Inhibitory activities of surian or toon (Toona sinensis) leaves fraction origin Sumedang, West Java, Indonesia against wood rot fungi (Ganoderma boninense IPBCC 10.658, Trametes versicolor InaCC F200, and Phanerochaete chrysosporium IPBCC 93.259) were studied. The samples were macerated using methanol and acetone. The results of brine shrimp lethality test (BSLT) showed that the methanol extract had the highest cytotoxicity of LC50 at 29.76 ?g/mL and inhibit the growth of G. boninense (27.78%), T. versicolor (79.26%) and P. chrysosporium (81.11%). The methanol extract was then subsequently fractionated using n-hexane, diethyl ether, and ethyl acetate respectively. The diethyl ether fraction was found to have the highest inhibitory activity against T. versicolor (46.30%) and P. chrysosporium (81.11%). This fraction was further separated using column chromatography and analyzed using thin layer chromatography (TLC), which gave six fractions. Antifungal test exhibited that fraction 5 had the highest antifungal properties against T. versicolor (74.07%) and P. chrysosporium (80.37%). Inhibition of P. chrysosporium resulted in abnormal growth of hyphae morphology as indicated by changes in its growth direction and excessive hyphae branching. Additionally, the results of LC-MS/MS experiment indicated toon leaves fraction 5 which contains N-[2- (D-Glucopyranosyloxy)ethyl]-2-hydroxy-N-[2hydroxy3 (octadecyloxy) propyl] butanamide dan Brucine compounds that were regarded as the antifungal compounds.

Identification of active compounds and anti-acne activity from extracts and fractions of surian (Toona sinensis) leaves planted in Sumedang, West Java, Indonesia. Biodiversitas 19: 1406-1412. Surian or toon (Toona sinensis Merr) leave is one of the herbs containing many bioactive compounds as potential sources of anti-acne agents. The aim of this work is to identify and evaluate the anti-acne activity of Surian leaves extract and fractions against acne bacteria. Crude extracts of Surian leaves from various solvents (methanol, ethanol, and ethyl acetate) were screened for their antimicrobial activity using disc diffusion. The results showed that ethyl acetate extract demonstrated the highest inhibition against Propionibacterium acne (49.93%), Staphylococcus epidermidis (57.14%), Staphylococcus aureus (18.93%), and Eschericia coli (82.03%). The results of Brine Shrimp Lethality Test (BSLT) showed that the methanol extract had the highest cytotoxicity of LC50 at 29.76 ?g/mL. The extract was then fractionated using column chromatography, resulted in 6 fractions. The fraction 3 showed the best antibacterial effect against P. acne (28.27%), S. epidermidis (7.38%) and S. aureus (10.38%). Minimum inhibitory concentration (MIC) and minimal bactericidal concentration (MBC) test were carried out using liquid dilution, whereas dimethyl sulfoxide (DMSO) and Clindamycin were used as a negative and positive control, respectively. The MIC and MBC test assays of fraction 3 varied for different bacteria P. acne bacteria (5000 and 5000 ppm), S. aureus (1250 and 10000 ppm), E. coli (2500 and 5000 ppm), while for S. epidermidis the fraction 3 was able to inhibit with MIC value of 5000 ppm. Based on microscopic observation using Scanning Electron Microscope (SEM), the fraction 3 at the dose of 5000 ppm could promote the cellular destruction of P. acne. Further experiment using Liquid Chromatography-Tendem Mass Spectrometry (LC-MS/MS) showed various compounds associated with the anti-acne activity of the fraction 3, such as pheophorbide a, pheophytin a and dihomo-?-linolenic acid.

Biodiversitas 21: 252-257. Betel (Piper betle Linn), also called Golden Heart of Nature, is often used as traditional medicine. Nonetheless, betel plants originated from different places contains different phytochemicals profile, resulting in different utilization across ethnics. The research aimed to classify betel leaves from eastern Indonesia origins based on phytochemical profiles. In this study, the phytochemical profiles of 69 betel leave samples from 15 ethnics in eastern Indonesia were obtained using High-Performance Liquid Chromatography (HPLC) fingerprint analysis. This data was then used to classify the betel leaves samples using Principal Component Analysis (PCA). The results of the analysis show that the betel leaves from Eastern Indonesia can be divided into three clusters. Cluster 1 consisted of betel leaves originated from two ethnics, namely Komoro and Greri, from Papua, while Cluster 2 consisted of those originated of two ethnics, namely Sumber Baba and Demta, both were also from Papua. Cluster 3 consisted of betel leaves originated 11 ethnics, namely Asilulu (Maluku), Balesang (Central Sulawesi), Bungku (Central Sulawesi), Mulong Kuni (Central Sulawesi), Saluan (Central Sulawesi), Tialo (Central Sulawesi), Tolage (Central Sulawesi), Gebe (North Maluku), Makian (North Maluku), Mey Brat (West Papua) and Waigeo (West Papua). The location of P. betle plantation in this research accounts for clusterization of samples, Papua island give the highest biodiversity because we can find all the three cluster in Papua island.

