Abstract

Lablab bean (Dolichos lablab) seeds is a potential source of protein globulin.The bean’s protein content is 20.86 %, and the amount of globulin was more than 60% from the total protein, having major fractions of 7S and 11S. The objectives of this research were to explore the 7S globulin fractions,  to study interaction between 7S globulin fractions with xanthan gum, and to observe the functional properties of the product of the interaction. The research was conducted in 2 steps. The first step was to fractionate the 7S fractions from globulin. The second steps was to interact 7S globulin fraction with xanthan gum. The yield of these interaction were examined for its physicochemical and functional properties. The results showed that the 7S globulin fractions could be interacted by xanthan gum at pH 7. The interacted product of globulin 7S fraction 10 % with xanthan gum 0,75 % had good functional properties than globulin 7S fraction,  such as oil holding capacity, foaming capacity, and emulsion activity. Water holding capacity could not be detected because the yield became soluble. However,the foaming and emulsifying stability were still lower than those of soybean protein isolates. The research concluded that xanthan gum could be used to improve the physicochemical and functional properties of globulin 7S fraction.

Abstract

Diabetes mellitus (DM) merupakan suatu penyakit gangguan metabolik yang ditandai oleh peningkatan kadar glukosa darah melebihi batas normal atau hiperglikemia. Salah satu tumbuhan yang dimanfaatkan  masyarakat untuk pengobatan DM adalah  tanaman mahoni (Swietenia mahagoni). Bagian tumbuhan yang digunakan untuk pengobatan penyakit ini adalah bijinya. Namun potensi dari  ekstrak metanol biji mahoni belum banyak diketahui. Pada penelitian ini menggunakan tikus model DM dengan induksi MLD-STZ (Multiple Low Dose-Streptozotocin). Tikus dikelompokkan dalam 5 kelompok, yaitu kelompok kontrol negatif, kontrol positif, dan  kelompok  terapi 100, 250 dan 400 mg/KgBB yang sebelumnya telah diinduksi MLD-STZ. Metode yang digunakan dalam penelitian ini meliputi pembuatan tikus DM dengan injeksi MLD-STZ secara intraperitonial, pamberian terapi ekstrak metanol biji mahoni secara oral, uji kadar MDA serum. Hasil penelitian menunjukkan bahwa kadar MDA (Malonialdehide) serum yang didapatkan berdasarkan kelompok, yaitu 3,126; 10,245; 9,245; 7,203; 4,677 µg/ml. Dosis optimum untuk menurunkan kadar MDA adalah 400 mg/KgBB. Pemberian terapi ekstrak metanol biji mahoni mampu menurunkan kadar MDA serum tikus putih pasca induksi MLD-STZ. Kata Kunci: diabetes mellitus, MDA, MLD-STZ, pankreas

Abstract

Diabetes mellitus (DM) merupakan suatu gejala yang timbul pada seseorang dimana keadaan hiperglikemia (kadar gula yang tinggi) yang kronik dengan disertai berbagai kelainan metabolik akibat gangguan hormonal. Salah satu tumbuhan yang digunakan masyarakat untuk pengobatan DM adalah mahoni (Swietenia mahagoni). Bagian yang digunakan dari tumbuhan tersebut adalah bijinya. Namun mekanisme ekstrak metanol biji mahoni belum banyak diketahui. Pada penelitian ini menggunakan tikus model DM dengan induksi MLD-STZ. Tikus dikelompokkan dalam 5 kelompok, yaitu kelompok kontrol negatif, kontrol positif, dan  kelompok DM dengan terapi 100, 250 dan 400 mg/KgBB. Metode yang digunakan dalam penelitian ini meliputi pengekstrakan biji mahoni, induksi DM dengan injeksi MLD-STZ secara intraperitonial dan uji aktivitas SOD serum. Hasil penelitian menunjukkan bahwa  nilai SOD serum yang didapatkan berdasarkan kelompok, yaitu 98,99; 61,12; 70,05; 79,29 dan 84,49 unit/ml. Pemberian terapi ekstrak metanol biji mahoni pada tikus model DM mampu menaikkan aktivitas SOD. Kata Kunci : diabetes mellitus (DM), SOD (superoksida dismutase), STZ (streptozotocin)

Abstract

Yogurt or yogurt is fermented milk products are rich in various vitamins and amino acids, has potential as a potential source of exogenous antioxidants. Formaldehyde is an organic compound or the aldehyde group of the most simple alkanal are toxic and carcinogenic, presenting  in the body can act as a source of compounds reactive oxygen species (ROS) and free radicals that damage cells and tissues are organs. The purpose of this study was to determine the effects of formaldehyde as a toxic compound, and yogurt as detoksikan of antioxidant activity in this regard is the enzymatic antioxidant activity superoksid dismutase (SOD) and levels of reduced glutathione (GSH). The results showed that exposure to various doses of formaldehyde in food rat (Rattus norvegicus) respectively is 0 ppm, 25 ppm 50 ppm 75 ppm and 100 ppm is very real effect (P <0.01) toward decreased antioxidant enzymatic activity in this regard is the reduction of activity superoksid dismutase (SOD) and GSH levels).The results also showed that treatment of yogurt supplementation at 2 ml per cow per day in rats (Rattus norvegicus) were exposed to formaldehyde are not able to change the effect of formaldehyde exposure to the decrease of antioxidant activity (SOD activity and GSH levels) with the significance level remains highly significant (P <0.01)

Abstract

Glucosidase effects. This study aims to determine the effects of mangosteen rind juice inhibition against α-glucosidase enzyme activity and determine the levels of xanton in mangosteen rind juice. Measuring the level of xanton in mangosteen rind juice using High Performance Liquid Chromatography (HPLC). Enzyme inhibition assay against α-glucosidase activity performed in vitro. The research showed that Mangosteen rind juice samples had a retention time of 8.507 and xanton concentration in each mL juice mangosteen rind of 4.238 μg/mL or 385.272 μg/g juice mangosteen rind. The results showed that xanton in mangosteen rind juice can inhibit the activity of α-glucosidase enzyme. Based on the calculation of percent inhibition mangosteen rind juice against α-glucosidase enzyme activity by 64.71 %.

