Abstract

The aim of this research was to investigate the effect of brown seaweed (Sargassum prismaticum) extract to decreased MDA levels and improvement of pancreatic tissue histology on rats Rattus norvegicus type 1 diabetes mellitus (DM). Type 1 diabetic rats induced by intraperitonial (i.p) injection of Multiple Low Dose-Streptozotocin (MLD-STZ) with 20 mg/kg (bw) dose for 5 consecutive days. MDA was measured by UV-Vis spectrophotometry method at a wavelength of 540 nm using TBA (Thiobarbituric acid) reagent. Histology of pancreatic tissue performed using Hematoxylen-Eosin staining method. In this research, rats were grouped into 6 treatment groups: the group of control rats, type 1 diabetic rats exposed to MLD-STZ, and type 1 diabetic rats exposed to MLD-STZ than treated by brown seaweed extract orally with 0.8 g/tail dose by 2 mL per day for 1, 3, 5, and 7 days. The results of this research obtained average MDA levels of each group of consecutive treatments were 2.507; 6.770; 5.543; 4.444; 3.734 and 2.980 mg/mL. Therapy with Sargassum prismaticum extract for 7 days can reduce MDA levels of type 1 diabetic rats by 55.98% indicating level of (6.770 ± 0.027) mg/dL to be (2.980 ± 0.017) mg/dL and able to improved pancreatic tissue histology type 1 diabetic rats. Abstract

The aim of this research was to investigate the ability of brown seaweed (Sargassum prismaticum) extract that can increase the activity of SOD and improvement of hepar tissue on rats Rattus norvegicus type 1 diabetes mellitus the results of multiple low dose-streptozotocin (MLD-STZ) induction with 20 mg/kg (bw) dose for 5 consecutive days. As many as 24 rats were devided into 6 groups of 4 each tail. Group treatments were the group of control, type 1 diabetes mellitus, and type 1 diabetes mellitus that given by therapy of Sargassum prismaticum extract orally with variations of given therapy on the different  days to 1, 3, 5, and 7. SOD activity were measured by Elisa Reader Kit on the wavelength at 450 nm and histology description with hematoxylin-eosin staining. The results of this research obtained that SOD activity after given by Sargassum prismaticum extract have been raised and the results as follow as 53,80 (unit/mL), 63,78 (unit/mL), 73,67 (unit/mL), 83,85 (unit/mL). Sargassum prismaticum therapy can fixed the damage of DM type 1 hepar tissue on rats that have been induction by MLD-STZ. Keyword : Diabetes Mellitus, Hepar, Histology, SOD

Abstract

This research aims to determine the effect of Spirulina sp. extracts in the form of spray on pancreatic tissue histology, and protein profiles of skin wound rats (Rattus norvegicus) induced by Multiple Low Dose streptozotocin (MLD-STZ). The injection of MLD-STZ at a dose of 20 mg/kg BW causes Diabetes Mellitus (DM) type 1. In this study, rats were divided into 3 groups: negative control (no STZ induction), positive control (MLD-STZ-induced), and therapy group (MLD-STZ-induced and therapy). Therapy was given in the form of spray-based herbal bioactive Spirulina sp. with a dose of ± 200 μL right on rat wound 3 times/day during 2 weeks after being DM as a result of MLD-STZ induced. The method used for the analysis of protein is SDS-PAGE and pancreatic tissue histology using hematoxylin-eosin staining methods (HE). The results of the study showed that post-therapy causes the difference of protein band profiles of the three treatment groups, emergence a new protein band with a molecular weight of 93 kDa in skin wounds of diabetic rats, and this protein bands disappear in skin wound rats after treated by herbal spray of Spirulina sp.. In addition, results of histological pancreatic tissue diabetic group has reduction of pancreatic β cells and islet of Langerhans in the cavity width. As well, there is improvement in the therapy group of pancreatic histology.Keywords: diabetes mellitus, pancreatic histology, skin protein bands

