Abstract

Diabetic foot ulcer (DFU) has been associated with genetic and environmental factors, which could potentially have a role in DFU development. Single nucleotide polymorphisms (SNPs) in genes linked to DFU, including inflammation. Bibliometric studies on the SNP on genes affecting DFU still have not been evaluated. This study aims to depict bibliographically and understand the topic trend of genetic polymorphism and the risk of DFU publications. A bibliometric methodology was applied in this study. The data were extracted through the Scopus database from 2011 to 2021. VOS viewer was used to classify and summarize Scopus articles. The 35 articles were evaluated. India topped the list of countries with the most publications, and Tehran University of Medical Sciences was the primary institution. Singh K and his team were the first contributing authors with 44 citations. Keywords analysis indicated that the research hotspots were DFU, type 2 diabetes mellitus (T2DM), diabetic foot ulcers, polymorphisms, hypoxia, vascular endothelial growth factor (VEGF), diabetic foot, diabetes mellitus, and oxidative stress. This study summarizes the current state, trends in genetic polymorphism and risk with DFU research. It may provide researchers with insight into the genetic polymorphism and risk associated with DFU research, as well as useful information for identifying possible collaborators and partner institutions.

Abstract

Introduction. The biological cause of osteoporosis, a metabolic bone disease, is osteoclastic bone resorption that is not offset by osteoblastic bone synthesis. Fractures become more likely as a result of the bones being brittle and weak. Common genetic variants that indicate hereditary susceptibility factors to osteoporosis in the general population, as well as mutations affecting specific genes that cause uncommon monogenic causes of osteoporosis, are the two main types of osteoporosis. Bone defects can now be caused by numerous additional genes. In this study, we aimed to identify variants of this pathogen across continents using genome-based and bioinformatics methodologies. Methods. We integrated osteoporosis-associated variants into this study using various bioinformatics-based techniques by using GWAS data from the National Human Genome Research Institute (NHGRI). Results. We found that the variant rs3742909 is likely to cause osteoporosis. SMOC1 gene expression in whole blood tissue also appears to be affected by this variant. We found that this genomic variant requires additional research to validate functional and clinical studies in patients with osteoporosis. Conclusions. We suggest that better understanding of disease susceptibility, including osteoporosis, can be achieved through the merging of genome-based databases and bioinformatics. Our goal is to validate the findings of this study both in vitro and in vivo during the preclinical stage.

Abstract

Posyandu is the front line in public health services, especially for mothers, children, and the elderly at the village level. However, along with the development of the era and the needs of the community, there are significant challenges related to the limitations of the tools and the effectiveness of the services provided. This study aims to identify and implement innovations in improving tools and improving services at the Aster Posyandu located in Srihardono, Pundong, Bantul, Special Region of Yogyakarta (DIY). Through a participatory approach, this community service activity involves Posyandu cadres and the local community in the process of evaluating service management, improving, and training in the use of digital health tools. The results of this intervention showed a significant increase in the quality of service, especially in terms of measurement accuracy, service time efficiency, and community satisfaction. In addition, innovations in the management system and service flow have also succeeded in reducing queues and accelerating the process of recording health data. Improvements in tools and service innovations at Posyandu have a positive impact on increasing health independence, as well as strengthening the role of Posyandu as the spearhead of health services at the village level.

Abstract

A bibliometric analysis was conducted to assess the interest and significance of research on the Health Belief Model. The bibliographical information provides insights into the Health Belief Model concerning parents of children receiving the COVID-19 vaccine. As of January 25, 2023, data for the study were obtained from the Scopus database. Our analysis identified 876 articles with the keywords: Health Belief Model, Parents, and COVID-19 Vaccine. Articles were exclusively selected in English, and the analysis was performed using the R Studio 4.2.2 application for Bibliometric Analysis. China hosts the top five most productive authors, with Peking University contributing the highest number of articles (66 in total). The Vaccines Journal emerges as the primary publisher, while the majority of collaborative research on COVID-19 involves authors from the United States. Frequently used keywords include Human, Female, Male, Adult, COVID-19, and Article. The increasing trend in Health Belief Model articles related to the COVID-19 vaccine from 2020 to 2023 underscores the widespread interest in and appeal of this subject for discussion. Given the study's limitation, including only publications released before January 25, 2023, it is plausible that some more recent publications were unintentionally overlooked.

