Abstract

Liver cancer development is highly dependent on angiogenesis, the formation of new blood vessels that supply nutrients and oxygen to cancer cells, facilitating tumor growth and metastasis. Vascular Endothelial Growth Factor Receptor 2 (VEGFR2), a receptor tyrosine kinase, represents a promising anti-angiogenesis therapeutic target. Bioactive compounds from tulsi (Ocimum sanctum L.) possess potential anticancer properties, but their specific mechanisms against VEGFR2 in liver cancer require investigation. This study evaluated the interaction potential of tulsi bioactive compounds against VEGFR2 protein structure (PDB ID: 2XIR) through in silico molecular docking analysis. Drug-likeness evaluation was conducted based on Lipinski's rule of five and ADMET profiling. Molecular docking analysis revealed comparative binding performance between two tulsi compounds against VEGFR2. Cirsimaritin demonstrated significant inhibition potential with free binding energy (∆G) of -9.06 kcal/mol, inhibition constant (Ki) of 226.95 μM, and stabilizing interactions with residues PHE1047, VAL848, LEU840, and CYS1045. The native ligand exhibited superior binding affinity with ∆G of -12.68 kcal/mol and Ki of 508.94 pM, indicating greater therapeutic potential for anti-angiogenic liver cancer treatment. Overall, this study is useful for the development of the potential of tulsi bioactive compounds as angiogenesis inhibitors and alternative natural ingredient-based therapies for liver cancer. Further in vitro and in vivo studies are needed to validate the anticancer activity and mechanism of angiogenesis inhibition by these compounds.

Abstract

Ascorbic acid and its derivatives with nicotinamide are often used as skin lightening, anti-aging, and anti-pigmentation agents. Combining these two active substances may increase the risk of instability due to degradation. This research was conducted to determine the effect of nicotinamide concentration on the degradation process of ascorbic acid and its derivatives, and to determine the degradation products formed using liquid chromatography-mass spectrometry (LC-MS). The study was conducted by combining nicotinamide concentrations ranging from 5 to 15 micrograms per milliliter with incubation times of 1, 2, 3, and 24 hours to react with ascorbic acid and its derivatives. The spectral shape and maximum wavelength were analyzed using a UV-Vis spectrophotometer. Samples that showed changes in chemical structure were subsequently analyzed by LC-MS to identify degradation products. The results showed that only the combination of ascorbic acid and nicotinamide shifted the maximum wavelength and generated a new peak at 310 nm after 24 hours of incubation. Nicotinamide can accelerate the degradation process of ascorbic acid. The LC-MS analysis results show that the degradation products formed from the combination of ascorbic acid and nicotinamide samples are dehydroascorbic acid and 3-hydroxy-2-pyrone. These findings highlight the importance of incorporating stabilizing agents into cosmetic formulations containing unstable active ingredients, such as ascorbic acid, to ensure product stability during storage and use.

Abstract

Colorectal cancer (CRC) remains a leading cause of cancer-related deaths worldwide, with limited effective therapies due to drug resistance and adverse effects. Zerumbone, a sesquiterpene isolated from Zingiber zerumbet Smith, has emerged as a promising natural anticancer agent. This review examines zerumbone's anti-colorectal cancer properties, including induction of apoptosis and cell cycle arrest, inhibition of invasion and metastasis, anti-angiogenic activity, and anti-inflammatory effects. Additionally, zerumbone demonstrates antioxidant properties, modulates gut microbiota composition, and targets multiple signaling pathways involved in CRC pathogenesis. Structure-activity relationship studies reveal the critical role of the α,β-unsaturated carbonyl group in its bioactivity. Despite promising preclinical evidence, clinical validation remains necessary to establish zerumbone's therapeutic potential for colorectal cancer management.

Abstract

South Lampung Regency is the second largest cocoa bean producer in Lampung Province. This natural resource is very abundant, one of which is in Tarahan Village, South Lampung, but until now it has not been optimally processed and is only sold in the form of dry brown beans which have low selling value. This is because the villagers does not know how to process cocoa beans into cocoa products. Therefore, this service program aims to assist the target community in processing cocoa beans into ready-to-sell products (jam and mask) to improve people welfare. This activity consisted of providing material and training in making jam and chocolate masks for 1 day. After the training, the villagers were able to understand and practice making the chocolate jam and mask. This program is expected to increase chocholate selling value as local regional commodities.

Abstract

Diabetes melitus tipe 2 merupakan penyakit metabolik kronis yang memerlukan terapi jangka panjang dengan efek samping yang minimal. Salah satu target terapi potensial adalah enzim α-glukosidase, yang berperan dalam pemecahan karbohidrat menjadi glukosa. Dalam studi ini, tujuh senyawa dari Erythroxylum laurifolium dievaluasi menggunakan pendekatan in silico untuk menilai potensinya sebagai inhibitor α-glukosidase. Hasil evaluasi menunjukan bahwa meskipun sebagian besar senyawa tidak memenuhi kriteria Lipinski’s Rule of Five, efektivitasnya tetap terjamin karena aksi lokal di lumen usus. Senyawa prosianidin B1 memiliki nilai energi bebas Gibbs (ΔG) sebesar –6,93 kkal/mol dengan konstanta inhibisi 8,30 µM, diikuti oleh prosianidin B2 dengan ΔG –5,74 kkal/mol dan konstanta inhibisi 61,90 µM. Afzelin, yang memenuhi tiga dari empat kriteria RO5, menunjukkan ΔG sebesar –5,14 kkal/mol dengan konstanta inhibisi 169,65 µM. Berdasarkan afinitas pengikatan dan potensi farmakologis, prosianidin B1, prosianidin B2, dan afzelin adalah kandidat kuat sebagai inhibitor α-glukosidase. Penelitian lebih lanjut diperlukan untuk mengonfirmasi efektivitas dan keamanannya melalui studi in vitro dan in vivo.
