Abstract

The position of Pasie Nan Tigo Village on the Coast of Sumatra causes the area to be very risky to coastal abrasion, floods, earthquakes, and even by tsunamis. Therefore, the PKM activities undertaken are aimed at increasing the understanding and readiness of the community in the village in dealing with disasters so that the impact of disasters can be reduced. The activity was carried out using the lecture/counseling method followed by a discussion. The results of the activities showed that in general the community was well-informed in facing disasters. Understanding of natural phenomena especially related to the impact of the earthquake and tsunami such as: the receding of sea water, cracked houses, sudden emergence of water sources, strong and repeated earthquakes and the running of animals towards the land to be a sign of what they should do. The decision to be prepared to evacuate is generally depending on the information released by the government. The community is also equipped with saving valuable family documents such as diplomas, certificates, ID-Card, family card, etc. by doing digital scanning and backing up. Photos and results are stored in a cloud system in the form of an email or in the google drive facility. Communities are also taught about making disaster signs and making agreements especially in families to determine safe gathering places by using the shortest evacuation route map or buildings that can function as shelters.

Abstract

Calli is one of the materials used for genetic transformation. Studies on calli induction in some pepper genotypes were conducted in the Plant Tissue Culture Laboratory of the Faculty of Agriculture, Andalas University, Padang. This study aimed to obtain calli which can be used as material for genetic transformation. We used three genotypes of chili (Kopay, Kampung and Trisula). Explants were derived from first leaf, cotyledon, epycotyl and hypocotyl obtained from 24 days old chili germination. The study found that all the explants were able to produced calli. From 17 combinations of medium, the two combinations that produced 100% calli were MS + BAP + IAA + L2 vitamin and MS + 2,4 D. Mediums that can produce organogenic calli were MS + BAP 4 mg / L, IAA 0.5 mg / L, L2 vitamin ( Pyridoxin hydroklorit 0.5 g / L  and Thiamin hydroklorit 0.1 mg / L). The colour of calli produced were yellowish green and compact. The study also found that structures develop with a globular phase, followed by a heart shape, torpedo, cotyledonary and eventually form buds. While MS + 2.4 D 4 mg / L produced yellowish white calli and crumb, it could not grow organogenic calli.

Abstract

Salacca (Salacca edulis L.) is a tropical plant with a high economic value. The sexes of salacca can be grouped as dioecious, monoecious, dan hermaphrodite. Farmer's success in cultivation is especially determined by the male to female ratio. The present ratio of male and female plants is 1:4. Determining sex based on morphological characteristics is not effective and difficult for young plants (seedlings), also to lysozim marker and chromosome. Marker Assisted Selection is a strategy to determine sex using a marker. Random Amplifed Polymorfism DNA (RAPD) is an amplification technique that uses markers. Of 305 primers tested, 4 primers, OPO-17 produced 297 bp product, OPAP-20 produced 554 bp as the specific fragment for the female and UBC-454 produced 934 bp as the specific fragment for the female and UBC-454 produced 946 bp as male, primer UBC-78 with the size of fragment 562 bp as the specific fragment for the female.

Abstract

Bunga matahari merupakan tanaman yang stategis karena selain dapat digunakan sebagai bahan pangan, juga dapat menghasilkan minyak terutama minyak makan yang menempati posisi terbesar ketiga dunia setelah kedelai dan kelapa sawit. Pengembangan budidaya bunga matahari di Indonesia masih kurang optimal terutama produksi bunga matahari. Dalam peningkatan hasil produksi salah satunya perlu benih unggul. Identifikasi keragaman biji perlu dilakukan agar mendapatkan karakter genotipe tanaman yang berbeda dalam proses perakitan kultivar untuk mencegah duplikasi. Adapun tujuan dari penelitian ini untuk mengetahui karakter dan variabilitas karakter biji bunga matahari. Aksesi tanaman yang digunakan yaitu Ha1, Ha15, Hu1, Hu2, dan Hu3. Metode penelitian ini terdiri atas kegiatan pengolahan lahan, penanaman, perawatan, pengendalian OPT, dan pemanenan bunga matahari. Setelah diperoleh biji hasil panen, dilakukan karakterisasi benih dengan peubah kuantitatif yaitu panjang biji, diameter biji, bobot 100 biji, persentase biji hampa. Peubah kualitatif terdiri atas warna biji, bentuk biji, garis tepi dan warna garis. Analisis data yang digunakan adalah statistik deskriptif sederhana. Analisis data melalui perbandingan varians fenotipe dengan standar deviasi varians fenotipe, varians, standar deviasi. Hasil yang didapatkan pada karakter kualitatif dan kuantitatif menunjukkan adanya keragaman. Peubah yang memiliki nilai variabilitas luas adalah karakter diameter biji, warna biji, bentuk biji, garis tepi dan warna garis biji, sedangkan peubah panjang biji, bobot 100 biji dan persentase biji hampa menunjukkan variabilitas sempit.

