Abstract

The Regional Prominence Product Development Program (RPPDP) aims to determine the impact of thecoronavirus disease2019(COVID-19)pandemic on Micro, Small, and Medium Enterprises (MSMEs) based on the creative economy of seafood culinary in Bulak, Surabaya. This program was conducted to find out the solution that can be taken by government to recover and develop the creative economy of seafood culinary entrepreneur due to the COVID-19 pandemic impacts. Data were collected by interview/questionnaire, literature review, and implementation of recovery of the the creative economy of seafood culinary. The period of this study was from March (the beginning of the COVID-19 pandemic in Indonesia) to the middle of December 2020. The results of RPPDP showed that there were two elements influenced the seafood culinary entrepreneur due to the COVID-19 pandemic, namely Human Resources and innovation processing and marketing products. The government can take steps to overcome these problems, such as providing business capital assistance, giving discounts on electricity bills, deferring loan payments, assisting MSMEs who closed their businesses as result of the pandemic to transform or start new businesses. In conclusion, innovation of seafood culinary during the pandemic should be in accordance with the market demand, innovating traditional souvenirs to be work from home snacks, and online marketing using non-cash payment.

Abstract

Vibrio parahaemolyticus infection poses a significant challenge in intensive shrimp culture, leading to substantial economic losses. Its common treatments using synthetic antibiotics have been linked to increased risks of antibiotic resistance and residual effects. Therefore, finding environmentally safe and effective natural alternatives is deemed essential. Tengkawang Bukit (Shorea beccariana) stem extract contains antibacterial compounds, including asiatic acid, oleanolic acid, and lupanone, all classified as terpenoids. This study aims to evaluate the antibacterial potential of S. beccariana stem extract through phytochemical screening and a disc diffusion test against V. parahaemolyticus. The screening results confirmed the presence of bioactive compounds, including alkaloids, flavonoids, saponins, tannins, and steroids. The disc diffusion test showed an increase in the inhibition zone with increasing extract concentration, with inhibition diameters ranging from 9.04 ± 0.48 mm to 10.75 ± 0.26 mm. The 17% extract yield indicates a high availability of active compounds. These findings suggest that tengkawang bukit stem extract has potential as a natural antibacterial alternative for controlling V. parahaemolyticus in shrimp culture. This approach could help reduce reliance on synthetic antibiotics and promote sustainable fisheries. Infeksi Vibrio parahaemolyticus merupakan salah satu kendala utama dalam budidaya udang yang menyebabkan kerugian ekonomi signifikan. Penggunaan antibiotik sintetis sebagai pengobatan telah menimbulkan kekhawatiran terkait resistensi dan residu, sehingga diperlukan alternatif alami yang aman dan efektif. Batang tengkawang bukit (Shorea beccariana) diketahui mengandung senyawa antibakteri seperti asam asiatik, asam oleanolik, dan lupanon yang tergolong ke dalam senyawa terpenoid. Studi ini bertujuan untuk mengevaluasi potensi antibakteri ekstrak batang S. beccariana melalui skrining fitokimia dan uji difusi cakram terhadap V. parahaemolyticus. Hasil skrining menunjukkan adanya kandungan senyawa bioaktif berupa alkaloid, flavonoid, saponin, tanin, dan steroid. Uji difusi cakram memperlihatkan zona hambat yang meningkat seiring dengan peningkatan konsentrasi ekstrak, dengan diameter hambatan berkisar 9,04 ± 0,48 mm hingga 10,75 ± 0,26 mm. Rendemen ekstrak sebesar 17% menunjukkan ketersediaan senyawa aktif yang melimpah. Hasil riset ini mengindikasikan bahwa ekstrak batang tengkawang bukit berpotensi sebagai alternatif antibakteri alami dalam pengendalian V. parahaemolyticus pada budidaya udang, sehingga dapat mengurangi ketergantungan terhadap antibiotik sintetis dan mendukung keberlanjutan perikanan.

