Abstract

Ashitaba (Angelica keiskei) is one of the plants cultivated in the Sembalun region, Lombok, NTB, which has the potential to be developed as a source of antioxidants. This study aims to determine the antioxidant activity of the methanol extract of ashitaba herb by measuring the inhibition against DPPH (1,1-diphenyl-2-picrylhydrazyl) radicals using a UV-Vis spectrophotometer. Ashitaba herb samples were collected in Jorong Hamlet, Sembalun Bumbung Village, Sembalun District, East Lombok Regency at coordinates 8°23'5.273" LAT - l16°32'12.016" EAST. The sample was then processed into simplisia with a simplisia yield of 14.75%. The simplisia was dechlorinated with n-hexane solvent and then extracted with 80% methanol solvent using the sonic method, giving a methanol extract of ashitaba herb yielding 20.30%. DPPH radical inhibitory activity was measured at a maximum wavelength of 516 nm and an operating time (OT) of 30 minutes. The methanol extract of ashitaba herb has an IC50 value of 129.40 ± 7.36 ppm, which shows moderate antioxidant activity, with a standard compound comparison of ascorbic acid with an IC50 value of 2.37 ± 0.06 ppm, which is classified as very potent.

Abstract

To determine the free-radical scavenging activity and total phenolic content of the ethanol extract of Poinsettia leaves (Euphorbia pulcherrima Willd.) from Lombok Island, we used the 1,1-diphenyl-2-picrylhydrazyl method and expressed the total phenolic content as gallic acid equivalent (GAE) per gram of ethanol extract. The results show the strong antioxidant activity (79.77 ppm) of the ethanol extract of red Poinsettia leaves and the moderate antioxidant activity of green Poinsettia leaves (118.350 ppm). The ethanol extracts of red and green Poinsettia leaves possessed an average of 63.276 ± 0.228 and 22.715 ± 0.090 mg GAE/g extract total phenolic contents, respectively. In addition, the ethanol extracts of red and green Poinsettia leaves exhibited moderate and strong free-radical inhibitory activities, respectively. These findings indicate that color affects the antioxidant ability of leaves and their phenolic content.

Abstract

Inflammation is a manifestation of the immune response to eliminate antigens and a process to respond to tissue damage. There are various drugs to control and suppress inflammation, referred to as anti-inflammatory agents. One method to test anti-inflammatory activity is the inhibition of Bovine Serum Albumin (BSA) protein denaturation. This review article will discuss the stages and optimal conditions in the inhibition of BSA protein denaturation method using various leaf extract samples. Literature was gathered from several journal database websites such as PubMed, ScienceDirect, and Google Scholar with keywords "anti-inflammatory," "inhibition of BSA protein denaturation," and "leaf extract," spanning publication years from 2002 to 2023. The results of anti-inflammatory activity tests can be expressed as either percentage inhibition or IC50 values. Optimal conditions were obtained by preparing a 0.2% BSA solution with Tris Buffer Saline (TBS), maintaining a 9:1 composition ratio between the BSA solution and the sample, and heating at an optimal denaturation temperature of 70 °C. Before conducting tests using this method, it is crucial to ensure that the suspected active compounds with anti-inflammatory activity are not degraded at this heating temperature.

Abstract

Pineapple (Ananas comosus) is a source of protease enzymes. The protease enzyme present in pineapple is the bromelain enzyme. Bromelain enzymes are widely used in various industrial fields. The purpose of this experiment was to isolate the bromelain enzyme from pineapple plants, to test the activity of the enzyme, and to determine the optimum temperature of the enzyme. Isolation of the bromelain enzyme from pineapple was carried out by precipitating the pineapple fruit filtrate using table salt (NaCl). Then the crude extract of the enzyme obtained was tested on pieces of meat with variations in the testing temperature (room temperature, hot, and cold). The yield of the crude extract of the enzyme obtained from the experiment was 40%. The results of testing the bromelain enzyme activity of pineapple showed that room temperature was the best temperature for the enzyme to tenderize meat compared to hot and cold temperatures.

