Abstract

The aim of the research is to describe the blood of Tilapia that had been cultured in a fishpond since June to December 2015. The ponds were located in Marpoyan Subdistrict, Pekanbaru city. Twenty samples were collected as much as 20 fish that consisted of 10 male and 10 female with the size ranging from 9.5 – 18.2 cm. The blood of the fish was taken with syringe via caudal vena as much of 1 ml. The blood parameters then were calculated in term of the number of hematocrit, leucocyte, erythrocyte, and leukocyte. The results showed that the amount of hematocrit and leukokrit were ranged between 10 – 31 % and 1.78 – 3.10 % consecutively. The total cell of erythrocyte ranged between 1,170,000 – 2,190,000 cell/mm3, and the total of leukosit were about 81,000 – 105,000 cell/mm3. In addition, the types of leukocyte found at this study were hepatocyte (53- 86%), thrombocyte (25 – 39%), monocyte and neutrophil between 1 – 3 %.

Abstract

This research has been carried out from November to December 2019 at the Laboratory of Parasites and Fish Diseases at the Fisheries and Marine Faculty of Riau University. The aim of the study was to obtain the best dose of mango leaf solution for the treatment of striped catfish (P. hypophthalmus) infected with E. tarda, seen from the erythrocyte picture. The method used was an experiment with a Completely Randomized Design (CRD) of a factor of five treatment levels and replications three times. Test fish used were striped catfish measuring 8-12 cm, with a total of 150 fish. The treatments used for treatment were 1000 ppm, 1200 ppm, 1400 ppm, negative controls (not infected and not treated), and positive controls (infected and not treated). Parameters measured and observed among others; total erythrocytes, hemoglobin levels, hematocrit values, erythrocyte cell morphology and survival rate. The results showed that apple mango leaf solution (M. indica) was able to treat conjoined striped catfish infected with E. tarda by Emmersion method. The best treatment of 1200 ppm is characterized by an average total erythrocyte, mean total erythrocytes 149.33 x 104 cells / mm3, hemoglobin level 9.03 g / dL, hematocrit value 35.67% and the survival of the test fish after treatment is 86.67%. The results of water quality measurements during the study, namely temperature 27.00-27.46 0C, pH 6.1-6.77, dissolved oxygen 3.53-3.87 mg/L, and ammonia 0.02 mg/L.

Abstract

Curcumin is one of the secondary metabolite compounds products that can increase the immune system of fish against bacterial attack. The purpose of this research was to find out the changes in the erythrocytes of a striped fish that were infectious bacteria A. hydrophila and treated with feeding containing curcumin and know the best dose of curcumin extract in the feed. This research was conducted in September to November 2019. The test fish used was the seed of a striped fish that measures 8-10 cm by 150 tails. Method used was the method of experimental and complete random design a factor of 5 levels of treatment and 3 times repeated, the treatment was Kn (feeding without the extract of curcumin and not infected with bacteria A. hydrophila), Kp (feeding without being given curcumin extract and infectious bacteria (A. hydrophila), P1 (addition of curcumin in feed at a dose of 0.7 g/kg), P2 (0.9 g/kg), P3 (of 1.1g/kg). The results shown that P3 treatment was the best treatment with a < 0.05 P where in a total of erythrocytes was 2.46 x 106 cells/mm3. Hemoglobin 9.4 g/dl, hematocrit value of 32.3%, and survival rate was 90%. Curcumin was able to treat fish from an A. hydrophila infection.

Abstract

Koto Masjid Village is the largest production center in striped catfish fish farming activities in Riau Province. One of the inhibiting factors is the disease caused by bacteria, this study aims to determine the types of pathogenic bacteria that have the potential to infect striped catfish. This research was conducted in August - November 2019, samples of striped catfish used in the study amounted to 15 tails with an average length of 15-25 cm taken from 5 ponds in Kampung II, Koto Masjid Village, XIII Koto Kampar Subdistrict, and then identified bacteria by Conventional Laboratory in Fish Parasites and Diseases, Faculty of Fisheries and Marine, Riau University, Pekanbaru. This study uses a survey method that is by taking samples from the field in Pu and then the isolation of bacteria is carried out from the kidney organ in the TSB media then purified on the TSA media. The results of the study found 3 types of pathogenic bacteria namely Aeromonas sp., Edwardsiella sp., and Pseudomonas sp. Water quality during the study was pH 6.6-70, DO 4.05-4.35 mg/L, NH3 0.035-0.2 mg/L and Temperature 28-300C.

