Abstract

Kesehatan merupakan salah satu kebutuhan penting setiap orang karena dengan tercapaianya kesehatan akan memungkinakan terlaksananya kegiatan atau aktifitas yang lain. Kesehatan merupakan hak setiap orang dengan tidak mengenal tingkat ekonomi seseorang. Ekonomi yang minim diprediksi menjadi salah satu hambatan dalam mendapatkan kesehatan yang maksimal. Peran tenaga kesehatan sangat diperlukan dalam mambantu pasien mendapatkan kesehatan yang optimal. Salah satu kegiatan yang dapat membantu masyarakat terutama dengan ekonomi yang minim adalah pengobatan gratis. Pengobatan gratis yang merupakan salah satu kegiatan pengabdian kepada masyarakat telah dilakukan di balai desa Potronayan kecamatan Nogosari, kabupaten Boyolali pada ahad 31 Juli 2022. Selain pengobatan gratis, kegiatan yang lain adalah pemeriksaan tanda-tanda vital, pelayanan obat dan konseling obat serta pelayanan gizi. Kegiatan ini diikuti oleh 105 pasien. Pasien kebanyakan adalah wanita (73,3%) dengan usia terbanyak adalah antara 51 tahun sampai 60 tahun sebanyak 32,38%.

Abstract

Kabupaten Boyolali memiliki 19 kecamatan. Masing-masing kecamatan menghasilkan komoditas tanaman pangan yang berbeda beda. Salah satu komoditas yang dihasilkan di Kabupaten Boyolali adalah komoditas padi-padian, sayuran, dan empon-empon. Salah satu daerah penghasil empon-empon adalah desa Potronayan. Desa Potronayan merupakan desa binaan UMS yang terletak di pinggir Boyolali sebelah utara bandara Adisumarmo dengan jumlah penduduk sekitar 4.984 jiwa yang mayoritas penduduknya berpenghasilan dari bercocok tanam, salah satunya adalah tanaman yang merupakan jenis obat herbal seperti jahe, temulawak, kunyit, dan kencur. Rimpang (empon-empon) ini banyak digunakan sebagai ramuan obat tradisional dan sebagai bumbu dalam masakan. Selain itu empon-empon juga banyak digunakan untuk pengobatan maupun pencegahan gangguan kesehatan. Kegiatan ini bertujuan untuk (1). Meningkatkan harga jual empon-empon, (2). Membuat diversifikasi produk berupa herbal healthy drink, (3). Mempromosikan produk menggunakan platform online E-Commerce yang dapat diakses melalui internet. Kegiatan ini juga merancang dan mengaplikasikan green economy. Green economy meningkatkan kesejahteraan manusia dan kesetaraan sosial, sekaligus mengurangi risiko lingkungan secara signifikan. Hasil Kelompok Wanita tani berupa rimpang yang berkhasiat tersebut sebelum dijual bisa diolah menjadi suatu produk herbal healthy drink yang mempunyai daya jual lebih tinggi daripada bahan mentahnya.

Abstract

Interleukin-8 is a pro-inflammatory cytokine that plays a role in the inflammatory response. Sea cucumbers (Holothuria sp.) are used as alternative medicines that have anti-inflammatory activity. The study aims to determination of Sea cucumber (Holothuria sp.) as anti inflammation activty based on capability of reducing interleukin-8 (IL-8) on Rats induced by Carrageenan. Experimental research used was a post-test-only control group design. A total of 25 rats were divided into 5 groups, namely negative control, positive control (Na. Diclofenac), and treatment groups given Sea cucumber extract (doses of 45, 90, and 180 mg/ g BW). Carrageenan-induced mouse paw edema model was used for induction of inflammation. IL-8 of Paw tissue were measured by an ELISA reader at 450 nm wavelength. The result showed that treatrment groups has anti-inflammation activity based on IL-8 levels compared to negative and positive controls. The lowest levels of IL-8 showed at a dose of 90 mg/g BW. Based on One-way ANOVA, there were no significant differences of between groups. Sea cucumber has anti-inflammation activity by reducing IL-8 at all dose tested

