Abstract

Transcription factors have an important function in regulating gene expression and plant responses to stresses. The ERF, bZIP, WRKY, MYB, and NAC are stress inducible transcription factors. The OsNAC6 is a member of the NAC transcription factor family in rice and its expression is induced by abiotic stresses, wounding and blast disease. Characterization of OsNAC6 gene sequences would give a better undersanding on how OsNAC gene functions biologically. The objectives of this research are to isolate the OsNAC6 cDNA from Nipponbare, Batutegi, and Rojolele cultivars, to characterize their DNA sequences, and to compare their sequences to other NAC genes from other plants available in GenBank DNA databases. Isolated cDNA and sequencing of the fragments resulted in a 912 bp DNA sequences. Translation of the sequences yielded a protein consisted of 303 amino acid residue. Blast analysis of amino acid sequences indicated identity of isolated cDNA from three Indonesian rice cultivars are the OsNAC6 gene. Deduced amino acid residues from amplified cDNAs of Nipponbare, Batutegi, and Rojolele cultivars shared 100% sequence identities to rice OsNAC6 (Acc. # BAA89800), 71-100% sequence identity to a number of OsNAC protein from Oryza sativa and 63-83% sequence identity to NAC protein from other plants.

Abstract

Demostration rice plot of IPB 3S and IPB 9G varieties were at Mekarharja village, Purwaharja district, Banjar city. The demostration plot area of one hectare is spread over three hamlets including Randegan, Cibentang, and Pasirleutik hamlets. Stasiun Lapang Agro Kreatif (SLAK) program aims to develop IPB 3S and IPB 9G varieties of rice in an effort to increase farmer resources and rice production. Application of IPB 3S dan IPB 9G rice cultivation technology through training and field assistance to farmers. Farmers are given training and assistance to utilize cow dung and rice straw, seed treatment and tillage technology, and balanced fertilization technology. There are farmers’ knowledge increases about the use of cow dung and rice straw to increase rice production, and balanced seed treatment and fertilization. Farmers can apply cultivation technology easily because the materials used are easy to obtain and simple technology implementation procedures. The application of rice cultivation technology that has been carried out by most farmers in the village of Mekarharja. Farmers’ knowledge increases about the management of agricultural and livestock waste which can be used as organic fertilizer. The submitted science and technology was accepted and applied well by the farmers.

Abstract

Rice is one of the major staple foods in the world, especially in Asia. Improving yield potential of superior cultivars is important to meeting the demand for rice production, which is increasing due to human population increase, climate change, and degradation of agricultural resources, such as land and water. In this study, a BC3F2 population developed from an intraspecific cross between Ciherang and a new plant type line (B11143D) was used in a quantitative trait locus (QTL) analysis. Ciherang is a high yielding rice cultivar with good grain quality which has been planted in 37% of the irrigated rice area in Indonesia. The objective of this study was to identify QTL(s) for yield components on chromosome 12, which can be used to improve the elite cultivar Ciherang or other popular cultivars through marker-assisted breeding. A total of two hundred BC3F2 lines were evaluated in the greenhouse during this study. The population was observed for eight agronomic traits including days to heading (dth), plant height (ph), flag leaf length (fll), panicles per plant (ppl), panicle length (pl), grains per panicle (gpp), 1000-grain weight (gw), and yield (yld). Four simple sequence repeats (SSR) markers (RM3472, RM28048, RM28195, and RM1986) were used for targeted mapping on chromosome 12. Linkage analysis identified a QTL for 1000-grain weight located on chromosome 12 at position 53.5 cM–73 cM.

Abstract

One essential objective of rice breeding is to obtain high-yielding varieties. This study aimed to (1) determine the effect of genotype (G), environment (E), and genotype by environment (G×E) interaction on agronomic traits and yield of 12 lowland rice genotypes, (2) estimate variance components and repeatability (3) identify promising rice genotypes with good agronomic performance and high yield potential. The trials were conducted in three irrigated lowland locations from June to November 2020, using a randomized complete block design with three replications. The results showed that the G×E interaction effect was significant on days to flowering, days to harvest, plant height, number of tillers, and panicle length. The genotype's main effect was significant on yield. Four IPB lines (IPB189-F-13-1-1, IPB189-F-23-2-2, IPB193-F-17-2-3, and IPB193-F-30-2-1) had a higher average yield than Ciherang and Inpari 32 varieties. The IPB189-F-23-2-2 had a panicle length stability across the three test locations and a higher average yield than the checks.