Abstract

Research was conducted from October to December 2017, at the labory Examination of Fish Disease, Fisheries and Marine Faculty of Universitas Riau. The Purpose of this study was to determine the sensitivity of harumanis mango seed (Mangifera indica L) as antimicrobial to A. hydrophila, the range of Minimal Inhibitory Concentration (MIC) and the lethal dose (LD50) harumanis manggo seed (M. indica L) solution against to catfish (Pangasius pangasius) (7-9 cm) by immersion. The research method used doses; D1: 100%; D2: 90%; D3: 80%; D4: 70%; D5: 60%; D6: 50%; D7: 40%; D8: 30%; D9: 20% D10: 10%, and K : Control (Oxytetracyclin).The results showed that mango seed solution was sensitive to A. hydrophila bacteri and was able to inhibit A. hydrophila up to 0.09% dose with inhibit zone of 7.31 mm. Minimum Inhibitory Concetrantion (MIC) is 0.1% (1000 ppm) number of colonies 270.66 CFU/ mL.The dose of LD50 is 2115,85 ppm

Abstract

 Ekstrak daun R. apiculata memiliki manfaat sebagai suplemen herbal yang dapat ditambahkan ke dalam pakan, karena mengandung senyawa flavonoid, tanin, saponin, terpenoid, dan steroid yang dapat meningkatkan kesehatan ikan jambal siam (P. hypophthalmus). Penelitian ini telah dilaksanakan pada bulan Agustus sampai Oktober 2019. Tujuan penelitian adalah untuk mendapatkan dosis terbaik ekstrak daun R. apiculata yang ditambahkan ke dalam pakan untuk meningkatkan kesehatan ikan jambal siam yang dilihat dari total eritrosit, kadar hemoglobin, kadar hematokrit, dan morfologi eritrosit. Metode yang digunakan adalah metode eksperimen dengan Rancangan Acak Lengkap (RAL)satu faktor empat taraf perlakuan terdiri dari P0;(tanpa penambahan ekstrak). P1;(1,5 mg/kg). P2;(1,7 mg/kg). dan P3;(1,9 mg/kg) dan dilakukan ulangan sebanyak tiga kali sehingga diperlukan 12 unit percobaan. Pemeliharaan dilakukan selama 60 hari di dalam keramba dengan ukuran 1x1,5x1 m3. Pemeriksaan darah dilakukan sebanyak tiga kali,yaitu  pada awal pemeliharaan, hari ke-30, dan hari ke-60. Dosis terbaik terdapat pada perlakuan P2 (1,7 mg/kg) dilihat dari total eritrosit 2,57x106 sel/mm3, kadar hemoglobin 13,13 g/dL, nilai hematokrit 38,43%, pertumbuhan bobot mutlak 38,93 g, kelulushidupan 97,33%, dan menunjukkan morfologi eritrosit yang normal didukung oleh kualitas air selama penelitian seperti DO 6,4-6,9mg/L, pH 6,1-6,8, Suhu 28,2-29,5 0C, dan Amoniak 0,04-0,08 mg/L.

Abstract

This research was conducted from July-August 2017. The purpose of the research is to know the effect of adding the chicken feather meal in fish food which was fermented by Bacillus sp. The fermented chicken feather meal was added to the food for adjusting the leukocytes differentiation of Silver Pompano (Trachinotus blochii). The method was an experimental method with a Completely Randomized Design (CRD), one factor with six treatments level. The for 180 fishes. This research used six treatments such as, Kn (negative control): without fermented chicken feather meal by Bacillus sp. in the feed and were not infected by Vibrio alginolyticus, Kp (positive control): without fermented chicken feather meal byBacillus sp. in the feed and were infected by V. alginolyticus, P1-P4 respectively: fermented chicken feather meal by Bacillus sp. in the feed 5%, 10%, 15% and 20%. The collected data is; leukocytestotal, leukocytes differentiation, the average of weight, and survival rate. The results showed that there were a significant effect of adding the fermented chicken feather meal by Bacillus sp.  in the feed to the leukocytes total and leukocytes differentiation of Silver Pompano (Trachinotus blochii). The best treatment was 10% of Bacillus sp.(P2) wich was shown by the80,53x103 cells of leukocytes total, 80,33% of lymphocytes, 9,67 % of monocytes, 10.00 % of Neutrophils, , 5,30 g was the average of weight and100 % of survival rate.

