Abstract

Virus infection is one of the major constraints in garlic production since the viruses are readily accumulated on vegetative propagation material (bulbs). This research aimed to detect garlic common latent virus (GCLV), shallot latent virus (SLV), onion yellow dwarf virus (OYDV), and leek yellow stripe virus (LYSV) infecting local garlic as seed bulb and imported garlic as consumption bulb. Seed bulb samples were obtained from seed breeders in several garlic growing centers in Indonesia. In contrast, consumption bulb samples were obtained from plant quarantine warehouses and three local markets in Bogor. Some bulb samples were used for morphological observations, and some were germinated in the laboratory until the leaves emerged. Leaves were collected for virus detection by RT-PCR using specific primers for GCLV, SLV, OYDV, and LYSV. Seed and consumption bulbs have differences in their morphological characteristics, especially in the type of neck hardness and the size of the bulb diameter. OYDV and LYSV infections were successfully detected in seed and consumption bulbs, while SLV was only found in consumption bulbs. Nucleotide sequence analysis showed that SLV from consumption bulbs formed one group, GCLV from seed bulbs formed one group, while OYDV and LYSV from seed and consumption bulbs were in different groups, indicating that the viruses came from different strains. Further research through high-throughput detection methods and providing virus-free planting material are needed to anticipate the spread of new strains of garlic viruses in Indonesia.

Abstract

Aphelenchoides fragariae, commonly known as strawberry crimp nematodes, primarily target the aerial parts of plants, affecting both internal and external structures. In Indonesia, where strawberries are predominantly cultivated in highland regions, the presence of strawberry crimp disease has been confirmed. Infected plants exhibit symptoms such as stunted growth, reddened foliage, crimped or curled leaves, and malformed buds and blooms. Aboveground damage caused by the nematodes includes contorted shoots, undersized leaves, and reddish petioles, often accompanied by discolored patches on the foliage. These symptoms significantly impair the growth and productivity of strawberry plants, highlighting the nematode’s potential as a serious pest in these regions. The identification of A. fragariae was achieved through a combination of morphological and molecular characterization methods. Species confirmation relied on PCR amplification of the nematode’s cytochrome oxidase subunit I (COI) gene, using primers (COI F and COI R) designed in the laboratory. The amplification yielded a specific fragment of approximately 550 base pairs, which was sequenced for further analysis. Sequence alignment revealed identity levels ranging from 82.8% to 99.7%, confirming the presence of A. fragariae. The resulting sequences were deposited in GenBank under the accession numbers LC804455 (A. fragariae isolate RB) and LC804456 (A. fragariae isolate LB), providing a valuable resource for future studies on this nematode species.

Abstract

Infeksi virus dapat menyebabkan gejala yang bervariasi pada tanaman. Penilaian tingkat keparahan gejala virus tanaman di lapangan umumnya menggunakan metode konvensional berbasis pengamatan visual. Seiring dengan perkembangan teknologi digital saat ini, telah dikembangkan pula metode penilaian terkini berbasis analisis citra digital. Namun informasi terkait kepresisian dan keakurasian kedua metode penilaian tersebut masih sangat terbatas khususnya penilaian keparahan gejala yang disebabkan oleh virus. Penelitian ini bertujuan untuk mengevaluasi hasil penilaian keparahan penyakit berdasarkan metode konvensional dan analisis citra digital berbasis NDVI (Normalized Difference Vegetation Index) serta membandingkan akurasi kedua metode penilaian tersebut dalam mengestimasi variabel produksi tanaman. Survei lapangan dilakukan dengan melakukan perekaman citra dan pengamatan secara visual terhadap gejala serangan virus pada tajuk tanaman cabai. Dilanjutkan dengan pengukuran bobot buah sampel tanaman, dan analisis citra digital tanaman. Hasil penelitian menunjukkan bahwa penilaian keparahan gejala virus menggunakan metode analisis citra digital berbasis indeks vegetasi NDVI lebih presisi (standar deviasi < 0,04) dibandingkan metode pengamatan visual (standar deviasi > 4,00), khususnya pada fase generatif. Selain itu, metode indeks vegetasi NDVI juga lebih akurat dalam mengestimasi bobot buah cabai karena koefisien regresi bertanda positif dengan nilai R2 > 0,90.

