Abstract

Penelitian ini bertujuan untuk mengetahui pengaruh ekstrak kedelai (Glycine max) terhadap jumlah pertumbuhan folikel ovarium pada mencit (Mus musculus). Sampel terdiri dari 20 ekor mencit yang secara acak dibagi menjadi lima kelompok. Kelompok kontrol (K) tidak diberi ekstrak kedelai. Kelompok P1 diberi ekstrak kedelai dengan dosis 0,05 mg/kgBB. Kelompok P2 diberi ekstrak kedelai dengan dosis 0,010 mg/kgBB. Kelompok P3 diberi ekstrak kedelai dengan dosis 0,015 mg/kgBB. Kelompok P4 diberikan ekstrak kedelai dengan dosis 0,020 mg/kgBB. Ekstrak kedelai diberikan selama 14 hari. Analisis data menggunakan uji One Way ANOVA dan dilanjutkan dengan uji Honestly Significant Difference (HSD). Hasil penelitian menunjukkan kelompok kontrol (K) memiliki perbedaan signifikan dengan kelompok P1, P2, P3 dan P4. Hasil rata-rata jumlah folikel primer tertinggi terdapat pada perlakuan 4 (P4) dengan dosis 0,020 mg/kgBB dan jumlah rata-rata folikel sekunder tertinggi pada perlakuan 3 (P3) dengan dosis 0,015 mg/kgBB. Penelitian ini dapat disimpulkan bahwa ekstrak kedelai yang diberikan pada mencit betina dapat meningkatkan jumlah pertumbuhan folikel ovarium dan jumlah rata-rata folikel tertinggi adalah pada perlakuan 3 (P3) dengan dosis 0,015 mg/kgBB.

Abstract

Penelitian ini bertujuan untuk mengetahui pengaruh perbedaan waktu ekuilibrasi 2 jam, 3 jam, 4 jam dan 5 jam terhadap motilitas, viabilitas, dan abnormalitas spermatozoa Sapi Rambon sebelum pembekuan. Penelitian ini menggunakan Rancangan Acak Lengkap (RAL) dengan empat perlakuan yaitu waktu ekuilibrasi P1 (2 jam), P2 (3 jam), P3 (4 jam) dan P4 (5 jam), satu ekor pejantan sapi rambon, dan lima kali pengulangan. Hasil analisis statistik Anova satu arah dan dilanjut uji Duncan mendapatkan hasil motilitas (P1) 76.00±2.2/3; (P2) 73.00±2.7/3; (P3) 67.00±2.7/3; dan (P4) 52.00±2.7/2. Hasil viabilitas (P1) 86.00±1.41; (P2) 78.00±2.73; (P3) 72.00±5.70 dan (P4) 57.00±2.73. Hasil abnormalitas (P1) 3.7±0.75; (P2) 3.8±0.75; (P3) 4.4±1.08 dan (P4) 5.2±0.57. Kesimpulan dari penelitian ini menunjukan hasil yang berbeda nyata (p<0.05) antara waktu ekuilibrasi dengan kualitas spermatozoa Sapi Rambon sebelum pembekuan dan hasil dari penelitian ini menunjukan persentase motilititas tertinggi terdapat pada waktu ekuilibrasi 2 jam, persentase viabilitas tertinggi terdapat pada waktu ekuilibrasi 2 jam dan persentase abnormalitas terendah terdapat pada waktu ekuilibrasi 2 jam.

Abstract

Penelitian ini bertujuan mengetahui persentase keberhasilan penetasan telur penyu lekang (Lepidoehelys olivacea) pada sarang semi alami Banyuwangi sea Turtle Foundation (BSTF) di Pantai Boom Banyuwangi. Data diperoleh dari observasi dan rekap data milik BSTF. Persentase keberhasilan penetasan diperoleh dari total telur penyu lekang selama periode bertelur tahun 2018. Persentase penetasan telur penyu lekang pada sarang semi alami BSTF periode tahun 2018 adalah 75%. Total telur penyu yang diinkubasikan adalah 4495 butir, total telur yang menetas adalah 3392 butir, dan jumlah telur yang gagal menetas adalah 1103 butir.

