Abstract

Pantai Samudra Indahmerupakan kawasan pesisir yang merupakan salah satu tempat aktivitas masyarakat seperti lalu lintas kapal angkutan maupun pembangunan tempat wisata yang berpotensi menimbulkan pencemaran terhadap lingkungan pesisir. Pada penelitian ini telah dilakukan penentuan kadar logam berat pada air dan sedimen di perairan Pantai Samudra Indah. Tujuan penelitian ini adalah untuk mengetahui kandungan logam berat Cadmium (Cd), Timbal (Pb) dan Merkuri (Hg) yang terakumulasi pada air dan sedimen. Lokasi pengambilan sampel dilakukan pada 3 stasiun yang masing-masing  terletak di dekat PLTU, dekat lalu lintas kapal dan  dekat kawasan wisata. Penelitian ini menggunakan metode yang mengacu pada Standar Nasional Indonesia (SNI) nomor 06-6992.2-2004 menggunakan Atomic Absorption Spectrophotometer (AAS) Series 200 dan 220. Hasil analisis laboratorium, logam berat Pb, Cd, dan Hg pada air dan sedimen masih di bawah nilai ambang batas yang ditetapkan oleh KLH No. 51 Tahun 2004. Konsentrasi logam berat Pb tertinggi pada air di Stasiun I sebesar 0,0031 dan sampel sedimen tertinggi terdapat di Stasiun I sebesar 0,8684. Sedangkan konsentrasi logam Cdpada sampel air diStasiun I, II dan III tidak terdeteksi. Pada sedimen tertinggi terdapat pada Stasiun I sebesar 0,7449 dan konsentrasi logam berat Hg tertinggi yang terdapat pada sampel air pada Stasiun II sebesar 0,369. Sedangkan pada sedimen tertinggi pada Stasiun III sebesar 0,0642

Abstract

Research has been carried out on the characterization of biochar pores produced from bagasse through a pyrolysis process at a temperature of 110oC for 4 hours, using the Scaning Electron Microscopy-Energy Dispersive X-Ray (SEM-EDX) method. The biochar of sugarcane pulp produced is then tested for its ability to absorb nitrogen content (% N-total), with the source of N is urea fertilizer through the Kjedhal method based on SNI (2801: 2010), and the source of nitrogen from a solution of ammonium sulfate (NH4)2SO4 by the titrimetric method. The SEM measurement results showed that the biochar pores were uniform and micro-sized, while the peat pores were macro in size and irregular. EDX results show that biochar is composed of 100% carbon, while peat soil consists of carbon compounds (C), Oxygen (O) Silica (Si) and Aluminum (Al). Both of biochar and peat have moisture content of 0.38% and 11.79%, ash content of 2.42% AND 24.44%, volatile content of 0.53% and 5.23% of bound or fixed carbon (cf) of 97.02% and 58.53%. The results of the biochar capacity test applied to peat soils can increase the pH of the peat from 5 to 6.5 with the ability to absorb N-total from urea (NH2)2CO and N-total fertilizer sources from ammonium sulfate (NH4)2SO4 solution of 38% - 66%, and 56.5% -84.5%.

Abstract

Unsur hara nitrogen (N) merupakan unsur hara makro yang diperlukan oleh tanaman. Mikroorganisme yang memiliki kemampuan untuk meningkatkan penggunaan N yang tersedia dalam tanah disebut dengan bakteri penambat nitrogen. Bakteri penambat nitrogen ini dapat dijadikn sebagai sumber pupuk yang sebagai pupuk hayati. Pupuk hayati bermanfaat untuk memulihkan dan meningkatkan kesuburan tanah. Tujuan penelitian ini adalah untuk memperoleh isolat bakteri penambat nitrogen sebagai pengekskresi amonia dari rizosfer mangrove di Kuala Singkawang. Pengambilan sampel tanah menggunakan metode komposit sampling. Isolasi dilakukan secara Langsung dan pengayaaan menggunakan metode tuang pada media Burk’s. Isolat bakteri penambat nitrogen yang diisolasi dari rizosfer mangrove berjumlah 16 isolat yang memiliki kemampuan sebagai penambat nitrogen. Hasil uji penegasan penambat nitrogen menunjukkan 2 dari 16 isolat sangat aktif dalam penambatan nitrogen. Hasil dari uji pengekskresi amonia menunjukkan dari 16 isolat ada 5 isolat yang mampu menghasilkan amonia. Genus yang didapatkan pada penelitian yaitu mengarah pada genus Nitrosococcus dan Bacillus.