Abstract

Colletotrichum acutatum is known as the major causal agents of anthracnose disease of chilli pepper in Indonesia. This research was aimed to study genetic variability of C. acutatum in Java and Sumatera based on morphological and molecular characteristics and to evaluate pathogenicity of several isolates. Observation on morphological characteristics involved colour and growth of each colony as well as shape and size of conidia. Molecular identification was performed by DNA amplification using specific primer for C. acutatum, i.e. CaInt2/ITS4 followed by sequencing and nucleotide sequence analysis. Pathogenicity test for each isolate on Capsicum annuum and C. frutescens was conducted in the laboratory using detached chilli. Morphology variability was observed from 40 isolates of C. acutatum. Most of the isolates have white/beige colonies with fusiform conidia of 6.11–9.73 µm in length and 2.24­–2.73 µm in width. Pathogenicity of 3 C. acutatum isolates varies from moderate to high based on lesions size on infected chilli. Abstract

Application of Guano Filtrate on Infection of Pepper yellow leaf curl virus in Chilli Plants Pepper yellow leaf curl virus (PYLCV) is a member of Begomovirus genus, which causes yellow leaf curl disease in chili plants in Indonesia. This virus is transmitted by vector Bemisia tabaci (Hemiptera: Aleyrodidae). Research was carried out to determine the potential of guano in suppressing yellow leaf curl disease in chili pepper. Field experiment was conducted using factorial design in a randomized block design with 2 factors. The first factor was chili cultivar (‘Gelora’, ‘Bara’, and ‘Pelita 8’) and the second factor was treatment of guano filtrate (before virus inoculation, 1 week after virus inoculation, 2 weeks after virus inoculation, virus inoculation without guano, without virus inoculation nor guano). Virus inoculation was carried out using B. tabaci. In general, symptoms were developed 1 to 3 weeks after inoculation although the incubation period varied between chili cultivars. Green mosaic with leaf curling was mostly found in ‘Gelora’, whereas yellowing with leaf cupping was mostly found in ‘Bara’ and ‘Pelita 8’. Begomovirus infection on plant showing symptoms has been confirmed by polymerase chain reaction method. Application of guano did not cause suppression on disease incidence and severity. Similarly, it did not affect plant height and flowering period.

Abstract

Infeksi virus pada tanaman bawang merah di Indonesia merupakan salah satu faktor yang dapat menyebabkan penurunan produktivitas tanaman, terutama karena sifatnya yang terbawa umbi. Deteksi virus untuk memantau insidensi penyakit di lapangan atau memeriksa benih sehat membutuhkan metode yang akurat dan dapat diandalkan. Pada penelitian ini dilakukan deteksi anggota Potyvirus, yaitu Onion yellow dwarf virus (OYDV) dan Shallot yellow stripe virus (SYSV) menggunakan metode double antibody sandwhich enzyme-linked immunosorbent assay (DAS-ELISA) dan reverse transcription polymerase chain reaction (RT-PCR) dari sampel daun bawang merah. Sampel daun dikumpulkan dari beberapa daerah penanaman bawang merah di Brebes dan Cirebon. Infeksi OYDV dan SYSV dideteksi menggunakan metode DAS-ELISA, selanjutnya sampel lapangan yang memberikan reaksi positif dipilih untuk deteksi lebih lanjut menggunakan metode RT-PCR dan perunutan asam nukleat dengan sikuensing. Berdasarkan metode DAS-ELISA, infeksi OYDV dan SYSV berhasil dideteksi dari sampel lapangan sebagai infeksi tunggal dan infeksi campuran. Infeksi Potyvirus terkonfirmasi dengan metode RT-PCR dari sampel lapangan yang memberikan reaksi positif dalam ELISA. Analisis sikuen nukleotida menunjukkan infeksi OYDV baik pada sampel yang bereaksi positif dengan antiserum OYDV maupun SYSV. Hasil tersebut mengindikasikan terjadinya reaksi silang antara antiserum OYDV dan SYSV.

