Abstract

Mikroalga epifit merupakan organisme bentik yang hidup menempel pada berbagai habitat atau substrat makroalga. Kelimpahan dan keanekaragaman jenis mikroalga berpotensi sebagai bioindikator pencemar kualitas perairan. Penelitian ini bertujuan untuk mengetahui struktur komunitas mikroalga epifit berasosiasi pada padina sp. di perairan Desa Sepempang, Kabupaten Natuna. Penelitian dilaksanakan pada bulan Januari-Juni 2019. Pengambilan sampel makroalga dan pengukuran parameter lingkungan dilaksanakan secara in situ dan pengambilan data fisika-kimia dilkukan dalam tiga hari pada musim timur pada bulan Januari dengan metode survey dan penentuan stasiun pengambilan sampel secara purposive sampling yang terdiri dari empat stasiun dengan rona lingkungan yang berbeda yaitu zona daerah dermaga, zona daerah pemukiman, zona budidaya ikan dan zona non aktivitas. Hasil penelitian mendapatkan komposisi kelimpahan 9 kelas, 41 ordo, 58 famili dan 81 genus. Mikroalga yang dominan ditemukan adalah kelas bacillariophyceae dengan persentase kelimpahan (70,03%), genus yang mendominan ditemukan adalah Navicula, synedra, nitzschia, cocconeis, pinnularia dan grammatophora. Nilai indeks keanekaragaman (H') berkisar 3.61-3.93, indeks keseragaman (E') berkisar 0.85-0.91, indeks dominansi (C) berkisar 0.02-0.04 dan nilai indeks similarita Sorensen berkisar 81.53-92.62%. Parameter lingkungan perairan Desa Sepempang yang mempengaruhi kelimpahan dan pertumbuhan mikroalga epifit yaitu suhu, kecepatan arus, kedalaman dan oksigen terlarut.

Abstract

Mikroalga merupakan salah satu komponen penyusun perifiton yang hidupnya melekat pada substrat, baik substrat alami maupun substrat buatan. Perifiton memiliki peran salah satunya sebagai bioindikator. Penelitian ini bertujuan untuk menganalisis komunitas mikroalga perifiton pada substrat berbeda yaitu substrat alami (batu) dan substrat buatan (kaca objek glass) serta untuk menganalisis status mutu perairan berdasarkan pendekatan perifiton. Lokasi penelitian yaitu perairan Ranu Pakis Lumajang dengan 3 stasiun pengambilan sampel secara purposive sampling. Metode penelitian adalah survei dan dilakukan pada musim barat tahun 2019 dengan waktu pengambilan sampel setiap dua minggu sekali selama enam minggu. Parameter yang dianalisis meliputi identifikasi mikroalga perifiton, penghitungan kelimpahan, indeks biologi, dan pengukuran kualitas air. Data yang diperoleh dianalisis secara statistik menggunakan one way ANOVA dan regresi linear berganda menggunakan Ms. Excel dan SPSS 23.0. Hasil penelitian menunjukkan bahwa mikroalga perifiton yang ditemukan berasal dari empat divisi yaitu Chrysophyta (47%), Chlorophyta (27%), Cyanophyta (26%), dan Pyrrophyta (0,2%). Secara keseluruhan kelimpahan yang diperoleh berkisar antara 5.567-34.841 sel/cm2 (substrat alami) dan 14.367-42.563 sel/cm2 (substrat buatan). Indeks keanekaragaman di perairan Ranu Pakis tergolong tinggi sehingga perairan tergolong bersih (stabil), kecuali pada stasiun 3 tergolong tercemar ringan (kurang stabil). Akan tetapi, tidak ada dominansi pada seluruh stasiun tersebut. Hasil pengukuran parameter kualitas air menunjukkan bahwa secara umum nilai kualitas air masih berada di bawah ambang baku mutu yang ditetapkan oleh pemerintah PP No.82 Tahun 2001 untuk kegiatan perikanan kelas III. Komponen kualitas air seperti karbondioksida, kecerahan, nitrat, orthofosfat, dan suhu secara simultan berpengaruh terhadap kelimpahan mikroalga sebesar 39,7%.Periphyton microalgae  live by attaching to the substrate, both natural and artificial substrate. Perifiton plays role as an aquatic environment bioindicator. This study aimed to analyze periphyton microalgae community in different substrate, natural substrate (stone) and artificial substrate (objeck glass). Furthermore, the microalgae periphyton are used to analyze the environmental status quality. Survey method was used and sampling location is in three site of the Ranu Pakis environment by purposive sampling. Research was carried out in 2019 every two week during six weeks on west season. Identification of periphyton microalgae, density counting, biological indexes, and water quality measurements was taken during the research. All data was analyzed statistically by using one way ANOVA and multiple linear regression by using Ms. Excel and SPSS 23.0. The results depict periphyton microalgae consists of Chrysophyta (47%), Chlorophyta (27%), Cyanophyta (26%), and Pyrrophyta (0,2%). The density of microalgae periphyton was ranging from 5.567 to 34.841 sel.cm-2 (natural substrate) and 14.367 to 42.563 sel.cm-2 (artificial substrate). Biological index indicates that Ranu Pakis environment was belonging to stable (unpolluted) in site 1 and 2, while 3 was belonging to less stable (less polluted). However, there was no domination in all sites. Water quality values provide information that in general it still under of quality standards determined by PP No. 82 year 2001 for fisheries activites class III. Water quality components  including carbondioxide, transparency, nitrate, orthophosphate, and temperature simultantly influence the microalgae density of 39,7%.

