Abstract

Calamansi fruits (Citrus microcarpa) or musk lime is one horticulture crops has been known as beverage and food flavorings. This research aimed to determine the effect of 2,4-D and BAP on callus induction of explants calamansi fruits (Citrus microcarpa). Parameters observed is percentage of explants, time appeared callus, callus width, and texture of the callus. Research results and a growing percentage of the width callus explants were analyzed by ANOVA and a further test DMRT at 5% level. The next to appear callus timing parameters and texture of callus analyzed in description. The result shows that a combination of hormones D4B1 and D4B2 produces a callus emerged fastest of 3.3 days after culture. The highest percentage of callus formation that is 100% contained in the treatment of D2B0, D2B0.5-D4B0. The mean width of callus was highest in treatment D4B2with a width of 0.97 cm. Embryogenic callus resulting by treatment of D2B1, D2B2, and D3B0. However, the best a combination of hormones is treatment D2B2 (2 mg/l, 2,4-D, dan 2 mg/l BAP) producing embryogenic callus edvanced to suspension cultures.

Abstract

Compentency is expected in biotechnology subjects learning requires student to make it more in learning. It requiresd a real effort both from lectures and from students. Class Action Research has been conducted (PTK), which aims to improve the activity and the results of studying Biotechnology with the type STAD cooperative learning based audio visual media in class of PGMIPA-U Biology FKIP UR. The result of data analysis cycle I and cycle II showed an increase in student learning activities for all indicator. The improvement of the student activity everage from I cycle about 69,46% up to II cycle about 89%. An increase in student activity 20.5%. Learning outcomes of students in I cycle and II cycle also increased. The average value of learning outcomes students increased from pre-cycle 71.35 to 75.55 in I cycle and then from I cycle 75.55 to 84.15 in II cycle. Generally its can be said that the use of cooperative learning model type STAD based Audio visual in biotechnology teaching can anhance students learning outcome.. Abstract

Orchids are a very popular ornamental plants, but orchids have a very slow vegetative growth, thus requiring special treatment in the grown.To  spur grow than development can be used through tissue culture techniques of multiplication. But the success of the technique of tissue culture is inseparable with give a substance on the plant growth regulator. This study aims to determine the growth  an influence  D.phalaenopsis Fitzg orchids of giving a gaint swith IBA administration and Kinetin according to in vitro  and as well as to obtain a concentration combination of  IBA and Kinetin on the best orchid D.Phalaenops is Fitzg explants.  Base don’t he results of the study indicate that the combination of IBA and Kinetin concentration giving a influence percentage of live explants,   number of shoots and number of roots orchid D. phalaenopsis Fitzg explants.Combination treatment of IBA 1.0 ppm and 0.1ppm Kinetin is a combination of the best treatment to produce a number of shoots orchids.The giving of IBA 0.5ppm in the medium without the addition of kinetin concentration can increase the number of roots of the orchid D.phalaenopsis Fitzg.

