Abstract

This study aimed to determine the concentrations of Follicle Stimulating Hormone (FSH) and Luteinizing Hormone (LH) in New Zealand White (NZW) rabbits after superovulation induction using FSH. Six female NZW rabbits and one male NZW rabbit were used in this study. Criteria for female NZW rabbits used include having previously given birth, not being pregnant, and having a body weight of 2-3 kg. The rabbits were divided into two treatment groups; the first group was injected with physiological NaCl (NZW1, n= 3), while the second group was injected with FSH (NZW2, n= 3). Injections of physiological NaCl or FSH were administered five times at 12-hour intervals and first injection was given at 20:00 p.m. In NZW1, each injection was given physiological NaCl with volumes of 0.2:0.2+0.2;0.4+0.4 mL, respectively whereas in NZW2, each injection was given FSH with doses of 4:4+4:8+8 mg, respectively. Twelve hours after the last FSH injection, NZW2 rabbits were injected with 100 IU hCG and mated with male rabbit, while NZW1 rabbits were mated without hCG administration. On days 1, 3, and 5 after mating, blood samples were collected for FSH and LH hormone examination using the Enzyme-Linked Immunosorbent Assay (ELISA) method. The FSH concentrations on days 1, 3, and 5 between NZW1 vs NZW2 were 34.781.46 vs 39.753.18; 41.434.90 vs 41.425.14; and 44.471.51 vs 30.130.19 mIU/mL, respectively (P0.05). The LH concentrations on days 1, 3, and 5 between NZW1 vs NZW2 were 33.784.01 vs 40.1812.36; 29.342.24 vs 31.3110.13; and 42.0911.38 vs 30.578.87 mIU/mL, respectively (P0.05). It is concluded that FSH administration does not affect the concentrations of FSH and LH in NZW rabbits. The FSH concentration five days after mating tends to fluctuate, while the LH concentration tends to decrease as the days increased.

Abstract

This study aimed to determine the effect of aerobic and anaerobic incubation, and the addition of CaCO on the growth and antibacterial activity of Lactic Acid Bacteria (LAB) isolated from laying hens intestines (strain ISA Brown) after given 4% AKBISprob. The LAB used in this study was the isolates strains that produced the strongest antibacterial activity with inhibition zone 5 mm. The treatments consisted of two incubations condition (aerobic and anaerobic) with five concentrations of CaCO3 (1%, 2%, 3%, 4%, and 5%). Each treatment consisted of three repetitions. Total Plate Count (TPC) method and the agar diffusion method were used to calculate the amount of LAB and antibacterial activity, respectively. The results showed that anaerobic conditions had a significant effect (P0.05) on the increase of LAB growth, but both of aerobic and anaerobic conditions (P0.05) did not show significant effect on antibacterial activity. The addition of CaCO3 had a significant effect (P0.05) on the increase in growth and antibacterial activity of LAB. Based on the results of the study, it can be concluded that anaerobic incubation can increase the growth of two isolate of LAB isolates (P31S72 and P31H62), however aerobic and anaerobic incubation have no effect on antibacterial activity. In addition, the administration of 3% CaCO3 can increase the growth and antibacterial activity of all LAB isolates.

Abstract

The purpose of this research was to identify avian influenza (AI) virus using serological and molecular methods on poultry which suspected as AI infected in Aceh province. This study used 37 samples of tracheal and cloacal swabs and organs from various species of poultry that were collected from several districts/cities in Aceh. Samples were collected and put into transport media and stored at 4 C before sending to the laboratory. Samples were inoculated in specific pathogen-free of embryonated chicken egg with the age of 9-11 days for further serological and molecular examination. From 37 samples which infected to embryonated chicken egg then followed by hemagglutinin agglutination test/hemagglutinin inhibition revealed that 7 samples were positively infected with AI virus. The amplification result of specific matrix gene primer was followed by electrophoresis on 2% agarose gel which were obtained in the form of a deoxyribonucleic acid (DNA) band at 276 bp for matrix gene and 1.725 bp for H5 gene for all isolates test. In conclusion, the virus which caused the death of various types of poultry in Aceh province is avian influenza A virus subtype H5.Key words: avian influenza virus, H5N1, serologic, matrix, heamaglutinin

Abstract

Penelitian ini bertujuan mengetahui pengaruh pemberian kombinasi ekstrak jaloh dan kromium terhadap pembentukan heat shock protein-70 (HSP-70) dan kadar kortisol ayam yang mengalami cekaman panas. Dalam penelitian ini digunakan ayam broiler sebanyak 12 ekor yang dibagi atas empat perlakuan: Perlakuan I (EC), cekaman panas + dan kombinasi 1.000 mg ekstrak jaloh dengan 1.000 g kromium per liter air minum, perlakuan II (E), cekaman panas + 1.000 mg ekstrak jaloh per liter air minum, perlakuan III (KD), cekaman panas + 0,0 mg ekstrak jaloh dan 0,0 g kromium, dan Perlakuan IV (KL), tanpa cekaman panas dan 0,0 mg ekstrak jaloh dan 0,0 g kromium. Masing-masing perlakuan terdiri atas tiga ulangan. Metode pemberian cekaman panas dilakukan dengan meningkatkan suhu dalam kandang pada 331 C selama lima jam per hari dalam waktu lima belas hari. Pemberian kombinasi ekstrak jaloh dan kromium dilakukan dengan cara melarutkannya dalam air minum dan diberikan selama dua jam (pemberian pukul 10.00) sebelum suhu di dalam kandang mencapai 331 C. Pengambilan sampel serum dan jaringan paru dilakukan pada hari ke-15 pelaksanaan penelitian (ayam umur 36 hari). Pengambilan sampel darah dilakukan sebelum ayam dipotong dan jaringan organ paru diambil setelah ayam dipotong. Pada sampel serum dilakukan pemeriksaan kortisol dengan enzymelinkedimmunosorbantassay (ELISA) dan deteksi HSP-70 di dalam jaringan paru dilakukan menggunakan metode imunohistokimia. Hasil penelitian ini menunjukkan bahwa pemberian cekaman panas pada suhu 331 C selama lima jam per hari dapat meningkatkan pembentukan HSP-70 dalam paru dan kadar kortisol dalam serum. Pemberian ekstrak jaloh secara tunggal lebih efektif menurunkan jumlah HSP-70 pada jaringan paru dibandingkan jika dikombinasi dengan kromium.