Abstract

Meskipun enzim selulase rekombinan banyak dikembangkan untuk meningkatkan efisiensi pemecahan serat, informasi mengenai bagaimana modifikasi seperti mutasi dan penambahan tag SKIK (Ser-Lys-Ile-Lys) dapat memengaruhi aktivitas enzim dan produksi protein total masih terbatas. Oleh karena itu, penelitian ini bertujuan mengevaluasi pengaruh mutasi dan penambahan tag SKIK terhadap aktivitas dan protein total enzim selulase rekombinan yang diproduksi oleh Escherichia coli BL21 (DE3). Penelitian dilakukan secara eksperimental menggunakan rancangan acak lengkap (RAL) dengan empat perlakuan, yaitu P1= wild type (WT), P2 = WT + SKIK, P3 = mutan, dan P4 = mutan + SKIK, masing-masing dengan empat ulangan. Produksi enzim dilakukan melalui kultur E. coli rekombinan yang membawa plasmid pET-22b-CellE dengan induksi laktosa monohidrat, kemudian dianalisis menggunakan uji aktivitas selulase metode 3,5-dinitrosalisilat (DNS) dan pengukuran protein total dengan metode Bradford. Data dianalisis menggunakan ANOVA satu arah dan uji lanjut Duncan pada tingkat signifikansi 5% (p < 0,05). Hasil penelitian menunjukkan bahwa perlakuan mutan + SKIK menghasilkan nilai tertinggi baik pada aktivitas enzim (10,66 U mL-1) maupun protein total (0,85 mg mL-1). Penambahan SKIK meningkatkan ekspresi dan kelarutan protein, sedangkan mutasi memperbaiki efisiensi katalitik enzim. Kombinasi keduanya memberikan efek sinergis dalam meningkatkan perfoma enzim selulase rekombinan. Penelitian ini merupakan yang pertama mengombinasikan penambahan tag SKIK dengan mutagenesis untuk meningkatkan ekspresi dan aktivitas katalitik enzim selulase pada E. coli. Temuan ini tidak hanya relevan untuk peningkatan produksi enzim pada industri bioteknologi, tetapi juga berpotensi diaplikasikan dalam formulasi pakan ikan. Although recombinant cellulase enzymes have been widely developed to enhance fiber-degradation efficiency, information regarding how modifications such as mutagenesis and the addition of the SKIK (Ser-Lys-Ile-Lys) tag influence enzymatic activity and total protein production remains limited. Therefore, this study aimed to evaluate the effects of mutagenesis and SKIK tagging on the activity and total protein yield of recombinant cellulase expressed in Escherichia coli BL21 (DE3). The research was conducted experimentally using a completely randomized design (CRD) with four treatments: P1 = wild type (WT), P2 = WT + SKIK, P3 = mutant, and P4 = mutant + SKIK, each consisting of four replications. Enzyme production was carried out using recombinant E. coli harboring the pET-22b-CellE plasmid induced with lactose monohydrate, and the resulting enzyme was analyzed for cellulase activity using the 3,5-dinitrosalicylic acid (DNS) method and total protein concentration using the Bradford assay. Data were analyzed using one-way ANOVA followed by Duncan’s post-hoc test at a 5% significance level (p < 0.05). The results showed that the mutant + SKIK treatment produced the highest values for both enzymatic activity (10.66 U mL-1) and total protein (0.85 mg mL-1). The addition of the SKIK tag enhanced protein expression and solubility, whereas mutagenesis improved catalytic efficiency. The combination of both modifications produced a synergistic effect, resulting in superior performance of the recombinant cellulase enzyme. This study is the first to combine SKIK tagging with mutagenesis to enhance the expression and catalytic activity of cellulase in E. coli. These findings are not only relevant for improving enzyme production in biotechnology industries but also hold potential applications in fish feed formulation.

