Abstract

Background: Breast cancer cell's growth and survival are supported by breast cancer stem cells (bCSCs) mammosphere formation. bCSCs represent a subpopulation of undifferentiated cancer cells which associated with self-renewing abilities, tumor initiation, drug resistance, and metastasis. Overexpression of the B Cell Lymphoma 2 (BCL-2) family in many tumor cells contributes to apoptosis resistance. Trisindoline is an indole trimer alkaloid natural compound that provides a cytotoxic effect on cancer cells. A new modification of trisindoline has been synthesized into trisindoline 5 in 2021. Objective: This study purposed to investigate the effect of trisindoline 5 compounds against BCL-2 gene expression in bCSCs in vitro. Methods: The bCSCs MDA-MB-231 were divided into control and treatment groups which were further analyzed in gene expression using the qPCR Livak method. Results: Based on gene expression analysis, the results showed that trisindoline 5 may decrease the expression of BCL-2 in MDA-MB-231 cells. Conclusion: This study concludes that trisindoline 5 could downregulate the antiapoptotic BCL-2 gene expression in bCSCs in vitro.

Abstract

Background: Cancer stem cells (CSCs) are the subpopulation of cancer cells that have been demonstrated as the cause of tumor formation. The most common cancer for women and the second leading cause of death in breast cancer. Numerous genes have been involved in breast cancers, including p53. In cancer cells, p53 as well-known as the tumor suppressor gene plays an important role in directing cells with DNA damage into apoptosis. Trisindolines are heterocyclic nitrogen compounds consisting of an isatin core bearing two indole moieties that provide cytotoxic effects on cancer cells. Recently research had led to the development of the new modification of trisindoline compound into trisindoline 5. Objective: This study aims to investigate the effect of trisindoline 5 compounds against p53 gene expression in CSCs MDA-MB-231 in vitro. Methods: CSCs MDA-MB-231 were divided into control and treatment groups which were further analyzed in gene expression using the qPCR Livak method. Results: Based on gene expression analysis, trisindoline 5 increases the expression of p53 in CSCs MDA-MB-231. Conclusion: This study informed that trisindoline 5 could upregulate the expression of tumor suppressor gene p53 in CSCs MDA-MB-231 in vitro.

Abstract

Lahan terbuka menjadi kebutuhan utama bagi kehidupan masyarakat, terutama setelah COVID-19. Pandemi yang berlangsung selama lebih dari dua tahun makin menyadarkan masyarakat bahwa ruang terbuka adalah hal yang penting untuk diupayakan keberadaannya. Di antara beberapa hal yang bisa dilakukan adalah pemanfaatan lahan kosong di sekitar bantaran sungai dengan mengubahnya menjadi kawasan yang ramah lingkungan. Salah satu sungai yang dapat dikembangkan untuk menjadi sarana rekreasi terletak di Desa Kemantren, Kecamatan Tulangan, Kabupaten Sidoarjo. Kawasan ini diharapkan menjadi tempat wisata bertemakan "Urban Tourism" yang dapat dinikmati oleh masyarakat desa ataupun warga sekitar. Pemanfaatan lahan ini akan membawa dampak yang positif bagi Dusun Kemantren, karena selain warganya dapat menikmati Urban Tourism, mereka juga dapat melakukan kegiatan ekonomi di tempat wisata baru. Kas dusun pun akan terisi dengan potensi-potensi yang mengikutinya. Untuk mewujudkan fungsi tersebut, diperlukan suatu perencanaan yang memenuhi prinsip-prinsip penataan lanskap yang baik, mulai dari studi literatur, survey lokasi, analisis tapak, hingga proses perencanaan itu sendiri. Diawali dengan sketsa ide dan dilanjutkan dengan pengembangan model 3D, hasil desain dan perencanaan telah dilaksanakan sebagian dengan membuat lampu penanda dinamis yang diletakkan di dekat jalan masuk pemukiman. Selain itu, pengelolaan sampah yang menjadi perhatian utama menjadi dasar pembagian tempat sampah menjadi dua, organik dan anorganik. Sampah organik akan masuk ke dalam proses pengomposan dan sampah anoganik dapat disetorkan ke bank sampah atau diproses sebagai bahan dasar kerajinan tangan yang memiliki nilai guna dan ekonomi. Perancangan dan pembuatan Urban Tourism di Dusun Kemantren, Sidoarjo ini mengkolaborasikan institusi pendidikan dengan pemerintah desa, sehingga penerapan ilmu dapat maksimal dengan dilakukannya pengabdian masyarakat, dan pihak desa juga dapat merasakan manfaatnya secara nyata.

