Abstract

Diabetes Mellitus is characterized by leveling up glucose in human blood and affects increasing of free radicals in body as well as leading to cellular oxidative stress. Experimentally, this condition is able to be characterized by increasing malondialdehyde (MDA) level in cell and histological changing in pancreas appearance. Consumption of antioxidant substances was reported able to reduce the MDA quantity as free radicals. Black soybean or Glycine max (L) Merr. was reported contains important antioxidant agents such as anthocyanin and isoflavone. This paper discloses recent investigation on application of black soybean water extract to reduce the MDA level on diabetes mellitus-rat model induced by STZ (DM) and also reports the pancreas histological changing of the DM rats. Investigation revealed that black soybean water extract significantly affect decreasing of MDA level by 4.9%, 27.1% and 45.7% in three different doses theraphy (500, 750, and 1000 mg/kg BW). Histologically, it also clearly indicates repairing of pancreas tissue of the DM rats.

Abstract

Diabetic Nephropathy is a kidney disease which occurs due to complication of diabetes mellitus  as a consequence of the damage of the kidney endothelial cells. Hyperglicemia condition in patients with diabetes mellitus that induces an oxidative stress, were related to endothelial cell damage. Oxidative stress as a result of hyperglycemia will activate a number of signal transduction pathways resulting in increase of  free radicals. D-alpha tocopherol as one of antioxidant substance, that can act as an inhibitor of free radical chain reactions, play an important role in the reduction of the oxidative stress effect. Effect of D-alpha-tocopherol in reducing oxidative stress is identified by the levels of malondialdehyde (MDA) in kidney and histology of kidney. This study used five groups mice; they were a control group, a diabetic group which was induced with MLD-STZ, and a therapeutic groups with a varieties doses of D-alpha tocopherol (100 mg/kgBW, 200 mg/kgBW and 300 mg/kgBW). The results showed that the D-alpha tocopherol was able to reduce the levels of malondialdehyde (MDA) and repair the histology of kidney of mice induced by MLD-STZ.

Abstract

Diabetes Mellitus (DM) is a chronic metabolic disorder characterized by high blood sugar levels as a result of insufficiency of insulin function. Xanton a bioactive compound in the mangosteen rind is convinced to possess antidiabetic effects. There for, in this study effect of juice mangosteen rind (Garcinia mangostana L.) on blood sugar levels, and histological features of pancreatic rats with the induction of streptozotocin was investigated. This research using experimental rats species of Rattus norvegicus Wistar strain male. Rats were divided into 3 groups, the first group was the control without any treatments, group II was sick rat group, and group III was therapy rat group treated with juice mangosteen rind (a dose of 110 mg/kg body weight) by sonde for 2 consecutive weeks. Then,blood sugar levels of all rat groups were measured along with observation of their pancreatic histological. The results showed that the blood sugar level in therapy rat group (104.7 ± 10.9 mg/dL) is not significantly different with that of rat control group (108.5 ± 19.5 mg/dL), revealing effectiveness of therapy. Conservely, blood sugar level remained high in sick rat group (163.8 ± 16.2 mg/dL). Juice mangosteen rind can improve conditions of the pancreatic histology rat with the induction of streptozotocin.

Abstract

UVB generates the production of Reactive Oxygen Species (ROS) and decrease antioxidants enzymatic excessively. Both of these biological effects caused photo-aging. Excessive ROS production afforded overexpression of AP-1 as a major regulator of photoaging. This paper figured out the potential of Aloe vera extract as topical gel treatment on the UVB-induced skin photo-aging in twenty male wistar hairless rats (Rattus novergicus) divided into 2 groups. First group was induced by UVB and the second was induced by UVB and topical gel extract Aloe vera treatment. Each group was given treatment for 4 weeks. The expression of MDA and SOD were measured with immunohistochemistry. The result showed that the topical gel therapy decreased the MDA expression and increased SOD expression significantly (p < 0.01). The conclusion from this study was Aloe vera extract had potentially as an alternative topical treatment of photoaging.