Abstract

Diabetes mellitus (DM) is a disease caused by disorders of carbohydrate metabolism. DM can also be caused by the destruction of β cells of the islet of Langerhans in the pancreas that functions as an insulin-producing. In patients with DM when injured will be more difficult in the wound healing process. This study aims to determine the effect of Spirulina herbal spray against changes cuts, reduction in iNOS expression in β-cells of the pancreas and protease activity in wound induction results MLD STZ rats (Multi Low Dose Streptozotocin) at a dose of 20 mg / kg for 5 consecutive days in a row. In this study using rats (Rattus norvegicus) were divided into three groups with each group consisting of 5 rats, the three groups are groups of healthy mice with skin wound on his back, a group of diabetic rats with wound and groups diabetic rats with wound and to get therapy of Spirulina herbal spray during 14 days. The results showed that administration of Spirulina herbal spray can reduce the length of incision wounds in diabetic rats from 4,28 cm until 0,5 cm, able to decrease the expression of iNOS in pancreatic β-cells of DM type 1 rats and also able to decrease of 48,23% protease activity in wound DM type 1 rats.Keywords: protease activity, DM type 1, iNOS expression, herbal spray Spirulina, wound.

Abstract

Diabetes melitus (DM) adalah penyakit metabolit yang ditandai dengan timbulnya hiperglikemia akibat gangguan sekresi insulin atau peningkatan resistensi insulin seluler terhadap insulin. Terapi herbal spray Spirulina sp. merupakan salah satu pengobatan alternatif penyakit DM. Penelitian ini dilakukan untuk membuktikan pengaruh terapi herbal spray dari Spirulina sp. terhadap kadar Malondialdehid (MDA) pada luka sayatan tikus  hasil induksi Multiple Low Dose-Streptozotocin (MLD-STZ) dosis 20 mg/kg BB selama 5 hari berturut-turut. Kadar glukosa darah diukur menggunakan glukometer digital dan dinyatakan DM apabila kadar glukosa darah  > 200 mg/dL. Hasil penelitian menunjukkan bahwa pemberian herbal spray Spirulina sp. 3x sehari selama 2 minggu pada tikus hasil induksi MLD-STZ menunjukkan penurunan kadar glukosa darah (148,8 ± 34,30) mg/dL, dan penurunan kadar MDA rata-rata (0,55 ± 0,024) μg/mL, serta dapat mempercepat penutupan luka sayatan dari 4,28 cm menjadi 0,5 cm. Abstract

Proses pengkajian terhadap Al-Qur’an di Indonesia bermula pada Abad ke-16 M, ditandai dengan munculnya karya tafsir dari Ulama Indonesia yang bernama Abd al-Ra’uf al-Sinkili, dengan tafsirnya berjudul Tarjuman al-Mustafid. Karya Abd al-Ra’uf al-Sinkili tersebut dikatakan oleh banyak peneliti cenderung mengadopsi pemikiran dari Tafsir Jalalayn, bahkan ada yang mengatakan bahwa Tarjuman al-Mustafid tersebut merupakan terjemahan Tafsir Jalalayn dalam Bahasa Melayu. Setelah masa tersebut kajian Al-Qur’an di Indonesia muncul kembali pada abad ke-19, dengan karya tafsir yang dihasilkan oleh Ulama asal Indonesia yang hidup di Tanah Makkah, dan karyanya juga diselesaikan di Tanah Makkah pula, yaitu Imam Nawawi al-Bantani, dengan karya tafsirnya yang berjudul Tafsir Munir. Kemudian kajian Al-Qur’an di Indonesia berkembang dengan baik pada abad ke-20 M, pada masa tersebut banyak sekali karya-karya tafsir tercipta, dan tentunya sudah beralih dari pemikiran klasik menuju pemikiran yang modernis. Pada abad ke-20 tersebut, penulisan karya tafsir terbagi menjadi beberapa model dan fokus kajian, ada yang fokus pada satu surat tertentu, satu juz tertentu, dan juga menafsirkan Al-Qur’an lengkap 30 Juz. Dan pada akhir-akhir dekade, muncul karya tafsir yang mengusung model tematik, membahas tafsir Al-Qur’an terhadap satu tema yang diangkat.