Abstract

Determination of isoniazid concentration for therapeutic drug monitoring must produce valid values, so validation of the analytical method of LC-MS/MS needs to be carried out. This research aims to obtain valid method of LCMS/MS in order to determination isoniazid in rats serum. Sample preparation was carried out using the liquid-liquid extraction method using acetonitrile and methanol (2:1). The chromatography system was developed with a C18 column (ACQUITY UPLC® HSS T3 1.8 µm, 2.1 × 100 mm), the mobile phase was 70% acetonitrile with 0.1% formic acid (A), water with 0.1% formic acid (B), as well as methanol (C) with a flow rate of 0.3 mL/min, with gradient elution, detection in ESI+ mode with multiple reaction monitoring (MRM) at the transition 138 m/z → 121 m/z. The research results showed that the method was accurate with a % recovery of 96 - 113% and an RSD of 2.4 ± 0.9%. The precision value was obtained with an average RSD of 3.7 ± 2.4% for intra-day and 4.1 ± 2.3% for inter-day. Linearity showed good results with a correlation coefficient of 0.9956 in tests with 5 different concentrations. The LOQ and LOD values obtained from the linear regression equation were 0.1987 µg/mL and 0.0656 µg/mL respectively. Abstract

Previous research stated that galangal (Alpinia galanga) extract has a potential as cytotoxic agent with active compound of 1’-Acetoxychavicol Acetate (ACA). The objective of this study was to determine the selectivity of ethanol extract, ethyl acetate fraction, and methanol fraction of of galangal, and ACA on cancer cell lines. Cytotoxic activity was carried out using the MTT method on T47D breast cancer, WiDr colon cancer, HeLa cervical cancer, and Vero normal cell lines. The results showed that galangal ethanol extract and its fractions had selectivity index equal to or less than 2 on cancer cells. Meanwhile, ACA had selectivity index more than 3 on T47D cell and HeLa cell. ACA showed a strong cytotoxic activity against cancer cells T47D, HeLa, and WiDr with IC50 values of 3.14, 7.26, and 12.49 μg/ml, respectively. Abstract

Protein merupakan senyawa organik kompleks dengan bobot molekul besar dan sebagai sumber nutrisi yang paling baik untuk pertumbuhan. Susu adalah salah satu hasil ternak yang dikenal sebagai bahan makanan bernilai gizi tinggi dan sering dikonsumsi untuk memenuhi kebutuhan protein tubuh salah satunya ialah protein. Metode kjeldahl digunakan untuk menganalisis kadar protein kasar dalam bahan makanan secara tidak langsung, misalnya pada asam amino, protein dan senyawa lain yang mengandung nitrogen, karena yang dianalisis dengan cara ini adalah kadar nitrogennya. Analisis Kualitatif yang telah dilakukan pada sampel susu bubuk merek X didapatkan hasil positif mengundung protein dengan timbul-nya warna ungu atau biru violet akibat pereaksi biuret yang bereaksi dengan gugus amida asam bersama gugus lainnya. Analisis kualitatif dilakukan menggunakan metode metode Kjedahl, dapat disimpulkan bahwa sampel mengandung protein 4,56 % + 0,1% mendekati klaim produk 5%.

Abstract

Cervical cancer is the leading cause of death in women in the world. Cases of cervical cancer are the majority causes of death occurring in developing countries. The existing medicines which have a lot of functions as anticancer, however, still can damage the normal cells in our body. Cancer treatment using chemotherapy also has some side effects. The condition encourages researchers to explore anticancer cure made of natural ingredient. This research was conducted to find the potency of ethanol extract of herb kitolod (Isotoma longiflora (L) Presl.) as a cervical cancer cure to heLa cell. The method being used for the cytotoxic test on HeLa cells was MTT assay. Absorbant performed by ELISA reader at a wavelength of 595nm. Phytochemical screening which had been conducted showed a significant result that kitolod plants contain secondary metabolites compounds such as flavonoids, steroids, saponins, tannins and alkaloids. The results of cytotoxic test of the kitolod herb ethanol extract on HeLa cells by using MTT assay method acquired a value of IC50 of 227 μg/mL. This study showed that the ethanol extract of the herb kitolod had the potency to inhibit cervical cancer cells (HeLa cells) in a moderate ability.

