Abstract

Grifola frondosa or maitake, not only can be used as a food ingredient but also as medicine. Other than nutrients, fruit body and mycelium of G. frondosa also contains bioactive compounds, such as terpenoids, alkaloids, flavonoids, and beta glucans extracellular polysaccharides (β-1,3 glucans and β-1,6 glucans). Extracellular polysaccharide harvested more often in mycelium form which is cultivated in liquid medium. Liquid medium which is commonly used for the growth of mycelium is MYPB (Malt Yeast Peptone Broth). Mycelium production on MYPB as a medium can be increased by adding additional ingredients, one of which is sunflower (Helianthus annuus L.). The treatment was the addition of sunflower seeds on medium MYPB. The main parameters were mycelium’s dry weight and the weight of raw extract of G. frondosa. Supporting parameters were final pH medium, extracellular polysaccharides (qualitatively), terpenoid, alkaloid, and flavonoid compounds in the raw extract of G. frondosa. The addition of 250 g/l sunflower seeds in the medium MYPB was the optimum treatment that can produce 1,379 g/100ml of mycelium and 0,299 g/100ml G. frondosa extracts.

Abstract

Background Tea parasitic plant (Scurrula oortiana L.) is one of medicinal plants (herba medicina) containing several active compounds but its utilization is still constrained by some problems, so that there needs other approaches by utilizing potential of BR-S1 (A) isolate endophytic fungi which grows in these plants to control MRSA pathogenic bacteria. Objective This research aimed to examine the effectiveness of the four types of BR-S1 (A) endophytic fungi extract  from the origin of tea parasites to the growth of MRSA bacteria.Methods The researchers used laboratory experimental method by Kirby Bauer disk diffusion method and the data obtained were analyzed statistically. Result The research result showed that MRSA bacteria had various sensitivity on four types of BR-S1 (A) isolate endophytic fungi; with its highest sensitivity on extract of ethyl acetate and its lowest sensitivity was extract of n-hexane (no inhibition). The difference of sensitivity test results was suspected to be related to active compound content found in ethyl acetate extract in form of flavonoid and tannin class compounds which had been proven to inhibit growth of pathogenic bacteria.  Conclusion It can be concluded that  the four types of BR-S1 (A) endophytic fungus extract originating from tea parasites, only ethyl acetate extract was the most effective in inhibiting the growth of MRSA bacteria, this is due to the content of two active compounds from the flavonoid and tannin groups.

Abstract

Ganoderma sp., one of fungi belonged to the family Ganodermataceae, order of Aphyllophorales, class of Basidiomycetes is very widely spread.  This fungus is a soil-borne, and has parasitic and saprophytic features which are interesting because of their two contradictory roles namely harmful and beneficial effects.  As a plant parasite, Ganoderma is able to cause root and stem rot of tropical perennial crop plantation and forest which cause serious losses.  The fungus is well known as white rot fungus which is able to cause a wood rotten by lignin destruction.  On the contrary, the fungus has some advantages such as medical potentials.  In this study, some collections and characterization of Ganoderma spp. found in Banyumas has been conducted.  The technique used for sampling Ganoderma spp. mushroom is a survey with Purposive Random Sampling method.  Samples were taken from nature in the area of Banyumas Regency and its natural substrate were noted. Characterization based on the macromorphology and micromorphology was done for grouping and identification.  Micromorphology observations were done by slicing and painting samples with plants microtechnique method.  The result showed that there were 43 Ganoderma spp isolates, distributed from low to high lands of 100 - 500 m above sea level.  The most fungi were found on the high lands, in Baturraden sub district, which included 16 isolates.  The interesting finding is the discovery of Ganoderma spp. which attacked flamboyant plant (Delonix regia) in West Purwokerto sub district.  Based on micromorphology observations, these fungi form a basidium and basidiospore inside and below as well as the surface of the fruit body.  Some species were also found having such phenomenon so it needs further research on the role of basidiospora in the cycle of stem rot disease, especially basidiospora role in the initiation of diseases infection.  As the soil infecting mushroom, study of the disease spreading is focused more on the role of the mycelium found in the remaining plants as infecting the material.  Basidiopore role in the spread of the disease is rarely studied, but actually basidiospore is a potential source of genetic variation.

