Abstract

Transcription factors have an important function in regulating gene expression and plant responses to stresses. The ERF, bZIP, WRKY, MYB, and NAC are stress inducible transcription factors. The OsNAC6 is a member of the NAC transcription factor family in rice and its expression is induced by abiotic stresses, wounding and blast disease. Characterization of OsNAC6 gene sequences would give a better undersanding on how OsNAC gene functions biologically. The objectives of this research are to isolate the OsNAC6 cDNA from Nipponbare, Batutegi, and Rojolele cultivars, to characterize their DNA sequences, and to compare their sequences to other NAC genes from other plants available in GenBank DNA databases. Isolated cDNA and sequencing of the fragments resulted in a 912 bp DNA sequences. Translation of the sequences yielded a protein consisted of 303 amino acid residue. Blast analysis of amino acid sequences indicated identity of isolated cDNA from three Indonesian rice cultivars are the OsNAC6 gene. Deduced amino acid residues from amplified cDNAs of Nipponbare, Batutegi, and Rojolele cultivars shared 100% sequence identities to rice OsNAC6 (Acc. # BAA89800), 71-100% sequence identity to a number of OsNAC protein from Oryza sativa and 63-83% sequence identity to NAC protein from other plants.

Abstract

Penyakit bilur kacang tanah yang disebabkan oleh peanut stripe virus (PStV)merupakan salah satu penyakit utama pada pertanaman kacang tanah (Arachishypogaea L.). Penggunaan varietas tahan merupakan alternatif paling efektif untukmengatasi penyakit tersebut. Rekayasa genetika merupakan metode efektif untukmendapatkan varietas kacang tanah yang resisten PStV. Tujuan penelitian ini adalahuntuk (1) mengetahui respons tanaman kacang tanah transgenik yang membawa gen cpPStV terhadap infeksi PStV dan (2) menguji stabilitas transgen sampai tujuh generasisilang-dalam. Tanaman kacang tanah transgenik cv. Gajah generasi T0, T1, T2, T3, T5,T6, dan T7 diinokulasi secara mekanik dengan PStV. Terdapat tiga jenis respons kacangtanah transgenik terhadap infeksi PStV, yaitu resisten, recovery, dan rentan. Padatanaman resisten gejala tidak muncul. Pada tanaman recovery, gejala chlorotic ringmottle muncul pada satu daun atau lebih, selanjutnya gejala tidak tampak pada daundaunyang tumbuh kemudian. Pada tanaman rentan, gejala severe blotch muncul padasuatu daun, selanjutnya gejala tersebut tetap muncul pada seluruh daun yang tumbuhkemudian. Transgen cp PStV tetap stabil setelah mengalami tujuh generasi silangdalam.Sejumlah galur murni kacang tanah transgenik yang resisten PStV telahdiidentifikasi.

Abstract

Aluminum (Al) is one of the major limited factors in crop production on acid soils. Aluminum tolerant plants can beselected from plant breeding program by one of the physiological parameters representing Al tolerance character,such as root re-growth capability during recovery from the Al-stress. In this study we determined the concentrationand time exposure of Al stress that was able to differentiate the response of three local upland rice varieties(Grogol, Hawarabunar and Krowal) and an Al-sensitive rice variety (IR64) to Al-stress, and evaluated the effectivenessof root re-growth (RRG) characters as an Al tolerance parameter in rice. The study consisted of three experiments,which were 1) nutrient culture experiment with different Al concentration treatments in growth chamber, 2) potexperiment in greenhouse using Jasinga yellow red podzolic acid soil containing 26,66 me/100 g Al and pH 4,6 asplanting media, and 3) phenotyping of F2 population using RRG character. The results showed that Al treatment at15 ppm for 72 h was able to distinctly differentiate between Al-tolerant (Grogol and Hawarabunar) and Al-sensitivevarieties (Krowal and IR64). Planting of the rice varieties on acid soils showed similar result as that of the nutrientculture. Phenotyping of F2 population using RRG character indicated the existence of RRG value variation. Thesevariations demonstrated that RRG character can be used as an Al tolerance parameter in rice and therefore can beeffectively applied to screen rice F2 population that segregate to Al tolerance character.

Abstract

Indonesia was known to have high diversity of mangosteens (Garcinia mangostana) and the related species. Inorder to elucidate the genetics variability of the diversity, thirty three accessions were examined by using isozymeanalysis. The genetic diversity and relationships among several mangosteens and other Garcinia sp wereestablished by using four isozymes. The level of polymorphism as revealed by isoenzyme was 88%. Althoughmangosteen is believed to reproduce exclusively through apomixis, our result show that considerable geneticdiversity exists within G. mangostana and between other Garcinia species. Based on 27 bands there were 5-42%dissimilarity level among mangosteen accessions, while the other species has 75% dissimilarity. The dendrogramis built based on isozyme marker analysis to separate clusters of mangosteen and other Garcinia sp. The datashowed that G. mangostana is a close relative of G. malaccensis, G. porrecta, G. celebica, and G. hombroniana. Theconcurrence analysis on isozyme analysis result showed the very good fit of Rolf correlation value (0.914). Thisresult indicated that isozymes could group G.mangostana and the related species.