Abstract

This research deals with enzymatic extraction of virgin coconut oil (VCO) by protease from biduri plant (Calotropisgigantea) in various concentrations of 0.00; 0.05; 0.10 and 0.15% (w/w), and incubation times of 2; 3 and 4 hours. Theresults showed that the biduri protease was effective in extracting VCO. The higher concentration of biduri protease,the viscocity of VCO increased. Furthermore, the VCO quality produced by using the enzymatic extraction was betterthan that of spontaneous fermentation based on acid value and % FFA.ABSTRAKPenelitian ini bertujuan untuk mempelajari ekstraksi virgin coconut oil (VCO) secara enzimatis menggunakan proteasedari tanaman biduri (Calotropis gigantea) dengan variasi konsentrasi 0,00; 0,05; 0,10 and 0,15% (b/b), dan lamainkubasi 2; 3 dan 4 jam. Hasil penelitian menunjukkan bahwa protease biduri efektif digunakan untuk mengekstrakVCO. Semakin tinggi konsentrasi protease biduri, viskositas VCO semakin meningkat. Kualitas VCO yang diproduksisecara enzimatis lebih baik daripada VCO yang diproses secara fermentasi spontan ditinjau berdasarkan parameterbilangan asam dan % FFA (free fatty acid).

Abstract

The aim of this study is to construct the hGAD65 gene and to identify the hGAD65 clone by using PCR & RFLP. The samples were derived from normal person & DM patient’s blood. Blood DNA was isolated by salting out method and then amplified by PCR with a pair of specific primer, GAD65-F-BamH1-807 & GAD65-R-Xho1-945. The PCR-product was cloned into vector pET-28a and the pET28a-hGAD65-clone was transformed into E.coli BL21 competent cells. The pET28a-hGAD65-clone was confirmed by PCR and RFLP by BamH1 & XhoI. The PCR product of pET28a-hGAD65-clone was one band of 159bp and has two bands 5.3 kb and 159 bp by RFLPwith both restriction enzymes. The GAD65 protein is expressed in 65kD of pET28a-hGAD65-clone. PET28a-hGAD65-clone was able to recognize by gold standard monoclonal antibody specifically. These results indicated that the hGAD65 gene inserted into pET28a properly and provided the GAD65 protein expression. Key words: hGAD65, PCR, pET-28a, RFLP

Abstract

Antioxidant activity, amino acids profile and microbial quality of raw bebontot of chicken meat of spent laying henwere investigated. The samples were prepared using meat dices (1.5 x 2.0 cm) mixed with fresh ground coriander,garlic, galangal, white pepper, salt, sugar and coconut oil then wrapped in Areca catechu palm dried sheaths andfinally fermented spontaneously by drying under the sun for 5 days. The results showed there was a decreasing inpH value, moisture content and 1,1-diphenyl-2-picrylhydrazyl radical scavenging activity of final products, whileits total phenolic content was increasing. Glutamic acid was the most abundant amino acid in products after 5days fementation, followed by tyrosine, aspartic acid, lysine, leucine and histidine. The total plate count and lacticacid bacteria counts were decreasing to 9.39 log cfu/g and 8.98 log cfu/g; the Micrococcaceae was decreased to5.31 log cfu/g; the yeast and moulds counts were increased to 8.58 log cfu/g and 6.51 log cfu/g at the final stageof fermentation. It can be concluded that bebontot chicken meat in this study is a good source of natural phenolicantioxidant, and the present microorganisms will provide the source for the selection of strains well adapted to theenvironment and able to compete with contaminant bacteria.

