Abstract

Curah  hujan  mempengaruhi  salinitas  air  laut,  yang  pada  gilirannya  dapat  menyebabkan  rendahnya pertumbuhan Kappaphycus alvarezii.  Penelitian ini bertujuan untuk mengetahui perubahan morfologi, ukuran dan jumlah sel, pertumbuhan serta rendemen karaginan K. alvarezii pada salinitas yang berbeda.  Penelitian dilakukan  menggunakan  metode  kultur  jaringan,  selanjutnya  thallus  yang  telah  tumbuh  diekstraksi karaginannya. Hasil penelitian menunjukkan bahwa salinitas yang berbeda berpengaruh nyata terhadap jumlah sel K. alvarezii (p<0,05), tetapi tidak terhadap rata-rata pertumbuhan harian (p>0,05). Rendemen karaginan pada semua perlakuan salinitas relatif tinggi dan sesuai dengan persyaratan untuk ekspor. Kata kunci:  salinitas, sel, pertumbuhan, karaginan, K. alvarezii Rainfall affects the salinity of sea water, which in turn can lead to the low growth of Kappaphycus alvarezii. This research aimed to determine changes in the morphology, size and number of cells, growth and carrageenan yield of K. alvarezii at different salinitity levels. The research was conducted using tissue culture method, furthermore, from the grown thallus then the carageenan was extracted. The results showed that different of salinity levels significantly influenced the number of cells K. alvarezii (p<0.05), but not against the average daily gain (p>0.05). The carrageenan yields at all salinity treatment were relatively high and eligible for export.  Key words: salinity, cell, growth, carrageenan, K. alvarezii

Abstract

Bakteri memainkan peran penting dalam ekosistem mangrove terutama dalam mengurai searasah daun. Keberadaan bakteri ini sangat dipengaruhi oleh berbagai faktor lingkungan. Penelitian ini bertujuan untuk mengisolasi dan karakterisasi bakteri dari perairan mangrove dari dengan vegetasi yang berbeda yakni Avicennia marina, Rhizophora apiculata, Avicennia alba dan Sonneratia alba. Pengambilan sampel dilakukan di perairan mangrove desa Bulu Kerto Kraton Pasuruan. Proses karakterisasi bakteri dilakukan di laboratorium mikrobiologi Fakultas Kedokteran Universitas Brawijaya Malang. Metode penelitian yang digunakan adalah eksploratif sedangkan parameter uji adalah isolasi bakteri dan uji morfologi, biokimia, serta deteksi pertumbuhan bakteri yang dominan. Hasil penelitian diperoleh 7 isolat bakteri Avicennia marina yaitu Nitrococcus sp., B. subtilis, B. pumilus, Pseudomonas putida. P.stutzeri, Micrococcus luteus, Vibrio sp, 6 isolat bakteri Rhizophora apiculata yaitu B. megaterium, Nitrococcus sp, Staphylococcus sp., P.putida, Lactobacillus sp., B. Subtilis,  8 isolat bakteri Avicennia alba yaitu B.megaterium, B.pumilus, B. subtilis, Nitrococcus sp., Pseudomonas putida, P. stutzeri, B. mycoides, Micrococcus sp.. dan 6 isolat bakteri di Sonneratia alba yaitu Bacillus megaterium, Nitrococcus sp., B.subtilis, Planococcus citreus, B. mycoides, Lactobacillus plantarum. Hasil ini menunjukkan bahwa jenis bakteri dipengaruhi oleh vegetasi mangrove yang ada di suatu perairan. Kata kunci: bakteri, perairan mangrove, vegetasi Bacteria play an important role in the ecosystem, especially in degrading mangrove leaves (litter). The presence of these bacteria is greatly influenced by various environmental factors. This research aims to isolate and characterized bacteria from mangrove waters with different vegetaion namely Avicennia marina, Rhizophora apiculata, Avicennia alba and Sonneratia alba. Characterization of the bacteria were done at microbiological laboratory in Medical Faculty, Brawijaya University. Several tests were done in this explorative research i.e. morphological test, biochemistry, and bacteria growth. The results showed there were 7 isolated bacterias found at Avicennia marina area i.e. Nitrococcus sp, B. subtilis, B. pumilus, Pseudomonas putida. P. stutzeri, Micrococcus luteus, Vibrio sp.; 6 isolated bacterias found at  Rhizophora apiculata area i.e. B. megaterium, Nitrococcus sp, Staphylococcus sp., P. putida, Lactobacillus sp., B. subtilis; 8  isolaed bacterias from Avicennia alba area i.e. B. megaterium, B. pumilus, B. subtilis, Nitrococcus sp., Pseudomonas putida, P. stutzeri, B. mycoides, Micrococcus sp., and 6 isolated bacterias from Sonneratia alba area i.e. Bacillus megaterium, Nitrococcus sp, B. subtilis, Planococcus citreus, B. mycoides, Lactobacillus plantarum. Abstract

