Abstract

Previous studies have shown that the formulations of clove bud essential oil (CBEO) with a concentration of 5% in O/W cream and 2.5% in W/O cream have anti-inflammatory properties. The formulations are developed by adding enhancers, namely oleic acid and propylene glycol. The optimum compositions of oleic acid and propylene glycol are 50%:50% in W/O cream and 30%:70% in O/W cream. This study aimed to determine the effects of different compositions of creams on their anti-inflammatory activities. The creams were evaluated for their anti-inflammatory properties using mice with croton oil-induced inflammatory. The mice were sacrificed, and the back skin was removed to make histopathologic preparations at the end of the three-day inflammatory induction to obtain data of epidermal thickness and the number of cells with COX-2 expression. The results showed that the addition of oleic acid and propylene glycol as enhancers increased the anti-inflammatory activity of both O/W and W/O creams. Since the O/W cream decreased the thickness of the epidermis and the number of cells with COX-2 expression more than the W/O cream, the formulation of CBEO in O/W cream is concluded as a better anti-inflammatory than that of W/O cream.

Abstract

The fatty acids, like EPA and DHA, were known as anti-inflammation. It works on inflammatory tissue, brought by edema plasma, and changed into resolvins, protectins, and maresins by the enzyme reaction. Anguilla bicolor and Sea cucumber fish are known to contain EPA and DHA. This study analyzes the inhibition Cox-2 effect of the combination of Anguilla bicolor oil with Sea cucumber extract as an active ingredient in a cream preparation. This experiment used 7 groups of male BALB/C strain mice: normal control; negative control; positive control; A. bicolor oil cream; H. leucospilota extract cream; a combination of A. bicolor : H. leucospilota (2:1) cream; a combination of A. bicolor : H. leucospilota (1:2) cream. The anti-inflammatory activity was evaluated by the amount of inflammatory cell, the thickness epidermis, and the amount of cell expression COX-2  on mice's back skin tissue induced by croton oil (0,1 %). After 3 days, histopathological skin tissues were made. The data were analyzed statistically by One Way ANOVA followed by LSD to know each group's differences at a significance level of 0.05. The experiment results showed that the formula with the best anti-inflammatory activities is the combination of A. bicolor and H. leucospilota (2:1) cream.  The decrease of the amount of inflammatory cell (75.97%), the thickness of the epidermis (43.88%), and the amount of COX-2 cell expressions (60.52%) from the formula did not differ significantly with the positive control (p>0,05). It can be concluded that A. bicolor and H. leucospilota have anti-inflammation activity based on the experiment results.

Abstract

Indonesia is a country known for its source of biological wealth, one of which is beluntas leaves. Beluntas leaves have the potential to be an antibacterial, so it is appropriate to be formulated in the form of medicinal preparations, especially gels. This study aims to find out the influence of variations between carbopol gel base 940 and Methylcellulose Hydroxypropyl (HPMC) on the physical properties of gel preparations beluntas leaf extract (Pluchea indica (L.), and know the influence of gel of extract of beluntas leaves on antibacterial activity. The extract is obtained by the maceration method using ethanol solvent 96%. Each formula uses 15% of the extract of beluntas leaves. Gels are made in four gel base variations namely F0 (0.5% carbopol, 1% HPMC), FI (1% carbopol, 1.5% HPMC), FII (1.5% carbopol, 2.5% HPMC), and FIII (2% carbopol, 3% HPMC). Gels evaluated for their physical properties include organoleptic, viscosity, pH, homogeneity, scattering power, adhesion and Freeze-thaw cycling. Then the gel tested antibacterial activity against bacteria Staphylococcus aureus and Pseudomonas aeruginosa by cup-plate diffusion method. The data obtained were analyzed with One Way Anova and LSD with a 95% confidence level. The results showed that beluntas leaf extract gel meets the organoleptic requirements, homogeneity, good gel adhesion (> 4sec), good gel viscosity (2000-50.000 cps), and good gel pH (4.5-6.5). However, the gel does not meet the requirements of good scattering power (5-7 cm) and Freeze-thaw cycling. Based on the test results that have been done with some of the parameters above, The best composition of carbopol 940 and HPMC in the beluntas leaf extract gel which has antibacterial activity against Staphylococcus aureus and Pseudomonas aeruginosa  is 1% carbopol and 1.5% HPMC. Abstract

Abstract

Green tea extract cream contains epigallocatechin gallate (EGCG) as the active ingredient for anti-inflammatory. Epigallocatechin gallate is easyly oxidized and able to reduce its effectivity as an anti-inflammatory. Therefore, an addition of antioxidants in order to increase its stability is required. The purpose of this study was to determine the effect of adding the antioxidant Vitamin C on the effectivity of green tea extract as an anti-inflammatory. This study uses 6 groups of male mice strain BALB/C which were given treatment as follows: normal control, negative control, base cream, green tea extract (0.2%), Vitamin C cream (1%) and green tea extract cream with addition of Vitamin C. The anti-inflammatory activity was evaluated based on the expression of COX-2, inflammatory cells and the thickness of the epidermis in the skin tissue of mice after given crotton oil (0.1%) on the back for the induction of inflammation. After treatment cream for 3 days, mice were sacrificed for histopathological tissue preparations made with hematoxylin eosin staining and immunohistochemistry COX-2. Data were analyzed statistically with one way Anova followed by t-test to determine differences between groups at a significance level of 0.05. The test results indicate that cream of green tea extract is higher in decreasing inflammatory parameters in comparison with cream of Vitamin C, except in the thickness of epidermal parameter. Green tea extract cream with the addition of Vitamin C is higher in reducing inflammatory parameters than cream of green tea extract or cream of Vitamin C. The decline percentage of cells that express COX-2, inflammatory cells and the thickness of the epidermis in the each of groups were cream of green tea extract:57.95%;53.75%;34.83%, cream of Vitamin C:48.76%;34.96%;34.27%, cream of green tea extract and Vitamin C:61,89%;65,54%;46.30%, respectively. Based on the results of this study, it can be concluded that anti-inflammatory activity of green tea extract cream increased due to the addition of 1% vitamin C as an antioxidant.