Abstract

Marine macroalgae are potential sources as carbohydrate alternative, food additive, and bioactive compounds with antioxidant activity. The biological activity is strongly influenced by the type and phenolic compounds concentration. Tropical brown algae Padina pavonica Hauck was found with a large abundance in Kabung Island, West Kalimantan. The objective of this study was to determine the phytochemical screening, total phenolic content, and antioxidant activity of P. pavonica Hauck extract. The phytochemical screening and determination of total phenolic compounds were carried out using two different solvents with ethanol and ethyl acetate. Whereas antioxidant activity was evaluated using the DPPH (1,1-diphenyl-2-picrylhydrazyl) free radical scavenger method with UV-vis spectrophotometer. The qualitative analysis of phytochemical constituents revealed the presence of alkaloids, steroids, flavonoids, phenols, and saponins. Furthermore, for total phenolic compounds, the ethanolic extract had a higher content (20.34 mgGAE/g) dry sample than ethyl acetate extract (7 mgGAE/g). The ethanolic extract also had a higher of potential antioxidant activity which IC50 was 144.47 ppm compared to ethyl acetate extract (IC50 was 191.98 ppm).

Abstract

Macroalgae is marine biological resources that play a crucial role and have an important economic value. They synthesize bioactive compounds with different kind of biological activities, such as antioxidant, antitumor, including antibacterial. In the health sector, infectious diseases from bacteria is one of the problems that still increasing. In addition, E. coli besides having the capacity to be pathogenic agent, it also showed mutlidrug resistence (MDR). Antibiotic resistance is a serious problem worldwide. The increase of this phenomenon leads to the exploration of new antibiotics using natural resources as an effort to overcome the problem. Padina pavonica Hauck belongs to Phaeophyceae spreeding along Kabung Island waters, but its existence has not been widely used. The objective of this study was to evaluate antibacterial activity from ethanol and ethyl acetate extract of P. pavonica Hauck from Kabung Island, West Kalimantan against E. coli. The extraction was  made using ethanol and ethyl acetate solvents, and the antibacterial activity test was realized with concentration series of 5 ppm, 10 ppm, 15 ppm, and 20 ppm. The quantitative assessment of the antibacterial test showed that both ethanol and ethyl acetate extracts of P. pavonica Hauck had potential antibacterial activity against E. coli. The inhibition zone of ethanol extract was 11.6 mm, while ethyl acetate extract was 12.6 mm, respectively. These two solvents were included to the bacteriostatic category.