Abstract

Zinc (Zn) is an essential micronutrient that is very useful for plants as well as humans. Breeding high zinc rice varieties and pigmented varieties will increase the added nutritional value of the rice consumed. Knowing the distribution of zinc content based on pigmented rice is necessary to see the opportunities for obtaining pigmented rice lines with high zinc. This study aimed to determine the potential distribution of zinc based on the pigment of brown rice. The research was carried out at the Breeding Laboratory of the Indonesian Center for Rice Research from February to July 2022. A total of 970 lines derived from various biparental crosses and 10 varieties were used as research material. The samples were brown rice from a field research plot arranged in an augmented design with 5 blocks. Analysis of zinc content used the X-ray Fluorescence method using the XRF Machine Supreme8000. The results showed that there was a significant difference in Zn content based on CPC color. White rice had the widest range of zinc content compared to brown and black rice. The maximum zinc content found in white rice was 55.82 mg kg-1 and the maximum yield potential was 138.29 g per hill. The black rice genotypes in all categories had a zinc content exceeding the Inpari IR Nutri Zinc potency. Black rice type C was the one with the widest variety. Types of black rice with high zinc content and high yield potential are Black B and Black E types. ABSTRAKZinc (Zn) merupakan mikronutrisi esensial untuk tanaman dan juga manusia. Perakitan varietas  padi zinc tinggi pada beras-beras berwarna akan meningkatkan nilai tambah gizi beras yang dikonsumsi. Keragaman kandungan zinc berdasarkan warna beras perlu diketahui untuk melihat peluang diperolehnya varietas beras berwarna dengan keunggulan zinc tinggi. Penelitian ini bertujuan untuk mengetahui potensi zinc berdasarkan warna beras pecah kulit. Penelitian dilaksanakan Laboratorium Pemuliaan BB Padi dari bulan Februari sampai Juli 2022. Sebanyak 970 galur hasil persilangan biparental dan 10 varietas digunakan sebagai materi penelitian. Sampel berupa beras pecah kulit dari plot penelitian lapangan dengan rancangan augmented 5 blok. Analisa kandungan zinc menggunakan metode X-ray Fluorescence mesin XRF Machine Supreme 8000. Hasil menunjukkan bahwa terdapat perbedaan nyata kandungan zinc berdasarkan warna BPK. Beras putih memiliki rentang kandungan zinc yang paling luas dibandingkan beras merah dan hitam. Potensi zinc maksimum terdapat pada beras putih dapat mencapai 55,82 mg kg-1 dan potensi maksimum hasil 138,29 g per rumpun. Genotip padi semua kategori tipe beras hitam memiliki rentang kandungan zinc di atas potensi Inpari IR Nutri Zinc. Tipe beras Hitam C adalah beras yang memiliki varian paling luas. Tipe beras hitam yang memiliki peluang kandungan zinc sangat tinggi dan potensi hasil tinggi adalah tipe Hitam B dan Hitam E.

Abstract

The direct seeding of rice (DSR) system has been widely practiced in various Asian countries because it is considered more efficient in water and labor and provides better benefits than transplanting systems. However, improvements in the management of this system practice are still needed, especially in cropping patterns and the use of appropriate varieties. This study examines the physiological characteristics, yield components, and yields of rice genotypes in transplanting and DSR systems. This research was carried out in the field with transplanting and DSR cultivation systems (drill and broadcast sowing) and used four superior lines and one national variety. A total of 25 treatment combinations were designed using a split-plot randomized complete block design (Split plot-RCBD) with three replications for each treatment. The results showed that rice grown with DSR had no different level of greenery and photosynthesis rate than rice in transplanting cultivation, but produced lower panicle length, the number of grains per panicle, the number of filled grains, and faster flowering time. Genotype IPB193-F-17-2-3 produced the highest productivity in wet DSR (drill and broadcast sowing) and dry DSR (drill sowing). Genotype IPB193-F-38-2-1 had the highest productivity in transplanting techniques and dry DSR (broadcast method). The productivity of the Ciherang variety in the DSR system (drill method) was not different from the transplanting system. This finding indicates that the direct seeding system using the drill method is promising for further development.

Abstract

Rice is important staple food in the world. One important input in rice production is seed vigor, is determined by seed characteristics such as seed size and shape. The Digital image processing could facilitate observations of seed size faster, large samples, produce accurate, and robust data. However, this study was designed to investigate size of seed, endosperm and embryo of 55 tropical rice genotypes using image analysis and correlation anlysis between characters. A total of 400 seeds, 40 endosperms, and 8 embryos containing each genotype were investigated for their size characteristics. The results of the investigation showed that genotype differences had a significant effect on size of seed, endosperm and embryo. The length of tropical rice seeds is included in the medium-very long seeds and has a significant positive correlation (α <0.01) with the weight of 1000 grain grains, seed area, shape of the seed (length-to-width ratio), and endosperm length. Embryo size characters (length, width, area, and circumference) have a significant positive correlation (α <0.05) with seed area. The results can be considered in the crop improvement of tropical rice plants in the future.