Abstract

Rice panicle branching architecture is an important trait affecting yield, but its responses across different seasons have not been extensively studied yet. The aim of this study was to evaluate the panicle branching responses of fifteen rice genotypes on different seasons. This study was conducted from April 2017 to March 2018 at Babakan Experimental Station, IPB University, Dramaga, Bogor. Fifteen genotypes as treatments were grown in the dry and wet seasons to study the genotypes × seasons interaction effects on panicle branching. The experiment was laid out in a randomized complete block design with four replicates. Individual analysis and combined analyses over seasons were performed. The results revealed that the genotype, season, and genotype × seasons had significant effects on panicle length, number of primary branches, length of primary branches, and length of secondary branches. The results of the study may be useful for rice breeders in conducting selection based on panicle branching. Keywords: dry and wet season, genotypes × seasons, rice breeding

Abstract

ABSTRACTPreliminary study: Gene transfer from transgenic potato Katahdin RB to non transgenicpotato. One of the concerns associated with the release of transgenic crops, is the possibilityof the gene flow from transgenic crops to neighboring crops of the same species or to relatedspecies. In plants, gene flow is a routine process occur through the natural hybridization. Theopportunity for gene flow occur depends principally on two factors, the degree of sexualcompatibility between donor and recipient species, and the physical distance between thetwo. The experiment was conducted to determine whether the gene flow from transgenicpotato Katahdin RB to non transgenic was occurred, based on selection using a 50 mg/lkanamycin, and to estimate gene flow mediated by natural hybridization at different isolationdistances. Preliminary result indicated that a rapid and simple method using MS0 liquid mediawith kanamycin 50 mg/l was effective for screening the seeds. There was a gene flow fromtransgenic potato Katahdin RB to non transgenic, based on a rapid and simple selectionmethod using 50 mg/l of kanamycin as selectable marker. The isolation distance used in thestudy were 0.8, 1.6, 2.4, 3.2, 4.0, 4.8, 5.6, 6.4, 7.2, 8.0, 8.8, 9.6, 10.4, and 11.2 m from the row oftransgenic potato Katahdin RB. The gene flow through natural hybridization at a isolationdistances of (0.8 - 1.6 m), (2.4 ? 4 m), and (4.8 ? 6.4 m) from transgenic to non transgenic plantswere 13.78, 10.92, and 3.82%, respectively. At a distance of 7.2 ? 8 m, the frequency of gene flowwas declined to 0%. The frequency of gene flow from transgenic potatoes to non transgenicpotatoes markedly decreased by increasing the isolation distance, and was negligible at 7.2 m.Key words : natural hybridization, transgenic potato RB, kanamycin selection

Abstract

Linkage of 23 Microsatellite Marker on Chromosome 6and 7 to Downy Mildew Resistance on Maize. Roberdi,Hajrial Aswidinnoor, Asep Setiawan, Sutrisno, Marcia B.Pabendon, and M. Azrai. Downy mildew caused byPeronosclerospora is one of most important maize diseasein several countries, including Indonesia. Parental andprogenies selection based on conventional breeding is timeconsuming and laborious. Development of molecularbiology produces many DNA markers used for selection, oneof them is microsatellite. The aim of this research to identifymicrosatellite markers associated with downy mildewresistance on maize progeny MR-4 X AMATLCOHS-9-1-1-1-1-1-2-B, on chromosome 6 and 7. This research was consistedof two activities, phenotypic and genotypic analysis.Phenotypic analysis used 175 progenies BC1F2 and both ofparents. This analysis included planting of spreading row,inoculums preparation, inoculation of spreader rows, testmaterial planting, inoculation of test material andobservation. Genotypic analysis used 175 progenies BC1F1and both of parents. This analysis included DNA genomeisolation, PCR analysis, electrophoresis, gel staining and datascoring. Percentage of downy mildew infections on MR-4was 76%, while these on AMATLCOHS-9-1-1-1-1-1-2-B was16%, and on 175 progenies had range from 10.1-100%. Out of23 SSR, 12 markers could be mapped in chromosome 6 and11 markers in chromosome 7. QTL analyses showed thatchromosome 7 contain one QTL in position between phi082and phi116I marker as far as 18.6 cM with 2.6 LOD value.