Abstract

Amylase enzyme is a starch hydrolyzing enzyme that has wide applications in the food, beverage and biotechnology industries. One source of the amylase enzyme that is easily obtained is plant seeds and sprouts. This research is a preliminary study to qualitatively test the activity of the crude extract of the amylase enzyme from plants. Amylase enzyme was obtained from green bean sprouts, fresh and dried peanut seeds, and fresh and dried corn seeds. The crude extract of the amylase enzyme was tested for its activity using 1% starch as a substrate with variations in temperature and amount of substrate. The result of starch hydrolysis in the form of reducing sugar was detected using Benedict's reagent. The experimental results showed that the amylase enzyme extracted from green bean sprouts and fresh and dried corn kernels showed activity in breaking down starch at 25oC using 6 mL substrate concentration, while dried peanut seeds showed no activity. The enzyme activity is influenced by temperature and the amount of substrate.

Abstract

Continuous and repeated use of cooking oil at high temperatures accompanied by air and water contact can result in a hydrolysis reaction. The quality of the oil can be determined by carrying out the procedure for determining the free fatty acid content (FFA). Used cooking oil can be purified using an adsorbent/absorbent. Sugarcane bagasse and corn cobs have been widely used as adsorbents, but there has been no study comparing the increase in oil quality with these two adsorbents. This study aims to compare the color of the used cooking oil immersion with bagasse adsorbent and corncob charcoal adsorbent and determine the value of FFA levels of used cooking oil before and after soaking with bagasse adsorbent and corncob charcoal arsoben. The method used is fatty acid adsorption method and alkalimetric titration. From the observations, it was found that the color of the used cooking oil was fading after soaking with 3 types of adsorbents. The color of the immersion with a mixture of 1:1 adsorbent corncob charcoal and bagasse has the highest brightness level. FFA levels of used cooking oil after soaking also decreased from 0.207% to 0.141%; 0.132 %; and 0.094 %. The FFA content of used cooking oil by immersion in a 1:1 mixture of corn cob charcoal and bagasse was the lowest, which means the oil has the highest quality. It can be concluded that a mixture of 1:1 adsorbent of bagasse and corncob charcoal is the best adsorbent.

Abstract

High intensity of ultraviolet (UV) radiation causes several adverse effect such as erythema, sunburn, pigmentation, early senescence, and skin cancer. These effects can be prevented by using sun protection product. Wali (Brucea javanica L. Merr) is one of natural plants that contain phenol and flavonoid. These compounds are proven to have sunscreen effect. The purposes of this studies are to formulate and evaluate the effectivity of oil-in-water-type sunscreen cream using 1% ethanol extract of Wali seed. The sunscreen cream is developed by using a fusion method and the evaluation of the effectivity is conducted in vivo on the skin of mice (Mus musculus). The formulated sunscreen cream is then tested for its physical properties, including homogenity, pH, spreadability, adhesion, and stability. The effectivity of the formulated cream is also tested by observing the formation of erytema on the skin of mice (Mus musculus). Erythema appearing on the mouse skin is analyzed with ImageJ and SPSS v23 using the one way anova method. Abstract

Radikal bebas merupakan molekul sangat reaktif yang dapat menyebabkan kerusakan sel, kerusakan jaringan dan proses penuaan dini. Perlindungan terhadap serangan radikal bebas membutuhkan senyawa antioksidan. Juwet (Syzygium cumini L.) merupakan salah satu tumbuhan lokal yang dapat menjadi sumber antioksidan alami. Penggunaan daun juwet sebagai antioksidan akan lebih mudah dalam bentuk sediaan emulgel. Tujuan dari penelitian ini adalah untuk mengetahui aktivitas antiradikal bebas ekstrak daun juwet serta menformulasikannya menjadi emulgel. Penelitian yang dilakukan meliputi ekstraksi dengan metode maserasi menggunakan pelarut etanol 70%, lalu dilakukan uji kualitatif fitokimia dan uji aktivitas antiradikal bebas pada ekstrak. Setelah itu dilakukan formulasi emulgel, uji sifat fisik serta uji aktivitas antiradikal bebas pada emulgel dengan metode DPPH. Hasil penelitian yang diperoleh menunjukan bahwa esktrak etanol daun juwet positif mengandung flavonoid, tanin dan polifenol serta memiliki aktivitas antiradikal bebas yang sangat kuat dengan nilai IC50 sebesar 40,36 ?g/mL dan sediaan emulgel ekstrak daun juwet memiliki aktivitas antiradikal bebas yang kuat dengan nilai IC50 sebesar 94,44 ?g/mL. Hasil uji sifat fisik dari formula sediaan emulgel telah memenuhi syarat pH, homogenitas,daya sebar, dan daya lekat.

