Abstract

Peningkatan keamanan pangan menggunakan bahan alami, seperti kunyit, kencur, dan temulawak sebagai suplemen pada pakan disebabkan karena bahan tersebut memiliki sifat yang ramah lingkungan, biodegradable, dan mudah ditemukan di alam. Penelitian ini bertujuan untuk mendapatkan dosis suplemen herbal dan padat tebar yang efektif dalam meningkatkan status kesehatan ikan ditinjau dari profil hematologis. Rancangan yang digunakan adalah Rancangan Acak Lengkap pola Faktorial. Perlakuan yang diberikan adalah dosis suplemen herbal (D1= 100 mL/kg pakan, D2= 200 mL/kg pakan, dan D3 = 300 mL/kg pakan) dan padat tebar ikan P1 = 50 ekor/ m3 dan P2 = 75 ekor/ m3. Ikan patin (Pangasius hypophthalmus) yang digunakan berukuran 8-12 cm, pemeliharaan dilakukan selama 60 hari di keramba jaring apung berukuran 1,5x1x1m. Pengambilan sampel darah dilakukan sebanyak tiga kali, yakni sebelum perlakuan, kedua pada hari ke-30 dan hari ke-60 pemeliharaan. Hasil penelitian menunjukkan bahwa penambahan dosis suplemen herbal 200 mL/kg pakan dan padat tebar 75 ekor/m3 yang dipelihara selama 60 hari memberikan hasil tertinggi terhadap profil hematologis, yaitu total eritrosit sebesar 2,81 x 106 sel/mm3, nilai hematokrit 39,00%, kadar hemoglobin 10,73 g/dL, total leukosit 11,41 x 104 sel/mm3 dan kelulusanhidup 100%. Hasil penelitian dapat disimpulkan bahwa kombinasi padat tebar 75 ekor/m3 dan dosis suplemen herbal 200 mL/kg pakan adalah yang terbaik untuk peningkatan kesehatan ikan patin.

Abstract

This study aimed to observe the inhibition capability of Rhizophora sp. leaf extract towards the Streptococcus agalactiae and Edwardsiella tarda bacteria. Rhizophora sp. leaf was extracted using ethanol. Inhibition action of Rhizophora sp. leaf extract towards Streptococcus agalactiae and Edwardsiella tarda was tested on TSA solid media. The concentration of Rhizophora sp. leaf extract used were 1000, 2000, 3000, 4000, 5000, 6000, 7000, 8000, 9000, 10000 ppm, and novobiocin antibiotic was used as a control. Each treatment was conducted three times. MIC (minimum inhibition concentration) was performed to determine the minimum dose that could inhibit the bacterial growth. The results showed that the inhibition value of Rhizophora sp. leaf extract at 2000 to 10000 ppm towards Streptococcus agalactiae was 8.60-16.30 mm, and 6.97-12.27 mm towards Edwardsiella tarda, whereas the inhibition value of novobiocin for both bacteria was 18.00-20.45 mm. The results of MIC value at dose of 2.000 ppm of Rhizophora sp. leaf extract was towards Streptococcus agalactiae with bacterial density of 165x108 CFU/mL, and towards Edwardsiella tarda 75x108 CFU/mL, respectively. In conclusion, Rhizophora sp. leaf extract had more bacteriostatic activity against Gram-positive bacteria (Streptococcus agalactiae) rather than the Gram-negative bacteria (Edwardsiella tarda).

Abstract

This study was conductedto assess the physiological responses of commoncarp due to administrationof ich vaccines and kept at different water temperatures. Completely Randomized Factorial Designwasusedin this study. The number and the size of the experimental fish was720 and 5-7 cm, respectively. Fish wasvaccinated by immersing the fish in water that contained 3 mLL-1 of ich vaccines for 15 minutes. Then, fishwas kept for 21 days at various temperatures: at 18ºC which was room temperature without heater andfish had not been vaccinated and at 20, 24, and 28ºC where fish was given the vaccines and the aquariumis facilitated with electric heater. Atday15post immunization, challenge test with 5.000 cells of life therontfish-1was performed.Immobilization test, differential leukocyte, prevalence rates, and survival rates weremeasured. The measurements of these parameters were performed four times:before the treatment (day0), day7, day14, and day21, respectively. The results showed that the vaccine could reduce the stress inthefish that were kept at 20, 24, and 28ºC, and the prevalence of ichthyophthiriasis. In addition the vaccineincrease the fish survival rates after challenge test. The highest survival rate (100%) was found in fish thatwerekept in 28ºC.