Biodiversitas 25: 942-949. Gedi (Abelmoschus manihot (L.) Medik) leaves are popularly consumed as a green vegetable by Eastern Indonesians. This work aimed to carry out the phytochemical compounds and antioxidant capacities of five extracts, i.e., water, 70% ethanol, absolute ethanol, ethyl acetate, and hexane of Abelmoschus manihot leaf. Qualitative analysis was done as a preliminary screening for flavonoid, tannin, saponin, alkaloid, triterpenoid, steroid, and quinone. Quantitative analysis determined the flavonoid, phenolic, tannin, quercetin, and steroid content. In-vitro antioxidant analysis was conducted to determine free radical scavenging activity and lipid peroxidation inhibitory activity. The results showed that flavonoid was only found in 70% ethanol extract; steroid was obtained in all extracts, while alkaloid, saponin, tannin, quinone, and triterpenoid were absent in all extracts. The results of the quantitative analysis showed that the highest flavonoid was in 70% ethanol extract (39.2 mg GAE/g), phenolic in ethyl acetate extract (34.792 mg QE/g), tannin in 70% ethanol (1.395 mg/g), quercetin in absolute ethanol extract (0.246 mg/g), and steroid content in hexane extract (201.948 mg/g). Half maximal inhibitory concentration (IC50) of each extract to inhibit malondialdehyde (MDA) formation was lower than free radical scavenging activity. The IC50 values of MDA formation in absolute ethanol and hexane extracts were 6.77 ppm and 7.67 ppm, respectively. The IC50 of vitamin E was 3.67 ppm. Further study is needed to determine the mechanism of Abelmoschus manihot leaf extracts as antioxidants used as a functional food.

Nutritional composition of gedi (Abelmoschus manihot) leaves powder in comparison to common natural galactagogue plants in Indonesia. Biodiversitas 25: 2583-2589. Galactagogue is a substance believed to help initiate, maintain, and increase breast milk production. Low or no breast milk production and perceived insufficient milk are the most common reasons that cause a mother to stop breastfeeding earlier. To help this problem, natural galactagogue consumption is highly recommended. Gedi (Abelmoschus manihot (L.) Medik) plant is locally used in Indonesia as food ingredient and medicinal plant. The leaves have been believed for generations to increase breast milk production. This study aimed to evaluate the macro- and micronutrient content of gedi leaves powder, compared to torbangun (Coleus amboinicus) and katuk (Sauropus androgynous) leaves powder as common natural galactagogue plants in Indonesia. Clean fresh leaves were oven-dried and ground. Carbohydrates, crude fiber, protein, fat, ash, energy, water, vitamins A, B1, B2, B6, B9, C, E, and K, mineral calcium, iron, magnesium, sodium, zinc, phosphorus, and potassium were analyzed using standard methods. The analysis result showed that gedi leaves powder contained highest carbohydrate (51%), crude fiber (10.3%), vitamin K (1,110 µg/100 g), mineral magnesium (700 mg/100 g), sodium (89.6 mg/100 g), and calcium (2,671 mg/100 g) content, which were significantly different compared to torbangun and katuk leaves powder (P<0.05). Total energy (344 kcal/100 g), protein (25%), ash (13.6%), zinc (4.8 mg/100 g), potassium (2,314 mg/100 g), and iron (8.9 mg/100 g) content of gedi leaves powder were between torbangun and katuk leaves powder. This study showed that gedi leaves powder had a high carbohydrate, crude fiber, vitamin K, and mineral, especially calcium and magnesium, and sufficient protein and energy content, compared to torbangun and katuk leaves powder as common natural galactagogue plants in Indonesia. Further research in the extraction, bioactive compound, and in vivo study has to be carried out to scientifically prove the galactagogue effects of gedi leaf powder.