Abstract

Aim: This study aims to determine the relationship of HIV-1 genetic diversity with acquired drug resistance mutation in HIV-1 patients who receive first-line antiretroviral drugs at RSUP Dr. M. Djamil Padang. Methods: The study was conducted on 20 people with HIV who were diagnosed based on HIV-1 antibody screening tests using simple / rapid methods at the Central Laboratory of RSUP Dr. _ _ M. Djamil Padang and had received first-line ARV therapy for at least 6 months . Furthermore, proviral HIV-1 DNA extraction was carried out and amplification using a PCR (nested PCR technique) with a specific primer against reverse transcriptase at the Biomolecular Laboratory of the Microbiology Section of the Faculty of Medicine, Andalas University .  Result: The results showed that HIV-1 genetic diversity and changes in ARV regimens with the discovery of drug resistance mutations obtained for first-line ARVs were expected to be the basis for the management of HIV/AIDS patients. Conclusion: The conclusion of this study is that HIV-1 genetic diversity found is a form of recombinant CRF-AE and CRF- BC and subtypes A and B , a acquired drug resistance found is M184L and T215N (types of mutations that are resistant to NRTI- class ARVs)

Abstract

Lactic Acid Bacteria (LAB) were isolated from cocoa beans three days fermentation Criollo variety. That were eleven isolates, all of isolates were named Gr3 (1, 2, 3, 4,…,11). All of isolates were tested to antimicrobial activity against indicator pathogenic bacteria (Escherichia coli  14237, Staphylococcus aureus NBRC 13276, Bacillus subtilis BTCCB 612, Listeria monocytogenesis dan Salmonella typhii),  than that was chosen potential one isolate had highest inhibition zones to pathogens.  One potential isolate was done determination weigh protein bacteriocin by Sodium Dodecyl Sulfate Polyacrylamide Gel Electrophoresis (SDS-PAGE). Result of antimicrobial activity test showed that, 11 isolates had inhibition zone to pathogenic bacteria, that were 7 mm till 11 mm at 48 hours observation. That was found one isolate potential, it was Gr3.7.  Gr3.7 isolate had high inhibition zones to against growth pathogenic bacteria, that was 11mm to Salmonella typhii and Listeria monocytogenesis at 48 hours observation. Determination crude weight protein test was estimated 24 kDa protein by using SDS-PAGE.  To Found purifying weight protein bacteriocin must be done purification processing. Probiotic candidate (Gr3.7) can be used as natural antibiotics, because it had positive responce to against Salmonella typhii.  Salmonella typhii was known pathogen in intestine,  beside that it was used as food preservative, because it had positive response to against Listeria monocytogenesis.

Abstract

One of the unique local wisdoms implemented by the local community in Nagari Sikucur, Padang Pariaman West Sumatera is “banned fish”. However, changes and shifts in people's lifestyles affect the pattern and values adopted by the local community, including the local wisdom of the community in that area. In order to describe the local wisdom in that area, a qualitative study was conducted. This study was aimed to answer some research questions figuring social phenomena related to local wisdom in West Sumatera people. The results showed that management of banned fish in Nagari Sikucur has been implemented as informal public policy in order to preserve fish population in their rivers. Such agreement has successfully conserved the ecosystem while another impact also attracted people in seeing such ecotourist system. Management of such local wisdom is applied in well practice, accepted and bind to the whole society member. People are benefited with such local wisdom by having clean water in the river that can be used for their agricultural activities.

Abstract

Land in the coastal area has not been optimally utilized for agricultural land due to a number of issues, both economically and in terms of cultivation practices. An idea for maximizing the land's potential so that it can be more productive and deliver concrete benefits to the community is required. Sunflower cultivation is one technique to make use of coastal land. The goal of this study was to see how well sunflowers grew and yielded on unused land in coastal areas. The study used a randomized block design, with treatments of three sunflower kinds and two accessions repeated three times. The findings revealed that sunflowers have the potential to be grown on undeveloped land along the coast of the United States.

Abstract

Sunflower is a commodity that has the potential to be developed as a raw material for producing vegetable oil because it contains high levels of vitamin E. This plant has wide adaptability and requires areas with full sun. However, only certain varieties are able to grow and develop optimally in various agroecosystems. So we need information regarding the suitability of varieties with specific conditions of a location. This study aims to obtain information about varieties that are adaptive to new openings in the highlands of Alahan Panjang, Solok Regency, West Sumatra (1,620 masl) based on growth and yields achieved. The experiment was designed using a Randomized Block Design (RAK) with 3 groups and 5 levels of treatment, namely sunflower varieties. The varieties include IPB BM 1, Ring of fire, Russian Mammoth, Hoppy Black Dye, Black Russian, and Kanigara. The varieties of Russian Mammoth, Ring of Fire, and Hopi Black Dye have better growth when viewed from their adaptability in the research location. Meanwhile, based on the weight of seeds/plant, the percentage of pithy seeds, and the weight of 100 grains the Russian mammoth variety had the best results.

Abstract

The large number of children suffering from ADHD and autism today is a result of the increasing use of pesticides in agriculture. This applies to women farmers of childbearing age who work in agricultural settings or women whose work is related to the use of pesticides. Pesticides are a mixture of several chemicals used to control pests and protect plants. With the use of pesticides, yields obtained increased significantly. This study aims to examine the effect of exposure to pesticides on female farmers of childbearing age on growth and development disorders of children born. The review was conducted according to state-of-the-art methods using selected reporting item guidelines for reviews and meta-analyses (PRISMA). We reviewed the literature from several publications and analyzed the effect of exposure to pesticides on female farmers of childbearing age on developmental disorders of the children they were born with. The results of this study are the effect of exposure to pesticides on female farmers of childbearing age on birth defects. The use of pesticides causes pesticide poisoning. Accumulation of pesticides at a certain threshold has a negative impact on the body, namely brain disorders, tumors, cancer, even in pregnant women can cause birth defects. Apart from adults, pesticides also have a negative impact on children