Abstract

An experiment on the immunization of Jambal Siam ( Pangasius hypophthalmus) withIchthyophthirius multifiliis vaccine was conducted in the Laboratorium of Parasitic and Fish Diseases,Faculty Fisheries and Marine Science of Riau University, and Laboratorium of Fish Health of FacultyFisheries and Marine Science, IPB. The objective of the study was to enhance immune system of the fishfry on the ichthyophthiriasis diseases. The vaccine was prepared by prolonged treatment of theron in waterbath of 47°C for 30 minutes. The vaccine was administrated to fish by immersion in aquaria. A completelyrandomized design (CRD) in factorial pattern (3 X 3 X 3) was carried out with dose and time as factors.Doses of treatment and time of immersion were of 1 ml/L, 2 ml/L and 3 ml/L as well as 15, 30 and 45minutes of treatment time respectively. To evaluate the effectiveness of vaccine in fish, a challenge test tofish was done at day 15 with theron 90.000 cell/ aquaria until rearing day 25 after vaccination. It revealedthat the best performance (p<0.5) were of dose 3 ml/L and time treatment of 15 minutes in that survivalrate of fish was 100% followed by dose 3 ml/L with time 45 minutes (63,3%). The total erythrocyte count,hematocrit level and hemoglobin of tested fish were fluctuated. The water quality were recorded including;dissolved oxygen range from 3,74 - 4,98 ppm; temperature 25 - 30°C and acidity of 4 - 6, quality were ofnormal range for fish. The optimal vaccine dose is 3 ml/l with 15 minute immersion.

Abstract

Senyawa kimia yang terkandung dalam kulit kayu manis (Cinnamomum burmannii) berfungsi sebagai antibakteri dan imunostimulan yang dapat meningkatkan respons imun non spesifik pada ikan. Penelitian ini dilakukan pada bulan Maret-Agustus 2023 di Laboratorium Parasit dan Penyakit Ikan, Fakultas Perikanan dan Kelautan, Universitas Riau. Tujuan penelitian adalah untuk mengetahui status kesehatan ikan jambal siam (Pangasianodon hypophthalmus) yang diuji tantang dengan Aeromonas hydrophila. Metode yang digunakan adalah metode eksperimental dengan Rancangan Acak Lengkap (RAL), satu faktor, lima perlakuan, yaitu Kontrol negatif (tanpa pemberian larutan kulit kayu manis dan tanpa diuji tantang A. hydrophila); Kontrol positif (tanpa pemberian larutan kulit kayu manis dan diuji tantang A. hydrophila); P1, P2, dan P3 dosis penambahan larutan kulit kayu manis 15, 30, dan 45 mL/kg pakan dan diuji tantang dengan A.Hydrophila. Masing-masing perlakuan memiliki tiga ulangan. Ikan uji berukuran 8-12 cm dipelihara dalam akuarium berukuran 40x30x30 cm3 dengan padat tebar 1 ekor/3L. Pakan diberikan tiga kali sehari dengan dosis 5% dari bobot biomassa. Ikan diuji tantang pada hari ke-32 secara intramuskuler sebanyak 0,1 mL/ekor dengan kepadatan bakteri 108 CFU/mL. Hasil penelitian diperoleh nilai hematologi ikan pada perlakuan P2 pascauji tantang berada dalam kondisi normal (total eritrosit 2,30 x 106 sel/mm3 , kadar hemoglobin 10,53 g/dL, nilai hematokrit 45,00%, total leukosit 10,85 x 104 sel/mm3, aktivitas fagositosis 32,33%, kelulushidupan 93,33% dan tingkat perlindungan relatif 91.07%). Sebagai simpulan, penambahan kulit kayu manis dengan dosis 30 mL/kg pakan dapat mencegah Aeromoniosis dan meningkatkan kekebalan ikan terhadap infeksi A. hydrophila.
