Abstract

Prebiotics is indigestible foods that can not be digested but can stimulate the growth and activity of bacteria in the digestive tract effecting human health. Alginate oligosaccharides (AOS) can be used as a source of prebiotic. That compounds can be produced enzymatically by cutting long chain alginates using alginate lyase. The aim of this study was to produce alginate lyase enzyme then producing Alginate oligosaccharides (AOS) as a prebiotic ingredients. The alginate lyase enzyme can be produced from Bacillus megaterium bacteria using a discontinuous fermentor. The enzyme was  optimum temperature of 45°C and an optimum pH of 7.0. Alginate oligosaccharides production was performed with the addition of different enzyme concentrations 25, 50, 75, and 100 U. The result of the addition of enzyme (25, 50,75 U) showed that the value of polymerization degrees (DP) were between 4-5. However, the addition of enzyme (100 U) was in the range of  DP 3-4. Bacterial probiotic growth test results of Bifidobacteria and Lactobacillus showed that 1% added AOS media were able to increase the growth of probiotic bacteria compared to themedia without addition of AOS. The addition Alginate lyase activity of 50 U in AOS production is the best treatment of both probiotic bacteria.

Abstract

Seaweed is main commodity with abundant amount of Indonesian waters. Seaweed contains naturallybioactive compounds that acts as antioxidant and tyrosinase inhibitor which inhibit melanin formationand the latest approach used to skin lightening. The aim of this research were to determine chemicalcharacterization such as heavy metal, secondary metabolite (phytochemical compounds), moisture, vitaminC and E, antioxidant activity and total plate count (TPC) on dry simplisia and slurry of S. plagyophyllumand E. cottonii. Total microbe of S. plagyophyllum amounted 2.3×102 colony⁄g with moisture content 16,71%and E. cottonii amounted 2.2×103 colony⁄g with moisture content 19.79%. Brown seaweed, S. plagyophyllum,did not contain dangerous heavy metal. Vitamin C value in both spesies of seaweed were 212.95 mg/kg forS. plagyophyllum and 15,95 mg/kg for E. cottonii. Vitamin E of S. plagyophyllum amounted 363.86 mg/kgand 0.23 mg/kg for E. cottonii. IC50 value of S. plagyophyllum was 109 ppm and 130.62 ppm for E. cottonii.S. plagyophyllum slurry contain bioactive compound such as alkaloids, steroids, flavonoids, saponins andtannins. E. cottonii slurry contain bioactive compound alkaloids and terpenoids.

Abstract

The horn snail (Telescopium telescopium) is a gastropod living in mangrove forests and is considered as a pond pest. This study was aimed to determine the effects of alkaline and acetic acid pretreatment on the characteristics of collagen from the horn snail meat. The immersion of the meat in NaOH 0.20% for 10 hours gave a significant reduction in non-collagen protein content (α = 0.05). Further immersion with acetic acid 0.05% for 6 hours also significantly affected (α = 0.05) the swelling degree and the collagen solubility. The extraction yield was 1.08±0.21%, with 72.23% of brightness, and the viscosity was 16.34 cP. The proline content of the collagen was 9.21%.

Abstract

Rumput laut Gracilaria sp. sering dimanfaatkan sebagai sumber agar-agar komersial. Penelitian ini bertujuan untuk menentukan karakteristik dari Gracilaria sp. sebagai bahan baku pangan fungsional. Metode ekstraksi pada penelitian ini yaitu maserasi tunggal menggunakan berbagai pelarut seperti: etanol, etil asetat, dan n-heksan, dengan perbandingan Gracilaria sp. dan pelarut yaitu 1:6. Parameter analisis terdiri atas proksimat, logam berat, rendemen, fitokimia, penentuan total fenol dan total flavonoid menggunakan metode spektrofotometer UV-Vis, serta aktivitas antioksidan menggunakan metode DPPH. Hasil penelitian menunjukkan kadar air 83,28% (bb), protein 9,36%, lemak 0,60%, abu 24,83% dan karbohidrat 11,05% (bk). Kadar logam berat Hg, Pb, Cd di bawah ambang batas dan tidak ditemukan logam berat As sesuai dengan SNI 2690:2015. Pengujian komponen bioaktif ekstrak etanol Gracilaria sp. secara kualitatif didapatkan alkaloid, fenol, saponin, flavonoid, triterpenoid. Kadar total flavonoid dan total fenol berturut-turut sebesar 21,78±0,32 mg QE/g dan 124±2,13 mg GAE/g. Perbedaan pelarut dalam ekstraksi berpengaruh nyata (α=0,05) terhadap aktivitas antioksidan, dengan aktivitas tertinggi adalah ekstrak etanol dengan nilai IC50 sebesar 22,15±1,63 μg/mL (sangat kuat). Hasil penelitian ini membuktikan bahwa Gracilaria sp. aman dan dapat digunakan sebagai bahan baku pangan fungsional.