Abstract

The level of pond productivity is influenced by the environmental conditions of maintenance caused by the physiochemical and biological quality of pond waters. Biological parameters of waters consist of the abundance of Phytoplankton and Bacteria. The purpose of this study was to determine the correlation between the abundance and types of Phytoplankton and Bacteria in pond water to the productivity level of whiteleg shrimp (Penaeus vannamei) ponds in different DOCs cultivated with an intensive system. The research method used observed maintenance parameters, namely the diversity and abundance of Phytoplankton including Green Algae (GA), Blue Green Algae (BGA), Diatome, and Dinoflagellate groups. Other parameters include Total Bacteria Count (TBC), Total Vibrio Count (TVC) Yellow, and Total Vibrio Count (TVC) Green. These research parameters will be subjected to Multiple Linear Regression analysis on the total harvest, feed conversion value (FCR), and survival rate (SR) of whiteleg shrimp obtained from each pond. The results of the study showed that the abundance of Phytoplankton based on the group from the lowest to the highest value was the Dinoflagellate group of 0 cells/mL with the GA group of 1.52 x 106 cells/mL. Parameters found that were significant to the level of pond productivity were the abundance of TBC and BGA (P <0.05). The conclusion is that the level of pond productivity is influenced by the abundance of Phytoplankton and bacteria based on the diversity of their species.

Abstract

Indonesia has a tropical climate with high year-round solar exposure, which increases the risk of oxidative stress and premature skin aging caused by ultraviolet radiation. Red seaweed Gracilaria verrucosa is known to contain bioactive compounds with potential antioxidant properties; however, its application as a substitute for synthetic antioxidants in cosmetic formulations remains limited. This study aimed to evaluate the antioxidant activity of G. verrucosa extract and its potential as an alternative to Butylated Hydroxytoluene (BHT) in body lotion formulations. Extraction was performed using 96% ethanol through maceration for 48 hours, resulting in an extract yield of 2.47%. The study employed an experimental method with a Completely Randomized Design consisting of five treatments, including a control without antioxidants (K0), BHT 0.1%, and G. verrucosa extract at concentrations of 3%, 5%, and 7%, with four replications. Phytochemical screening revealed the presence of alkaloids, flavonoids, saponins, phenolics, and steroids, while tannins were not detected. The IC₅₀ value of G. verrucosa extract was 63.57 ppm, indicating strong antioxidant activity. The IC₅₀ values of body lotion formulations were 1,266 ppm (K0), 137.77 ppm (BHT 0.1%), 175.95 ppm (EGV 3%), 131.82 ppm (EGV 5%), and 106.17 ppm (EGV 7%). The incorporation of G. verrucosa extract into body lotion exhibited antioxidant activity comparable to or higher than that of BHT 0.1%, indicating its potential as a safe and sustainable natural antioxidant for marine-based cosmetic formulations.

Abstract

Coffea canephora or robusta coffee was known as a medicinal plant that admitted various biological properties. However, its application as a medicinal plant in fish disease treatment has not been reported. This study aimed to investigate the effect of dietary robusta coffee leaf (RCL) extract on the survival rate and hematology of Cyprinus carpio infected by Aeromonas hydrophila. Two hundred fish were randomly divided into five treatment groups consisting of uninfected fish (UF), infected fish (IF), and a diet supplemented with different concentrations of RCL extract (30%, 40%, and 50%). The negative control group was injected by NaCl 0.9% and other treatment groups were injected by A. hydrophila. Feeding treatment was carried out for 7 days after clinical symptoms appeared. The results showed that RCL extract increases the survival rate and hematological parameters (number of erythrocytes, hemoglobin, and lymphocytes) significantly (P0.05). The highest survival rate (100%) was recorded in groups that accepted 50% of RCL extract. This current study indicates that RCL extract could be considered a potent medicinal plant for treating fish disease caused by A. hydrophila at the optimum dose of 50%.Keywords:AeromonasCoffea canephoraExtractHematologyCyprinus carpio