Abstract

Identifikasi Polerovirus yang Menginfeksi Tanaman Cabai di Jawa Infeksi polerovirus pada tanaman cabai semakin banyak dilaporkan dari berbagai bagian dunia termasuk di Indonesia. Infeksi Pepper vein yellow virus (PeVYV) pada tanaman cabai di Bali, Jawa, dan Aceh telah dilaporkan pada kurun waktu 2012 sampai 2020. Belum lama ini gejala infeksi polerovirus berupa daun kuning dengan penebalan tulang daun ditemukan pada pertanaman cabai di wilayah Jawa Tengah, yaitu di Brebes, Magelang dan Kulon Progo. Deteksi molekuler dengan metode RT-PCR menggunakan primer universal polerovirus O3R/PL4F yang dilanjutkan dengan analisis sekuens membuktikan adanya infeksi PeVYV dari sampel asal Magelang dan Kulon Progo. Identifikasi lebih lanjut menunjukkan bahwa isolat PeVYV tersebut ialah PeVYV-2, PeVYV-6, dan PeVYV-8. Insidensi infeksi PeVYV ini menunjukkan perluasan distribusi Polerovirus di Indonesia.

Abstract

Pepper yellow leaf curl disease (PYLCD) is the primary disease that affects chili plants, resulting in a loss of quality and quantity. The identification of chili plants resistant to PYLCD is a solution to support optimal chili cultivation. The purpose of this study was to estimate genetic parameters to obtain information on the genetic control of chili resistance to PYLCD. The genotypes used in this study consisted of four parental, four hybrid genotypes, and 180 individuals of the F2 population. The results showed that the genotypes "F6074" and "IPBC12" were categorized as resistant, while the genotypes “IPBC5” and “YUNI” were categorized as susceptible. The distribution of resistance level in chili peppers to yellow curly leaf disease tends to follow a ratio of 13:3, with a dominance of susceptible traits. The assumption for this phenomenon is that resistance to PYLCD is controlled by at least two genes. Both genes have dominant and recessive epistatic interactions. Abstract

Aphid-borne viral infection is a major limiting factor for groundnut production in South Sulawesi, Indonesia. The study aimed to elucidate the effectiveness of integrated management packages (IMP) in controlling aphid-borne viruses infecting groundnut in the field. Field trials were conducted in a plot without (M0) and with plastic mulch and maize as a plant barrier (M1), while two different groundnut varieties, Bougainvillea leaf extract, and insecticides were incorporated as the subplot treatments. The treatments in M1 plot significantly reduced the aphid population (17.4–79.9%) and AUDPC incidence (51.7–87.3%), and severity (61.0–92.9%), respectively. The treatments increased dry pod production by 11.9–63.3% depending on subplot treatment compared to treatments in the M0 plot (p>0.05). Bean common mosaic virus strain PSt and Cowpea Polerovirus 2 were positively detected in groundnuts as well as in aphids using RT-PCR. Further, nine weed species in the vicinity of the field trial were found to be positively infected with BCMV-PSt, indicating their crucial role as reservoir hosts. Interestingly, the effectiveness of Bougainvillea leaf extract treatment in the M1 plot was comparable to that of the insecticide treatment, indicating that this IMP is a more cost-effective, eco-friendly groundnut virus control than the currently used method.

Abstract

Infeksi patogen tanaman merupakan salah satu penyebab menurunnya produksi bawang merah di Indonesia. Salah satu penyakit utama pada bawang merah adalah busuk pangkal batang yang disebabkan oleh infeksi Fusarium oxysporum. Infeksi Fusarium patogenik pada umbi benih bawang merah dapat menyebabkan kejadian penyakit busuk pangkal batang pada tanaman bawang merah. Oleh karena itu perlu dilakukan uji kesehatan umbi benih, untuk mendeteksi keberadaan Fusarium patogenik pada umbi benih bawang merah. Uji kesehatan umbi benih bawang merah dilakukan dengan menggunakan metode blotter test. Selanjutnya isolat Fusarium yang muncul dari hasil blotter test diuji sifat patogenisitasnya pada umbi dan tanaman bawang merah. Penelitian ini bertujuan untuk mendeteksi keberadaan Fusarium terbawa umbi benih bawang merah pada beberapa varietas dan sifat patogenisitasnya. Hasil penelitian menunjukkan varietas bawang merah dengan persen infeksi Fusarium tertinggi pada varietas Bima (74,5%) dengan nekrosis basal plate (79%). Hasil uji patogenisitas menunjukkan dari 60 isolat Fusarium diperoleh dari uji blotter test, 46,6% merupakan isolat Fusarium patogenik.