Abstract

Perairan Pantai Samudra Indah dipengaruhi oleh aktivitas manusia seperti aktivitas rumah tangga, perikanan, pelayaran dan aktivitas industri PLTU (Pembangkit Listrik Tenaga Uap). Limbah yang dihasilkan dari aktivitas industri PLTU mengandung bahan yang membahayakan bagi lingkungan. Keberadaan PLTU di kawasan Pantai Samudra Indah berpotensi melepaskan logam berat ke perairan. Logam berat dapat bersumber dari batubara yang digunakan sebagai bahan bakar PLTU. Analisis kandungan logam berat pada sampel ikan menggunakan metode Spektrometer Serapan Atom (SSA). Hasil penelitian menunjukkan kandungan logam berat Pb pada sampel ikan adalah <0,0204 μg/g, logam berat Cd berada pada kisaran 0,0337 – 0,0572 μg/g dan logam berat Hg pada kisaran <0,0138 – 0,2347 μg/g. Nilai Maximum Tolerable Intake atau batas konsumsi maksimum agar tidak keracunan logam berat Pb, Cd dan Hg untuk orang dengan berat badan 50-80 kg adalah 0,34 – 0,54 kg per minggu untuk ikan kelarau (Selaroides leptolepis), 5-79 – 9,27 kg per minggu untuk ikan kambing (Choerodon rubescens), ikan ketang (Scatophagus argus) dan ikan kerapu (Epinephelus amblycephalus).

Abstract

Dunaliella salina is a green microalgae that has the ability to produce β-carotene used in various fields, such as food supplements, natural colorant, antioxidants, anti-cancer, and anti-aging. D. salina is capable of producing large amounts of carotenoids under stressful conditions, including light and nutrient. Light is the main factor that stimulate the production of carotenoid pigments and media composition plays an important role for growth, biomass, and β-carotene production. The accumulation of β-carotene in microalgae is closely related to the type and quality of light and the composition of the culture media. This study aimed to determine the production of β-carotene in microalgae D. salina using LEDs and different culture media to select the best culture conditions for producing high value compounds. The results showed that D. salina cultured using red LED light and technical Walne media was able to produce β-carotene with the highest amount of 767,499 mg/100 g. In blue LED light and Walne Pro-analysis media, the β-carotene content was 380,522 mg/100 g, while the ZA+NPK media has the lowest value. In this study, Walne Pro-analysis media became the best culture medium for D. salina. Therefore, natural sources of β-carotene can be obtained from D. salina, so it could reduce the use of synthetic carotene in meeting global demand.

Abstract

Thalassia hemprichii merupakan jenis lamun yang ada di perairan Lemukutan. T. hemprichii berpotensi menghasilkan senyawa antibakteri. Penelitian ini bertujuan untuk mengetahui aktivitas antibakteri dari bakteri berasosiasi lamun T. hemprichii dari perairan Lemukutan. Hasil uji aktivitas antibakteri menunjukkan 6 dari 8 isolat bakteri berasosiasi T. hemprichii aktif terhadap bakteri uji. Aktivitas antibakteri ditunjukkan dengan terbentuknya zona bening di sekitar isolat bakteri berasosiasi T. hemprichii. Isolat bakteri LM07 menunjukkan aktivitas antibakteri yang kuat terhadap 4 bakteri uji. LM07 menunjukkan aktivitas antibakteri terhadap Staphylococcus aureus, Vibrio cholera, Pseudomonas aeruginosa dan Salmonella typhimurium. Thalassia hemprichii is a species of marine seagrass in the Lemukutan waters. It has a potency as a antibacterial source. The aim of the research was to obtained bacteria associated T. hemprichii. The result of this research showed six of eight bacterial strains have antibacterial activity. A growth inhibition zone formed as a clear zone around bacterial strains. LM07 showed the highest antibacterial activity. This bacteria inhibited the growth of S. aureus, V. cholera, P. aeruginosa dan S. typhimurium.