To reduce organic waste in catfish maintenance media (Clarias gariepinus) and waste management wastes a cultivation management technology is needed, one of these technologies is bioremediation using Bacillus subtilis as a bioremediator. This study aims to analyze the parameters of organic matter content including TOM (Total Organic Matter), Protein, Carbohydrates and Fat. The study was conducted in January-February 2019. This study used a factorial completely randomized design (Factorial RAL) with the addition of Bacillus subtilis 4 treatments including controls (K = Control, D1 = 100 ml / L, D2 = 10 ml / L and D3 = 1 ml / L) at 24th, 48th, 72nd, 96th and 120th hours, consisting of three replications. The results obtained from this study indicate that the concentrations of each parameters in treatments D1, D2 and D3 were lower than those of controls, indicating that Bacillus subtilis can reduce organic waste from catfish ponds. The best treatment was W5D2 as bioremediation of catfish organic waste with the remaining TOM 16.23% with an efficiency value of 84%, remaining protein 86.66% with an efficiency value of 13%, remaining carbohydrate 63.91% with an efficiency value of 36% and remaining Fat 52.69% with an efficiency value of 47%.

Abstract

The objective of this study was to observe the interactions between salt-soluble proteins from beef and seaweed (Eucheuma cottonii) flour and its effect to the quality of the produced protein gel. The study was conducted as experimental research with two factorials completely randomized design. The effect of different NaCl (2.0%; 2.5%; 3.0%; and 3.5%) and Eucheuma cottonii flour (5%; 10%; and 15%) concentrations was observed to the physical and proximate quality of the protein gel matrix. The observed gel quality variables include pH, gel strength, color (L, a*, and b*), moisture, protein, and fat content. The gel was gelatinized at 90°C for 20 min and cooled down at 4°C for 12 h. The results showed that there was no interaction effect found on all of observed variables. However, a highly significant effect (P<0.01) was found on the ash content with different salt concentrations, while different Eucheuma cottonii flour concentrations improved the gel strength (P<0.01) and significantly gave darker (P<0.01) color. The effect of Eucheuma cottonii flour on the physicochemical properties of the gel was concentration dependent. Therefore, this study concludes that Eucheuma cottonii flour showed a potential as binding or filling agent to form strong gel matrix in processed meat products.