Abstract

The IPB rice technology dissemination program implemented in Pangumbahan Village, Ciracap District, Sukabumi Regency, aimed to improve the knowledge and skills of fostered partner farmers through technology transfer and innovation to increase land utilization and support the intensification of rice production. This service activity involved four active IPB students, four IPB lecturers, and the Sabilulungan Farmer Group Union (Gapoktan) as fostered partners. The method of activity implementation consisted of observation, mapping of potential and problems, program implementation, and monitoring and evaluation of the program. The implementation of the IPB rice technology dissemination program began with field observations, including a survey of field conditions. Focus group discussion (FGD) activities were conducted to map the potential and problems in the community. Program implementation consisted of training and assistance in transferring technology and innovation in rice cultivation. Monitoring and evaluation were conducted through in-depth interviews and discussions with the assisted farmers. Through the establishment of demonstration plots of IPB 3S and IPB 9G rice varieties, the community can increase its knowledge and skills regarding the development of their commodities. The implementation of technology transfer through training and practice of the IPB Prima rice cultivation technique can encourage more efficient and productive rice cultivation.

Abstract

AbstractLeaf fall disease caused by Corynespora cassiicola fungi is one of the most important diseases in rubber plant. Rubber clone AVROS 2037 is considered resistant to this pathogen while clone PPN 2444 is susceptible. These two rubberclones were used to identify genes or transcripts differentially expressed during interaction between rubber plants and the fungi, using cDNA-Amplified Fragment Length Polymorphism (cDNA-AFLP) method. Induced genes/transcripts expression was examined to compare differencies between plants uninfected and infected with C. cassiicola at 24, 36, 48 and 72 hours after inoculation. cDNA-AFLP analysis was performed using restriction enzyme VspI and TaqI so the adapters and primers also have the recognition site of these enzymes. By using 29 specific primers, 35 out of approximately 1450 fragments were differentially expressed between AVROS and PPN 2444 clones. All of these fragments were cloned and sequenced. The homology-based grouping of these sequences resulted in 19 contigs and nine individual sequences. Among these, 10 contigs and five sequences have significant sequence homology with known genes in gene bank data base, such as Ran binding protein, protein transporter and transcriptional regulators of some organisms; arginase, GTP-binding protein, heat shock protein (HSP) and aconitase. Ran binding protein, GTPbinding protein and protein transporter were known as membrane proteins while arginase and HSP usually expressed as a response to wounding or toxin treatment. The present of arginase is usually related to the availability of nitric oxide (NO) in plant tissue. NO is well known as a signal molecule on plant defense response. AbstrakPenyakit gugur daun yang disebabkan oleh fungi Corynespora cassiicola merupakan salah satu penyakit penting tanaman karet. Klon karet AVROS 2037menunjukkan sifat resisten terhadap patogen tersebut sedangkan klon PPN 2444 merupakan klon yang rentan. Kedua klon karet tersebut digunakan untuk mengidentifikasi gen atau transkrip yang diekspresikan secara diferensial selama terjadi interaksi antara tanaman karet dengan C. cassiicola menggunakan teknik cDNA-Amplified Fragment Length Polymorphism (cDNA-AFLP). Ekspresi gen/transkrip dipelajari untuk membandingkan perbedaan antara tanaman yang tidak diinfeksi dengan yang diinfeksi patogen pada waktu 24, 36, 48 dan 72 jam setelah inokulasi. Analisis cDNA-AFLP dilaksanakan dengan menggunakan pasangan enzim restriksi VspI dan TaqI sehingga adapter dan primer memiliki sekuen pengenalan kedua enzim restriksi tersebut. Dengan menggunakan 29 pasang primer spesifik, sebanyak 35 dari sekitar 1450 fragmen memiliki ekspresi berbeda antara klon AVROS 2037 dan PPN 2444. Semua fragmen yang berbeda tersebut kemudian diklon pada vektor kloning dan disekuen. Hasil sekuensing dikelompokkan berdasarkan homologi sekuennya dan menghasilkan 19 contigs serta sembilan macam sekuen yang tidak mengelompok. Sebanyak 10 dari 19 contigs dan lima dari sembilan sekuen tersebut memiliki homologi dengan produk gen yang telah dikenal yang terdapat di pangkalan data GenBank, seperti putative Ran binding protein, protein transporter, regulator transkripsi, arginase, GTP-binding protein, heat shock protein (HSP) dan aconitase. Beberapa di antaranya seperti putative Ran binding protein, protein transporter dan GTP-binding protein dikenal sebagai protein membran, sedangkan arginase dan HSP merupakan protein atau enzim yang ekspresinya pada tanaman antara lain dipengaruhi oleh adanya pelukaan dan perlakuan toksin. Keberadaan arginase sering berhubungan dengan ketersediaan nitric oxide (NO) pada jaringan tanaman. NO dikenal sebagai salah satu sinyal molekul dalam mekanisme pertahanan tanaman.