Abstract

Zat goniotalamin pada tanaman gajah beranak (Goniothalamus tapis) merupakan obat alternatif penyembuhan kanker. Penelitian bertujuan untuk mendapatkan zat goniotalamin melalui kultur kalus dan kultur suspensi sel. Metode penelitian eksperimen Rancangan Acak Lengkap (RAL) dengan kombinasi 2,4-D (1-10 mgL-1) dan BAP (0,5-2 mgL-1) menggunakan eksplan batang muda, terdiri dari 17 perlakuan dan 3 kali ulangan. Analisis data menggunakan Analysis of Variances dan uji lanjut Duncan Multiple Range Test (DMRT) taraf 5%. Hasil menunjukkan bahwa kultur kalus G. tapis pada media 5,0 mg L-1 2,4-D + 1 mg L-1 BAP adalah yang terbaik dengan waktu muncul kalus 28,33 hari dan persentase pembentukan kalus 100%. Kalus untuk kultur suspensi sel bertekstur remah dan berwarna kuning kehijauan. Kultur suspensi sel menghasilkan pertumbuhan sel yang cepat, tidak lembek berair dan mudah dipisahkan. Hasil kualitatif Kromatografi Lapis Tipis kultur suspensi sel sangat jelas, bersih dan terdapat potensi kandungan zat goniotalamin pada perlakuan 2,4-D 5 mg L-1 + BAP 0,5 mg L-1, 2,4-D 5 mg L-1 + BAP 1 mg L-1, 2,4-D 5 mg L-1 + BAP 2 mg L-1, 2,4-D 10 mg L-1 + BAP 0,5 mg L-1 dan 2,4-D 10 mg L-1 + BAP 1 mg L-1. Hasil kuantitatif zat goniotalamin dengan Kromatografi Cair Prestasi Tinggi terdapat pada perlakuan 2,4-D 5,0 mgL-1 + BAP 1 mg L-1 yaitu 9,57 mg g-1.The goniothalamine compound on Goniothalamus tapis is an alternative cancer medicine. This study aimed to obtain gonotalamin through callus culture and suspension cell culture. The experiment research method was Completely Randomized Design (CRD) with a combination of 2.4-D (1-10 mg L-1) and BAP (0.5-2 mg L-1) using young stem explants consisting of 17 treatments with 3 replications. Data analysis used ANOVA and DMRT at 5%. The results showed that G. tapis callus culture on 5.0 mg L-1 2.4-D + 1 mg L-1 BAP was the best treatment medium with callus emergence time of 28.33 days and percentage of callus formation 100%. The callus used for suspension cell culture was friable and greenish-yellow in color. Suspension cell culture resulted in rapid cell growth, was not fleshy, and easily separated. The  quality test by Thin Layer Chromatography (TLC) from suspension cell culture resulted very clear, clean, and potential content of goniothalamin found in treatments 2.4-D 5.0 mg L-1 + BAP 0.5 mg L-1, 2.4-D 5.0 mg L-1 + BAP 1 mg L-1, 2.4-D 5.0 mg L-1 + BAP 2 mg L-1, 2.4-D 10 mg L-1 + BAP 0.5 mg-1 and 2.4-D 10 mg-1 + BAP 1 mg-1. The quantitative results of the best goniotalamine compounds in cell suspension cultures using High-Performance Liquid Chromatography (HPLC) on medium 2,4-D 5.0 mgL-1 + BAP 1 mg L-1 ie 9.57 g-1.

Abstract

Pineapple (Ananas comosus (L.) Merr.) is one tropical fruits are highly favored by the Indonesian people and the potential for export commodities. This research is aimed to determine the effect of NAA and kinetin with different concentrations of Murashige Skoog medium (MS) to mikropropagasi Bogor pineapple (Ananas comosus (L.) Merr.) cv. Queen. The research design used was completely randomized design (CRD) factorial with two factors, each factor consists of 4 degree of concentration are: NAA hormone concentrations (0-1 ppm) and kinetin (0-3 ppm) with 3 replications. Parameters observed that the percentage of explants growing, lengt of shoot ,number of shoots and number of roots. The data were analyzed further test ANAVA and DMRT at 5% level. The results showed that the percentage of explants all treatments grew 100%, lengt of shoot is 7,73 cm in control, the highest number of shoots in treatment N0,25K3 is 13.67, and the highest number of roots in the treatment N1K0 is 11.67 weeks after initiation. The results concluded that the combined treatment N0,25K3 is the best treatment for micropropagation of pineapple cv. bogor Queen.

Abstract

Abstract: The aim of this research was to determine the composition of the best combination of rubber seed shells and rice husk in manufacture of briquettes. The research was conducted in July to September 2019. The results of research were used as a worksheet design for student of conventional biotechnology grade XII senior high school. The research design was divided into two stages, namely the stage of making briquettes in combination of rubber seed shells and rice husk at the Laboratory of Teacher Training and Education Biology of Riau University, Laboratory of Mechanical engineering of Riau University, Laboratory of Minister Energy and Mineral Resources of Riau Province. The research using RAL consisting of 7 treatments and 3 replication. The student worksheet (LKPD) used ADDIE (Analyze, Design, Development, Implementation dan Evaluation) only reaches the stage design. Making briquettes in combination of rubber seed shells and rice husk consist of the preparation of raw materials, the carbonization stage, the stage reducing size, the stage of making briquettes dough and the stage of molding briquettes. The parameters in this research are water content, ash content, heating value, density and compressive firms. The result of the research was found that the P0 (9:1) treatment was the effective in making combination of briquettes rubber seed shells and rice husk. Abstract