Abstract

Penerapan teknologi bioflok, sistem budidaya heterotrofik dengan pertukaran air minimal, saat ini sedang dikembangkan dalam kegiatan budidaya udang vaname (Litopenaeus vannamei) karena dapat memberikan produktivitas yang lebih tinggi, konversi pakan yang lebih rendah, dan kualitas air yang lebih stabil. Pada teknologi bioflok, bakteri heterotrofik di dalam sistem budidaya cenderung membentuk agregat (bioflok) yang dapat dikonsumsi oleh udang sebagai sumber protein alami. Dalam penelitian ini, studi SDS-PAGE dilakukan pada daging L. vannamei sepanjang siklus produksi tambak intensif dengan sistem budidaya yang berbeda (Fitoplankton, Semi-Bioflok, dan Bioflok) untuk menyelidiki pengaruh sistem budidaya yang berbeda tersebut terhadap pola pita protein daging L. vannamei. Penelitian ini berlangsung pada beberapa lokasi di Kabupaten Tuban, Jawa Timur. Tujuan penelitian ini adalah untuk memperoleh karakteristik pita protein daging L. vannamei yang berasal dari sistem pemeliharaan yang berbeda. Hasil penelitian menunjukkan bahwa sistem bioflok menghasilkan pola pita protein dengan variabilitas yang lebih tinggi karena sistem ini dapat mempertahankan kualitas air yang baik selama pemeliharaan dan menyediakan sumber protein alternatif berupa bioflok bagi L. vannamei selain pakan pelet.Kata kunci: udang vaname (L. vannamei), sistem bioflok, daging, pita protein, SDS-PAGEAbstractApplication of biofloc system, a heterotrophic culture with minimal water exchange, has gained interest particularly in relation to providing high productivity, low feed-conversion ratios, and a stable culture environment, especially in shrimp. In biofloc system, heterotrophic bacterial tends to form noticeable aggregates (biofloc), which can be consumed by shrimp as a natural protein source. In relation to this, the impact of different culture systems on white shrimp (L. vannamei) muscle protein patterns was investigated. In this research, an SDS-PAGE study was performed on white shrimp muscle tissue along the production cycle in intensive shrimp culture ponds with different culture systems (Phytoplankton, Semi-Biofloc, and Biofloc). The study was located in different areas of the District of Tuban, East Java. The aim of this study was to accomplish systematic characterization of the white shrimp muscle protein pattern, which derives from each of the ponds with different rearing systems. The result of the study showed that the biofloc system yields variability of protein pattern as it maintains good water quality throughout the culture and provides an alternative protein source (as biofloc) for the shrimp besides the pellet.Key words: white shrimp (L. vannamei), biofloc system, muscle, protein pattern, SDS-PAGE

Abstract

Sipermetrin pada dasarnya digunakan untuk mengendalikan hama, penyakit dan gulma pengganggu pada kegiatan pertanian.  Tetapi oleh pengolah Jambal Roti digunakan untuk mencegah pembusukan sehingga daya simpan produk lebih lama dan kerugian bisa dikurangi.  Penelitian ini bertujuan untuk mengetahui pengaruh sipermetrin terhadap kadar ureum dan kreatininTikus Wistar (Rattus norvegicus). Tikus diberi perlakuan paparan sipermetrin dengan dosis 0.00 mg/kg (K-), 0.05 mg/kg, 0.60 mg/kg, 1.10 mg/kg, 1.60 mg/kg, 2.15 mg/kg dan daging ikan yang mengandung sipermetrin 1.73 mg/kg (kontrol positif : K+). Kadar ureum dan kreatinin darah tikus ditetapkan berdasarkan metode Enzymatic Photometric.  Hasil menunjukkan bahwa sipermetrin berpengaruh terhadap peningkatan kadar ureum dan kreatinin Tikus Wistar.  Kadar ureum sudah melewati normal pada hari ke-7 pemeliharaan untuk perlakuan sipermetrin dosis 2.15 mg/kg.  Rata-rata kadar kreatinin pada hari ke-14 semua perlakuan sudah melewati batas kadar normal, kecuali pada perlakuan K- dan 0.05 mg/kg. Kadar ureum dan kreatinin tertinggi yaitu 27.7±0.98 mg/dL dan 1.03±0.018 mg/dL diperoleh pada perlakuan sipermetrin dosis 2,15 mg/kg.