Abstract

In certain conditions of cancer, isoenzymes of glutathione-S-Transferase (GST) showed increased activity. Consequently, anticancer drugs would be inactive due to increase of phase II metabolism through conjugation with glutathione. In order to increase the effectiveness of anti-cancer drugs, a GST inhibitor is required. One of the natural sources have antioxidant activity is snakehead fish. The study aims to determine GST inhibition activity of the combination of extract of snakehead fish (SF) and sea cucumber (SC). This activity was determined using rat liver and lung microsomal-cytosolic fractions obtained through the Lundgren method. GST activity was measured spectrophotometrically using a simple kinetic program at a wavelength of 345 nm with a DCNB (1.2-dichloro-4-nitrobenzene) as a substrate. The results showed an increase in the percentage of inhibition GST enzymes of the liver and lungs of rats by extracts and their combinations. The data of IC50 values showed that the smallest value for liver GST was found in SC with the value of 117.7 µg/mL and for lung GST enzymes, it was a combination of SC-SF (1:2) with a value of 45.6 µg/mL. This showed that these two samples had the greatest activity in inhibiting lung and liver GST enzymes and the potential as a complementary therapy for cancer patients.

Abstract

Breast cancer remains one of the most prevalent malignancies worldwide. The use of combinations of natural products is increasingly recognized as a promising strategy for cancer treatment. Turmeric rhizome (Curcuma longa L.) and soursop leaves (Annona muricata L.) are two natural materials known for their anticancer potential. This study aimed to identify the phytochemical constituents of turmeric rhizome and soursop leaf extracts and to evaluate the anticancer activity of their combination against T47D breast cancer cells. Turmeric and soursop leaves were extracted with 96% ethanol using the maceration method. Raw material standardization was performed by measuring water content, ethanol-soluble content, and water-soluble content. The extract was standardized by thin-layer chromatography. T47D and Vero cell lines were used in this study. Compound identification was performed using Liquid Chromatography–Quadrupole Time-of-Flight Mass Spectrometry (LC/Q-TOF-MS). The candidate combination ratio was determined using the simplex lattice design approach in Design Expert 13 software. Cytotoxicity and antiproliferative effects were assessed using the MTT assay, while antimetastatic potential was evaluated through the scratch assay. Apoptosis and cell cycle arrest were analyzed by flow cytometry. IC_50 data were analyzed using one-way ANOVA, and post hoc testing was performed using Tukey’s multiple-comparison test. Antiproliferation and scratch assay data were analyzed using two-way ANOVA followed by the Bonferroni test. Apoptosis and cell cycle assay data were analyzed using one-way ANOVA followed by the Tukey post hoc test. Phytochemical profiling indicated the existence of fifteen chemicals in both turmeric rhizome and soursop leaf extracts. The candidate combination ratio of turmeric rhizome to soursop leaf extracts was 1:21, exhibiting cytotoxic activity with an IC50 value of 32.2 ± 2.5 ug/mL. The combined extract was associated with antiproliferative, anti-migratory, and pro-apoptotic responses and induced G2/M phase cell cycle arrest in T47D cells.

Abstract

Indonesian Migrant Workers (PMI) in Shah Alam, Selangor, Malaysia, face limited access to formal healthcare services, thus heavily relying on herbal-based traditional medicine. Low literacy regarding the safe use of herbs potentially leads to misuse and negative interactions with modern medicine. This International Partnership Community Service (PkM-KI) activity aims to increase the knowledge and awareness of PMI and the community around the surau (Islamic prayer room) regarding the rational and evidence-based use of traditional medicine through a community-based educational approach. The program was conducted over three days through workshops, focus group discussions, and the distribution of bilingual Malay–Indonesian educational booklets containing an introduction to local medicinal plants, proper usage methods, and herbal safety aspects. The evaluation involved 24 respondents using a pre-test and post-test design to measure changes in participants' knowledge levels. Statistical analysis using the paired t-test revealed a highly significant increase in knowledge (p < 0.05), rising from 45.7% before the activity to 97.5% after the intervention. These findings indicate that the surau-based educational approach is effective as a contextual and sustainable medium for disseminating herbal health literacy. This activity not only strengthens PMI's understanding of safe traditional medicine use but also encourages the integration of local wisdom with modern pharmaceutical approaches. This program has the potential to serve as a cross-border community service model for improving herbal-based health literacy among migrant communities and local societies.