Abstract

ABSTRAKKomponen bioaktif dari bahan alami telah banyak diteliti untuk digunakan sebagai obat antikanker. Penelitian mengenai komponen bioaktif dari keong matah merah (Cerithidea obtusa) sebagai antikanker belum pernah dilaporkan. Penelitian ini bertujuan: (1) Menentukan cara ekstraksi keong matah merah dengan beberapa pelarut (air, air + methanol, aseton, etil asetat, dan methanol), dan (2) Uji aktivitas zat aktif dari keong matah merah dalam melawan sel lestari kanker (K562, A 549, dan HeLa/cervix cancer). Dari penelitian dapat ditentukan bahwa waktu ekstraksi terbaik yang dipilih untuk penelitian adalah 72 jam dan perbandingan antara bahan baku dengan pelarut adalah 1:6 (w/v). Hasil penelitian menunjukkan bahwa nilai terbaik adalah ekstrak keong matah merah dengan pelarut aseton yang mempunyai daya hambat terbesar pada sel kanker (90.62% untuk HeLa/cervix cancer, 79.84% kanker paru/A 549, dan 76.71% untuk leukemia/K562) pada konsentrasi 25 ppm.Kata kunci: Antikanker, Cerithidea obtuse, ekstraksi, sel tumor lestari.ABSTRACTBioactive components derive from natural substances have been widely studied for the use as an anticancer drug. A study on a bioactive component from matah merah mollusks (Cerithidea obtusa) as an anticancer has not yet been reported. This research was intended to find the best method of extraction for the bioactive components of this animal. The research were divided into of two stages: (1) an extraction of matah merah mollusks by applying various solvents (water, water and methanol, acetone, ethyl acetate, and methanol), and (2) anti-proliferation ability of matah merah mollusks against tumor cell line (HeLa/cervix cancer, A 549, and K562). The results showed that the best extraction time was is 72 hours with the ratio of main material and solvents being 1:6 (w/v). The result also found that acetone of 25 ppm was the best solution resulted the strongest inhibiting power against cancer cells (90.62% against HeLa/cervix cancer, 79.84% against lung cancer/A 549, and 76.71% against leukemia/ K562.Key words: Anti-cancer, Cerithidea obtuse, extraction, tumor cell line.

Abstract

Hipokotil buah mangrove hitam (R.mucronata)telah terbukti mempunyai efek yang baik pada jaringan hati secara biokimia dan gambaran histopathologi[U1] [A2]  pada dosis 5 mg/kg pada tikus galur Sprague Dawley.  Penelitian ini bertujuan untuk menentukan efek toksisitas secara sub-akut dari ekstrak hipokotilbuah mangrove hitam(R.mucronata)pada tikus dengan dosis 0; 15; 10[A3] 5 mg/kg berat badan.  Rancangan percobaan yang digunakan pada penelitian ini adalah rancangan acak lengkap  dengan uji lanjut Duncan.  Hasil penelitian menunjukkan bahwa ekstrak tidak menyebabkan penurunan berat badan dan kematian tikus.  Hasil analisis secara biokimia pada darah untuk SGPT[U4] [A5] , total bilirubin, BUN, dan kreatinin masih dalam batas normal.  Gambaran secara histopathologi menunjukkan bahwa terjadi perubahan di jaringan ginjal pada  dosis 105 mg/kg berat badan.  Hasil penelitian menujukkan bahwa dosis 15 mg/kg dari ekstrak hipokotilbuah mangrove hitam(R.mucronata) dianggap aman sebagai nutrasetika. Kata kunci: buah bakau (R.mucronata), sub-akut, toksisitas