Abstract

Bahan bakar yang semakin berkurang maka dalam penelitian ini membahas tentang dual fuel system dimanaalternatif bahan bakar yang akan digunakan adalah etanol E98, dikarenakan nilai oktan etanol sangat tinggi makaperlu modifikasi pada rasio kompresi karena nilai oktan yang tinggi memerlukan rasio kompresi yang tinggi juga, agartidak terjadi adanya knocking, jadi pada pengujian kali ini akan dilakukan penambahan rasio kompresi dari kompresistandart mesin 9:1, CR 10,5:1 dan CR 11,5 sedangkan penambahan uap etanol pada penelitian ini di modifikasikandengan bukaan katup yaitu bukaan 0°, 30°, 60° dan 90°. Menaikkan kompresi rasio terdapat beberapa cara dimanasalah satunya dengan metode dom piston seperti yang akan dilakukan pada penelitian ini. Penelitian dual fuel yangmenggunakan etanol sebagai bahan bakar alternatifnya banyak dilakukan akan tetapi terjadi penurunan daya padamesinnya, maka pada penelitian ini diharapkan akan mendapatkan hasil nilai daya yang lebih bagus atau optimaldalam pembakaran. Dari penelitian yang telah dilakukan dengan penambahan rasio kompresi dan masuknya uapetanol yang sudah di atur dengan bukaan valve, maka didapatkan daya maksimum rasio kompresi standartdidapatkan 9,3 HP pada RPM 8000 bukaan katup 90° dan didapatkan torsi maksimum 9,7 N.m pada bukaan katup90° di RPM 6000, sedangkan pada CR 10,5:1 didapatkan daya maksimum 9,5 HP pada bukaan katup 90° di RPM 8000dengan torsi maksimum sebesar 9,41 N.m pada RPM 5500 bukaan katup 0°, pada variasi rasio kompresi terbesaryaitu 11,5:1 didapatkan daya maksimum sebesar 9,7 HP di RPM 8500 dengan bukaan katup 90° dan didapatkan torsimaksimum 9,79 N.m dengan bukaan katup 60° pada RPM 6000, jadi dapat disimpulkan variasi terbaik rasio kompresipada pengujian ini terdapat pada CR 11,5:1 dan bukaan katup 60° karena didapatkan daya maksimum terbesar.

Abstract

In the process of maintaining security and maintenance of the approach lightingsystem (ALS) at the airport area in Natuna which was built on the Natuna coast. So weneed a patrol boat that can meet the criteria in terms of the strength of the ship's structureto maneuver properly and accept internal and external loads. Structural capabilities thatare sufficiently safe are permitted according to BKI rules for ships made of fiberglassreinforced plastic. Case of slamming loading, it will be analyzed whether it is sufficientlysafe in terms of the strength of the construction structure on the 13-meter patrol boat thatloading may occur due to ship operations in the area around the airport. The initialcalculation is to calculate the scantling with a scantling weight of 2,226 tons and then drawthe construction in accordance with the lines plan and general arrangement of the 13 meterpatrol boat. Followed by the provision of a static load from slamming loading of 15,237KPa when the ship is at full speed of 20 knots to find out the response from the ship'sconstruction made of FRP. The value analyzed is the maximum stress (von mises) using theFinite Element Method (FEM) with static structural analysis and slamming loading in theform of pressure on the software as the method chosen in this study. Then raises the stressvalue in the ANSYS software, the maximum stress value that occurs in the construction is13.808 MPa and from this maximum stress value the safety factor value can be calculatedto find out how safe the ship's construction is if it is given a slamming load on the ship. Thesafety factor value in this study is 7,097. From the value of the safety factor it is knownthat the ship is still able to accept slamming loads at full speed.

Abstract

- Wind speed is the movement of air from areas of high pressure to areas of lowpressure. As the use of electric energy increases due to the growing human population in aregion, there is a need for renewable energy sources to convert renewable energy intoelectricity. Wind energy is highly potential if utilized, considering its relatively low utilizationfor electricity generation in Indonesia. In the initial stage of using wind energy for electricitygeneration, data on the wind potential of a region is required, thus an analysis of wind speedin several areas on Java Island is necessary. The wind speed data in this Final Projectincludes several regions in the Java Province, namely: Bandung, Banyuwangi, Bekasi,Gresik, Kudus, Malang, Semarang, Surabaya, Surakarta, and Tuban. The method used inmodeling wind speed involves the Weibull distribution, Rayleigh distribution, and Gammadistribution. Goodness-of-fit tests are applied to examine the best probability functions ofeach distribution to be analyzed. The distribution that meets the criteria of Coefficient ofDetermination, Chi-Square, and Root Mean Square Error is the Weibull distribution, asevidenced by the probability plots of the Weibull distribution, which closely match the actualdata from various regions. Areas with potential for wind energy to be converted intoelectricity are evaluated based on the percentage of wind speeds exceeding 2 m/s. The areaswith the highest potential are Tuban with a percentage of 56.89%, Malang with 56.71%, andBandung with 50.20%.

