Abstract

Bay leaves (Syzygium polyanthum) contain the flavonoid quercetin which can be used as an antihyperlipidemic drug. The development of antihyperlipidemic drug formula in the form of fast disintegrating tablet (FDT) is needed for patients who experience dysphagia. FDT preparations require an optimal super disintegrant concentration to produce a good drug formula. This study aims to develop the FDT formula of bay leaves extract using the super disintegrant croscarmellose sodium (CCS) intra and extra-granular. FDT formulation using the wet granulation method with variations of CCS concentrations; F1: 2%, F2: 3.5%, and F3: 5% for extra granular, and 2% for intra granular. The formulation process, in-process control (IPC) granules, weight uniformity tests, and various physical properties tests of tablets were carried out. Data were statistically analyzed using one way ANOVA test (α=95%). The results of statistical tests of IPC granules, uniformity of weight, and tablet size of all FDT formulas were not significantly different (p>0.05). The CCS concentration for extra granular significantly affected the wetting time, disintegration time, hardness, and the value of friability percentage of FDT (p<0.05). The combination of intra and extra-granular CCS (2%:5%) gave the most optimum physical properties of bay leaf extract FDT.

Abstract

Clove extract is a natural ingredient abundant in Indonesia that has various benefits, including being an antioxidant and anti-inflammatory. Although Research has been conducted, it has been limited to discussing the effects of clove extract on muscle damage. This study aims to determine the effect of clove extract on skeletal muscle damage due to excessive eccentric exercise in mice. We used experimental research methods by a post-test only with control group design. Fifteen male mice were randomized into 5 groups. Group I: Normal control, Group II: Negative control, treatment group with a dose of 100 mg/kg BW, treatment group with a dose of 150 mg/kg BW, and treatment group with a dose of 250 mg/kg BW. Measurement of Nicotinamide Adenine Dinucleotide Phosphate Oxidase (NOX) and Toll Like Receptor4 (TLR 4) levels were carried out using the ELISA method via Quadriceps muscle samples, and skeletal muscle damage was viewed through an x1000 light microscope. The results of this study carried out using ANOVA showed results of p 0.004 and 0.006, which means that there was a decrease in oxidative stress as measured using NOX and a decrease in the inflammatory stimulator TLR 4 in skeletal muscle damage due to eccentric exercise.ANOVA test results show that there are differences in influence between treatment groups and show that clove extract has benefits as an antioxidant and anti-inflammatory in muscle damage. According to this study, clove extract has the potential to be an antioxidant and anti-inflammatory which can reduce NOX levels and reduce the inflammatory, mediator TLR4 in muscle damage caused by excessive eccentric exercise.

Abstract

Nanostructured Lipid Carries NLCs have gained attention in formulation science and nanotechnology due to their biocompatible material and ability to enhance skin penetration. The goal is to create products that can effectively deliver active compounds to desired skin layers with minimal side effects. This review focuses on the use of functionalized NLCs in dermocosmetics, specifically looking at their composition, manufacturing method, characteristics, and role in delivering active compounds. The review obtained articles from databases such as Scopus, Pubmed, and Google Scholar, covering the years 2014-2024. Data search was conducted in May 2024. The search used keywords such as "NLC OR formulation OR dermal OR activity”. By considering factors such as NLC composition, manufacturing method, and impact on compound delivery, researchers aim to optimize NLCs for use in topical preparations. This research contributes to the development of dermocosmetics that can address various skin problems more effectively. The search results from 32 articles gave an overview of the widely used NLC components: solid lipids: glycerol monostearate, compitrol 888 ATO, and tristearin; liquid lipids: myglyol, transcutol, and oleic acid; surfactants: poysorbat 80 and polysorbate 20. The widely used methods were ultrasonication and high pressure homogenization. Frequently performed characterizations include particle size, polydispersity index (PDI), zeta potential, and Entrapment Efficiency (EE). The pharmacological activity of active components loaded into NLCs increased than without NLCs. The active ingredient formulated in NLC can produce better activity compared to the active substance formulated directly in the conventional preparation. The application of nanostructured lipid carrier (NLC) nanoparticles on the skin is very beneficial. These nano systems have shown promising results and more commercial formulations, so it is expected to be done in further research.