Abstract

Pengembangan pemaknaan terhadap Al-Qur’an sangat penting bagi perkembangan khazanah keilmuan Al-Qur’an dan tafsir dalam dunia Islam. Perlu diketahui bahwa pengembangan makna bukan berarti menghapus makna awal sebuah ayat atau kata dalam al-Qur’an. Akan tetapi pengembangan ini merupakan upaya melengkapi literasi di bidang Al-Qur’an dan Tafsir. Tulisan ini akan menjabarkan pengembangan makna tafassahu fi al-majalis yang tertuang dalam Q.S. Al-Mujadilah (58): 11 dengan menggunakan pendekatan ma’na cum maghza yang digagas oleh Sahiron Syamsuddin. Sesuai dengan misi dari munculnya pendekatan tersebut, tulisan ini merupakan sebuah upaya untuk memberikan kontribusi dalam pengembangan penafsiran al-Qur’an di dunia Islam, di Indonesia khususnya.

Abstract

There is an increasing amount of evidence that oxidative stress related to hypertension can damage the function of diverse structures such as the aorta and kidney. It is a well-established fact that chlorogenic acid and anthocyanin found in purple sweet potato generates bioactive compound with antihypertensive and anti-ACE activities in RAA system. The present study sought to investigate anti-ACE activities in RAA system of extract ethanol of purple sweet potato (EP) in deoxycorticosterone acetate (DOCA–salt)–induced hypertensive rats (Rattus norvegicus). The rats were orally administrated a 95% ethanol extract of purple sweet potato (var. ayamurasaki) (EP) in a daily dose of 200 and 400 mg/kg body weight also chlorogenic acid (CA) of 700 mg/kg bw for 4 weeks. Activity of renin, ACE, and Ang II concentration were assessed. Inhibiting activity of renin, ACE, and decreasing Ang II concentration after treatment was observed in the DOCA-salt hypertensive rats compared to normotensive group rats; (P<0.05). This is the first report that demonstrates the lowering of blood pressure and anti-ACE in RAA-system effects of an ethanol extract of purple sweet potato, containing chlorogenic acid, in a DOCA–salt model of hypertension.

Abstract

The latex of milkweed plant (Calotropis gigantea) is known to have proteolytic activities. The research was conducted to evaluate the use of ammonium sulphate for partial purification of such enzymes. The phosphate buffer extract of the latex was first centrifuged to precipitate gum and non-proteinous matter. The supernatant was then treated with ammonium sulphate to achieve the respective degree of saturation, namely 35, 50, 65 and 80%. It showed that the use of ammonium sulphate at 65% degree of saturation was the optimum condition, and produced a partially purified enzyme with proteolytic activity which was not significantly different from that obtained from the use of the same salt at 80% degree of saturation. Nonetheless, the supernatant still showed a substantial degree of proteolytic activity, indicating that the latex contained a various types of proteases. Abstract

Endogenous polygalacturonase (PG) enzymes in cocoa pulp have not been studied yet. The research, aimed to extract and characterize endogenous PG enzymes in cocoa pulp, was carried in two steps: (i) extraction of endogenous PG enzyme and (ii) enzyme characterization i.e. enzyme activity assay and determination of enzymatic kinetics (Km and Vmax), molecular weight (MW) by electrophoresis, and determination of its optimum temperature and pH. The results showed that the characteristics of endogenous PG in cocoa pulp were as followed: the relative activity of 4,58± 0,06 Units/ml,  Vmax of 6,69 Units/ml, Km of 0,37 %, MW was 32,84 and 63,58 kDa, and the combination of temperature and pH optimum were 42,5oC and 4,6.