Abstract: This study aims to describe the supporting facilities for the implementation of learning and the relationship between problem-based learning through e-learning and students' critical thinking skills. This research is a descriptive study with a qualitative approach. The subjects of the research were students of class XI SMA Islam As-Shofa Pekanbaru. The problem based learning based of students' critical thinking skills with indicators of analyzing, synthesizing, recognizing and solving problems, concluding, and evaluating. The collection of data on critical thinking skills is a survey with observation. Based on the results of the research, SMA Islam As-shofa stated that it has adequate facilities for e-learning. Students on average have facilities in the form of hardware, software, and a learning community. It can be concluded that the implementation of learning with the Problem Based Learning (PBL) model through e-learning on the aspect of students' critical thinking skills is running well in accordance with the relationship of e-learning activities with problem based learning and has been accompanied by facilities that support adequate learning.Key Words: E-learning, Problem Based Learning, Critical Thinking, Facilities

Abstract

Abstract: The research aims to know influence of yeast concentration sukun (Artocarpus altilis) bioethanol level and produce a design biology student worksheet at senior high school biotechnology material. Experimental research by conducted experiment for 9%, 10%, 11%, 12% and 13% of sukun with 5 treatments 3 replicates whewe the substrate was hudrolyzed first with 5% sulfuric acid and continued with the developments of student worksheet from experimental research (only analyze and design stages). The research data were analized using variances analysis techniques and continued with Duncan’s Multiple Range Test (DMRT) of the 5% level. The results of the study indicate that yeast concentration affects the bioethanol. The highest average bioethanol content was found at 11% which was 9.70%. All research data can be used to design senior high school biology student worksheet in conventional biotechnology material class XII with project based learning models. However, before the worksheet design is used on a wider scale it is recomended that a feasibility test and effectiveness be carried out on a limited scale.Key Words: Yeast Concentration, Artocarpus altilis, Bioethanol, Student Worksheet Desain

Abstract

Abstract: Tempoyak is a typical traditional food from Bengkulu which is from durian fruits meat the making of tempoyak is arded by addition of salt, but salt concentration is not known by the public. The research aims to determine effectiviness of salt addition to the quality of tempoyak durian (Durio zibethinus Murr) and produce the design of biology student worksheet in hight school class XII Biotechnology subject. Experimental research by conducting tempoyak durian fermentation experiment with the addition of 1 gram, 2 grams, 3 grams, 4 grams and 5 grams with 5 treatments of 3 repetitions of 200 gram durian and continued with the development of student worksheet learning from experimental research data (only at the Analysis and Design stage). The results showed that the addition of salt to tempoyak durian had an effect on the decrease in pH value. The lowest pH value for salt addition was in A2 and A3 treatments, while in A4 treatment it increased to an average pH value of 4.4 which had an organoleptic value in brownish yellow color, flavored tempoyak and stinging, slightly salty and slightly acidic and soft texture. The results of this study can be used as learning resources oriented to the Project Based Learning model of Biology subjects in class XII biotechnology material. All data from the research results can be used to design the Senior High School Biology student worksheet Learning in Conventional Biotechnology class XII with Project Based Learning Learning Models. However, before the student worksheet design is used on a broader scale it is recommended that a feasibility test and effectiveness be carried out on a limited scale.Key Words: Durio zibethinus Murr, Tempoyak, Salt, Student Worksheet

Abstract

Abstract : Tempe is a food produced by a simple fermentation technique by Rhizopus oligosporus. During this tempe on the market only the soy-based. While the last few years soybean production in Indonesia continues to decrease and not able to meet the needs. Therefore, it is necessary to develop the manufacture of tempe that uses other raw materials whose nutritional value is not inferior to the tempe of soybean such as jackfruit seeds. The purpose of this research is to know the effect of the old boiling effect on the quality of Tempe seed of jackfruit (Artocarpus heterophyllus) tempe and the result of this research can be used for the development of LKS SMA of Conventional Biotechnology material. This research consists of 2 stages, namely the first stage, making tempe while the second stage of LKS development from the research. This research was done using experimental method with Completely Randomized Design (RAL) which consist of 5 treatments and 3 replications. The result of this research is concluded that in terms of content, protein content and fat content and organoleptic, K2 treatment is a good time in making tempe at Protein content while in the K4 treatment a good time in making tempe at fat content with 75 minutes boiling duration. The results of the research can be developed as LKS SMA Conventional Biotechnology material.Keywords: Tempe, jackfruit seed, conventional biotechnology, LKS.