Abstract

Penyakit Hashimoto tiroiditis adalah penyakit autoimun di mana sistem kekebalan tubuh menyerang kelenjar tiroid, menyebabkan peradangan dan kerusakan pada jaringan tiroid. Proses peradangan pada tiroiditis Hashimoto (TH) dipahami sebagai faktor risiko potensial untuk perkembangan kanker tiroid. Sampai saat ini, banyak kemajuan telah dibuat dalam pengetahuan dan pemahaman tentang TH. Namun demikian, mekanisme yang menyebabkan rusaknya toleransi sistem kekebalan tubuh, dengan akibat respon autoimun terhadap kelenjar tiroid dan timbulnya penyakit, masih belum jelas. Pada penelitian ini dengan pendekatan bioinformatika yang kami akses di laboratorium bioinformatika tanggal 5 Juli 2023 menggunakan aplikasi GWAS, Haploreg v4.2, dan GTeX Portal, kami mengidentifikasi varian gen yang berperan dalam TH dan ekspresi gen tersebut dalam jaringan tubuh. Hasilnya diperoleh SNPs rs3184504 yang bersifar missense dengan variasi gen SH2B3. Hasil ekspresi gen SH2B3 dengan menggunakan GTEex Portal di 10 besar jaringan tubuh yaitu pada jaringan limpa, limfoma yang ditransformasikan sel-ebv, paru-paru, fibroblas yang dikultur sel, adiposa-subkutan, adiposa-visceral (omentum), arteri-koroner, tiroid, jaringan payudara-mammae, dan arteri-aorta. Hasil ekspresi SH2B3 di kelenjar tiroid lebih besar pada pria dan wanita. Selain itu dengan menggunakan aplikasi Ensembl kami juga berhasil mengidentifikasi pola sebaran SNPs rs3184504 di beberapa benua yaitu tertinggi (100%) di Asia Timur, selanjutnya di Afrika (98%), Asia Selatan (93%), Amerika (75%) dan terendah di benua Eropa (54%).

Abstract

The prevalence of hypertension in Indonesia is still quite high. One of the causes of the high incidence of hypertension is patient’s nonadherence in their treatment, such as forgetting to take medication. The patient's medication adherence determines the success of hypertension therapy. AD-Pharm Hypertension Mobile Health Application contains several features, one of which is a reminder to remind patients to take medication. The purpose of this study was to determine the effect of the AD-Pharm Hypertension Mobile Health Application on the level of medication adherence of hypertension patients in 4 public health centers in Yogyakarta.  This research method is a quasi-experimental pre-post design with a control group. The sampling technique was consecutive sampling involving 100 respondents, of which 50 respondents were the control group and 50 were the intervention group. The intervention that is given is AD-Pharm hypertension mobile health application. Data were collected using the MARS-5 questionnaire which were analyzed using Mann Whitney test and the Wilcoxon test. The results of the analysis showed that there was a significant difference (p=0.001) of medication adherence between the pre and post-test of the intervention group from 23.96±1.525 to 24.46±1. While in the control group there was no significant difference (p=0.059), from 24.04±1.142 (pre-test) to 24.16±0.976 (post-test). Based on the results of this study, it can be concluded that there is a significant effect on the use of AD-Pharm Hypertension Mobile Health Application on the level of medication adherence of hypertension patients.

Abstract

Non-Hodgkin lymphoma (LNH) is a heterogeneous malignancy of the lymphatic system with CHOP and RCHOP (Rituximab, Cyclophosphamide, Doxorubicin, Vincristine, Prednisolone) chemotherapy regimens as main options. Local data on patient characteristics undergoing these treatments are still limited, especially at Dr. Sardjito General Hospital. This study evaluates the characteristics of LNH patients receiving both regimens at Dr. Sardjito (2020–2023). It is a descriptive observational study with a cross-sectional design. Data from patient medical records were analyzed descriptively. Of 176 patients, 80 received CHOP and 96 R-CHOP. Most were male (CHOP: 70%; R-CHOP: 60.4%) and aged >50 years, with peak age of 51–60 in CHOP (40%) and >60 in R-CHOP (44.8%). Comorbidities were found in one-third of patients. Advanced stage predominated (CHOP: 31.3% stage 4; R-CHOP: 38.5% stage 2), and most completed 6 chemotherapy cycles. Rituximab ≥550 mg was widely used in R-CHOP; cyclophosphamide >1100 mg, doxorubicin 70–79 mg, and vincristine ≥2 mg were standard in both regimens. Prednisone 100 mg was common. Diffuse Large B-Cell Lymphoma (DLBCL) dominated R-CHOP (98.96%), while CHOP patients included DLBCL (38.75%), FL, PTCL-NOS, and others. Findings highlight the importance of individualized treatment based on histopathology and disease stage.