Abstract

Diethylnitrosamine (DEN) is a metabolic product of various foods and beverages in which there are preservatives in the form of nitrate and nitrite. Nitrosamines are found in various products such as alcoholic beverages, processed meats, cigarettes, and cosmetic products. DEN is toxic and carcinogenic to organs such as the liver, lungs, kidneys, skin, stomach, and blood. In addition to giving DEN compounds, there are also CCl4 compounds as tumor promoter agents known as genotoxic substances that can increase the risk of genetic errors and stimulate cells to become malignant neoplasms. Administration of DEN and CCl4 induction can damage various organs of the body, one of which is the kidney. In a healthy body, angiogenesis plays a role in wound healing and the formation of new tissue. However, angiogenesis also contributes to carcinogenesis or the uncontrolled growth of cancer cells, and is malignant when the body is contaminated with toxic substances. Therefore, to determine the effect of the administration of DEN and CCl4 observations were made using the immunohistochemical method, to see differences in the histology of the kidneys of experimental animals, the positive control group, and the negative control group based on the VEGF angiogenesis factor. From these results, there are significant differences. In the negative group, there was no necrosis and high-value VEGF expression, but in the positive group, there was necrosis and a decrease in the value of VEGF expression compared to the negative control group.

Abstract

Aggregatibacter actinomycetemcomitant (Aa) is gram-negative bacteria composed of lipopolysaccharides (LPS) in its outer membrane. This bacteria is normally found in dental plaque and periodontal pockets causing parotid gland damage when it is present in excess amounts. LPS is an endotoxin and a complex glycolipid composed of polysaccharides (hydrophilic) and lipid A (hydrophobic) which can induces Reactive Oxygen Species (ROS), and increase the levels of malonaldialdehyde (MDA) and tissue damage. The aims of this research are to find the expression of iNOS parotid gland cell and to determine levels of MDA using thiobarbituric acid (TBA) spectrophotometric test at 533 nm. The results showed a significantly difference (P<0.05) of MDA levels in control rats (3.135 ± 0.0436 ppm) and Aa rats (6.307 ± 0.0473 ppm). iNOS expression is confirmed by the brown spots formed on the parotid gland, affording the enhancements of iNOS expression in Aa rats and control rats of 2.292 ± 0.038 and 0.166 ± 0.003, respectively (P<0.05). Keywords       : Aggregatibacter actinomycetemcomitans, Reactive Oxygen Species, malondialdehyde, iNOS

Abstract

Penelitian ini bertujuan mengetahui efek inhibin B terhadap ekspresi protamine P2 di dalam kepala spermatozoa pada kauda epididimis. Penelitian ini menggunakan 24 ekor tikus jantan berumur 4 bulan yang dikelompokkan secara acak ke dalam 4 kelompok (KO, KI, KII, dan KIII), setiap kelompok terdiri atas 6 ekor tikus. Kelompok KO merupakan kelompok kontrol hanya diinjeksi dengan PBS. Kelompok KI, KII, dan KIII diinjeksi dengan inhibin B dengan dosis masing-masing adalah 25, 50, dan 100 pg/ekor. Tikus diinjeksi sebanyak 5 kali secara intra peritoneal dengan interval waktu pemberian 12 hari selama 48 hari. Injeksi pertama, inhibin B dicampur dengan 0,05 ml PBS dan 0,05 ml CFA. Injeksi kedua hingga kelima, inhibin B dicampur dengan 0,05 ml PBS dan 0,05 ml IFA. Pada hari ke-6 setelah injeksi inhibin B terakhir, hewan coba dikorbankan secara dislocatio cervicalis lalu jaringan kauda epididimis dikoleksi dan difiksasi dengan paraformaldehid 4%. Setelah melalui proses dehidrasi, jaringan blok di dalam parafin dipotong dengan ketebalan 6 mikron dan diwarnai secara imunohistokimia dengan menggunakan antibodi anti protamine P2. Pengamatan secara imunohistokimia menunjukkan adanya ekspresi protamine P2 di dalam kepala spermatozoa pada semua kelompok perlakuan. Akan tetapi, seiring dengan penambahan dosis inhibin B menyebabkan terjadinya penurunan tingkat ekspresi protamine P2 di dalam kepala spermatozoa pada kauda epididimis. Hasil penelitian menunjukkan bahwa injeksi inhibin B dengan dosis 100 pg/ekor menurunkan secara nyata jumlah ekspresi protamine P2 di dalam kepala spermatozoa pada epididimis (P0,05) dibanding KO.