Abstract

The main purpose of this study was to determine the effect of the interaction between glucose concentration and incubation time on the growth of fungi S. commune and also the weight of β-glucan was used to determine the best glucose concentration and incubation time for the growth of fungi S. commune and β-glucan production. The research was conducted by experimental method of completely randomized factorial design (CRD factorial) with two factors carried out in three repetitions. The treatments provided included variations in glucose concentration (G) with three levels, namely 10 g/L (G1), 20 g/L (G2), and 30 g/L (G3), and variations in incubation time (W) with three levels, namely incubation time of 20 days (I1). , incubation time of 25 days (I2), and incubation time of 30 days (I3). Variables which used in this research independent and dependent variables. The independent variables were glucose concentration and incubation time, while the dependent variables were fungal mycelium growth and β-glucan production. The main parameter observed was β-glucan weight. The supporting parameters were dry biomass weight and the final pH medium. Data analysis was performed by Analysis of Variance (ANOVA) at 95% accuracy levels, followed by Duncan's test (Ducan Multiple Range Test). The results showed that glucose concentration and incubation time were significantly affected to the growth and production of β-glucan fungus S. commune. Glucose concentration of 30 g/L and incubation time of 25 days was the optimum glucose concentration and incubation time for the growth of S. commune fungus, and also glucose concentration of 30 g/L and incubation time of 25 days was the optimum glucose concentration and incubation time for β-glucan production of S. commune.

Abstract

Trametes versicolor, dan Russula sp. merupakan jamur liar (wild mushrooms) dari kelompokBasidiomycota. Kedua jamur tersebut dapat menghasilkan senyawa bioaktif yang memiliki efek terapis.Salah satu senyawa bioaktif yang dihasilkan adalah β-glukan. Tujuan penelitian ini yaitu untukmengetahui potensi T. versicolor dan Russula sp. dalam memproduksi β-glukan, dan untukmendapatkan konsentrasi glukosa yang optimum dalam menghasilkan β-glukan pada mediumfermentasi cair. Penelitian dilakukan di Laboratorium Mikologi, Fakultas Biologi, Universitas JenderalSoedirman. Metode yang digunakan adalah metode eksperimental menggunakan Rancangan AcakLengkap (RAL). Perlakuan adalah jenis jamur dengan konsentrasi glukosa yang berbeda. Parameterutama adalah bobot kering miselium dan bobot kering β-glukan, sedangkan parameter pendukungnyayaitu pH awal dan pH akhir medium. Data dianalisis menggunakan analisis ragam pada tingkatkesalahan 0,5% dilanjutkan dengan uji Duncan. Hasil penelitian menunjukkan bahwa perlakuanpenggunaan dua jenis jamur yaitu T. versicolor dan Russula sp. dengan konsentrasi glukosa yangberbeda memberikan hasil yang berbeda nyata, baik pada pertumbuhan maupun pada produksi β-glukan. Russula sp. mampu menghasilkan β-glukan tertinggi pada medium cair dengan konsentrasiglukosa 30 g/l, diperoleh bobot β-glukan sebesar 490,00 ± 45,82 mg/100 ml.

Abstract

Tingkat kesehatan masyarakat dewasa ini menunjukkan tern penurunan dengan berbagai penyebab, seperti peningkatan radikal bebas dan pola konsumsi nutrisi yang menurun. Hipertensi dan diabetes masih menjadi permasalahan yang paling umum pada masyarakat pedesaan. Pentingnya monitoring dan pengecekan berkala menjadi salah satu cara preventif untuk mencegah berkembangnya penyakit hipertensi dan diabetes.Tujuan studi ini adalah untuk mengevaluasi dan memonitoring serta mengevaluasi hubungan kejadian hipertensi dengan diabetes serta kaitannya dengan penngkatan usia pada pedagang. Penelitian ini dilakukan dengan menggunakan 35 sampel warga Desa Mandiraja Wetan yang berprofesi sebagai pedagang, pengambilan sampel yaitu purposive random sampling, dengan parameter tekanan darah, glukosa darah dan parameter pendukung yaitu usia. Data dianalisis dengan uji korelasi bivariat (pearson correlation) dengan nilai signifikansi p < 0.05, menggunakan software SPSS ver.26.0. Hasil penelitian menunjukkan terdapat hubungan antara kejadian hipertensi dengan peningkatan kadar glukosa darah serta usia (p < 0.05). Rata-rata tekanan darah tertinggi 150.71/88.31 mm/Hg, kadar glukosa darah tertinggi pada pria 193 mg/dL, wanita 305 mg/dL dengan rata-rata usia pria 57.80 tahun, dan wanita 51.65 tahun.

