Abstract

Penelitian ini bertujuan untuk mengetahui pengaruh dan dosis yang terbaik pada pemanfaatan diatomae Chaetoceros ceratosporum dalam formula pakan terhadap peningkatan respon imun seluler udang windu (Penaeus monodon Fab.). Penelitian ini menggunakan Rancangan Acak Lengkap (RAL) dengan 4 perlakuan dan 3 ulangan. Sebagai perlakuan adalah pemanfaatan Chaetoceros ceratosporum dalam formula pakan iso protein 39,02% dan iso energy 3,58 kkal g pakan dengan dosis yang berbeda yaitu: A (0%), B (3,04%), C (6,08%) dan D (9,12%). Parameter yang diamati adalah total hemosit, total diferensial hemosit (hyaline, semi granular dan granular), dan aktivitas vibriocidal. Berdasarkan hasil penelitian menunjukkan bahwa pemanfaatan diatomae Chaetoceros ceratosporum dalam formula pakan dapat meningkatkan respon imun seluler, dan dosis terbaik berkisar 5,15% - 6,51%. Abstract

Interaksi auksin-sitokinin dianggap penting untuk mengatur pertumbuhan dan perkembangan dalam jaringan tanaman. Penelitian ini bertujuan untuk menentukan komposisi auksin dan sitokinin yang tepat untuk regenerasi filamen kalus rumput laut K. alvarezii. Kultur filamen kalus dilakukan pada media cair dengan formulasi ZPT indole acetic acid (IAA) : kinetin : zeatin, dengan komposisi konsentrasi sebagai berikut : A (0,4:0:1) ppm; B (0,4:0,25:0,75) ppm; C (0,4:0,5:0,5) ppm; D (0,4:0,75:0,25) ppm; E (0,4:1:0) ppm; kontrol (tanpa ZPT). Penelitian menggunakan rancangan acak lengkap dengan pengulangan masing-masing perlakuan 3 kali. Parameter yang diamati adalah laju pertumbuhan harian, sintasan, kecepatan regenerasi, panjang tunas dan perkembangan morfologi. Hasil yang diperoleh menunjukkan bahwa formula ZPT terbaik adalah formula A dengan laju pertumbuhan harian 1,929%/hari, sintasan 83,33%, kecepatan regenerasi 41,67% dan rata-rata panjang tunas 44,59 µm. Tunas mulai terbentuk pada 15 hari masa kultur. Kata kunci: formula zat pengatur tumbuh, auksin-sitokinin, K. alvarezii, mikropropagasi

Abstract

Uji hematologi sangat bermanfaat sebagai alat diagnostik didalam menetapkan status kesehatan ikan. Salah satu aspek dari infeksi adalah terjadinya perubahan gambaran darah. Pemeriksaan darah dapat digunakan sebagai indikator keparahan suatu penyakit tertentu. Beberapa parameter yang dapat memperlihatkan perubahan patologi pada darah meliputi jumlah leukosit, deferensial leukosit, eritrosit. Dari hasil penelitian diketahui jumlah rata-rata sel darah merah dalam darah ikan botia sehat pada masing-masing perlakuan K-, K+, A, B, C, D berturut-turut adalah 2.470.000, 2.500.000, 2.560.000, 2.350.000, 2.440.000, dan 2.430.000. Sementara itu pada ikan botia yang terinfeksi bakteri A.hydrophila diketahui jumlah rata-rata eritrosit pada masing-masing perlakuan K-, K+, A, B, C, dan D berturut-turut adalah 2.420.0000, 1.770.000, 1.780.000, 1.890.000, 1.780.000, dan 1.870.000. jumlah rata-rata eritrosit ikan botia yang terinfeksi bakteri A.hydrophila, dan kemudian diobati dengan ciprofloxacin pada masing-masing perlakuan K-, K+,A, B, C, dan D berturut-turut adalah 2.420.000, 1.630.000, 2.090.000, 2.280.000, 2.500.000, dah 2.540.000. Sementara jumlah rata-rata sel darah putih dalam darah ikan botia yang sehat pada masing-masing perlakuan K-, K+, A, B, C, D berturut-turut adalah 97.000, 90.000, 88.000, 87.000, 93.000 dan 88.000. Sementara itu jumlah rata-rata sel darah putih pada ikan botia yang terinfeksi bakteri A.hydrophila, pada masing-masing perlakuan K-, K+, A, B, C, D berturut-turut adalah 97.000, 174.000, 174.000, 179.000, 185.000 dan 185.000. Sedangkan dari hasil penelitian terhadap ikan botia yang terinfeksi A.hydrophila dan kemudian diobati dengan ciprofloxacin diperoleh hasil perhitungan rata-rata leukosit pada masing-masing perlakuan berturut-turut adalah sebagai berikut  980.000, 189.000, 125.000, 137.000, 133.000, dan 136.000. Abstract