Abstract

Estuaries are biogeochemical hot spots; they receive large inputs of nutrients from land and oceans to support high primary productivity rates. Estuaries also as a place for waste disposal allowing the accumulation that endangers the environment and organisms. Water quality assessment can be done by analyzing physical, chemical, and biological characteristics using microalgae. This study aimed to analyze the microalgae community as aquatic environment bioindicator. Survey method was used and sampling location was in four different sites of Peniti Estuary by purposive sampling. Identification of microalgae, density, biological indexes, and water quality measurements was taken in this research. The result showed microalgae community consists of 68 genera and Euglenophyceae was a component of microalgae with the highest percentage of abundance (60.93%). The most commonly found genera were Trachelomonas, Phacus, Lepocinclis, and Sphaerellopsis. The abundance of microalga was ranging from 0.5-2141.5 ind/L. Biological indexes indicate that Peniti estuary environment was belonging to moderately polluted and water eutrophication. The abundance of microalgae was influenced by physico-chemical factors such as temperature, current, nitrate and phosphate content.

Abstract

Phytoplankton plays an important role in marine ecosystems as  primary producer, as the basis of food chains and the food web, and are widely used as bioindicators to monitor water condition. The study of phytoplankton is the primary interest to explore aquatic resources for blue biotechnology applications in conditions as a live feed, antibacterial, antiviral, antioxidant, pharmaceutical, cosmetics, possibly food and health industry. The study aims to assess the composition, abundance and diversity of phytoplankton community in Lemukutan Island waters of West Kalimantan. The present study found 31 genera of phytoplankton, consisted of Bacillariophyceae (28 genera) and Dinophyceae (3 genera). In term of contribution, Bacillariophyceae were found to be dominant (93.035%) than dinophyceae (6.965%). The abundance of phytoplankton varied between 636.91 to 2034.48 cell.L-1. The diversity index (H’), the evenness (E) index, and the dominance (C) index ranged from 1.959–2.579, 0.582–0.868, 0.094–0.283, respectively. The result showed that the diversity index was moderate, the evenness index was high, and the dominance index was low.

Abstract

Free radicals contribute to human health problem resulting in various human diseases, including central nervous system injury, cancer, inflammations, and the decrease of organ function related to oxidation. This condition has encouraged the effort of finding new natural antioxidant and anti-inflammatory sources. Macroalgae act as excellent natural resources due to their bioactive potential with diverse applications in various fields. Eucheuma cottonii belonging to Rhodophyceae grow abundantly along Lemukutan Island waters, however, their existence has not been exploited. This study aims to evaluate antioxidant and anti-inflammatory activities of ethanol extract of E. cottonii from Lemukutan Island waters, West Kalimantan. The observation of antioxidant activity was done using the method of DPPH (1,1-diphenyl-2-picrylhydrazyl) with UV-vis spectrophotometer, while anti-inflammatory acyivity activity was determined using the RBCs membrane stability method. The ethanolic extract of E. cottonii had potential antioxidant activity with IC50 of 127.75 ppm and was classified as moderate category. Extracts showed also anti-inflammatory acyivity activity with the concentration of 219.83 ppm. Red macroalgae E. cottonii can be used as potential natural antioxidant and anti-inflammatory agent.

Abstract

Seagrass is one of the important ecosystems and habitats for marine organisms. Thalassia hemprichii is one of the seagrass species in Kabung Island Waters. Studies on seagrass ecosystem conditions must be carried out as initial information for integrated aquatic management. Analysis of seagrass ecosystem conditions includes density and coverage percentage. Seagrass data was collected using the line transect method with a length of 50 cm and quadrants transect of 1 m x 1 m perpendicular to the coastline toward the sea. The density of T. hemprichii at Stations I and II were 50.83 and 68.27 ind/m2, respectively. The average coverage percentage at Stations I and II were 5.05% and 5.85%, respectively. These ecosystem conditions could be categorized as rare density. Physico-chemical parameters of the waters of Kabung Island including temperature, salinity, pH, dissolved oxygen (DO), water visibility, depth, and current have been carried out. The Kabung Island waters have a salinity of 32-33‰, temperature 29-30oC, and pH 8.0-8.4. The depth and water visibility of the seagrass habitat was 0.6 m. The current velocity was 0.07-0.29 m/s. These parameters were optimum conditions for seagrass life. However, the dissolved oxygen in Kabung Island was low at 2.2-4 mg/L.