Abstract

Society is very dependent on fossil fuels, and currently Indonesia does not haveexcess oil reserves. According to the DEN, oil reserves will be exhausted in the next nineyears. As an alternative, wind turbines can be used. However, the factor hindering thisdeployment is the design of the wind turbines, which is lacking. MPPT is used in windturbines to optimize performance by determining the state of the wind turbine, thencalculating the power sampling algorithm, and then calculating the overall performance andefficiency of the wind turbine. State 1, Vci 1 m/s, Vr 6 m/s, and Vco 8 m/s provide the highestpower of the three. The H_Single Cp 0.47 and Tsr 4.5 turbines with a ratio of one rotor aresuitable for use in several areas on the island of Java. If you want a greater output powerfrom a wind turbine, then you can use a turbine with dual rotors, namely the HCR turbine,because it is capable of producing almost twice as much as a turbine with one rotor. Thearea with the most potential to generate electricity is in the Tuban area. Overall efficiencywith a maglev wind generator (model WKG-1000-200r) gives a Savonius efficiency of 18.2%,an H-Darrieus efficiency of 22.1%, an HCR efficiency of 63%, and an H-Single efficiency of42.3%.

Abstract

The maintenance of the swaying light system requires fast handling to meet the proceduralneeds of aviation. Therefore it is necessary to carry out an in-depth analysis of seakeeping andoperability to reduce the risk of ship delays when needed when sudden swaying lights are repaired.Movements caused by external forces acting or sea waves will affect the comfort and safety of the crewand passengers of the ship itself. Ship movement response is a seakeeping parameter that must beconsidered to support the feasibility of ship design.In this final project, the seakeeping analysis processuses Maxsurf Motion software to generate ship movement values. The variations in the direction ofarrival of the waves used are 00, 450, 900, 1350, and 1800 at a speed of 20 knots. Meanwhile, variationsin significant wave height are 0,05; 0,3; 1; 2; dan 3,3 meter and wave period used are 5,6; 6,8; 7,5;10; and 11 second and ship shipping routes according to Natuna waters wave data from BMKG.Calculations are performed using Maxsurf Motions Advance software.The results that can beconcluded in this final project are the percentage of ship operability based on seakeeping criteriaaccording to Olson 1978 and the U.S Coast Guard where 94,06% is operable and 5,94% downtime. Inaddition, it can also be concluded that the operational wave height that is permitted and safe for shipssailing in the waters of the Natuna Sea is 3 meters.

Abstract

The condition of Indonesia as an island nation that has many islands with potential for tourismand investment in the field of natural resources is abundant. The construction of airports in the islandsis an effort to support this potential, but due to limited land, some airport equipment that is importantand not eliminable is placed in the water area. Patrol boats are very important, to assist in airportoperations, especially for maintenance of equipment placed in the waters. So a patrol boat with FRP(Fiberglass Reinforced Plastic) material was designed. From the research conducted, the ship is designedto have a speed of 20 knots with the main size of the ship, namely 13 meters long, 3.6 meters wide, 1.5meters high, 0.6 meters of water level obtained from the ratio comparison of existing comparison ships.From modeling using the size of the ship that has been obtained, the linesplan as found Picture 2, andthe general plan as found in Picture 8 are also obtained after the calculation stages. The patrol boat isdesigned using 2 Yanmar 6LY400 engines, with a ZF 115 ATS gearbox and a B5-105 type propeller, fromthe propulsion system the ship can reach a speed of 21.31 knots at 100% engine power. The patrol boatin this study is analyzed for stability with eight conditions where all conditions meet the rules and criteriaof HSC 2000 ANNEX 8.

Abstract

Tuberculosis (TB) is an infectious yet often overlooked disease that remains a significant global challenge. Pyrazinamide (PZA), a key drug in the first-line TB treatment regimen, is used to reduce the duration of therapy, making it a compound of great interest for further exploration. Two pyrazine-2-carboxamide analogs have been successfully synthesized and reported, followed by an in-silico evaluation of their potency as antituberculosis agents. Yamaguchi reagent was employed as a coupling agent between pyrazine-2-carboxylic acid and corresponding amine, yielding N-(cyclohexylmethyl) pyrazine-2-carboxamide (D) and N-(4-cyclooctyl) pyrazine-2-carboxamide (E) in 60% and 55%, respectively. The molecular docking analysis of compounds (D) and (E) demonstrated lower binding energies (-7.65 and -7.37 kcal/mol, respectively), in comparison with the standard TB drugs, pyrazinamide and isoniazid. Additionally, ADME and pharmacokinetics evaluations revealed that compounds (D) and (E) meet the essential criteria for oral drug candidacy. These findings suggest that the pyrazinamide analogs (D) and (E) hold significant potential as promising antimycobacterial agents for tuberculosis therapy.