Soybean husk (Glycine max L. Merrill) a soybean processing waste as raw material for tempe obtained after the process of boiling and soaking soybeans. The main problem in the use of soybean husk (Glycine max L. Merrill) as feed material is its crude fiber content which is fairly high. This study aimed to observe the fermented soybean husk using Aspergillus niger to improve the quality of the raw feed materials. This was conducted by using completely randomized design (CRD) analysis and repeated three times; the time optimization of Aspergillus niger in 2, 4, and 6 days based on chemical analyses (moisture, protein, fat, ash, crude fiber and feed containing carbohydrates (NFE) and physical assessment fermentation (smell, texture, moisture and hyphae) were analyzed descriptive qualitatively. The results showed that 4 days fermentation of soybean husk using A. niger is successful gives the highest score based on physical characteristics texture, aroma, moisture, and the formed hyphae and the most effective treatment for decrease in crude fiber is 13% and increase in NFE contained in the largest on 4 days fermented soybean husk by Aspergillus niger with a long time 4 days. Abstract

The purpose of this study was to analyze the optimal dose of Tembelekan leaf extract (Lantana camara Linn) which has antibacterial properties in inhibiting or killing Edwardsiella tarda bacteria. The method used in this research is Completely Randomized Design (CRD). The results of the research yield obtained from tembelakan leaf extract amounted to 8.9%. The positive phytochemical test results contained active compounds, namely flavonoids, alkaloids, tannins, triterpenoids, and saponins. GC MS analysis of tembelekan leaf extract showed that the most components were 4H-1-Benzopyran-4-one, 5,7-dihydroxy-6-methoxy-2-(4-methoxyphenyl)-,2,1,3-Benzothiadiazole, Benzisot hiadiazole , 3,4-Benzo-1,2,5-thia diazole. This compound is a class of isoflavonoids compounds. The MIC (Minimum Inhibitory Concentration) test results obtained the lowest absorbance value at a concentration of 100 mg/L with a value of 0.602 close to the positive control. Results of discs with doses of 50, 100, 150, 200, and 250 mg/L. The disc results showed that the dose of 200 mg/L had the strongest inhibition zone with a value of 12.63 mm. Tembelekan leaf extract which contains dominant compounds such as flavonoids and alkaloids can be useful as an antibacterial which inhibits the growth of E. tarda bacteria

Abstract

Indonesia has very large territorial water, so it supports the development of the economy in the field of fisheries, one of which is through aquaculture. Recently fish meal has become the most expensive protein ingredient in aquaculture feeds. We aim to proximately analysis of some common alternative flour as fish feed ingredient i.e. fish flour, shrimp head flour, soybean flour, and bran flour to get best-recommended flour for type cultured fish. Fish flour, shrimp head flour, soybean flour, and bran flour was purchased from the fish farmer in Malang who use alternative fish feed flour for their aquaculture. Proximate analysis carried out by protein content, Moisture content, Fat and Fiber. Proximate analysis was conducted in the Fishery Products Engineering laboratory, Brawijaya University on April 2018. The results showed that shrimp head flour had higher protein content than others flour that is about 48.71%, fat and carbohydrate content about 7.99%, and 18.34% respectively. Soybean flour contains protein, fat and carbohydrate about 31.29%, 20.75%, and 30.45% respectively. Carnivorous fish culture can use high protein flour as an alternative ingredient for feed such as fish flour and shrimp head flour; balance proximate composition for omnivorous fish cultures such as soy flour and high carbo flour such as bran flour for herbivorous fish culture.Keywords: Bran flour, Shrimp head flour, Soybean flour, Proximate analysis.