Abstract

Marine macroalgae are potential sources as carbohydrate alternative, food additive, and bioactive compounds with antioxidant activity. The biological activity is strongly influenced by the type and phenolic compounds concentration. Tropical brown algae Padina pavonica Hauck was found with a large abundance in Kabung Island, West Kalimantan. The objective of this study was to determine the phytochemical screening, total phenolic content, and antioxidant activity of P. pavonica Hauck extract. The phytochemical screening and determination of total phenolic compounds were carried out using two different solvents with ethanol and ethyl acetate. Whereas antioxidant activity was evaluated using the DPPH (1,1-diphenyl-2-picrylhydrazyl) free radical scavenger method with UV-vis spectrophotometer. The qualitative analysis of phytochemical constituents revealed the presence of alkaloids, steroids, flavonoids, phenols, and saponins. Furthermore, for total phenolic compounds, the ethanolic extract had a higher content (20.34 mgGAE/g) dry sample than ethyl acetate extract (7 mgGAE/g). The ethanolic extract also had a higher of potential antioxidant activity which IC50 was 144.47 ppm compared to ethyl acetate extract (IC50 was 191.98 ppm).

Abstract

Macroalgae is marine biological resources that play a crucial role and have an important economic value. They synthesize bioactive compounds with different kind of biological activities, such as antioxidant, antitumor, including antibacterial. In the health sector, infectious diseases from bacteria is one of the problems that still increasing. In addition, E. coli besides having the capacity to be pathogenic agent, it also showed mutlidrug resistence (MDR). Antibiotic resistance is a serious problem worldwide. The increase of this phenomenon leads to the exploration of new antibiotics using natural resources as an effort to overcome the problem. Padina pavonica Hauck belongs to Phaeophyceae spreeding along Kabung Island waters, but its existence has not been widely used. The objective of this study was to evaluate antibacterial activity from ethanol and ethyl acetate extract of P. pavonica Hauck from Kabung Island, West Kalimantan against E. coli. The extraction was  made using ethanol and ethyl acetate solvents, and the antibacterial activity test was realized with concentration series of 5 ppm, 10 ppm, 15 ppm, and 20 ppm. The quantitative assessment of the antibacterial test showed that both ethanol and ethyl acetate extracts of P. pavonica Hauck had potential antibacterial activity against E. coli. The inhibition zone of ethanol extract was 11.6 mm, while ethyl acetate extract was 12.6 mm, respectively. These two solvents were included to the bacteriostatic category.

Abstract

Phytoplankton plays an important role in marine ecosystems as  primary producer, as the basis of food chains and the food web, and are widely used as bioindicators to monitor water condition. The study of phytoplankton is the primary interest to explore aquatic resources for blue biotechnology applications in conditions as a live feed, antibacterial, antiviral, antioxidant, pharmaceutical, cosmetics, possibly food and health industry. The study aims to assess the composition, abundance and diversity of phytoplankton community in Lemukutan Island waters of West Kalimantan. The present study found 31 genera of phytoplankton, consisted of Bacillariophyceae (28 genera) and Dinophyceae (3 genera). In term of contribution, Bacillariophyceae were found to be dominant (93.035%) than dinophyceae (6.965%). The abundance of phytoplankton varied between 636.91 to 2034.48 cell.L-1. The diversity index (H’), the evenness (E) index, and the dominance (C) index ranged from 1.959–2.579, 0.582–0.868, 0.094–0.283, respectively. The result showed that the diversity index was moderate, the evenness index was high, and the dominance index was low.