Abstract

Penelitian ini bertujuan untuk menentukan konsentrasi gliserol yang optimal dalam pengencer tris dalam mempertahankan kualitas spermatozoa kambing Sapera. Penelitian ini menggunakan kambing Sapera jantan umur 1.5 tahun. Konsentrasi gliserol yang digunakan adalah 5%, 6% dan 7%. Semen ditampung dua kali dalam seminggu dengan menggunakan vagina buatan. Hasil penelitian menunjukkan rata-rata persentase motilitas dan viabilitas spermatozoa pada pengencer dengan gliserol 6% masing-masing 62.33% dan 75.83% terdapat perbedaan yang signifikan (p<0.05) dibandingkan pada pengencer dengan gliserol 5% masing-masing 57.67% dan 68.33% dan 7% masing-masing 59% dan 70.33%. Rata-rata abnormalitas pada pengencer dengan gliserol 5%, 6% dan 7% masing-masing 8%, 7.5% dan 7.67% tidak terdapat perbedaan yang signifikan (p>0.05). Dapat disimpulkan bahwa penambahan gliserol 6% dalam pengencer Tris berhasil melindungi spermatozoa dari cekaman dingin selama proses gliserolisasi, sehingga dapat mempertahankan motilitas, vibilitas dan abnormalitas spermatozoa.

Abstract

This study aimed to determine the number of reproductive efficiency parameters including service per conception, conception rate, calving rate, and non-return rate for broilers in Kalipuro District, Banyuwangi Regency. The secondary data was collected from the Banyuwangi District Agriculture Office. This research is also intended to dissect factors related to the S/C, CR, CvR, and NRR figures with descriptive analysis, including bio reproduction mastery factors, service factors, and feed factors. The primary data was collected from beef cattle breeders in Kalipuro District, Banyuwangi Regency. In addition, there were IB implementation factors whose data were obtained from questionnaires to inseminators in Kalipuro District, Banyuwangi Regency. The number of S/C broilers in Kalipuro Subdistrict, Banyuwangi Regency was good maintenance, in January-March 2020 of 1.71, 1.99, and 1.74, meanwhile, the number of CR were 58%, 50% 57%, CvR for January-March were 44%, 84%, 53% and NRR for January, February and March were 41%, 49%, and 46%, respectively, were below the standard rate.

Abstract

The aim of the study was to detect the fertility level of females Bawean deer (Axis kuhlii) through fecal steroid metabolic which were tested using Enzyme-Linked Immunosorbent Assay (ELISA). The study used five females Bawean deer in Taman Flora Surabaya, which were marked using R1, R2, R3, R4, and R5 necklaces. The feces of each deer was collected on the 1st day, 6th day, 11th day, 16th day, and 21st day. The samples which were collected then extracted using the freeze dry method. Samples were tested using ELISA. The results of the measurement of the levels of fecal steroid metabolic  in five Bawean deers showed that three Bawean deer (R1, R4, and R5) were in the luteal or pregnant phase as indicated by the increased of progesterone from 1st day to 21st day. While the other two Bawean deer (R2 and R3) were in the follicular or estrous phase as indicated by the decreased of level of faecal steroid metabolic  on 11th day for deer R2 and 16th day for deer R3. The results showed that the levels of the fecal steroid metabolic can be used to determine the follicular phase and luteal phase and the pregnancy status of female Bawean deer.

Abstract

The Leptin gene is the gene that produces the leptin hormone, which is released from adipose tissue and can increase the productivity of animals. This study aimed to identify polymorphic nucleotides, changes in amino acid components, and species of goats based on GenBank Leptin DNA sequence data. A total of five goat leptin DNA sequences were extracted from NCBI GenBank data. The leptin DNA sequence was aligned with Bioedit to locate SNPs and amino acid changes. The tree produces cultivars grown using Clustal Omega Ver. 1.2.4. Based on the DNA sequencing results of leptin genes in five goats, five SNPs were located in the coding sequence (CDS), SNPs g.17T/A, g.43T/A, g.74G/A, g.93C/A. and d. 386A/G. SNP was a missense mutation and a silent mutation. The analysis of phylogenetic trees of Leptin showed that there were three breeds of goats in one branch and two breeds of goats in different branches. These results provided the first report for further studies on the genetic diversity of leptin genes in different local goat breeds.