Abstract

Skin lotion merupakan produk  kosmetika yang berfungsi melembutkan dan menjaga kulit dari kekeringan. Tujuan penelitian adalah menentukan konsentrasi terbaik dari penambahan karaginan dan ekstrak Rhizophora mucronata pada skin lotion.  Tahapan dalam penelitian ini adalah formulasi skin lotion dengan penambahan karaginan (0%; 1,5%; 1%; 1,5%; 2%) dan skin lotion terbaik ditambah ekstrak Rhizopora mucronata (0,5% dan 1%). Hasil penelitian menunjukkan bahwa formula skin lotion terbaik adalah penambahan karaginan 1,5%.  Karakteristik sensori skin lotion berkisar antara agak suka sampai suka;  nilai pH 7,62; viskositas 2500 cP, stabilitas emulsi 100%, penyusutan berat 3,72%;  dan total mikrob kurang dari 2,5 x 102 Cfu (log 2,39).  Skin lotion yang ditambah 1% ekstrak Rhizopora mucronata mempunyai aktivitas antioksidan terbaik (130,494 ppm).

Abstract

Tujuan penelitian ini adalah menentukan komposisi kimia, kandungan mineral dan kelarutan  mineral  daging  remis  segar  dan  setelah pengolahan,  serta  menentukan  metode pengolahan  terbaik  dengan  kehilangan  mineral terendah  dan  kelarutan  mineral  tertinggi. Komposisi kimia remis diuji dengan metode termogravimetri, soxhlet dan kjeldahl. Kandungan mineral dan kelarutan mineral diuji dengan metode Atomic Absorption Spectrophotometer (AAS). Pengolahan menurunkan komposisi kimia remis, namun meningkatkan kandungan kadar abu setelah dilakukan metode pengolahan dengan cara perebusan garam. Metode pengolahan juga memberikan penurunan kandungan mineral kalsium, magnesium, fosfor, kalium dan seng pada remis.  Kandungan mineral natrium pada remis segar meningkat setelah direbus garam, namun menurun setelah direbus dan kukus.  Metode pengolahan juga memberikan peningkatan persentase kelarutan mineral natrium, kalsium, fosfor dan magnesium remis. Penelitian ini dapat memberikan rekomendasi kepada masyarakat bahwa untuk memperoleh asupan mineral yang paling baik dari remis, sebaiknya masyarakat mengolah remis dengan cara direbus garam dengan konsentrasi 1,5%.   Kata kunci : Corbicula javanica, kandungan mineral, kelarutan mineral, dan komposisi kimia.

Abstract

A puffer fish contains a secondary metabolite toxic called tetrodotoxin (TTX), which can be used as an anesthetic and medication to relieve chronic pain for cancer patients. The purposes of this study were to determine the LC50 value and chemical composition of extracts from various tissues of puffer fishes as a first step to determine the further isolation of tetrodotoxin. In this study various tissues from meat, liver, skin, heart, and ovary of Arothron hispidus and Diodon hystrix species were extracted, characterized, and tested levels of toxicity using Brine Shrimp Lethality Test (BSLT) methods. Between the two puffer fishes studied, the highest toxicity was found in Arothron hispidus ovary with LC50 value of 29.65 ppm. Chemical constituents of the ovary were alkaloids, carbohydrates, and amino acids. Toxic compounds found in the extracts of puffer fish ovaries were tetrodotoxin which is a compound of the alkaloid class of secondary metabolites. Abstract

The utilization of local food commodities such as corn and cassava with seaweed addition as a dietary fiber source for producing artificial rice through extrusion technology is an  alternative for food diversification. The research was carried out to find out the best composition (rice, corn, cassava, and seaweed) and temperature of extrusion process on making artificial rice and the influence of dietary fibre on sensory properties and physicochemical. The composition of rice, corn, and cassava in proportion  of 1:3:1 with 20% seaweed, Eucheuma cottonii, addition and temperature extruder of 90 °C were selected as the best product for artificial rice. The  sensory evaluation was 8.02±0.21 (people’s preference). In physicochemical properties, dietary fiber significantly affected on low bulk density and starch digestibility. This condition is very good for health especially in maintaining the stability of blood glucose in the body. Keywords: artificial rice, composition, extrusion, seaweed, dietary fibre, temperature
