that the sperm concentration of the prepubertal was1.75±1.14x106/ml,the young adult was 8.86±4.21x106/ml and increased to12.56±6.33x106/mlin the adult group.Sperm motility also increased with increasing maturity. The prepuberta lgroup showed that sperm motility was33.33±25.17%, increased to41.67±12.58% (young adult) and3.54±57.50% followed by the adult.

Abstract

Landak jawa (Hystrix javanica) memiliki keunikan pada rambutnya yang termodifikasi menjadi duri. Duri pada landak berperan sebagai pertahanan diri dari predator. Penanganan dan pengekangan landak cukup sulit dilakukan sehingga pengambilan data biologi ladak jawa menjadi terbatas. Penanganan dan pengekangan secara kimiawi menggunakan sediaan bius umum digunakan pada hewan liar. Studi ini menggambarkan metode penanganan dan pengekangan fisik untuk pengambilan contoh biologi secara noninvasif pada landak jawa (H. javanica). Penggunaan kandang jepit yang dimodifikasi dengan penambahan sekat bergerak berbahan logam pada bagian dalam efektif untuk penanganan dan pengekangan landak jawa. Data biologi dari landak jawa yang dapat diambil untuk menilai status reproduksi dengan metode tersebut meliputi gambaran sel epitel vagina dan suhu tubuh.

Abstract

This study was conducted to investigate the histological characteristic, type, and distribution of connective tissue in Sunda porcupine skin. The investigation was carried out in three adult of sunda porcupines at microscopic level using hematoxylin eosin, Masson thrichrome, Verhoeffs van Gieson, alcian blue pH 2.5 and periodic acid Schiff staining methods. Skin consists of epidermis, dermis hypodermis, and subcutaneous muscle. Quill follicles were the main and dominant structure as well as the specific characteristic on Sunda porcupine skin. The connective tissue was distributed well in basal membrane, dermis, quill follicle, and hypodermis with various intensity and density. The collagen was the main fiber found in the skin while the elastin fiber was not observed. The acid carbohydrate was found distributed well in the skin while the neutral carbohydrate was not detected in this study. In addition the fibers of connective tissue associated with the adipose tissue which found plentifully in quill follicles and hypodermis. The present results showed that the wide distribution of connective tissue might have an important role on the wound healing physiology of Sunda porcupine skin.

Abstract

This study utilized conventional and immunohistochemical methods, to describe the morphology and distribution of glucagon and insulin cells in the pancreas of adult male babirusa (Babyrousa babyrussa). The results of research showed that the endocrine portion (Langerhans islet) spread evenly on the acinar portion of pancreas. Observation using immunohistochemical method showed that glucagon cells were distributed on the perifer, whereas insulin cells in the center of the Langerhans islet of pancreas. In general, the number of insulin cells was higher than glucagon cells. It is interesting to note, in the pancreas babirusa also found that neonatal elements similar to the Langerhans islet, but have larger sizes (ranging between 200 -700 mm). The picture is similar to the large Langerhans islet was reported in ruminant.  These cells give a negative reaction to insulin and glucagon antisera.

Abstract

The presence of glycoconjugates in caput, corpus, and cauda epididymis of the Javan muntjacs was performed using lectin histochemical method. The epididymis organs were obtained from two adult male Javan muntjacs in hard antler stage and then immediately fixed in Bouin’s solution and processed in histological manner. Six types of biotinylated lectins namely Peanut agglutinin (PNA), Soybean agglutinin (SBA), Wheat germ agglutinin (WGA), Ricinus communis agglutinin (RCA), Concanavalin A (Con A), and Ulex europaeus agglutinin I (UEA I) were applied to detect the specific glycoconjugates in the epididymal tissue of muntjacs. The result showed the presence of glycoconjugates were detected by five types of lectins except UEA I in caput, corpus, and cauda epididymis with specific distribution and also the intensity of lectins binding. Glycoconjugates β-galactose, β-glucose, mannose, N-acetyl-galactosamine, N-acetyl-glucosamine, and sialic acid were stained with various intensity in epithelial cells, basal cells, stereocilia, spermatozoa, and stroma of caput, corpus, and cauda epididymis. The presence of sialic acid on the surface of spermatozoa detected by WGA are thought to be play a role on sperm protection during their maturation and transit along epididymal lumen. In conclusion, the difference of glycoconjugates distribution pattern and lectin binding in the epididymal caput, corpus, and cauda of the Javan muntjac indicates the regional differences in epididymal function in spermatozoa maturation.