Abstract

Introduction; Cholesterol is a metabolite of fat that contains sterol compounds and is primarily concentrated in cell membranes in the body and the circulatory system. Purpose; To determine the total cholesterol level in homemakers (IRT) based on the age range category of 25-80 years and the relationship between increasing total cholesterol levels with the age of IRT. Method; The research method is descriptive-analytical with a cross-sectional approach, with a total sample of 30 and the sampling technique is purposive sampling. Data on the relationship between total cholesterol levels and age were statistically analyzed by bivariate analysis with the Pearson correlation index on SPSS software. Results; Total of 30 IR respondents, the average cholesterol level was 198.07 mg/dL, and the average age was 47.87 years. 50% of respondents have good cholesterol levels, 26.67% in the border category, and 23.33% in the danger category, with the highest cholesterol levels being 297 mg/dL and the lowest being 130 mg/dL. Conclusion; Total cholesterol levels in IRT in Tanjung are dominated by women over 45 years, with cholesterol levels ranging from 130 to 300 mg/dL, and there is a strong relationship between increased cholesterol levels in the body and rising age

Abstract

Escherichia coli and Staphylococcus aureus are bacteria that often cause health problems and diseases in humans, such as digestive disorders (diarrhea), or skin disorders (boils). Gentamicin is one of the most widely used antibiotics from the aminoglycoside group. Gentamicin is widely used as an antibiotic for both minor infections due to bacteria or moderate infections due to fungal or viral infections. The purpose of this study was to determine the inhibition and effectiveness of gentamicin in inhibiting the growth of E. coli and S. aureus and to determine the pattern of sensitivity of these bacteria to gentamicin. This research method was a completely randomized design with one way ANOVA, and further test using Duncan's test, with a total of 10 experimental units for each bacterium with 2 replications, then the data were analyzed using SPSS v.25.0 software. The results showed that the antibiotic gentamicin could inhibit the growth of E. coli and S. aureus. Gentamicin inhibited the growth of E. coli bacteria with the smallest inhibition zone value was 17 mm, the highest was 22.5 mm, and against S. aureus the lowest was 15.5 mm and the highest was 18.5 mm with the percentage of inhibition ability (inhibition) sensitive category for E. coli was 90%, S. aureus 60%, while the intermediate category for E. coli was 10% and S. aureus was 40%.

Abstract

Kebersihan air untuk kepentingan masyarakat di Desa Mandiraja Wetan, khususnya RT 07 RW 02 menjadi hal yang sangat penting, mengingat penggunaan air yang banyak digunakan untuk konsumsi, usaha, dan MCK. Pentingnya kebersihan air tandon air perlu di evaluasi dan investigasi serta dimonitoring agar kualitas air di tandon air dapat terjaga. Metode pengabdian masyarakat ini adalah dengan wawancara langsung dan pemberian kuisioner berupa pertanyaan. Hasil wawancara kemudian disajikan dalam bentuk data persentase dan histogram (deskriptif analitis) yang dibuat dengan microsoft excel. Tujuan dari pengabdian masyarakat ini adalah untuk mengetahui profil dan status kebersihan tandon air serta agar warga dapat lebih memperhatikan kebersihan tandon air tersebut guna meningkatkan status Kesehatan. Hasil pengabdian masyarakat ini menunjukan bahwa ada sekitar 60 kepala keluarga (KK), dimana 75% masyarakatnya memiliki tandon air berupa toren, dan hanya 16,7% tidak memiliki tandon air, dan sisanya 8,3% tandon air dalam bentuk cor. Selain itu 40% masyarakat selalu membersihkan tandon air pada rentang 6-12 bulan, dan hanya 20% yang membersihkan pada rentang 1-3 bulan dan warga membersihkan tandon air dengan frekuensi 1-2 kali yaitu 52% dan hanya 5% 4 dengan 4 kali.