Abstract

This study aimed to determine the fermented milk of Lactobacillus casei Shirota strain potential on cholesterol levels, SOD activity, LPL enzyme activity, and cardiac histopathology. This study used 25 rats (Rattus novergicus) Wistar strain aged 2-3 months, weighing 150-200 grams. These rats were divided into 5 groups: negative control group, positive control group with high-cholesterol diet for 21 days, and therapy group with 1.0 ; 1.5 ; 2.0 mL/rat/day of fermented milk for 14 days. The analysis methods used in this study are One Way ANOVA and Tukey. The results showed that the fermented milk of Lactobacillus casei Shirota strain had a strong effect on blood cholesterol levels and increasing the SOD activity. This study also showed that the fermented milk of Lactobacillus casei with 2.0 mL/rat/day is the most effective dose to decrease the total cholesterol and increase the SOD activity.

Abstract

This study aims to determine anthocyanin antioxidant performance with a dose of 2 cc/day based on the expression of apoptotic cells on ischemic stroke rats cerebellum. Research was conducted using ischemic stroke rats (Rattus norvegicus) that prepared by ligated for 3 h on the Common Carotid Artery (CCA) and External Carotid Artery (ECA) followed by reperfusion that commonly known as MCAO (Middle Cerebral Artery Occlusion). The anthocyanin extract was characterized by LC-MS and its IC50 was measured by DPPH method. The rats were divided into five groups 1) negative control; 2) reperfusion 1 h; 3) reperfusion 72 h; 4) reperfusion 24 h, with anthocyanin therapy; 5) reperfusion 72 h, with anthocyanin therapy. The results of LC-MS showed that anthocyanin from purple sweet potato extracts contained Petunidin-3,5-O-diglucoside (Pt-DG) with an IC50 value of 22.16 μg/mL, categorized as very strong antioxidant. The results showed that apoptotic cells expression of cerebellum decreased significantly (p<0.01) after 72 h reperfusion with anthocyanins therapy until 2.42%. The current work proved that anthocyanin extract effectively suppresses the apoptotic cell’s expression of the cerebellum on stroke ischemic rats.

Abstract

Autoantibody TPO is a potential marker for early detection of autoimmune thyroid disease (AITD). Autoantibody TPO has a specifity and a sensitivity ranging from 82% to100% in comparison to other AITD serology markers. Concentration of autoantibody TPO in sera had a positive correlation with activities of chronic AITD. This research have been conducted to investigate the characteristic of polyclonal antibody TPO induced by autoantibody TPO from serum of AITD patients. The autoantibody TPO was isolated from serum of AITD patients and its band proteins was confirmed using western blotting, acquiring molecular weight of 51.81 kDa. The protein was induced to a rabbit (Oryctolagus cuniculus) and recognized as an antigen. Serum of rabbit produced polyclonal antibody which was characterized using ELISA and Western blotting. The result showed that autoantibody TPO isolated from serum of AITD patients was immunogenic, which induced the polyclonal antibody anti-TPO (anti-anti-TPO). It was found that the highest polyclonal antibody was obtained on 3rd serum bleeding with the absorbance of 0.3225 and a molecular weight of 51.81 kDa, which is able to specifically recognize autoantibody TPO. Keywords : Autoimmune thyroiditis, Autoantibody TPO, TPO, polyclonal antibody, ELISA, Western blotting

Abstract

Rheumatoid arthritis (AR), an autoimun disease, is characterized by the inflammation in the joint area caused an excessive of free radicals. An excessive of free radicals in the body cause oxidative stress, that increasing levels of malondialdehyde (MDA) as an indicator of lipid peroxidation and the decreasing levels of anti-oxidants. The treated with extract of brown seaweed (Sargassum sp.) intended to find out the MDA levels in serum and the histological of the foot joints rheumatoid arthritis rats. Malondialdehyde levels are determined through a TBA test (Thio Barbituric acid), meanwhile the histological of the rat foot joints was determined by Hematoxylen-Eosin staining (HE). The results showed the brown seaweed extract therapy (Sargassum sp.) was significantly (p <0.01) reduce levels of malondialdehyde (MDA) in the serum of 21,24% and improving histological foot joints rheumatoid arthritis rats.

Abstract

Diabetes mellitus type 1 (DMT1) is an insulin deficiency disorder caused by pancreatic damage characterized by chronic hyperglycemia. Animal modeling of DMT1 using inducting Multiple Low Dose Streptozotocin (MLD-STZ) elevat levels of free radicals and inflammation in pancreatic organs which then damage β cells. Ruellia tuberosa L. is a plant containing triterpenoids from the class of antioxidant and anti-inflammatory flavonoids. This study used three groups of rat: negative control group, positive control and 240 mg/kg bw therapy group. Study are based on changes MDA levels, blood glucose and histologic of pancreas. MDA level determining with TBA method, glucose levels using EasyTouch GCU and histopalogy of pancreas using Hematoxylin-Eosin staining. Statistical analysis of MDA and blood glucose (α = 0.05) showed significant decreases in MDA and blood glucose levels. Based on the results of histologic observations of pancreas descriptively showed a decreas in cell damage on Langerhans island. Thus, the provision of root extract of Ruellia tuberosa L. can inhibit the damage in rat DMT1 model so that it can be used as therapy of diabetes mellitus.