Abstract

Cervical cancer is one of the most prevalent cancer types, making the development of effective anticancer agents critical. Cisplatin (CIS) is a drug that has been used to treat cancer, but it also affects normal cells. Hibiscus sabdariffa L. extract (HSE), which has the potential as an anticancer agent, can be developed as a co-chemotherapy with CIS. This study aimed to determine the potential of HSE as a co-chemotherapy with CIS against HeLa cervical cancer cells and determine specific and non-specific parameters of the studied extract. Hibiscus sabdariffa L. (HS) simplicia powder was macerated with 96% ethanol. An in vitro evaluation was carried out on antiproliferative activity by calculating the doubling time. Additionally, a selectivity test was conducted to calculate combination index (CI) values based on the microtetrazolium (MTT) method. Apoptosis mechanisms were explored based on immunocytochemical methods using p53, caspase, and Bax antibodies, followed by an observation of apoptotic induction using a flow cytometer. The HSE and CIS selectivity index values obtained for Vero cells and HeLa cells were 209 and 278, respectively. The antiproliferation test results showed that the combination of HSE and CIS could better extend the doubling time of cells compared to the negative control. This combination also demonstrated a strong synergistic effect, with a CI value of 0.001. The extract as a co-chemotherapy with CIS was capable of increasing the expression of p53, caspase-3, and Bax. The flow cytometry analysis results indicated that HSE could induce apoptosis. Based on the results on the IC50 and CI value of HSE, as well as on the doubling time and apoptosis induction of HSE-influenced HeLa cells, it is concluded that HSE has the potential as a co-chemotherapy against cervical cancer.

Abstract

Hepar merupakan organ yang berperan dalam pengaturan homeostasis tubuh meliputi metabolisme, biotransformasi, sintesis dan imunologi. Penyebab penyakit hepar bervariasi, diantaranya adalah virus, obat-obatan, alkohol dan stress oksidatif yang merupakan faktor utama. Jumlah radikal bebas yang melebihi antioksidan menyebabkan terjadinya stres oksidatif. Sidaguri (Sida rhombifolia L.) mengandung senyawa fenol dan flavonoid yang berkhasiat sebagai antioksidan. Penelitian ini bertujuan untuk menguji efek hepatoprotektif ekstrak etanol daun sidaguri (EEDS) terhadap nilai SGPT-SGOT, gambaran histopatologi hepar tikus dan rasio berat hepar yang diinduksi carbon tetrachloride (CCl4). Dua puluh lima tikus dibagi menjadi 5 kelompok. Kelompok I merupakan kelompok normal yang hanya diberi makan dan minum, kelompok II merupakan kelompok kontrol yang diinjeksi CCl4 secara intraperitoneal dengan dosis 1 mL/KgBBdan kelompok (III, IV dan V merupakan kelompok perlakuan yang diberi EEDS peroral dengan dosis: 25, 50 dan 100 mg/KgBB) berturut-turut selama 21 hari. Pada hari ke 21 kelompok ini diinjeksi CCl4 1 mL/KgBB secara intraperitoneal 1 jam setelah perlakuan. Data rasio berat organ hepar dari masing masing kelompok dianalisis secara statistik menggunakan analisis varian (ANOVA), LSD post hoc test P<0,05 untuk menentukan perbedaan signifikan. Hasil penelitian menunjukkan bahwa rasio berat hepar pada kelompok II (kontrol) 216,52±15,04 berbeda signifikan dibandingkan dengan kelompok III, IV dan V berturut turut dengan nilai 436,56± 46,78; 438,86±44,79; dan 534,46±45,49. Hal ini menunjukkan bahwa pemberian EEDS dapat menaikkan rasio berat hepar. Data ini didukung dengan nilai SGPT dan SGOT yang berbeda bermakna dengan kelompok kontrol (p<0,05). Dilihat dari gambaran histopatologi hepar, pada  kelompok  II (kontrol), III, IV dan V berturut-turut tidak  menunjukkan adanya nekrosis karena CCl4. Kesimpulan dari penelitian ini bahwa EEDS dapat melindungi hepar berdasarkan rasio berat hepar dan nilai SGPT-SGOT, namun belum dapat disimpulkan bahwa EEDS memiliki efek hepatoprotektif dilihat dari gambaran histopatologi.

Abstract

An autoimmune disorder is an abnormality that causes a disease. It is caused by a weakened immune system. One of the autoimmune diseases is Sjogren’s syndrome, which affects the salivary and lacrimal glands and causes dry mouth, dry eyes, and dry skin. Sjogren’s syndrome influences humans of every age, with the symptoms occurring at the age of 45–55 years and rarely in children. One of the factors causing Sjogren’s syndrome is genetic disorders. To identify genes that can influence Sjogren’s syndrome in this study, we used several databases, including GWAS Catalog, HaploReg Version 4.1, GTEX portal, and Ensembl, particularly to identify the gene expression profiles of TNIP1, TNFAIP3, and IRF5 and the quantitative properties of locus’ expression. This research showed that the missense variants and splice donor rs2233290, rs2230926, and rs2004640 influenced the susceptibility of autoimmune diseases, especially Sjogren’s syndrome, in the fibroblast tissue, sigmoid tissue, sigmoid colon, skin, esophagus, and adrenal glands. The allele frequency of each variant was then assessed in African, American, European, and Asian populations. Our data showed that TNIP1, TNFAIP3, and IRF5 genes in African and American populations had higher frequencies than in the Asian population. This implies that the last of the aforementioned populations might be relatively susceptible to the autoimmune disease Sjogren’s syndrome.