Abstract

Sipermetrin  bersifat  neurotoksin  dan  dapat  mempengaruhi  kadar  enzim kolinesterase. Sipermetrin ditemukan terdapat pada produk Jambal Roti dengan kadar yang melebihi batas maksimum residu yang diperkenankan SNI dan CAC. Penelitian ini bertujuan untuk mengetahui pengaruh sipermetrin terhadap kadar enzim kolinesterase Tikus Wistar (Rattus norvegicus). Tikus diberi perlakuan paparan sipermetrin dengan dosis 0,00 mg/kg (K-), 0,05 mg/kg, 0,60 mg/kg, 1,10 mg/kg, 1,60 mg/kg, 2,15 mg/kg dan daging ikan yangmengandung sipermetrin 1,73 mg/kg (kontrol positif : K+). Kadar enzim kolinesterase darah tikus ditetapkan berdasarkan metode Enzymatic Photometric.  Hasil menunjukkan bahwa sipermetrin berpengaruh terhadap penurunan kadar enzim kolinesterase Tikus Wistar.  Kadar enzim kolinesterase terendah pada perlakuan paparan sipermetrin dosis 2,15 mg/kg yaitu 4059.9 U/L.

Abstract

The utilization of maggot as larvae from black soldier fly is an alternative substitution of fish meal as the primary protein source in fish feed. Maggot is cultivated in 3 different cultivation, the medium used is organic waste, tofu waste, and wheat pollard. The method that used in this research is Completely Randomized Design (CRD) with 3 treatments and 3 replication. This study aims to determine the effect of various types of maggot growth media, 1) with maggot biomass parameters, containers, and growing media conditions maggot, 2) then, exact proximate composition, total amino acids content of the prepupae samples were determined. Based on the results of the analysis, each dose of treatment (media) has an influence on the different maggot biomass. The best media treatment results are pollard flour. The proximate analysis showed that BSF larvae had a protein content of 50,88% and also fat content of 15%.

Abstract

Centella asiatica is a herbal medicine has been used for many studies in some animals as antioxidant properties. This study was aimed to enhance the level of SOD in Cyprinus carpio after oral administration of C. asiatica extract. In the present study, phytochemical screening of bioactive constituents, antioxidant assay, and their effect on SOD in C. carpio liver were investigated. C. asiatica extract was obtained by maseration for 24 h using ethanol as a solvent. C. asiatica extract with 50, 100, 150 and 300 mg kg-1 body weight were administered every 3 days for two weeks. On the last day of experiment, fishes were killed and liver were removed from the body. The results showed that C. asiatica extract had some bioactive compounds such as flavonoid, alkaloid, terpenoid, tanin, saponin and DPPH scavenging activity with an IC50 value of 125 μg mL-1. C. asiatica extract also enhanced the level of SOD in fish liver. The SOD level was significantly difference compared to the control group with P<005. These results indicate that the ethanolic extract of C. asiatica had a potential antioxidant properties by scavenging free radical and enhancing the SOD level as antioxidant deffense inside the body.

Abstract

Indonesian waters have six types of turtles that can live, spawn and breed. Sea turtle conservation becomes an important and urgent program to be done in order to protect and save sea turtle population in Indonesia. One of the factors that most affect the turtle population is the cause of degradation of pathogenic factors. Alas Purwo National Park, East Java, there is some communities that have activities turtle conservation. Conservation is done by securing and protecting turtle eggs. Turtle eggs that have hatched are released into the sea once it is ready. This study aims was to determine the type of bacteria and fungi that infect hatchlings and environmental factors that influence. This research is descriptive method to Hawksbill turtle (Eretmochelys imbricate) is by way of random sampling. Sampling of microbes in sea turtle was conducted using cotton swab method and then microbes was cultured and indentified in laboratory. The results showed The kind of parasites and microbes which were indentified in hatching and adult Hawksbill sea turtles were fungus with genus Aspergillus sp., Geotrichum sp., Fusarium sp., and Gliocladium sp. ; bacteria are Pseudomonas aeruginosa and Enterobacter cloaceae; and parasite is Chelonibia testudinaria barnacles.  The parameter average value of water in pond indicated 28.1 – 29.2°C for temperature, 32 - 34 ‰ for salinity, 7.78 – 8.2 for pH, and 3.86 – 4.21 mg/L for DO.