Abstract

Daun bakau (Rhizophora apiculata) mengandung senyawa flavonoid, steroid, saponin, dan tannin yang berfungsi sebagai antimikrobial dan dapat meningkatkan kesehatan ikan. Penelitian ini telah dilaksanakan pada bulan Agustus sampai dengan bulan Oktober 2019. Tujuan penelitian mendapatkan dosis terbaik ekstrak daun R. apiculata yang ditambahkan ke dalam pakan untuk meningkatkan kesehatan ikan yang dilihat dari total leukosit, diferensiasi leukosit, dan aktifitas fagositosis ikan jambal siam (Pangasius hypophthalmus).Metode yang digunakan adalah metode eksperimen dengan menerapkan Rancangan Acak Lengkap (RAL) satu faktor dengan empat taraf perlakuandan tiga kali ulangan, sehingga diperlukan 12 unit percobaan.Perlakuan tersebut adalah P0 (pakan tanpa mengandung ekstrak daun bakau (R. apiculata), P1, P2, dan P3 pakan yang diberi kandungan ekstrak daun bakau (R. apiculata) dengan dosis 1,5 mg/kg, 1,7 mg/kg, dan 1,9 mg/kg pakan. Ikan yang digunakan adalah ikan jambal siam (P. hypophthalmus) ukuran 8-10 cm dengan padat tebar 50 ekor/m3. Hasil penelitian menunjukkan bahwa dengan penambahan ekstrak daun R. apiculata ke dalam pakan memberikan pengaruh nyata terhadap total leukosit, diferensiasi leukosit, aktifitas fagositosis, kelulushidupan ikan dan pertumbuhan bobot mutlak ikan jambal siam (P. hypophthalmus). Dosis terbaik adalah 1,7 mg/kg pakan dengan total leukosit 11,17x104 sel/mm3, persentase limfosit 84,33%, neutrofil 6,67%, monosit 9,00%, aktifitas fagositosis 35,00%, kelulushidupan 94,67%, dan pertumbuhan bobot mutlak 38,93g. Kualitas air waduk selama penelitian,  suhu antara 28,2-29,50C, DO 6,4-6,9 mg/L, pH 6,1-6,8 dan NH3 0,04-0,08mg/L.

Abstract

Terminalia catappa is commonly present in tropical area  and its can be used for increasing pH in fish culture and it also be used as disinfectant.  A research aims to understand the dosages of T. catappa leaf extract to hamper the growth of Aeromonas hydrophila  has been conducted  from February  to July 2019.  A diffusion agar KIRBY BAUER method was applied to understand the sensitivity of T. catappa  toward the A. hydrophila. Treatments applied were P0 (Control, Oxytetracyclin (30 µg.Disk), P1 (100%), P2 (90%), P3 (80%), P4  (70%), P5 (60%), P6 (50%), P7 (40%), P8 (30%), P9 (20%), P10 (10%) P11 (9%), P12 (8%), P13  (7%), P14 (6%), P15 (5%), P16 (4%), P17 (3%), P18(2%), P19 (1%). Pure leaf extract was obtained from fresh leaf. The disk blank was wet with 50 L of leaf extract. A. hydrophila was cultured in TSA, TSB and GSP media, and it was grew up for 24 hours. Results shown that the pure leaf extract produce  13.9 mm diameter clear zone, while 2% of pure extract produce 6.5 mm diameter clear zone.

Abstract

Pathogenic bacteria is disease causing microorganisms that can attack fish and can cause mass death in cultured fish. The purpose of this study was to identify the types of pathogenic bacteria Cyprinus carpio that were reared in floating cages. This study used a survey method that is purposive sampling and the fish sample were obtained from, PLTA Koto Panjang. the fishes were identified in the Fish Parasite and Disease Laboratory, Fish and Marine Faculty, University of Riau. Fish samples used were 15-20 cm goldfish totaling 12 tails with 3 times taken from 4 different cages. The kidneys organ were examined and to find out the type the bacteria. Result shown identified of pathogenic bacteria consist of 3 types of bacteria namely Aeromonas sp., Pseudomonas sp., and Edwardsiella sp. Water quality during the study was Temperature: 27-300C, pH 6-7, DO 3.67-4.28 ppm, and ammonia 0.048-0.64 mg/L.