Abstract

Abstract This study was aimed to know the changes of the character of tuna fish (Thunnus sp.) which fermented by Lactobacillus plantarum starter culture. Factorand variable used was the time of ripening, namely: 0, 2, 4, 6, 8, and 10 days. The parameters observed included: pH, Total Acid, N-Total, and NAmino. The results showed that ripening time of 6 days was the best make processing of sausage against pH, N-Total, and N-Amino value were 4.200±0.177, 3.6472±0.0386, and 0.684±0.0390 respectively, while the higher of Total acid showed that ripening time that 8 days was 3.973±0.064

Abstract

Abstract Seaweed is vulnerable to be infected by disease due to unlikely environmental circumstance. This condition could cause impact on the optimum growth of seaweed. This research was carried out to understand the rate and percentage of ice-ice disease on Kappaphycus alvarezii. Seven units of seaweed rafts were used. Each of them was observed every 12 hours. Results of this research indicated that the percentage of infection was relatively higher during the day on the observation units which were located next to the shore (1,008%). Three days after the infection of ice-ice disease occurred, the mortality of Kappaphycus alvarezii was certain. Therefore, it was recommended that harvesting Kappaphycus alvarezii should be done one day after the indications of ice-ice disease were noticed.

Abstract

Abstract Edible films made from chitosan and starch materials have several physical limitations, particularly in terms of tensile strength and elongation. To address these limitations, glycerol and sorbitol are often added as plasticizers during the production process. Chitosan has also been reported to have plasticizing properties and can serve as an alternative through its modification into nano-sized particles, thereby increasing its reactivity. Therefore, this study aimed to characterize the physical properties of edible films made from black mangrove (Rhizophora mucronata) fruit starch and chitosan by adding nano chitosan suspension at different storage temperatures to determine the best treatment. A completely randomized factorial design was used, and the data obtained were analyzed using Analysis of Variance (ANOVA) at a 95% confidence level with IBM SPSS statistics 25. The treatments used included the addition of nano chitosan suspension (A) at various concentrations of 0%, 25%, 50%, and 75% with storage (B) for 0 days, 9 days at room temperature, and 9 days at cold storage (± 50C) in triplicates. The significant treatments were then further tested using Honestly Significant Difference (sig < 0.05). The results showed that the addition of nano chitosan suspension with storage method affected the physical properties of the edible films. The edible film made from black mangrove fruit starch-chitosan with the addition of nano chitosan (50%:50%:75%) stored at cold temperature for 9 days had the best physical properties to be applied as a package for fishery products (fillets) based on De Garmo analysis.   Highlight Research Nano chitosan can be applied as plasticizer in making edible film of black mangrove starch-chitosan. The concentration of nano chitosan and storage temperature influenced the thickness, tensile strength, elongation, young modulus, and water resistance of edible film. The edible film made from Black mangrove starch - chitosan with the addition of 75% concentration of nano chitosan when stored at cold temperature (5 0C) for nine days can increase tensile strength, young modulus, elongation, and water resistance. Components of edible films interact less with one another intermolecularly due to nano chitosan, which encourages the creation of hydrogen bonds between polymer chains.

Abstract

Mangroves are one of the beneficial plants that directly touch human life, especially coastal communities. The extract and raw materials from these mangroves are widely used as a source of food and for natural medicinal. The purpose of this study was to identify the chemical content and bioactive compounds of Rhizophora mucronata from Langge Village, Kecamatan Anggrek, North Gorontalo District. In this research, the research method used is descriptive method. To obtain the extract using the maceration method with methanol as a solvent. Chemical testing includes testing of moisture content, ash content, total fat, protein, carbohydrates, while the identification of bioactive compounds uses qualitative methods. The results of the chemical content test for water content of 52.38%; Ash content 0.22%; Total fat content 2.33%; protein content of 6.85% and carbohydrates 30.30% and the identified bioactive compounds are flavonoids, saponins, tannins, tripertenoids and steroids as well as phenol hydroquinone compounds.