Abstract

Potato late blight, caused by Phytophthora infestans is one of the most devastating plant diseases. Potato yield losses due to this disease ranged from 47-100%. Frequent intervals and high rates of fungicide spray, currently practiced by potato growers to control the disease are expensive. Host resistance is an alternative control measure that is more economically and environmentally sustainable. Development of late blight resistant plants was conducted by crossing RB transgenic Katahdin SP904 andSP951 as male and two susceptible (Atlantic, Granola) varieties as female parents. F1 progenies were molecularly characterized for the integration of the RB transgene and evaluated for their segregations. Crossing data of Atlantic x transgenic Katahdin SP904 and SP951 produced 71 (57.72%) berries with average number of seeds per berry 139.58 and 83 (41.29%) berries with average number of seeds/berry 85.23, respectively. Granola x transgenic Katahdin SP904 and SP951 crosses gave higher results in terms of berry set (79.55 and 84.44%, respectively) than Atlantic x transgenic Katahdin crosses. A total of 554 F1 progenies were analyzed for the presence of the RB PCR marker. An expected 619-bp and 840-bp band were amplified in the progenies that contain the RB gene. The RB gene was integrated in 65 (45.45%), 77 (47.83%), 47 (45.63%), and 71 (48.30%) F1 progenies of Atlantic x transgenic Katahdin SP904, Atlantic x transgenic Katahdin SP951, Granola x transgenic Katahdin SP904, andGranola x transgenic Katahdin SP951, respectively. Chisquare tests showed that all the four cross combinations followed a 1 : 1 segregation ratio.

Abstract

Efficacy of RB gene in transgenic potato Katahdin SP904and SP951 to West Java isolates of Phytophthorainfestans. A. Dinar Ambarwati, S.M. Sumaraw, AgusPurwito, M. Herman, E. Suryaningsih, and HajrialAswidinnoor. Potato late blight, caused by Phytophthorainfestans is one of the most devastating plant disease. Potatoyield losses due to this disease ranged from 47-100%. Amajor late blight resistance gene, called RB, previously wasidentified in the wild potato species Solanumbulbocastanum. RB gene has been integrated into cultivatedpotato Katahdin using Agrobacterium-mediated transformation,and showed durable and broad spectrum resistanceeither in laboratory assay or in confined field trial. Evaluationof transgenic Katahdin SP904 and SP951 was conducted toverify whether the RB gene with broad spectrum to allknown races of P. infestans in the United States and inToluca, Mexico was also effective against P. infestansisolates in Indonesia. Efficacy of RB gene was evaluated forfoliar and tuber resistance to West Java isolates. TransgenicKatahdin were more resistant in foliar than non transgenicplants, at 14 days after inoculation. Diseases intensity oftransgenic Katahdin SP904 and SP951 were 19.8-43.8%,whereas non transgenic Katahdin, Granola, and Atlanticwere 46.9-100%. In contrast to the foliar resistancephenotype, RB-containing tubers in transgenic Katahdin didnot exhibit increased resistance to Lembang, Pangalenganand Galunggung isolates. Tubers of transgenic KatahdinSP904, SP951, and non transgenic Katahdin showed lesionvolume of 0.93, 0.91, and 0.91 cm3, respectively. RB gene intransgenic Katahdin showed efficacy against late blight P.infestans in foliar, but did not showed efficacy in tuber.Transgenic Katahdin RB thus providing a potential source ofresistance for breeding programs.

Abstract

Adaptive rice varieties on P sub-optimum condition can be used by farmer to resolve the high macro fertilizer used, these varieties should be applied on rice field with P low input. Phosphor deficiency decreasing potentiality of result, therefore needs to develop rice promising lines that adaptive with P Sub-Optimum condition. The objective of this researched is to generate rice lines that adaptive on field with P sub-optimum condition and optimum through rice lines selection on the availability of difference P condition. The materials used are two population generation of F3, IR6008023/Bernas Prima and Gampai/Progol. The researched held at experiment station Muara, Bogor and Leuwiliang, West Java,  started from April 2012 untill December 2014. F3 generation planted on P sub-optimum condition at Muara station, until obtained F4 and F5 population generations. Population of those two generations has been selected using bulk modification method and planted on environment conditions of P optimum (36 kg ha-1P₂O₅)  and sub-optimum (without P fertilizer) so that generate F6 population generation. Rice lines 160 of F6 generation selected by panicle weight characteristic per hill. Those rice lines evaluated on two production environment, P optimum and P sub-optimum. Experiment on F6 generation was done using Augmented design. The result of the experiment shows that selection of bulk modification method effecive towards production environment of P optimum and  P sub-optimum. The potential yield of promising rice lines more gained from Pop 1 population (IR6008023/ Bernas Prima), while tolarance on P sub-optimum condition gained from Pop 2 population (Gampai/Progol). 5 promising rice lines with high yield on P sub-optimum production environment are : B1451E-PS-40, B1451E-PS-21, B1452E-PS-48, B1452E-PS-43 and B1452E-PS-8, whereas rice lines with high yield on P optimum are: B1452E-PO-13, B1451E-PO-27, B1452E-PO-45, B1451E-PS-44 and B1452E-PO-8.