Abstract

Pulau Kabung terletak di Kabupaten Bengkayang, Kalimantan Barat dengan jumlah penduduk 600 orang yang memiliki mata pencaharian utama sebagai nelayan. Kondisi cuaca yang ekstrim dengan gelombang tinggi mengakibatkan penurunan produktivitas hasil tangkapan, sehingga masyarakat melakukan aktivitas berkebun dengan memanfaatkan lahan yang ada. Cengkeh dan Pala merupakan komoditas perkebunan yang memiliki banyak manfaat di berbagai bidang. Namun, daun cengkeh yang berguguran dan limbah hasil pengolahan daging buah pala belum banyak dimanfaatkan dan cenderung menjadi sampah. Pelatihan pembuatan minyak atsiri dalam memanfaatkan limbah perkebunan telah dilaksanakan sebagai upaya pemberdayaan masyarakat. Dalam satu kali produksi membutuhkan bahan baku 200 kg dan proses destilasi selama 3-4 jam. Penyulingan daun cengkeh menghasilkan 4-5 L minyak, sedangkan daging pala menghasilkan 2-4 L. Minyak pala dijual dengan harga Rp. 20.000/botol ukuran 8 mL atau Rp. 750.000/L.

Abstract

Eko enzim adalah cairan hasil fermentasi limbah organik berupa kulit/daging buah-buahan dan sayuran dengan menggunakan substrat gula aren atau sari tebu (molase) dan air dengan perbandingan 3 : 1 : 10, selama waktu 3–6 bulan. Enzim yang dihasilkan berupa cairan kental berwarna coklat tua dan bau asam yang kuat. Enzim ini memiliki manfaat yang besar terhadap lingkungan baik udara maupun tanah, serta manfaat ekonomi bagi keluarga. Kegiatan Pengabdian pada Masyarakat (PKM) berupa pelatihan pembuatan eko enzim dari limbah sayur dan buah-buahan dilakukan pada masyarakat Kecamatan Teluk Batang Kabupaten Kayong Utara bertujuan untuk mengedukasi masyarakat khususnya ibu rumah tangga dalam mengeliminir pembuangan limbah dapur ke lingkungan yang dapat terakumulasi sehingga menyebabkan kerusakan lingkungan udara dan tanah, bahkan kadang menghambat saluran air pembuangan yang pada akhirnya dapat menyebabkan banjir saat musim hujan tiba. Kegiatan pelatihan dilakukan secara offline dengan mematuhi protokol kesehatan secara ketat. Kegiatan ini diikuti oleh 15 orang yang terdiri dari ibu rumah tangga, dengan metode edukasi melalui ceramah tentang manfaat enzim dari limbah organic sayuran dan buah-buahan serta praktek membuat eko enzim dari limbah rumah tangga yang masing-masing dibawa dari rumah untuk difermentasi. Antusiasme dan perhatian peserta dalam mengikuti kegiatan pelatihan ini sangat besar, terindikasi dari kemampuannya menjawab pertanyaan-pertanyaan post test yang diberikan oleh tim pelaksana setelah sebelum kegiatan diberikan pre test dengan kontent pertanyaan yang sama, peningkatan pengetahuan peserta dari yang rata-rata 30% menjadi 90-100%. Karena pelaksanaan pembuatan enzimnya dilakukan pada 19 Agustus 2021, cairan enzim dari proses fermentasi ini dapat dipanen pada 30 hari kemudian.