Abstract

Glukomanan merupakan polisakarida yang tersusun oleh unit ?-D-glukosa dan ?-D-mannosa yang terikat dengan gugus asetil melalui ikatan b-1,4 dan b-1,6 glikosida. Glukomanan terdapat pada dinding sel beberapa spesies tumbuhan terutama diekstraksi dari umbi genus amorphophallus spp. Secara komersial, pemanfaatan glukomanan sangat luas yaitu khususnya dalam industri farmasi digunakan sebagai bahan pengisi, pengikat tablet, pengental, gelling agent, film former, coating materials, emulsifier, stabilizer dan drug delivery system. Tujuan dari penelitian ini yaitu untuk menentukan kadar glukomanan yang diekstraksi dari umbi porang di Nusa Tenggara Barat. Penelitian yang dilakukan meliputi pembuatan tepung porang, ekstraksi glukomanan menggunakan pelarut etanol 60% dan uji kuantitatif glukomanan dengan metode DNS (Dinitro Salisilic Acid) menggunakan instrumen spektrofotometer uv-vis. Hasil penelitian yang diperoleh yaitu rendemen tepung glukomanan sebesar 66,26% dengan kadar glukomanan hasil pemurnian sebesar 38,5312±0,5%.

Abstract

Antidesma bunius L. Spreng is one of the plants that can use for antioxidants. The antioxidant activity of buni leaves caused by the presence of flavonoid compounds that potentially scavenge free radicals. This study aimed to identify the flavonoid compounds in buni leaves and compare the antioxidant activity between infusions of dried and fresh simplicia of buni leaves. This research procedure includes the collection of materials, determination of plants, made simplicia and infusions, identification of flavonoid compounds, and quantitative antioxidant activity test using the DPPH free radical method (1,1-diphenyl-2-picrylhydrazyl). The result obtained in the IC50 values from the linear regression equation between % inhibition and the concentration of the test solution. The IC50 values have been analyzed by a non-parametric statistical test with a 95% confidence level. The result of flavonoid identification showed the infusion contains flavonoid. The IC50 values of infusion from dried and fresh simplicia of buni leaves were 197,344±0,819 ppm and 303,646±3,731 ppm. Dried simplicia on antioxidant classification have weak antioxidant activity, while fresh simplicia have very weak antioxidant activity. The infusion from fresh and dried simplicia of buni leaves has significantly different antioxidant activity (p?0,05). Dried simplicia is more recommended for making infusions for an antioxidant. Keywords: Antidesma bunius, antioxidants, infusion   Abstrak Daun buni atau Antidesma bunius L. Spreng merupakan salah satu tanaman yang dapat dimanfaatkan sebagai sumber antioksidan. Tujuan penelitian ini adalah untuk mengidentifikasi kandungan senyawa flavonoid daun buni dan mengetahui perbandingan aktivitas antioksidan sediaan infusa simplisia kering dan simplisia segar daun buni. Prosedur penelitian ini meliputi pengumpulan bahan, determinasi tanaman, pembuatan simplisia, pembuatan infusa, identifikasi senyawa flavonoid, serta uji aktivitas antioksidan secara kuantitatif dengan metode radikal bebas DPPH (1,1-diphenyl-2-picrylhydrazyl). Data yang didapatkan berupa nilai IC50 yang diperoleh dari persamaan regresi linier antara % inhibisi terhadap konsentrasi larutan uji. Nilai IC50 dianalisis dengan uji statistik non parametric dengan taraf kepercayaan 95%. Hasil identifikasi senyawa flavonoid menunjukkan infusa daun buni positif mengandung senyawa flavonoid. Nilai IC50 infusa simplisia segar dan infusa simplisia kering daun buni berturut-turut 303,646±3,731 ppm dan 197,344±0,819 ppm. Infusa simplisia segar memiliki aktivitas antioksidan sangat lemah, sedangkan infusa simplisia kering memiliki aktivitas antioksidan lemah. Infusa simplisia segar dan infusa simplisia kering daun buni memiliki aktivitas antioksidan yang berbeda signifikan (p?0,05). Berdasarkan data yang diperoleh simplisia kering daun buni lebih direkomendasikan dalam pembuatan infusa sebagai antioksidan dibandingkan simplisia segar. Kata Kunci: Daun buni, antioksidan, infusa
